Classification into Two Types Based on the Morphological Characteristics of the Marine Diatom Ditylum brightwellii (West) Grunow in Yellow Sea, Korea (우리나라 황해의 해산 돌말류 Ditylum brightwellii (West) Grunow의 형태학적 특징에 의한 두 유형)

  • This study deals with the taxonomy of the marine diatom Ditylum brightwellii (West) Grunow: shape, size and ratio, marginal ridge, labiate process, areolation, and seasonal distribution are all considered. Ditylum brightwellii were divided into two type; prism- and cylinder-shaped. Comparisons between specimens from the Yellow Sea were made on, (1) shape- prism types are prismatic-shaped and/or regular square or right-angled tetragon, cylinder types are cylinder-shaped and/or long right-angled tetragon in the girdle view, respectively; (2) size and ratiodiameters of prism types are 82.1 ± 2 μm, while those of cylinder types are 21.2 ± 5 μm and ratio of pervalvar axis/diameter- prism types cells are 1.0-1.5, but cylinder types are 3.9-5.5; (3) marginal ridges- prism types have dotted, while those of the cylinder-type cells have piece, slotted or fimbriated on the marginal ridge; (4) labiate process- both prism- and cylinder-types are central part in valve face, but length of labiate processes of prism types are longer than those of cylinder types; (5) cell margin- the girdle margin show many shallow furrows in prism type and smooth in cylinder type; (6) areolation- both prism- and cylinder-types are a radial pattern in the valve center, but areolae of prism types are larger than those of cylinder types; (7) seasonal distribution- cylinder types are dominated in summer and prism types in the other seasons.

Easy and rapid quantification of lipid contents of marine dinoflagellates using the sulpho-phospho-vanillin method

  • To develop an easy and rapid method of quantifying lipid contents of marine dinoflagellates, we quantified lipid contents of common dinoflagellate species using a colorimetric method based on the sulpho-phospho-vanillin reaction. In this method, the optical density measured using a spectrophotometer was significantly positively correlated with the known lipid content of a standard oil (Canola oil). When using this method, the lipid content of each of the dinoflagellates Alexandrium minutum, Prorocentrum micans, P. minimum, and Lingulodinium polyedrum was also significantly positively correlated with the optical density and equivalent intensity of color. Thus, when comparing the color intensity or the optical density of a sample of a microalgal species with known color intensities or optical density, the lipid content of the target species could be rapidly quantified. Furthermore, the results of the sensitivity tests showed that only $1-3{\times}10^5cells$ of P. minimum and A. minutum, $10^4cells$ of P. micans, and $10^3cells$ of L. polyedrum (approximately 1-5 mL of dense cultures) were needed to determine the lipid content per cell. When the lipid content per cell of 9 dinoflagellates, a diatom, and a chlorophyte was analyzed using this method, the lipid content per cell of these microalgae, with the exception of the diatom, were significantly positively correlated with cell size, however, volume specific lipid content per cell was negatively correlated with cell size. Thus, this sulpho-phospho-vanillin method is an easy and rapid method of quantifying the lipid content of autotrophic, mixotrophic, and heterotrophic dinoflagellate species.

  • SEM and LM studies were made to examine the frustule of the marine epizoic diatom Pseudohimantidium pacificum Hustedt & Krasske and to examine the variability of frustule morphology. The length of the apical axis and the transapical axis were similar to other reports. but Number of labiate processes showed a variation. the Number of striae of Korean specimens were similar to those of other regions, but the areolae shape of striae showed elliptical type. Pseudohimantidium pacificum were palnktonic, tychopelagic or epizoic attached to the prosome, the genital segment and abdomen of corycaeus affinis. this species was newly recorded I Korean waters and the biogeographic boundary of this species can therefore be extended to the northern Pacific Ocean.

The Effects of Substituting Squid Meal and Macroalgae with Soybean Meal in a Commercial Diet on Growth and Body Composition of Juvenile Abalone Haliotis discus hannai (전복(Haliotis discus hannai) 용 배합사료내 오징어분 및 해조류 대체원으로서 대두박이 전복 치패의 성장과 체조성에 미치는 영향)

  • We examined the effect of substituting squid meal and macroalgae with soybean meal in a commercial diet on the growth and body composition of juvenile abalone Haliotis discus hannai. We randomly distributed 2310 juvenile abalone into 33 rectangular plastic containers and fed them five experimental diets in triplicate as follows. The control diet (Con) consisted of 12% squid meal, 8% corn gluten and 20% soybean meal as protein source, wherein 10% ${\alpha}$-starch, 20% wheat flour, and 5% dextrin were carbohydrate source. The experimental diets, 50% squid meal (SM50), 50% squid meal and 50% macroalgae (SM50+MA50), and 100% squid meal and 50% macroalgae (SM100+MA50) were substituted with the same respective amounts of soybean meal. The fifth experimental diet consisted of the control diet plus 1% diatom powder (DP). We prepared two domestic (Domestic A and B) and two imported (China and Japan) abalone feeds. Finally, we prepared Undaria and sea tangle. We found that the weight gain of abalone fed the Con, DP, and China and Japan diets was significantly greater than that of abalone fed Undaria and sea tangle. We conclude that the substituting squid meal and macroalgae with soybean meal in abalone feed has limited benefits, but supplementing diets with 1% diatom powder is effective in improving weight gain.

A New Marine Species of Miracula (Oomycota) Parasitic to Minidiscus sp. in Iceland

  • Obligate endoparasitic oomycetes are known to ubiquitously occur in marine and freshwater diatoms, but their diversity is still largely unexplored. Many of these parasitoids are members of the early-diverging oomycete lineages (Miracula, Diatomophthora), others are within the Leptomitales of the Saprolegniomycetes (Ectrogella, Lagenisma) and some have been described in the Peronosporomycetes (Aphanomycopsis, Lagenidium). Even though some species have been recently described and two new genera were introduced (Miracula and Diatomophthora), the phylogeny and taxonomy of most of these organisms remain unresolved. This is contrasted by the high number of sequences from unclassified species, as recently revealed from environmental sequencing, suggesting the presence of several undiscovered species. In this study, a new species of Miracula is reported from a marine centric diatom (Minidiscus sp.) isolated from Skagaströnd harbor in Northwest Iceland. The morphology and life cycle traits of this novel oomycete parasite are described herein, and its taxonomic placement within the genus Miracula is confirmed by molecular phylogeny. As it cannot be assigned to any previously described species, it is introduced as Miracula islandica in this study. The genus Miracula thus contains three described holocarpic species (M. helgolandica, M. islandica, M. moenusica) to which likely additional species will need to be added in the future, considering the presence of several lineages known only from environmental sequencing that clustered within the Miracula clade.

Seasonal Variation of Primary Productivity and Pigment of Phytoplankton Community Structure in the Seomjin Estuary (섬진강 하구역의 계절별 일차생산력 및 식물플랑크톤 색소 조성 변화)

  • Four in situ incubation experiments were conducted in May, August and November 2009, as well as February 2010 to determine the seasonal primary productivity and the community structure of phytoplankton at the Seomjin estuary. The primary production of phytoplankton ranged from 9 to 3560 mgC $m^{-2}\;d^{-1}$. Primary productivity was the highest in the summer season (August), which was influenced by improved optical and temperature conditions of the water, as well as the supply of nutrients derived from its surrounding watershed. Particularly, the upper station (SJ-1, SJ-2) of Seom-jin estuary showed a higher productivity, as a result of inflow of input nutrients originated from the terrestrial source. The fucoxanthin, as an index pigment of diatoms showed the highest concentration (0.74~9.51 ${\mu}g\;L^{-1}$) at all stations, occupying 30~80% to total Chl a concentrations. The phytoplankton species composition determination, using a microscope showed similar results to the pigment analysis, which indicated diatom Skeletonema costatum, as the dominant species. The primary productivity in Seom-jin estuary indicates temporally and spatially large variation, according to different environmental conditions. Also, Skeletonema costatum has euryhaline features with relatively higher contribution.

  • Four Rhizosolenia species of marine diatom were appeared at 30 sites in Korean coastal waters from September 2008 to February 2010. The cell shape, diameter, length of pervalvar axis, external process, segment and areolation characters of the species were examined. All four species were cylindrically shaped, but the external processes differed. Segment striations were regularly straight, and velum showed hexagonal, circular pore and narrow slit. Rhizosolenia formosa and R. hyalina were only occurred in September 2008 from the Yellow Sea, R. bergonii was appeared throughout the year from Korea Strait and Yangyang coasts, and R. setigera was abundantly showed in all seasons from all site. R. formosa is a newly recorded species in Korean coastal waters.

  • A study on the fine structure of the marine diatom has been carried out for taxonomical purpose during the period from Fib. 1982 to Feb. 1986 in Korean coastal waters. Fine structure of small Thalassiosira (Bacillariophyceae) was studied by means of light and scanning electron microscopy. Firstly, four species of Thalassiosira were identified and described. Of these Thalassiosira weissflogii, T.minima and T.oestrupii var venrickae were new records for Korean coastal waters.

  • A study on the fine structure of the marine diatom Thalassiosira has been carried out during the periods from January 2007 to March 2008 in Korean coastal waters. As the third series of the Thalassiosira species, a fine structure, description, distribution and taxonomic remarks of the six Thalassiosira species were observed by means of light microscope and scanning electron microscope. The critical features of Thalassiosira species were a shape of external tubes of marginal strutted processes and labiate process. Six species showed each different shape of external tubes, marginal strutted processes and labiate process. The shape of external tube was divided into five types: T shape of Thalassiosira curviseriata, small-rounded shape of T. lundiana, double-layer form and flame shape of T. nordenskioeldii, tulip shape of T. punctigera and tooth-shape of T. tenera. This external character may be able to key character for positive identification of the Thalassiosira species. Of these Thalassiosira lundiana, T. minuscula and T. tenera were new records for Korean coastal waters.

Chemical and Mineralogical Properties on the Ulsan Marine Deposited Clay (울산 퇴적 점토의 화학적, 광물학적 특성)

  • 울산지역에 널리 분포하고 있는 해성 퇴적토에 대하여 깊이별로 물리, 화학 그리고 광물학적 특성을 파악하였으며, 이 지역에서 채취된 시료에 대하여 깊이별로 퓨준압말시험을 실시하였다. 본 논문을 위하여 울산 해성 퇴적토의 생성과정을 추정하고, X-선 회절 분석, X-선 형광분석 및 에너지분산분광분석을 실시하여 깊이에 따른 변화를 비교하였다. 울산 해성 퇴적토의 점토광물에 대한 정량분석결과, 일라이트, 카올리나이트, 녹니석, 스멕타이트 순으로 많게 나타났음, 화학성분 분석결과, SiO$_2$, $Al_2$O$_3$그리고 Fe$_2$O$_3$가 대부분을 차지하는 것으로 나타났다. 주사전자현미경 관찰결과, 유공충과 규조류가 관찰되었다. 표준압밀시험 결과, 울산지역의 해성 퇴적 점토는 깊이에 따른 변형거동이 화학적, 광물학적 분석결과와 마찬가지로 깊이에 따른 변화를 보이지 않아 같은 시디에 퇴적인 것으로 판단된다.
