A Study on Effectiveness of Utilizing Local R&D Centers in Science and Technology ODA Projects : Focusing on the Black Pearl Cultivation Project of the Korea Institute of Ocean Science & Technology and Micronesia (과학기술 ODA 사업에서의 현지 R&D센터 활용에 관한 연구 : 한국해양과학기술원과 마이크로네시아 '흑진주 생산연구' 사례를 중심으로)

  • The purpose of this study is to demonstrate, through case studies, the usefulness of utilizing local R&D centers under science and technology ODA programs. For the past few decades, advanced countries have supported ODA projects of developing countries, but there have been negative opinions regarding the results. Through a case study of the black pearl cultivation project between the Korea Institute of Ocean Science & Technology and Micronesia, this study explains the usefulness of actively utilizing Korean R&D centers established and operational in recipient countries. Although black pearl cultivation is not an ODA project, the case study offers valuable insights as it is operated in a similar form and thus highly applicable to future projects. Based on the case study, four implications were derived to ensure the successful operations of science and technology ODA projects in the future. First, there is a need to improve relevance by making use of the technological capacities of local R&D institutes to develop projects that reflect the needs of recipient and donor countries. Second, trust must be established with local communities over the long term in order to enhance the effectiveness and efficiency of project operations. Third, the proportion of science and technology ODA projects must be expanded to acquire sustainability, and more support should be granted to ODA projects involving marine resources, which are an advantage for countries of Micronesia. Lastly, the locals should be offered employment opportunities and regular training programs to allow for the actual transfer of knowledge instead of mere techniques. The implications derived in this study will prove useful in pursuing science and technology ODA projects, especially with Micronesia.

Comparison of Nekton Assemblage Structures Between Estuary and Inshore Waters on the Mid-Western Coast of Korea (서해 하구역과 연안역 유영생물의 군집구조 비교)

  • To elucidate the species compositions of estuarine and inshore fauna and compare productivities between them, catch were collected by bag nets in the Han River estuary off Ganghwa Island and the inshore waters off Jangbong Island for 24 hours at the spring tide from March to December 2010. Community structures of nekton assemblages were different with area, month, taxonomic groups. Although the diversity of estuarine nekton assemblage was lower than that of inshore nekton assemblage, the productivity of nekton assemblage in estuary was higher than that in the inshore waters. Nekton assemblages were distributed according to habitat environment: the estuarine species, Coilia mystus, Chelon haematocheilus, Collichthys lucidus were prominent in the Han River estuary off Ganghwa Island, and marine species, Thryssa kammalensis, Sardinella zunasi, Portunus trituberculatus were prominent in the inshore waters off Jangbong Island. Palaemon gravieri, Exopalaemon carinicauda, Synechogobius hasta were inhabited in both waters. The health assessment of estuarine ecosystem based on scientific research will be contributed to the effective management of estuary with determination of order and direction in policy.

Antibacterial Effects of Pseudomonas aeruginosa MB I-3 against Listonella anguillarum (어류 병원세균, Listonella anguillarum에 대한 Pseudomonas aeruginosa MB I-3의 항균 효과)

  • To study the possible use of probiotics in fish farming, The in vitro and in vivo antibacterial effects of Pseudomonas aeruginosa MB I-3 (MB I-3) against the fish pathogenic bacterium Listonella anguillarum were evaluated. The inhibitory effects of MB I-3 against vibrios were investigated by the double layer method and the co-culture. The results showed that MB I-3 inhibited the growth of pathogenic vibrios including Listonella anguillarum, Vibrio alginolyticus, Vibrio cholerae, Vibrio fluvialis, Vibrio furnissii, Vibrio harveyi, Vibrio parahaemolyticus and Vibrio vulnificus. Extracellular substances obtained from the cultural supernatant of MB I-3 by ethyl acetate extraction showed inhibitory effects on L. anguillarum. The antibacterial substance of MB I-3 was evaluated to destroy the cell membrane of L. anguillarum in electron micrographs. The probiotic effects of MB I-3 was tested by exposing olive flounder (Paralichthys olivaceus) fry to L. anguillarum with or without MB I-3. The cumulative mortality of olive flounder fry infected with L. anguillarum was 24% in the group with MB I-3, while it was 46% in the control group without MB I-3. These results indicate that MB I-3 has potential applications as a probiotic for the control of fish pathogenic vibrios in fish rearing system.

Combined effects of copper and temperature on Hematological constituents in the Rock fish, Sebastes schlegeli (조피볼락, Sebastes schlegeli의 혈액학적 성분변화에 미치는 구리 및 온도의 복합적 영향)

  • Copper ($CuSO_4$) has been widely used to control algae and pathogens in fish culture ponds. However, its toxic effects on fish depend not only on its concentration in water but also on water quality. The susceptibility of the rockfish, Sebastes schlegeli to copper was evaluated at three water-temperatures (WT; 18, 23 and $28^{\circ}C$) for 4 days. After the exposure of two copper concentrations (100 and $200{\mu}g/L$), a hematological effect was exerted on rockfish, by causing changes in red blood cell count and hematocrit value at $28^{\circ}C$. Total protein levels of the fish showed a tendency of co-increase with glucose depend on the WT, after copper exposure. However, the plasma calcium and magnesium levels were significantly increased at $200{\mu}g/L$ copper, regardless of the WT. Enzymes activities including ALT and LDH in serum were also significantly increased depend upon the copper treatment only. This indicates that inorganic components and enzymes activities were sensitive indexes to stress by toxicant such as copper. The cortisol levels were significantly elevated by both WT rising and copper treatment in serum of rock fish. In conclusion, these changes can be seen as an initial response to temperature stress and as a sustaining response to copper exposure. The present findings suggest that a simultaneous stress by temperature change and copper exposure could accelerate an alteration of hematological and plasma biological parameters in the rockfish.

Osteological Development of the Larvae and Juvenile in Coreoperca herzi (Perciformes: Centropomidae) (꺽지 (Coreoperca herzi) 자치어의 골격발달)

  • Samples were obtained from broodstok in May, 2010, while naturally fertilized embryos were maintained, and the process of skeletal development was observed from larvae and juvenile. Prelarvae immediately after hatching showed an average total length of $6.85{\pm}0.63mm$ (n =5), Premaxillary and dentary were ossified, parasphenoid was ossified in the cranium, and centrum and caudal bone did not ossify. Prelarvae showed ossification with maxillary, articular, and epihyal, and branchiostegal rays of hyoid arch were ossified at 2 to 3 days after hatching with an average total length of $7.25{\pm}0.28mm$ (n=5). The vertebrae began to ossify in the direction of the tail, and neural spine began to ossify above the ossified vertebra. Postlarvae showed ossification of lateral ethmoid, alisphenoid, parietal, and caudal skeleton in the cranium when the average total length was $9.00{\pm}1.53mm$ (n=5) in 6 days after hatching. At 40 days after hatching, postlarvae ossified maxillary in the cranium, and ossified endopterygoid and ectopterygoid, etc. in the palate, when the average length of $23.3{\pm}0.28mm$ (n=5). At 120 days after hatching, with the average length was $37.5{\pm}2.83mm$ (n=5), caudal skeleton had one additional epural bone ossification, resulting in ossification of a total of 3 epural bone to complete ossification of all spicules.

  • Spermatogenesis and reproductive cycle of the razor clam, solen grandis, were investigated monthly by histological and cytological observations. Samples were collected from natural intertidal population at Oshik-do, Kunsan, Korea, for one year, beginning from January to December, 1993. solen grandis is dioecious. Morphological structures of the spermatozoon of this species ar esimilar to those of other bivalve spermatozoa having a primitive type; i.e., a small head, a cap-shaped acrosome and a short mid-piece with four mitochondria surrounding axial filament. The head of spermatozoon is approximately 2 \mu m in length and sperm tail is about 20 \mu m long. The axoneme of tail flagellum consists of nine pairs of peripheral microtubules at the periphery and a pair of central microtubules at the center. Four spherical mitochondria form the paranucleus. Spawning occures once a year between early June and July, and the main spawning was observed in July when seawater temperature reaches above 20 \circ C. The reproductive cycle of male razor clam can be divieded into fivesuccessive stages; early active (December to january), late active (January to march), mature (March to early August), partially spawned (June to July), and spent/inactive stage (August to December).

Phylogenetic Analysis Using Cytochrome c Oxidase Subunit I of Silver Croaker(Pennahia argentata) Mitochondria DNA (미토콘드리아 DNA의 cytochrome c oxidase subunit I을 이용한 보구치(Pennahia argentata) 계통 분석)

  • Silver croaker (Pennahia argentata) is a turbulent species that is widely distributed worldwide and is mainly found in the bottom of the ocean. In the study, we characterized the cytochrome c oxidase subunit I (COI) gene of the mitochondrial DNA (mtDNA) on P. argentata inhabiting Gwangyang Bay and analyzed the phylogenetic location of marine fish species. As a result of multiple arrangement of 605 bp COI sequences, high homology of mtDNA nucleotide sequences was confirmed in the silver croakers from Gwangyang Bay (98~100%). However, the nucleotide variation was different according to the catching points of the inland and the open seas of Gwangyang Bay. The nucleotide sequence variation in COI was high in P. argentata from the open seas of Gwangyang Bay (43.2~70.3%). Furthermore, the phylogenetic analysis of 13 fish showed that P. argentata from Gwangyang Bay were grouped into one clade with P. argentata reported in Taiwan, and the evolutionary distance was 0.036. In addition, it was identified that the evolutionary distance was close to that of fish belonging to the Mi-iuy croaker (Miichthys miiuy) and the Big-head pennah croaker (Pennahia Macrocephalus) (0.041~0.048). The result of these studies will be used as the key genetic information for fisheries resources monitoring and species diversity management according to the coastal environment.

Subtidal Macrobenthic Community in Shia Sea, Southwest Coast of Korea (한국 서남해역 시아해의 조하대 저서동물 군집)

  • In order to study the community structure and distribution pattern of macrobenthos in the subtidal area of Shia Sea between Haenam peninsula and Shinan-Jindo area at southwest coast of Korea, duplicate sediment samples were taken seasonally by van Veen grab (surface area, $0.1m^2$) from 49 stations between November 2001 and August 2002. The surface sediment facies were muddy sandy gravel near Mokpo-Gu channel, gravel in the Myoungrang channel with high tidal currents, and sandy muddy in the southern Uido area. Ignition loss in the sediment ranged from 0.5 to 1.5% with a mean of 0.6%. A total of 271 taxa were collected with a mean density of $166ind.\;m^{-2}$ and a mean biomass of $29.2g\;m^{-2}$. Polychaetes were dominant faunal group in terms of species richness and abundance, accounting for 34% and 40% of total species number and abundance, respectively. Molluscs accounted for 44% of biomass. Bio-Env analysis revealed that sediment composition and ignition loss affected the spatial distribution of subtidal macrobenthic community in the study area. Major dominant species were Photis longicaudata, Septifer keenae, Maldane cristata, Lumbrineris longifolia, Heteromastus filiformis and Coptothyris grayi. Based on cluster analysis, four station groups were classified. They were associated with sediment types. Bivalve, Striarca symmetrica, polychaetes, Glycera chirori, Heteromastus filiformis and decapod Latreutes planirostris were major contributing fauna in station group A, B, C, and D, respectively. These results suggest that macrobenthic community of the Shia Sea is normally influenced by sediment type and related ignition loss.

Spatial distributions of phytoplankton community in the coastal waters of South Sea, Korea during the early summer of 2018 (2018년 이른 여름 남해 연안해역 식물플랑크톤 군집의 공간분포 특성)

  • For this study, we carried out a field survey on the analysis for the spatial distributions of phytoplankton community in the eleven areas of the Korean South Sea during the early summer of 2018. The results from the study showed that the phytoplankton community consisted of 56 genera and 105 species showing by diatoms with 52.4%, dinoflagellates with 40.0% and other phytoflagellates with 7.6%. The cell density of the phytoplankton ranged from 5.5 to 593.2 cells mL-1. The species number and cell density of the phytoplankton were high in the eastern waters of the South Sea and low in the western one. The phytoplankton community showed the characteristics of being dominated by the diatoms except in the Geumpo of Namhae, Ocheon-dong of Yeosu and Oenarodo of Goheung. The dominant species of the phytoplankton community were the centric diatoms, Skeletonema costatum-like species (ls), except for the Ocheon-dong and Chungdo of Wando. However, the Ocheon-dong was dominated by toxic dinoflagellate, Gymnodinium catenatum by 41.1% dominance. On the other hand, Keumpo and Oenarodo was by dinoflagellate, Tripos fusus more than 12% dominance in the surface layer. The spatial distribution of the phytoplankton community in the coastal waters of the Korean South Sea in the early summer were determined by the supply of nutrients through precipitation.

Seasonal Photosynthetic Performance of Laminaria japonica sporophytes on the Southeastern Coast of Korea (한국 동남해안에서 다시마(Laminaria japonica) 포자체의 광합성의 계절변화)

  • Rates of photosynthesis and respiration of Laminaria japonica sporophytes cultivated on the southeastern coast of Korea were monthly measured in situ and under constant temperature and nitrogen concentration in laboratory from February to July 1996 in order to understand the seasonal photosynthetic performance of this alga. P-I (the relationship between photosynthsis and irradiance) parameters measured in situ varied seasonally. Photosynthetic capacity ($P_{max}$) reached its maximum in March (6.64 mg $O_2{\cdot}gdw^{-1}{\cdot}h^{-1}$) and gradually decreased thereafter. Photosynthetic efficiency (${\alpha}$), which ranged from 0.026 to 0.106, generally showed a similar pattern with the $P_{max}$, curve. Correlation between respiration and $P_{max}$, was not significant (Spearman's rank correlation, p>0.05); respiration rate, which varied between 0.25 and 0.83 mg $O_2{\cdot}gdw^{-1}{\cdot}h^{-1}$, showed no gradual decline from March. $P_{max}$ in situ significantly correlated with the relative growth rate of frond weight (Spearman's rank correlation, p<0.01) and this result means that the amount of accumulated materials in body increased with the increment of $P_{max}$. Compared $P_{max}$, in situ with that in the laboratory. The lower $P_{max}$ in February was probably caused by the effect of lower seawater temperature at this time. The decrease in $P_{max}$ after March, however, was primarily attributable to the thickness of blade as the plants got old. Comparison of P-I parameters measured for different size groups in April, explained the negative relationship between $P_{max}$ and frond thickness.
