Bioactive lipids in gintonin-enriched fraction from ginseng

  • Background: Ginseng is a traditional herbal medicine for human health. Ginseng contains a bioactive ligand named gintonin. The active ingredient of gintonin is lysophosphatidic acid C18:2 (LPA C18:2). We previously developed a method for gintonin-enriched fraction (GEF) preparation to mass-produce gintonin from ginseng. However, previous studies did not show the presence of other bioactive lipids besides LPAs. The aim of this study was to quantify the fatty acids, lysophospholipids (LPLs), and phospholipids (PLs) besides LPAs in GEF. Methods: We prepared GEF from white ginseng. We used gas chromatography-mass spectrometry for fatty acid analysis and liquid chromatography-tandem mass spectrometry for PL analysis, and quantified the fatty acids, LPLs, and PLs in GEF using respective standards. We examined the effect of GEF on insulin secretion in INS-1 cells. Results: GEF contains about 7.5% linoleic (C18:2), 2.8% palmitic (C16:0), and 1.5% oleic acids (C18:1). GEF contains about 0.2% LPA C18:2, 0.06% LPA C16:0, and 0.02% LPA C18:1. GEF contains 0.08% lysophosphatidylcholine, 0.03% lysophosphatidylethanolamine, and 0.13% lysophosphatidylinositols. GEF also contains about 1% phosphatidic acid (PA) 16:0-18:2, 0.5% PA 18:2-18:2, and 0.2% PA 16:0-18:1. GEFmediated insulin secretion was not blocked by LPA receptor antagonist. Conclusion: We determined four characteristics of GEF through lipid analysis and insulin secretion. First, GEF contains a large amount of linoleic acid (C18:2), PA 16:0-18:2, and LPA C18:2 compared with other lipids. Second, the main fatty acid component of LPLs and PLs is linoleic acid (C18:2). Third, GEF stimulates insulin secretion not through LPA receptors. Finally, GEF contains bioactive lipids besides LPAs.

반응셀 유도결합플라스마 질량분석분석기를 이용한 칼슘 동위원소비율의 측정과 동위원소희석법의 적용 (Application of Dynamic Reaction Cell - Inductively Coupled Plasma Mass Spectrometry for the Determination of Calcium by Isotope Dilution Method)

  • 반응셀이 장치된 유도결합플라스마 사중극자 질량분석기를 이용하여 자연적으로 존재하는 6개의 칼슘 동위원소의 검출특성에 관한 연구를 수행하였다. 동적반응셀 (dynamic reaction cell, DRC)장치를 이용한 실험에서 최적의 신호 대 잡음비를 얻기 위한 실험조건을 조사하였다. 본 실험을 통해서 반응기체로서 0.7 mL/min의 $NH_3$를 사용하고 rejection parameter (RPq)값을 0.6으로 사용함으로써, Ca의 질량위치인 m/z 40, 42, 43, 44, 46 그리고 m/z 48의 위치에서 잠재적인 간섭이온인 $Ar^+$, ${CO_2}^+$, ${NO_2}^+$, $CNO^+$ 등이 효과적으로 제거됨을 확인하였다. 검출한계는 동위원소 $^{40}Ca$, $^{42}Ca$, $^{43}Ca$, $^{44}Ca$, 및 $^{48}Ca$에 대해서 각각 1, 29, 169, 34, and 15 pg/g으로 얻을 수 있었다. 이러한 실험조건 하에서 국제비교분석을 위해서 영국의 LGC (Laboratory of the Government Chemistry, Queens Road, Teddington, England)로부터 공급된 합성식품분해물질 중의 Ca을 동위원소희석법을 적용하여 분석하였다. 동위원소희석법에 의한 측정의 불확도는 ISO/GUM과 EURACHEM지침서에 따라 평가하였다. 측정된 시료중의 Ca의 농도와 불확도는 ($66.4{\pm}1.2$) mg/kg이었다. 또한 본 실험방법의 유효성을 확인하기 위해 농도가 인증된 표준시료 NRCC (National Research Council of Canada)의 SLRS-3 (riverine water CRM)과 NIST (National Institute of Science and Technology)의 SRM1643d (trace element in water)를 분석하였다.

타투화장품 및 문신용 염료의 유해금속 함량 연구 (Hazardous Metal Content in Tattoo Cosmetics and Tattoo Inks)

  • Background: Along with the increase in consumer interest in and consumption of tattoo products, the controversy over harmful heavy metals associated with the use of tattoo cosmetics is also increasing. Therefore, investigation of hazardous metals in these tattoo products is needed. Objectives: This study was performed to provide useful data for establishing reasonable standards to securely manage tattoo cosmetics, tattoo stickers, and tattoo inks distributed in the market. Methods: Thirteen kinds of hazardous metal contents (Pb, As, Cd, Sb, Ni, Co, Cu, Cr, Se, Ba, Zn, Sn, and Hg) were analyzed for 23 tattoo cosmetics, ten tattoo stickers, and 16 tattoo inks. Hg was measured through the combustion-gold amalgamation method, and other hazardous metals were measured by inductively coupled plasma-mass spectrometry (ICP-MS) after acidic decomposition using a microwave apparatus. Results: The detected ranges of Pb, As, Cd, Sb, Ni, and Hg in tattoo cosmetics were 0.07~1.18, 0.06~0.41, ND~0.07, 0.01~3.44, 0.12~2.75, and ND~0.01 ㎍/g, respectively. All of the hazardous metals detected were below the recommended maximum standards of the Ministry of Food and Drug Safety. The mean amount of Pb detected in tattoo stickers for children was 0.24 ㎍/kg and Cd was not detected, meaning both metals met the recommended criteria. There was no statistically significant difference in all measured metals between children's tattoo stickers and adults' tattoo stickers. In the results of the study on the hazardous metal content of tattoo inks, four products (25%) for Pb, one product (6%) for As, 13 products (81%) for Ni, four products (25%) for Cu, and five products (31%) for Zn exceeded the recommended standards approved by the government. The highest predicted exposure amount for hazardous metals exceeding the recommended level of tattoo inks in a single tattooing was 5.69 ㎍/kg for Ni, 8.51 ㎍/kg for Zn, 0.44 ㎍/kg for Pb, 8.07 ㎍/kg for Cu, 0.44 ㎍/kg for As, and 71.36 ㎍/kg for Ba. Conclusions: It is necessary to prepare criteria for content limitation for the management of Co, Cr, Ba and Se tattoo cosmetics, and tattoo inks require thorough quality control.

Polyamide 조리기구로부터 Ethylenediamine 및 Hexamethylenediamine의 이행에 관한 연구 (A Study on Migration of Ethylenediamine and Hexamethylenediamine from Polyamide Utensils)

  • 식품유사용매 중 EDA 및 HMDA 분석법을 확립하고, 국내유통 재질 조리기구 124건에 대하여 식품유사용매별 이행량을 조사하였다. EDA 및 HMDA를 ethyl chloroformate로 디우레탄 유도체화한 후 GC/FID로 정량 또는 GC/MS로 확인하였다. 확립된 분석방법은 EDA 및 HMDA 각각에 대하여 LOD $0.3\;{\mu}g/mL$$0.1\;{\mu}g/mL$이었으며, 상판계수($r^2$) 0.999이상의 직선성 및 식품유사용매별로 88% 이상의 회수율을 확인할 수 있었다. 확립된 정량분석방법을 적용하여 식품유사용매별 이행량을 조사한 결과, EDA는 식품유사용매로 물을 사용한 경우 2품목에서만 각각 $1.31\;{\mu}g/mL$$2.06\;{\mu}g/mL$ 수준으로 검출되었으며, 나머지는 모두 불검출이었다. HMDA는 식품유사용매로 20% 에탄올을 사용한 경우 3 품목에서 $0.27{\sim}0.93\;{\mu}g/mL$, n-헵탄을 사용한 경우 10 품목에서 $0.26{\sim}0.44\;{\mu}g/mL$으로 검출되었으며, 나머지는 모두 불검출이었다. 본 연구에서 확인된 이행량은 유럽연합의 EDA 및 HMDA 각각의 이행기준인 $12\;{\mu}g/mL$$2.4\;{\mu}g/mL$과 비교 시 매우 낮은 수준이었다.

실리카퓸 및 강섬유를 이용한 포장용 포러스콘크리트의 품질특성에 관한 연구 (A Study on the Quality Properties of Porous concrete for Pavement Using Silica Fume and Steel Fiber)

  • 본 연구는 도로용 포장재로서 포러스콘크리트의 실질적인 현장적용을 위한 자료제시와 성능향상 방안을 도출하기 위하여 실리카퓸과 강섬유 혼입률에 따른 포장용 포러스콘크리트의 물리 역학적 특성, 내구성 및 흡음특성을 평가하였다. 시험결과, 모든 시험조건에서 포장용 포러스콘크리트의 공극률과 투수계수는 국내외 기준치 및 제안값을 만족시키는 것으로 나타났고, 강도특성의 경우 압축강도는 대한주택공사 도로기준의 기준치를 모든 배합요인에서 만족시키는 것으로 나타났으나 휨강도의 경우에는 강섬유 혼입률 0.6vol% 이상에서 일본콘크리트공학협회 제안값을 만족시키는 것으로 나타났으며, 실리카퓸과 강섬유를 동시에 혼입한 경우가 우수한 강도특성을 나타냈다. 또한 내구성은 역학적 특성과 유사한 경향성을 나타내 실리카퓸과 강섬유를 동시에 사용한 경우가 가장 우수한 내구성능을 나타냈으며, 실리카퓸 10wt.%와 각섬유 0.6vol.%를 함께 사용한 경우 Plain의 경우에 비해 골재비산저항성은 27%정도, 동결응해저항성은 약 60%정도 개선되는 것으로 나타났다. 또한 포장용 포러스콘크리트의 흡음특성은 소음저감계수가 0.48로 나타나 50%에 가까운 흡음성능을 발휘하는 것을 확인할 수 있었다.

유통 강황의 이산화황, 중금속 및 쿠르쿠민 함량 (Sulfur Dioxide, Heavy Metal and Curcumin Contents in Market-Available Turmeric (Curcuma longa L.))

  • Background: Turmeric (Curcuma longa L.) that is used as a food material has antioxidant, anticancer and anti-inflammatory properties. Recently the demand for functional foods and drugs has increased. The present study was carried out to determined of contents of residual sulfur dioxide, heavy metals, ash, acid insoluble ash and curcuminoids in turmeric from the Seoul Yak-ryeong market. Methods and Results: A total of 31 samples were obtained. Residual sulfur dioxide was not detected in any samples. Heavy metals (arsenic, cadmium, lead and mercury) were analyzed by ICP-MS (Inductively Coupled Plasma Mass Spectrometer) and a mercury analyzer and were detected in the ranges of 0.00 - 0.28, 0.00 - 0.07, 0.00 - 0.29 and 0.002 - 0.027mg/g respectively. No significant difference were observed between the average heavy metal contents of domestic and imported tumeric. However, average content of ash in domestic samples (7.8%) were significantly higher than that in imported samples (6.1%), whereas that of curcuminoids was significantly higher in imported samples (47.6mg/g) than in domestic samples (11.2mg/g). The average content of acid insoluble ash was not significantly different between two sample types (0.9% in each). Conclusions: There are no specific standards for turmeric used as food materials. Therefore, this study can be provided as basic data for the establishment of quality standards for turmeric.

전동차 고속차단기 고장 분석을 위한 FMECA 기법 (FMECA Procedure for Failure Analysis of Train High-Speed Circuit Breaker)

  • 전동차는 대용량 교통수단으로서 정시 운행 및 높은 안전성이 요구되기 때문에, 고장 분석을 체계적으로 수행하여 신뢰도를 향상시키기 위한 수단으로서 고장 영향의 심각도 및 치명도를 정량적으로 평가하는 FMECA (Failure Modes, Effects and Criticality Analysis) 기법이 적용되고 있다. 그러나, 아직까지 전동차에 특화된 FMECA 규격 및 절차는 정립되어 있지 않고 자동차 산업 등 다른 산업분야의 FMECA 규격을 그대로 적용하고 있기 때문에 전동차의 고유한 운영 및 유지보수 여건을 충분히 반영하지 못하고 있다. 본 논문에서는 산업계 각 분야에서 적용되고 있는 FMECA 규격에 대한 분석을 토대로, 전동차 분야에 적합한 FMECA 기법으로서 고장 영향 분석과 치명도 분석을 단계별로 나누어 수행하고 고장 영향의 심각도에 중점을 두어 치명도를 분석하는 기법을 제시하였다. 제안 기법을 전동차의 핵심 안전 장치인 고속차단기에 적용하여 도시철도 현장에서의 15년 동안의 전동차 유지보수 데이터를 이용하여 분석한 결과, 고속차단기 부품 중에서 특히 아크 슈트의 절손이 심각도 3등급, 치명도 5등급으로 위험도가 가장 높았으며, 뒤를 이어서 전자변 파손 및 접촉 불량, 실린더 파손 등이 심각도 3등급, 치명도 4등급으로 위험도가 높은 것으로 나타났다. 이상의 분석 결과는 전동차 고속차단기의 설계 및 유지보수 업무의 개선에 활용할 수 있다.

방사형 회전이 추가된 임팩트 크러셔를 이용한 콘크리트용 순환굵은골재 품질향상 및 검증 연구 (A Study on Quality Improvement and Verification of Recycled Coarse Aggregate for Concrete Using an Impact Crusher with Radial Rotation)

  • 본 연구는 고품질의 콘크리트용 순환골재를 생산할 수 있는 타격 완충 장치인 방사형 회전판을 하단에 설치한 임팩트 크러셔를 개발하고 이를 이용하여 생산한 순환굵은골재의 품질성능 향상 효과 및 콘크리트 실험을 통해 적용 가능 여부를 검증하는 것을 목적으로 한다. 그 결과, 절대건조밀도, 흡수율, 마모감량, 입자모양판정실적률, 0.08 mm 체 통과량 시험에서 손실된 양, 알칼리골재반응, 점토덩어리량, 안정성, 이물질함유량 등 모든 항목에서 개선된 품질을 나타내었고 순환골재 품질기준에서 규정하는 기준에 부합하는 것으로 나타났다. 또한, 순환굵은골재를 적용한 콘크리트의 공기량 및 슬럼프는 국내 기준에 모두 부합하며 순환굵은골재의 혼입률에 따른 콘크리트의 재령별 압축강도 특성 실험 결과에 따라 순환굵은골재의 혼입률 60% 까지는 적용이 가능할 것으로 확인되었다.

국내 알루미늄 노출실태 및 노출기준 개정 제안 (Proposals for Revising the Occupational Exposure Limits for Aluminum in Korea)

  • Objectives: This study was intended to investigate the revision status of the occupational exposure standards for aluminum at home and abroad; to investigate worker exposure at domestic aluminum manufacturing and handling workplaces; to conduct social and economic evaluation for the revision of domestic aluminum exposure limits. Methods: We investigated the current status of occupational exposure limits for aluminum at home and abroad, and analyzed supporting data. An exposure survey was conducted targeting domestic aluminum manufacturing and handling workplaces. Based on these, revised aluminum occupational exposure limits were proposed. Results: The major aluminum exposure limits at home and abroad show a notable difference. The toxicity of aluminum, which was revealed through animal experiments and epidemiological investigations. The average concentration of aluminum in the air at 12 workplaces was 0.016 mg/m3, and the maximum was 0.0776 mg/m3. When total dust and respiratory dust were measured side by side and simultaneously for the same process, 12.1% of the total mass concentration of aluminum dust was respiratory dust. As a result of measuring and comparing the size distribution of dust with an optical particle counter in real time, 48.1% of the total dust in the form of welding fume and pyro-powder was respiratory dust. Based on the literature review and workplace survey, three proposals for changing the aluminum exposure limit were proposed. Proposal (1): For all types, 10 mg/m3 is unified as the exposure limit except for soluble salts and alkyls. Proposal (2): 1(R) mg/m3 as the exposure limit for all forms except soluble salts and alkyl. Proposal (3): 1(R) mg/m3 for pyro-powder and welding fume, and 10 mg/m3 for metal dust, aluminum oxide, and insoluble compounds as exposure standards. A pyro-powder was defined as dry aluminum powder of 200 mesh size (74 ㎛) or smaller (larger size classified as metal dust). Reason for setting: In the workplace survey, the ratio of respiratory dust to total dust was analyzed to be about 1:10, so it was judged that the domestic standard and the ACGIH standard were compatible. Conclusions: In all scenarios according to the revision of the exposure standard, the B/C ratio was greater than 1 or only benefits existed, so it was evaluated as sufficiently reasonable as a result of the socio-economic evaluation.

LC-MS/MS를 이용한 소시지 중 보존료 동시분석 (Simultaneous determination of preservatives in sausages using liquid chromatography with electrospray ionization tandem mass spectrometry)

  • A simultaneous determination method was developed for nine preservatives (benzoic acid, sorbic acid, dehydroacetic acid, methyl-, ethyl-, isopropyl-, propyl-, isobutyl- and butyl-parabens) in sausage by liquid chromatography with electrospray ionization tandem mass spectrometry (LC/ESI-MS/MS). Each parameter was established by multiple reaction monitoring in negative mode. Separation was achieved on a phenyl-hexyl ($2.5{\mu}m$, $2.1{\times}150mm$, Waters) with A-20 mM ammonium acetate containing 0.1% acetic acid in water, B-Acetonitrile as mobile phase with gradient mode at a flow rate of 0.3 mL/min. The developed method was validated for specificity, linearity, accuracy and precision in sausages samples. Linearity was over 0.998 with calibration curve of the mixed standards. The mean recoveries from sausages fortified at the level of 2.0~10.0 mg/L were in range of 98.60~109.16% with RSDs lower than 8.93%. The limits of detection (LOD) and the limits of quantification (LOQ) were in the range between 0.0003~0.085 mg/L and 0.01~0.257 mg/L, respectively. Intra-day precision and inter-day precision were 0.45~6.16% and 2.81~13.33%, respectively. Using presently developed determination method, 33 field sausage samples from Gwangju city in Korea were screened over nine preservatives. As a result, no preservatives were detected in all samples.
