  • Objective: The effects of maternal undernutrition during midgestation on muscle fiber histology, myosin heavy chain (MyHC) expression, methylation modification of myogenic factors, and the mammalian target of rapamycin (mTOR) signaling pathway in the skeletal muscles of prenatal and postnatal goats were examined. Methods: Twenty-four pregnant goats were assigned to a control (100% of the nutrients requirement, n = 12) or a restricted group (60% of the nutrients requirement, n = 12) between 45 and 100 days of gestation. Descendants were harvested at day 100 of gestation and at day 90 after birth to collect the femoris muscle tissue. Results: Maternal undernutrition increased (p<0.05) the fiber area of the vastus muscle in the fetuses and enhanced (p<0.01) the proportions of MyHCI and MyHCIIA fibers in offspring, while the proportion of MyHCIIX fibers was decreased (p<0.01). DNA methylation at the +530 cytosine-guanine dinucleotide (CpG) site of the myogenic factor 5 (MYF5) promoter in restricted fetuses was increased (p<0.05), but the methylation of the MYF5 gene at the +274,280 CpG site and of the myogenic differentiation (MYOD) gene at the +252 CpG site in restricted kids was reduced (p<0.05). mTOR protein signals were down-regulated (p<0.05) in the restricted offspring. Conclusion: Maternal undernutrition altered the muscle fiber type in offspring, but its relationship with methylation in the promoter regions of myogenic genes needs to be elucidated.

  • This study investigated the effects of maternal undernutrition during late pregnancy on lamb birth weight. 45 Mongolian ewes, synchronized for oestrus and then mated, were divided into four groups and offered 0.86 MJME/kgw-0.75d-1 (control group; CG : ad libitum access to feed), $0.44MJME/kgw^{-0.75}d^{-1}$ (Restricted Group 3; R3), $0.33MJME/kgw^{-0.75}d^{-1}$ (Restricted Group 2; R2) and $0.20MJME/kgw^{-0.75}d^{-1}$ (Restricted Group 1; R1) respectively during late pregnancy (90-150 days). During restriction, maternal net body weight loss, insulin and NEFA concentrations and lamb birth weight were measured. The results indicated that loss of maternal body weight in R3, R2 and R1 was 4.42, 7.23, 11.13 kg respectively, which was significantly (p<0.01) higher than that in CG (0.93 kg). Insulin concentrations of the ewes in R1, R2 and R3 were lower and were significantly different (p<0.05) between restricted groups and CG at 124 d of pregnancy. NEFA concentrations in all groups tended to decrease from 90d of gestation to parturition and in R1 were significantly (p<0.05) lower than in CG at 124 d of gestation. Lamb birth weight in R1 was significantly lower than in R2, R3 and CG (p<0.05). In conclusion, with decreasing supply of maternal nutrition, the retardation of fetal growth became worse. When the plane of nutrition was below $0.33MJME/kgw^{-0.75}d^{-1}$, significant effects of maternal undernutrition on lamb birth weight were observed.

  • This study investigated the effect of maternal undernutrition during late pregnancy on the growth and development of ovine fetal visceral organs. One hundred Mongolian ewes were mated at a synchronized oestrus and divided into three groups and offered 0.175 MJ ME $kgw^{-0.75}\;d^{-1}$ (Restricted Group1; RG1), 0.33 MJ ME $kgw^{-0.75}\;d^{-1}$ (Restricted Group2; RG2) and ad libitum access to feed (Control Group; CG) during late pregnancy (90 days). Selected animals in each group were slaughtered immediately at d 90 of pregnancy and after parturition (neonatal lambs), and major visceral organs were removed and weighed separately. The results indicated that the weights of lung (p<0.01), spleen (p<0.01), heart (p<0.05), liver (p<0.05) and abomasum (p<0.01) in RG1 were significantly lighter than those of CG. For RG2, only the weights of the lung (p<0.05) and spleen (p<0.01) were significantly lighter than those of CG; when expressed as a percentage of body weight, significance was retained in the spleen (p<0.01) for both restricted groups, but the percentage of brain in RG1 was significantly higher than that in CG (p<0.01). For lung and spleen, the amount of DNA was significantly lower (p<0.01) in both groups of restricted neonatal lambs compared to CG; however, there was a significant difference only between RG1 and CG for protein: DNA ratio (p<0.01). The DNA content of kidney, abomasum and jejunum were decreased (p<0.05) in RG1 neonatal lambs, but protein: DNA ratio in the liver was decreased compared with that of CG (p<0.05). The plane of maternal undernutrition during late pregnancy had a significant effect on the growth and development of fetal visceral organs, which altered ontogeny of fetal organ growth and development. These perturbations in fetal visceral development may have significant implications on postnatal growth and adult health.

  • Objective: The experiment was conducted to evaluate the effects of maternal undernutrition during late pregnancy on the expressions of genes involved in growth and development in ovine fetal perirenal brown adipose tissue (BAT). Methods: Eighteen ewes with singleton fetuses were allocated to three groups at day 90 of pregnancy: restricted group 1 (RG1, 0.33 MJ metabolisable energy [ME]/kg body weight [BW]0.75/d, n = 6), restricted group 2 (RG2, 0.18 MJ ME/kg BW0.75/d, n = 6), and a control group (CG, ad libitum, 0.67 MJ ME/kg BW0.75/d, n = 6). The fetuses were removed at day 140 of pregnancy. All data were analyzed by using the analysis of variance procedure. Results: The perirenal fat weight (p = 0.0077) and perirenal fat growth rate (p = 0.0074) were reduced in RG2 compared to CG. In fetal perirenal BAT, the protein level of uncoupling protein 1 (UCP1) (p = 0.0001) was lower in RG1 and RG2 compared with CG and UCP1 mRNA expression (p = 0.0265) was decreased in RG2. The protein level of myogenic factor 5 (Myf5) was also decreased in RG2 (p = 0.0001). In addition, mRNA expressions of CyclinA (p = 0.0109), CyclinB (p = 0.0019), CyclinD (p = 0.0015), cyclin-dependent kinase 1 (CDK1) (p = 0.0001), E2F transcription factor 1 (E2F1) (p = 0.0323), E2F4 (p = 0.0101), and E2F5 (p = 0.0018) were lower in RG1 and RG2. There were decreased protein expression of peroxisome proliferator-activated receptor-γ (PPARγ) (p = 0.0043) and mRNA expression of CCAAT/enhancer-binding protein-α (C/EBPα) (p = 0.0307) in RG2 and decreased PPARγ mRNA expression (p = 0.0008) and C/EBPα protein expression (p = 0.0015) in both RG2 and RG1. Furthermore, mRNA expression of bone morphogenetic protein 4 (BMP4) (p = 0.0083) and BMP7 (p = 0.0330) decreased in RG2 and peroxisome proliferator-activated receptor co-activator-1α (PGC-1α) reduced in RG2 and RG1. Conclusion: Our observations support that repression of regulatory factors promoting differentiation and development results in the inhibition of BAT maturation in fetal perirenal fat during late pregnancy with maternal undernutrition.

  • A quantitative restriction of maternal diet without changes in quality of diet was given to the Sprague Dawley rats during the third week of gestation and lactation. Half the normal average daily intake of control group was given to deficient groups in this period. Female rats of control group were fed a commercial diet ad libitum throughout the experimental period. Dietary restriction started from birth to weaning in deficient I group and from the 15th day of gestation to weaning in deficient II group. Body and brain weight of offsprings of deficient groups were significantly lower than control group, but the ratios of brain weight to body weight in deficient groups were higher than the control group. Significant difference between deficient groups (I and II) was noticed at weaning. Brain DNA, RNA and total protein of offsprings of deficient groups were significantly lower than control group, but RNA/DNA, brain weight/DNA, and total protein/DNA show that cell number were more affected than the cell size by the maternal dietary restriction during the third week of gestation and lactation. Between the deficient groups, there was a significant difference in brain DNA and RNA, but no significant difference in total brain protein. • Objectives: Poor complementary feeding practices have consistently contributed to the burden of child undernutrition in Indonesia. This study aimed to estimate the prevalence and predictors of the time of the introduction of solid, semi-solid, and soft foods (ISSSF), minimum dietary diversity (MDD), minimum meal frequency (MMF), and minimum acceptable diet (MAD). Methods: We analyzed 4804 last-born infants aged 6-23 months from the 2017 Indonesia Demographic and Health Survey, which employed multistage cluster random sampling. The outcomes were calculated based on the 2021 World Health Organization/United Nations Children's Fund guidelines. The predictors of the 4 complementary feeding indicators were assessed using multivariate Poisson regression with robust variance adjusting for potential confounders and study design. Results: The prevalence of ISSSF, MDD, MMF, and MAD was 86.1%, 54.3%, 71.8%, and 37.6%, respectively, with younger children less likely to meet 3 out of the 4 outcomes. Parental education, the presence of a birth attendant, and maternal media consumption were among the predictors of MDD and MAD. Children from families with higher income were more likely to meet MDD than those from low-income households (adjusted prevalence ratio [aPR], 1.16; 95% confidence interval [CI], 1.05 to 1.28). Living in an urban area was positively associated with MMF (aPR, 1.09; 95% CI, 1.04 to 1.15) and MAD (aPR, 1.12; 95% CI 1.02 to 1.24). In eastern regions, the prevalence of children achieving MDD and MAD was lower than in those living in Java and Bali. Conclusions: It is crucial that more attention and efforts are made to improve the recommended practices throughout Indonesia, since the prevalence of adequate complementary feeding practices remains low.

  • A survey of 113 children, aged 4 to 6 years, of the San Cheon a day care center, located in the upland, over populated, poor area of Seoul City, was conducted from July 28 to September 6, 1982, to investigate the dietary and nutritional status, and prevalence of parasitic inpection. House size was 10.1 Pyung; number of households per house, 3.1; number of family member per household, 4.8; and number of family members living together per room, 3.7; on the average. Only 49% of households possess own houses and the others live in the rented houses. About 40% of mothers were engaged income-producing activities within or outside of the home. Mean values of height, weight, arm circumference, girth of chest, and girth of head ranged from 98.4 to 102.2% of KIST mean. However, 13% of the subjects were assessed as undernutrition for 'weight for height'. Mean value of hemoglobin was $12.9{\pm}2.9\;g/dl$ and mean hematocrit value was $35.2{\pm}5.5%$; 17.9% of the subjects were proven to be anemic according to the criteria of the WHO. The positive prevalence of parasitic inpection was 3.3%, significantly lower than that observed in any other area, probably because of community parasitic control project. Intakes of energy and nutrients except thiamin and ascorbic acid were far below RDAs. Carbohydrates for energy provided 71. 2 to 73.8% of total energy intake; protein accounted 11.3 to 12.2%; fat provided 14.5 to 16.6%. Energy intake was divided among breakfast, lunch, evening meal and snack, on the average, in a percentage of 22.0; 23.4; 24.4; 30.2. There was significant correlation between pattern of food consumption and nutrient intakes. The nutrient intakes and quality of foods were highly correlated to the maternal factors, especially mother's educational level. The study suggested that development of nutrition guides for mother and children would be needed to improve nutritional status of those young children.

  • Objective: This study investigated the effects of intrauterine growth restriction (IUGR) during late pregnancy on the cell growth, proliferation, and differentiation in ovine fetal thymuses. Methods: Eighteen time-mated Mongolian ewes with singleton fetuses were allocated to three groups at d 90 of pregnancy: restricted group 1 (RG1, 0.18 MJ ME/body weight [BW]0.75/d, n = 6), restricted group 2 (RG2, 0.33 MJ ME/BW0.75/d, n = 6) and control group (CG, ad libitum, 0.67 MJ ME/BW0.75/d, n = 6). Fetuses were recovered at slaughter on d 140. Results: The G0/G1 phase cell number in fetal thymus of the RG1 group was increased but the proliferation index and the expression of proliferating cell nuclear antigen (PCNA) were reduced compared with the CG group (p<0.05). Fetuses in the RG1 group exhibited decreased growth hormone receptor (GHR), insulin-like growth factor 2 receptor (IGF-2R), and their mRNA expressions (p<0.05). For the RG2 fetuses, there were no differences in the proliferation index and PCNA expression (p>0.05), but growth hormone (GH) and the mRNA expression of GHR were lower than those of the CG group (p<0.05). The thymic mRNA expressions of cyclin-dependent protein kinases (CDKs including CDK1, CDK2, and CDK4), CCNE, E2-factors (E2F1, E2F2, and E2F5) were reduced in the RG1 and RG2 groups (p<0.05), and decreased mRNA expressions of E2F4, CCNA, CCNB, and CCND were occurred in the RG1 fetuses (p<0.05). The decreased E-cadherin (E-cad) as a marker for epithelial-mesenchymal transition (EMT) was found in the RG1 and RG2 groups (p<0.05), but the OB-cadherin which is a marker for activated fibroblasts was increased in fetal thymus of the RG1 group (p<0.05). Conclusion: These results indicate that weakened GH/IGF signaling system repressed the cell cycle progression in G0/G1 phase in IUGR fetal thymus, but the switch from reduced E-cad to increased OB-cadherin suggests that transdifferentiation process of EMT associated with fibrogenesis was strengthened. The impaired cell growth, retarded proliferation and modified differentiation were responsible for impaired maturation of IUGR fetal thymus.
