Construction of Pseudoalteromonas - Escherichia coli shuttle vector based on a small plasmid from the marine organism Pseudoalteromonas (극지해양 Pseudoalteromonas 유래의 소형 플라스미드에 기반한 Pseudoalteromonas - Escherichia coli 셔틀벡터 제작)

  • A small plasmid (pDK4) from the Antarctic marine organism Pseudoalteromonas sp. PAMC 21150, was purified, sequenced and analyzed. pDK4 was determined to be 3,480 bp in length with a G+C content of 41.64% and contains three open reading frames encoding a replication initiation protein (RepA), a conjugative mobilization protein (Mob) and a hypothetical protein. PCR-amplified pDK4 was cloned in high-copy pUC19 to yield the fusion vector pDOC153. The chloramphenicol resistance gene was inserted into pDOC153 to give an ampicillin and chloramphenicol-resistant, Pseudoalteromonas - Escherichia coli shuttle vector (7,216 bp; pDOC155). The TonB-dependent receptor (chi22718_IV ) and exochitinase (chi22718_III ) genes from Arctic marine P. issachenkonii PAMC 22718 were cloned into pDOC155 to produce pDOC158 and pDOC165, respectively. Both vector derivatives were transferred into plasmid-free Pseudoalteromonas sp. PAMC 22137 by the triparental mating method. PCR experiments showed that the genes were stably maintained both in Pseudoalteromonas sp. PAMC 22137 and E. coli $DH5{\alpha}$ cells, indicating the potential use of pDOC155 as a new gene transfer system into marine Pseudoalteromonas spp.

  • Tropical forests whether fragmented or undisturbed or be they equatorial or deciduous, remain the storehouse of biodiversity for hundreds of thousands of plant and animal species. This unique characteristic continues to attract a wide range of scientists and international organizations to study and attempt to understand tropical forest ecosystems. Gene flow is mediated by pollen, seed and seedling dispersal, and factors affecting this gene flow include phenology, spatial distribution, population structures, seed predation, sexual and mating systems as well as physical and biological barriers to gene flow. Two methods are used in measuring gene flow: direct method that relies on the actual observation of seed and pollen dispersal, whereas indirect methods involve the use of genetic markers such as allozymes and DNA techniques. Political strife, extreme natural and artificial disasters, the lack of a comprehensive forestry research vision, coupled with difficult socio-economic conditions in Africa have made the environment quite difficult for sustained research activities on the part of those undertaking or wishing to undertake such studies. Gene flow studies in this region are few and far between. This review elaborates on the mechanisms of gene flow mediation in Sub-Saharan Africa.

  • Unlike rigid parts, flexible parts can be deformed by contact force during assembly. In robotic assembly, information about their deformation as well as possible misalignment between the holes and their respective mating parts is essential for successful assembly. This paper presents a method to design a visual sensing system for measuring parts deformation and misalignment in flexible parts assembly. This paper performs ray-trace analysis of the system. A series of experiments for flexible parts assembly by using the implemented system are performed.

Measurement of Shear Contact Characteristics on Mechanical Joints (기계 조인트의 전단 컨택 특성 측정)

  • An experimental method based on contact resonance is developed to extract the contact parameters of mechanical joints under various clamped conditions. Mechanical joint parameters of shear contact stiffness and damping were extracted for different physical joint parameters such as surface finish of the mating surfaces, the presence of lubrication, the effect of the clamping pressure, and shear load. It was found that the shear contact stiffness values decreased with increasing clamping load and increased with increasing shear loading. Contact damping ratio values were almost constant with clamping load, but decreased with increasing shear load. Moreover, rough surfaces exhibited the highest shear stiffness and contact damping compared to smooth surfaces.

Breeding and Screening of Lentinula edodes Strains Resistant to Trichoderma spp.

  • Trichoderma spp. cause large crop losses of the cultivated shiitake mushroom, Lentinula edodes. We bred several shiitake strains that are resistant to Trichoderma spp. using di-mon mating to establish a useful method for controlling the greenmold disease. We examined the competitive ability of L. edodes against Trichoderma spp. using a dual culture system to select resistant strains. By screening Trichoderma-resistant strains, we found that among 11 parental strains, 4 strains, including KFRI 36, were confirmed resistant strains. They showed especially strong resistance to T. harzianum, which formed deadlock after mycelial contact and then invaded into the territory of T. harzianum. KFRI 171 also showed resistance to T. atroviride strains. Among 13 strains, which were made by hybridization of shiitake strains, 5 were confirmed to be resistant to Trichoderma, including KFRI 58-1. Their resistance was not correlated to the resistant activity of their parents’ strains. Two strains lose resistance and two strains acquire resistance compared to their parents’ strains. In SEM observation, the mycelium of L. edodes at the interaction zone of Lentinula-Trichoderma was rugged and swollen by T. harzianum.

The Static Performance Analysis of Foil Journal Bearings Considering Coulomb friction (마찰을 고려한 포일 저널베어링의 정특성 해석)

  • In foil bearings, the friction between bumps and their mating surfaces is the major factor which exerts great influence on the bearing performance. From this point of view, many efforts have been made to improve the understanding of the influence of the friction on the foil bearing performance by developing a number of analytical models. However, most of them did not consider the hysteretic behavior of the foil structure resulting from the friction. The present work developed the static structural model in which hysteretic behavior of the friction was considered. The foil structure was modeled using finite element method and the algorithm which determines the conditions of the contact nodes and the directions of the friction forces was used to take into account the friction. The developed model was integrated into the foil bearing prediction code to investigate the effects of the friction on the static performance of the bearing. The results of analysis show that multiple static equilibrium positions are presented for the one static load under the influence of the friction, inferring its great effects on the dynamic performance. However, the effect of friction on the minimum film thickness which determines load capacity of the bearing is negligible.

Mechanical characteristics of involute-circular arc composite tooth profile (인벌류우트-圓弧 合成齒形의 諸特性)

  • In this study, full-rounded tip curve of rack and its mating fillet curve of pinion in Involute-circular arc composite tooth profile are derived. Mechanical characteristics are calculated analytically, i.e., Specific sliding, Nominal bending stress at working root circle and the Contact factor of the arc of contact in circular arc part to the arc of double contact. These characteristics compared with standard involute tooth profile are improved in circular arc part of composite tooth profile. To obtain more efficient composite tooth profile, we studied these characteristics with regard to the changes of unwound angle and radius of circualr arc. And a design method of composite tooth profile is suggested. Composite tooth profile are compared with standard involute tooth profile.

  • The changes of the plasma progestin concentations in normal pregnancy and artificially maintained pregnancy of the rabbits by hormonal replace therapy after induced ovulation and insemination on Day 1, followed by ovariectomy on Day 2, were checked by direct measurement, competitive protein-binding assay, from the time of mating through to parturition. The results obstained in this experiment can be summarized as follows: (i) The mean concentration of progestin in normally pregnant rabbits increased from $1.7{\sim}2.7$ ng/ml on Day 1 to $19.8{\sim}25.3$ ng/ml on Days $13{\sim}15$ and declined slowly thereafter until term. (ii) The progesterone levels in artificially maintained pregnancy by hormonal replacement therapy showed quite a similar pattern to that in normally pregnant rabbits.

Comparison of Ablation Characteristics of Carbon Nanotube reinforced Hybrid Al2O3 by using Ultrashort Pulse Laser (순수 알루미나와 탄소나노튜브 강화 알루미나 복합체의 극초단 펄스레이저 가공특성 비교)

  • In this paper, pure $I_{ph}$ and hybrid carbon nanotube reinforced $I_{ph}$ were sintered using the SPS(spark plasma sintering) method for high densification. A nanosecond laser (${\lambda}=1063nm$, ${\tau}P=10ns$) and a femtosecond laser (${\lambda}=1027nm$, ${\tau}P=380fs$) were installed on an optical system for the micromachining test. The ablation characteristics of the pure $I_{ph}$ and CNT/$I_{ph}$ composites, such as thermal effect and ablation depth, were investigated using FE-SEM and a confocal microscope device. Laser machining results for the two mating materials showed improved performances: CNT/$I_{ph}$ composites showed good surface morphology of hole drilling without a melting zone due to the composites' high thermal properties; also, the ablated depth of CNT/$I_{ph}$ was higher than that of pure $I_{ph}$.

A Stress Analysis on the Involute-Circular Arc Composite Tooth Profile Gear (인벌류우트-圓弧 合成齒形기어의 應力解析)

  • In a new involute-circular arc tooth profile which is composed of an involute curve in the vicinity of pitch point, a circular arc in the addendum part, and a curve in the dedendum part which is generated by the circular arc profile of mating gear tooth profile, the tooth contact stress is calculated analytically and the root fillet stress is calculated by the finite element analysis. The root fillet stress and the Hertzian contact stress of composite tooth profile gear are decreased with increasing the pressure angle and with decreasing the radius of circular arc and unwound angle. Compared with the standard involute gear, the root fillet stress is decreased by 2-15% and the Hertizian contact stress is decreased by 6-24%.
