We studied the development of schizandrin as a marker compound in Jangsu Omija. Schizandrin was validated for its LOD (limit of detection), LOQ (limit of quantitation), precision, accuracy, and recovery by HPLC relative to Omija powder. It showed a high linearity in the calibration curve with a coefficient of correlation ($R^2$) of 0.9990. The LOD and LOQ were 0.2 ${\mu}g/mL$ and 0.5 ${\mu}g/mL$, respectively. The intra- and inter-day precision of schizandrin calibration standards was 0.06~0.66% and 0.13~1.19%, respectively, and the intra- and inter-day accuracy of schizandrin was 98.35~103.30% and 98.35~103.00%, respectively. Overall, schizandrin was validated through our analytical methods as a marker compound in Jangsu Omija.
Objectives To check marker content for appropriate quality control of Woohwangchungsimwon sterilized to ensure microbiological safety and to observe antioxidant activity for any changes in efficacy. Methods To measure any effects of sterilization on the effective compounds, 8 ingredients of Woohwangchungsimwon were screened for any changes in marker content using HPLC-DAD. Using the colorimetric method on the microplate reader any changes in total phenolic compound and flavonoid levels were observed. Antioxidant activity was measured using the DPPH, ABTS, and FRAP. Results Of the ingredients of Woohwangchungsimwon, 8 were subject to quantitative analysis before and after sterilization. 21.6 mg and 1.93 mg of Glycyrrhizin was found in Glycyrrhiza uralensis Fischer pre and post sterilization, respectively. Decursin found in Angelica gigas Nakai increased from 0.16 mg to 0.29 mg after sterilization. Bilirubin found in Gallstone of Bostaurusvar. domesticus increased from 0.24 mg to 0.33 mg. Cinnamic acid found in Cinnamomum cassia Blume increased from 0.02 mg to 0.05 mg. Ginsenoside Rb1 found in Panax ginseng C. A. Meyer decreased from 0.02 mg to 0.14 mg. Paeoniflorin found in Paeonia lactiflora Pallas increased from 1.05 mg to 1.13 mg. Amygdalin found in Armeniacae Amarum Semen increased from 2.68 mg to 2.83 mg. L-muscone found in Musk increased from 0.63 mg to 0.76 mg. As for total phenolic compound and total flavonoid content, there was a 1.22 and 4.15-fold increase. DPPH and ABTS increased by 20.45% and 20.69%, respectively. FRAP activity was 2.78 times more active post stabilization. Conclusions This study confirmed that high temperature high pressure steam sterilization, a method used to ensure microbiological safety of Woohwangchungsimwon, does not affect marker content; in other words, does not affect quality of the Woohwangchungsimwon. It could also be seen that total phenolic compound and flavonoid content increased after sterilization. An antioxidant activity test showed that there was significantly increased activity of antioxidants.

Method development and validation of decursin for the standardization of Angelica gigas Nakai as a functional ingredient and health food were accomplished. The quantitative determination method of decursin as a marker compound of aerial parts of Angelica gigas Nakai extract (AAGE) was optimized by HPLC analysis using a C18 column ($3{\times}150mm$, $3{\mu}m$) with 0.1% TFA in water and acetonitrile as the mobile phase at a flow rate of 0.5 mL/min and detection wavelength of 330 nm. The HPLC/PDA method was applied successfully to quantification of the marker compound in AAGE after validation of the method with linearity, accuracy, and precision. The method showed high linearity in the calibration curve at a coefficient of correlation ($R^2$) of 0.9994 and the limit of detection and limit of quantitation were $0.011{\mu}g/mL$ and $0.033{\mu}g/mL$, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 1.10% and 1.13%, respectively. Recovery of decursin at 0.5, 1, 5 and $10{\mu}g/mL$ were 92.38 ~ 104.11%. These results suggest that the developed HPLC method is very useful for the determination of marker compound in AAGE to develop a health functional material.

Compound calcrete deposits are recognized from the Dadaepo Formation in Dusong peninsula, Pusan, and their textures, structures, morphology, and stratigraphic distribution are examined. The Dadaepo Formation in study area consists of fluvial plain deposits including floodplain-lake deposits, and the compound calcrete deposits occur above floodplain deposits and below lacustrine deposits. The compound calcrete deposits are composed of nodular to massive (micritic), laminar, nodular, nodular to massive (marly), and marly calcrete deposits. In these calcretes, circumgranular and curved cracks, alveolar structures, fitted peloids, tubular fenestrae with laminar micrite wall (rootlet casts), microstalactic calcite, and tepee structures are observed, which indicates that they are calcic paleosols. Considering their stratigraphic occurrences and pedogenic origin, compound calcrete deposits are interpreted to have formed on distal fluvial plain to palustrine environment by the repetetion of deposition and subsequent calcification due to alternation of lake expansion and contraction. The repetetion of calcrete formation suggests that arid climatic condition was alternated with humid condition in short-term period. Such alternated paleoclimatic condition is similar to those of the Sindong and Hayang groups, and indicates that short-term fluctuations from arid to humid climate were prevailing in overall Gyeongsang Basin. In stratigraphic sense, the compound calcrete deposits can be used as a marker deposit for the correlation of the Dadaepo Formation.

Centella asiatica (C. asiatica) has been widely used in food, cosmetics, and pharmaceutical industry as a functional material. In a previous study, we have investigated not only pharmacological effects such as antioxidative and anti-inflammatory effects, but also analyzed various functional ingredients. In this study, triterpenoids were analyzed using HPLC-DAD to determine marker compounds among functional ingredients. When triterpenoids were analyzed, asiaticoside from C. asiatica was determined as an optimal marker compound. Next, specificity, linearity, limited of detection (LOD), limited of quantification (LOQ), precision, accuracy, and range were evaluated using HPLC-DAD to determine asiaticoside contents in C. asiatica juice and extracts. The specificity was elucidated by chromatogram and retention time using an established analytical method. The coefficient of correlation obtained was 0.9996. LOD was 4.99 ㎍/mL and LOQ was 15.12 ㎍/mL. Intra- and inter-day precision of asiaticoside were determined to be 0.48~1.68% and 0.08~1.09%, respectively. Furthermore, the recovery rate of asiaticoside was 98.88% and the analytical range of Field-70E was determined to be 0.625~10 mg/mL. As a results of evaluating ABTS, DPPH, and FRAP antioxidative effect, Field-70E showed potent antioxidant activities. Results of this study could be used as basic data for quality standardization of C. astiatica juice and extracts.

Saposhnikoviae Radix (SR) and Peucedani Japonici Radix (PR) have been used as the main traditional herbal medicines in Korea, China and Japan. In this study, ultra-performance liquid chromatography coupled to quadrupole time of flight mass spectrometry (UPLC-QTOF/MS)-based metabolomics was applied to evaluate the quality of SR and PR using the marker compounds. In the S-plot of SR and PR, 5-O-methylvisammioside and peucedanol were selected as a marker compound for SR and PR, respectively. Also, an UPLC method was established and well validated for marker compounds of SR and PR. These results suggested that the established analysis method could be used one of the good methods for the classification and quality assessment of SR and PR.

Background: Compound K (CK) is a bioactive derivative of ginsenoside Rb1 in Panax ginseng (Korean ginseng). Its biological and pharmacological activities have been studied in various disease conditions, although its immunomodulatory role in innate immunity mediated by monocytes/macrophages has been poorly understood. In this study, we aimed to elucidate the regulatory role of CK on cellular events mediated by monocytes and macrophages in innate immune responses. Methods: The immunomodulatory role of CK was explored by various immunoassays including cell-cell adhesion, fibronectin adhesion, cell migration, phagocytic uptake, costimulatory molecules, reactive oxygen species production, luciferase activity, and by the measurement of mRNA levels of proinflammatory genes. Results: Compound K induced cell cluster formation through cell-cell adhesion, cell migration, and phagocytic activity, but it suppressed cell-tissue interactions in U937 and RAW264.7 cells. Compound K also upregulated the surface expression of the cell adhesion molecule cluster of differentiation (CD) 43 (CD43) and costimulatory molecules CD69, CD80, and CD86, but it downregulated the expression of monocyte differentiation marker CD82 in RAW264.7 cells. Moreover, CK induced the release of reactive oxygen species and induced messenger RNA expression of proinflammatory genes, inducible nitric oxide synthase, and tumor necrosis factor-alpha by enhancing the nuclear translocation and transcriptional activities of nuclear factor kappa-B and activator protein-1. Conclusion: Our results suggest that CK has an immunomodulatory role in innate immune responses through regulating various cellular events mediated by monocytes and macrophages.
Experiments were carried out to examine the rusty tolerance in 61 inbred lines of ginseng cultivated in field, and chemical components were analyzed to clarify the difference between healthy and rusty ginseng roots. Among them, 10 lines showed rusty tolerance (RT) while 10 lines showed rusty sensitivity (RS). The content of phenolic compound in RT was lower than that in RS in cortex, epidermis and branch & fine roots, but it was not difference between RT and RS in stele. The contents of K, Ca, Na in RT were lower than RS in cortex, and the content of Mg, Fe, Na, Mn, AI, Si in RT were lower than RS in epidermis, and the content of Fe in RT were lower than RS in branch & fine roots, but mineral contents were not difference between RT and RS in stele. The content of phenolic compound in healthy cortex was lower than that in rusty cortex in same 6-year roots, but the mineral contents were not difference between healthy and rusty cortex in same 6-year roots. In root of seedlings, the contents of phenolic compound, K and Na in RT were lower than RS. It was suggested that the contents of phenolic compound, K and Na might be marker to select rusty tolerance ginseng lines.
For the syntheses of tritium labelled polystyrene, the basic material for the preparation of tritium luminous compound, various methods of labelling such as Tesla discharge, Wilzbach exposure, gamma irradiation, and U V. irradiation were compared in view of getting high specific activity-product. The obtained polystyrene-T(G) by the method of Tesla discharge and by the method of U. V. irradiation had specific activity of 1~1.2 mCi/mg, and these two methods were the most encouraging. Mixing of 1 part of polystyrene-T(G) with 4 parts of ZnS:Cu phosphor, in weight, appeared to be the most suitable ratio in tile preparation of luminous compound in luminosity point of view. When 30 mg. of obtained luminescent mixture was applied on steel plate by using 1 ml. of the selected binder (i.e., 1g of commercial varnish in 100m1. of benzene) the luminosity maximum was ca. 20 micro Lambert. The prepared luminous compound was confirmed to be practically applicable for mine marker or dark-room light source.

In present study, we conducted quantification analysis of phenolic compound (paeonol), monoterpene glycoside (paeoniflorin), and tannin ((+)-catechin in the 70% ethanol extracts of non-processed Moutan Radicis Cortex (MRC) and processed MRC by roasting using a high-performance liquid chromatography coupled with photodiode array detector. Three marker components were separated on Gemini $C_{18}$ analytical column and the column was maintained at $40^{\circ}C$ using two mobile phase system consisting of 1.0% (v/v) aqueous acetic acid and 1.0% (v/v) acetic acid in acetonitrile. The flow rate and injection volume were 1.0 mL/min and 10 mL. In non-processed MRC sample, the concentrations of three marker compounds, (+)-catechin, paeoniflorin, and paeonol were 0.20, 1.18, and 2.12%, respectively. On the other hand, the concentrations of the three compounds in processed MRC samples were 0.03-0.24, not detected-1.08, and 0.76-1.82%, respectively.
