바지락 (Ruditapes philippinarum)의 번식량 측정 및 유생발달에 관한 연구 (Quantification of Reproductive Effort and Microscopic Observation on the Larval Development of Manila clam Ruditapes philippinarum (Adams and Reeve, 1850))

  • 이 연구에서는 바지락 (Ruditapes philippinarum) 의 번식량 및 유생발생을 관찰하였으며, 이를 위하여 수정란 및 유생을 21일 간 실내사육수조를 이용하여 배양하였다. 바지락은 2011년 8월 제주시 김녕에서 채집 실험 수조로 옮겨졌으며, 다음날 자연적인 방란 방정이 관측되었다. 바지락 수정란은 19시간의 배 발생과정을 거쳐 21일 후 포복피면자유생(pediveliger) 으로 발달하였다. 조직학적 방법을 이용하여 바지락의 생식소를 관찰한 결과, 방란, 방정 전 모든 개체는 성적으로 완숙한 상태였고, 산란 후 모든 개체에서 부분산란이 관찰되었다. 효소면역학적 방법 (ELISA) 에 의한 번식량 측정 결과, 암컷 바지락의 생식소 지수는 (GSI) 산란 전 28.6%에서 산란 후 17.3%로 11.3% 감소하였으며, 추정된 포란수는 6,998,658 eggs였다. 이 연구서 광학현미경 및 전자현미경을 이용하여 관찰된 바지락 유생 발달 및 번식량은 향후 바지락 인공종묘 산업 및 바지락 자원관리에 있어 중요한 자료로 활용될 것으로 사료된다.

  • The Manila clam Ruditapes philippinarum, is an important marine bivalve that is widely distributed along the west and north coasts of South Korea. It has been used in a variety of Korean foods owing to its superior umami taste. In the present study, we developed a flavoring with an excellent sensory preference from Manila clam using extrusion cooking processing. Optimization of extrusion cooking conditions was performed using response surface methodology (RSM). Barrel temperature (X1, 140-160℃) and screw speed (X2, 400-560 rpm) of the extruder were chosen as independent variables. The dependent variable was overall acceptance (Y, points). The estimated optimal conditions were as follows: overall acceptance (Y): X1=140℃ and X2=560 rpm. The indicated value of the dependent variable overall acceptance (Y) under the optimal conditions was 8.94 points, which was similar to the experimental value (8.82 points). Overall acceptance of the Manila clam flavoring was related to its umami and Manila clam tastes. The electronic nose and tongue results successfully segregated different clusters of the samples between the lowest and highest sensory scores. The sample with the highest sensory score had higher sourness, umami, and sweetness intensities, and the lowest sensory scored sample showed more off-flavor compounds.

  • Oxygen consumption by the Manila clam, Ruditapes philippinarum (Bivalvia: Veneridae) maintained in a culture tank for 7-10 weeks was measured with an automatic intermittent-flow-respirometer (AIFR). There was evidence for an endogenous tidal rhythm of 12.4 h in oxygen consumption, at irregular intervals of between about 7 and 18 hours during experiments of between 22 h 5 min and 70 h 32 min (duration) recording for fasting animals in constant darkness at constant temperatures in the absence of tidal rhythms.

  • Gonad maturation of Manila clam, Ruditapes philippinarum was induced in this study using a recirculation system over 8 weeks in early spring. Clams used in the experiment were collected in $15^{th}$ April 2010 from the west coast of Korea, as the surface water temperature remained $11^{\circ}C$. To induce gametogenesis and subsequent maturation seawater temperature was elevated $1^{\circ}C$ per day over 10 days to reach $20^{\circ}C$. For the experiment, clams were raised in 120 L quadrangle tank maintained with re-circulated seawater system over 57 days. Water quality parameters including the water temperature, salinity dissolved oxygen, ammonium ion and nitrate levels in the tanks were monitored daily. Mixture of concentrated microalgae including Tetraselmis, Isochrysis, Pavlova and Thalassiosira weissflogii was supplied to clams twice a day, and quantity of the daily ration was adjusted as 3% of clam body dry weight. Histology was applied to examine gonad maturation. Daily monitoring of the water quality parameters indicated that the recirculation system supplied suitable environment to Manila clam; the nitrogenous components stayed below toxic levels (< 0.2 mg/L). At the beginning of the study, clams were mostly in early developing stage. As the seawater temperature reached $20^{\circ}C$, 10 days after the experiment, 20% of clams reached late development at 12 days. First ripe clams were observed at 42 days and 40% of clams were in ripe and ready for spawning at the end of study, 57 days after the experiment. In this study, gametogenesis of Manila clam was successfully induced by elevating water temperature and supplying commercially produced microalgae in a recirculation tank system.

곰소만 바지락(Ruditapes phillippinarum) 양식장 주변 퇴적물 내 유기물과 미량금속 분포특성 (Distributions of Organic Matter and Trace Metals in Surface Sediments around a Manila Clam Ruditapes phillippinarum Farming Area in Gomso Bay, Korea)

  • Organic matter and trace metals were investigated in surface sediments of Gomso Bay, where there is dense Manila clam Ruditapes philippinarum farming activity, to evaluate contamination of sediments in intertidal shellfish farming area. We measured mean grain size (Mz), chemical oxygen demand (COD), ignition loss (IL), acid volatile sulfide (AVS), total organic carbon (TOC), and total nitrogen (TN), and trace metals (As, Cd, Cr, Cu, Fe, Hg, Mn, Pb and Zn) in intertidal and sublittoral sediments. The intertidal sediments were mainly composed of coarser sediments (sand, silty sand, and sandy silt), with Mz values ranging from 2.61 to 4.79 Ø. Mz and the content of organic matter in sediments were lower in the intertidal zone than in the sublittoral zone. The mean metal concentrations in surface sediments decreased in the order Fe > Mn > Zn > Cr > Pb > Cu > As > Cd > Hg. The metal concentrations in surface sediments showed a significant positive correlation with Mz and organic matter content, indicating that metal concentrations in the surface sediments of Gomso Bay are controlled by Mz and organic matter. The concentrations of organic matter and trace metals in the study region were lower than or similar to those in other intertidal zones in western coast and much lower than those reported in other shellfish farming areas in Korea. Our results suggest that intertidal Manila clam farming sediments from Gomso Bay are not contaminated by organic matter and trace metals.

Morphological and Molecular Confirmation of Parvatrema duboisi Metacercariae in the Manila Clam Ruditapes philippinarum from Gochang-gun, Korea

  • Gymnophallid metacercariae found in the Manila clam Ruditapes philippinarum ('Banjirak' in Korean) from Gochang-gun, Jeollabuk-do, Korea were morphologically and molecularly confirmed to be Parvatrema duboisi (Dollfus, 1923) Bartoli, 1974. The metacercariae were morphologically characterized by having a large oral sucker, small ventral sucker, genital pore some distance anterior to the ventral sucker, no ventral pit, and 1 compact or slightly lobed vitellarium, which were all compatible with P. duboisi. Some of the metacercariae were experimentally fed to mice, and adult flukes were recovered at day 7 post-infection. The morphology of the adult flukes was basically the same as that of the metacercariae except for the presence of uterine eggs; the uterus was filled with up to 40 eggs. The nucleotide sequences (1,193 bp) from ITS regions (ITS1, 5.8S rDNA, and ITS2) of the metacercariae showed 99.7% identity with P. duboisi and 75.7% identity with Gymnophalloides seoi deposited in GenBank. These results confirmed the presence of P. duboisi metacercariae in the Manila clam R. philippinarum in an estuary region of Gochang-gun, Korea.

Pathology Survey of the Manila Clam Ruditapes philippinarum from Hwangdo Tidal Flat in Cheonsu Bay on the West Coast of Korea

  • Manila clam Ruditapes philippinarum is present at high rates of density in tidal flats in Cheonsu Bay on the west coast of Korea, where clams often exhibit mass mortalities in late summer. We monitored the pathologic condition of clams at Hwangdo tidal flat (HD) to understand the parasitic impacts on clam fitness. Manila clams were fully ripe in July and spawned during August and September, as the histology indicated. The histology revealed that clams in HD tidal flats were heavily infected by the protozoa parasite Perkinsus olseni, as the monthly prevalence ranged from 53% (September) to 93% (August). In addition, Manila clams were co-infected by the metazoan parasite Cercaria tapetis and Parvatrema duboisi with the prevalence of 0-33% and 0-14%, respectively. Massive hemocyte infiltration and subsequent inflammation were commonly observed from the gills of P. olseni infected clams. Clusters of P. olseni trophozoites and heavy hemocyte infiltration were also observed from the female gonad, suggesting that P. olseni interferes with host gonad maturation. The larval trematode occupied almost the entire host gonad, resulting in gonad castration. In addition, Metacercaria of P. duboisi were observed from the subsurface of the mantle. Ray's fluid thioglycollate medium assay (RFTM) indicated that clams collected in August and September contained approximately 4.0×106 P. olseni cells/g gills. Condition Index (CI) declined gradually from spring to early summer, and the decline in CI was interpreted as a consequence of the heavy parasitism, as the parasites drain the host's net energy to be used in somatic growth and gamete production.

A network pharmacology and molecular docking approach in the exploratory investigation of the biological mechanisms of lagundi (Vitex negundo L.) compounds against COVID-19

  • Coronavirus disease 2019 (COVID-19) is an inflammatory and infectious disease caused by severe acute respiratory syndrome coronavirus 2 virus with a complex pathophysiology. While COVID-19 vaccines and boosters are available, treatment of the disease is primarily supportive and symptomatic. Several research have suggested the potential of herbal medicines as an adjunctive treatment for the disease. A popular herbal medicine approved in the Philippines for the treatment of acute respiratory disease is Vitex negundo L. In fact, the Department of Science and Technology of the Philippines has funded a clinical trial to establish its potential as an adjunctive treatment for COVID-19. Here, we utilized network pharmacology and molecular docking in determining pivotal targets of Vitex negundo compounds against COVID-19. The results showed that significant targets of Vitex negundo compounds in COVID-19 are CSB, SERPINE1, and PLG which code for cathepsin B, plasminogen activator inhibitor-1, and plasminogen, respectively. Molecular docking revealed that α-terpinyl acetate and geranyl acetate have good binding affinity in cathepsin B; 6,7,4-trimethoxyflavanone, 5,6,7,8,3',4',5'-heptamethoxyflavone, artemetin, demethylnobiletin, gardenin A, geranyl acetate in plasminogen; and 7,8,4-trimethoxyflavanone in plasminogen activator inhibitor-1. While the results are promising, these are bound to the limitations of computational methods and further experimentation are needed to completely establish the molecular mechanisms of Vitex negundo against COVID-19.

Sterilization of Freeze Dried Manila Clam (Ruditapes philippinarum) Porridge for Immuno-Compromised Patients

  • This study was conducted to evaluate the combined effect of gamma irradiation and different conditions (vacuum packaging, antioxidant and freezing) on the microbiological and sensory characteristics of freeze dried Manila clam porridge (MCP) for immuno-compromised patient food. MCP can be sterilized at 1 kGy to 10 kGy. The initial counts of total aerobic bacteria and yeast molds in the non-irradiated MCP were $2.4{\pm}0.5$ and $1.2{\pm}0.3{\log}\;CFU\;g^{-1}$, respectively, but gamma irradiation significantly decreased the total aerobic bacteria to below the detection limit ($1{\log}\;CFU\;g^{-1}$) (5 kGy). Moreover, gamma irradiation effectively eliminated yeasts/molds at dose below than 1 kGy. However, gamma irradiation accelerated the increase of lipid oxidation and therefore, decreased the sensory characteristics of MCP as irradiation dose increased. To improve the sensory qualities of gamma irradiated MCP, combination treatment (vacuum packaging, 0.1% vitamin C) were applied. There was no significant difference in the overall acceptance scores between the combined-treatment sample (5.6 points) and the non-irradiated samples (6.0). The results indicate that combination treatment (vacuum packaging, 0.1% vitamin C) may help to maintain the quality of MCP. Therefore, it considered that irradiation of MCP with combined treatment and this is an effective method for the consumption as a special purpose food such as for space travel or immuno-compromised patients.
