Characterization of an Apple Polygalacturonase-Inhibiting Protein (PGIP) That Specifically Inhibits an Endopolygalacturonase (PG) Purified from Apple Fruits Infected with Botryosphaeria dothidea

  • An apple polygalacturonase-inhibiting protein (PGIP), which specifically inhibits endopolygalacturonase (PG, EC 3.2.1.15) from Botryosphaeria dothidea, was purified from Botryosphaeria dothidea-infected apple (Malus domestica cv. Fuji) fruits. The purified apple PGIP had a molecular mass of 40 kDa. The N-terminal amino acid sequence of the purified protein showed high homologies to those of PGIP from pear (100%), tomato (70%), and bean (65%). We also purified polygalacturonase (PG) from B. dothidea. The PG hydrolyzes pectic components of plant cell walls. When the extracted apple pectic cell wall material was treated with purified apple PGIP and B. dothidea PG, the amount of uronic acid released was lower than that treated with B. dothidea PG alone. This result demonstrates that PGIP functions specifically by inhibiting cell wall maceration of B. dothidea PG Furthermore, we characterized the de novo function of the PGIP against PG on the solubilization and depolymerization of polyuronides from cell wall of apple fruits inoculated with B. dothidea. This result demonstrated that the PGIP of plants exhibits one of the direct defense mechanisms against pathogen attack by inhibiting PGs that are released from pathogens for hydrolysis of cell wall components of plants.

Anti-inflammatory effect of Malus domestica cv. Green ball apple peel extract on Raw 264.7 macrophages

  • We examined the anti-inflammatory effect of the peel extract of the newly bred Korean apple (Malus domestica Borkh.) cultivar Green ball. To test its possible use as anti-inflammatory functional material, Raw 264.7 macrophages were treated with pro-inflammatory lipopolysaccharide (LPS) in the presence or absence of Green ball apple peel ethanol extract (GBE). Notably, up to 500 ㎍/mL of GBE did not result in any signs of inhibition on cellular metabolic activity or cytotoxicity in Raw 264.7 macrophages. Supplementation with GBE to LPS-treated Raw 264.7 macrophage significantly suppressed various pro-inflammatory responses in a dose-dependent manner, including i) nitric oxide (NO) production, ii) accumulation of inducible NO synthase and cyclooxygenase-2, iii) phosphorylation of nuclear factor-kappa B (NF-κB) subunit p65, and iv) expression of pro-inflammatory biomarker genes, including tumor necrosis factor alpha, interleukin 1 beta, interleukin 6, monocyte chemoattractant protein-1, and prostaglandin E synthase 2.

Comparison of the Effects of Early and Conventional Defoliation on Fruit Growth, Quality and Skin Color Development in 'Fuji' Apples

  • We compared the quality of 'Fuji' apples (Malus ${\times}$ domestica) from trees whose leaves were not removed (no artificial defoliation; NAD) with apples from trees that underwent early defoliation (ED, treated in mid September and early October) and conventional defoliation (CD, treated in early and mid October). The experiment was conducted in three consecutive years using 15-year-old 'Fuji' apple grafted on Malus prunifolia. Fruits were harvested on November 7, 16 or 12 in 2011, 2012 and 2013, respectively. Compared to NAD treatment, ED and CD treatment reduced the fresh weight by 4.7% and 0.6%, respectively. The soluble solids content of NAD apples ($14.4^{\circ}Brix$) was slightly higher than that of CD ($14.1^{\circ}Brix$) and ED ($14.0^{\circ}Brix$) apples. Soluble sugar content, flesh firmness, water-core index, and titratable acidity were not affected by defoliation treatment regardless of treatment timing. The skin blush index of NAD apples (2.3) was inferior to that of CD (3.3) and ED (3.4)- treated apples. Furthermore, artificial defoliation treatments increased skin redness ($a^*$) and yellowness ($b^*$) and significantly improved the degree of skin blush compared to NAD fruits.

  • 본 연구는 사과나무의 수분이용, 신초 생장 및 작물계수에 미치는 토양수분영역과 잔디 피복의 영향을 조사하였다. 이를 위해 비가림 시설하에 3년생 사과 '후지'/M. 26 (Malus domestica Borkh.)을 재식한 3반복 double pot-lysimeter 실험을 수행하였다: (A) 토양 매트릭 포텐셜 -50 kPa의 점적관수 (IR50), (B) 토양 매트릭 포텐셜 -80 kPa의 점적관수 (IR80), 및 (C) 지하 0.45 m의 정체 자유수면 (WT45), 각 처리에 나지와 잔디 피복의 두 가지 지표상태를 두었다. 평균 월별 수분이용은 IR50 처리에서 IR80 처리보다 컸으며, WT45 처리에서 가장 컸다. 점적관수 처리 사과나무의 월별 작물계수는 시간의 1차 함수로 증가하였으나 ($r^2=0.953^{***}$, 잔디 피복 $r^2=0.862^{***}$, 나지), WT45 처리 사과나무의 작물계수는 변동하였다. 신초생장은 잔디 피복 IR50 처리에서 가장 컸으며, 나지 WT45 처리에서 가장 작았다. 토양의 Kjeldahl 질소는 처리에 관계없이 동일하였으나 8월 10일 측정한 엽중 Kjeldahl 질소는 WT45 처리보다 점적관수 처리에서 높았다.

First Report of an Apple Ring Rot Fungus Botryosphaeria kuwatsukai in Korea

  • Three fungal strains designated as KNUF-23-MG32, KNUF-23-YC8, and KNUF-23-MJ82 were isolated from the abnormal symptomatic apple trees during screening fungal pathogens collected in Jeollanam-do and Gyeongsangbuk-do, Korea. These fungal strains were found to have similar cultural and morphological characteristics close to the genus Botryosphaeria. Morphological characteristics were matched with B. kuwatsukai CBS 135219 but different with B. dothidea KACC 45481 and B. sinensis HMAS 246714T. Pathogenicity tests of strain KNUF-23-MG32 showed that this strain causes rot in Fuji apples. Molecular phylogenetic analysis based on the sequences of the internal transcribed spacer (ITS) regions, translation elongation factor 1-alpha (TEF1α), and partial β-tubulin (TUB2) showed that these strains are B. kuwatsukai. To the best of our knowledge, this is the first report of an apple ring rot fungus Botryosphaeria kuwatsukai in Korea.

  • 사과 겹무늬썩음병균이(Botryosphaeria dothidea) 생성하는 세포벽 분해효소인 endopolygalaturonase를 억제하는 polygalacturonase-inhibiting protein (PGIP)를 사과 과실로부터 분리하였다. 분리되어진 사과 PGIP는 사과 겹무늬썩음병균이 생성하는 PG에 대하여 혼합형의 저해를 나타내었다. PGIP의 반응 최적온도는 $40^{\circ}C$이며 최적 pH는 5.0이었다. 이 효소는 $60^{\circ}C$까지는 비교적 안정하였으나 $70^{\circ}C$에서는 효소의 활성이 완전히 억제되었으며 pH 4.0에서 8.0까지는 안정하였다. PGIP는 $K^+$, $Cu^{2+}$, $Mg^{2+}$, $Ca^{2+}$$Zn^{2+}$ 등의 금속이온과 SDS 그리고 CDTA에 의해 효소의 활성이 저해되었다.

Effects of Prohexadione-Ca, Ethephon, and Water Stress on Growth and Productivity of 'Golden Delicious'/M.9 Apple

  • Prohexadione-calcium (Pro-Ca), ethephon and transient water stress were evaluated in a factorial design, as potential inhibitors of early-season shoot growth of high density orchard management of apple (Malus domestica Borkh.) trees. In the experiment, water stress was imposed to one-half of the 7-year-old 'Golden Delicious'/M.9 apple trees in each of 5 blocks, by stopping irrigation for 3 weeks between 35 and 56 days after full bloom (AFB). Within each whole unit, the following Pro-Ca and ethephon treatments were randomly allocated at $2{\times}2$ factorial: a) 0 or 250 $mg{\cdot}L^{-1}$ a.i. Pro-Ca applied at 28 days AFB and b) 0 or 300 $mg{\cdot}L^{-1}$ a.i. ethephon applied twice (35 and 71 days AFB). All trees were hand thinned to king flowers prior to treatments. Vegetative shoot growth was markedly reduced by Pro-Ca, with its effect being obvious within 14 days after application, while ethephon and water stress treatments were less effective. Pro-Ca had no effect on fruit set and yield but slightly increased fruit size. Ethephon substantially reduced the fruit size and yield but had no effect on fruit set. Water stress reduced fruit set, fruit size and yield. With regard to fruit quality, Pro-Ca did not influence fruit shape, flesh firmness and soluble solids contents (SSC) but slightly reduced titratable acidity. Ethephon had no effect on fruit shape but increased firmness, SSC and acidity, while water stress did not influence these fruit quality attributes. Dry weight of dormant spur buds was reduced by both Pro-Ca and water stress, while increased by ethephon. The larger dormant buds led to the larger spur flowers at the tight cluster stage the following spring. Return flowering was promoted only by ethephon, especially on previous season's shoots. There were no significant interactions between Pro-Ca and ethephon or water stress on most variables observed in this study.

‘바이오 그린’ 기능수 처리가 사과 쓰가루 품종의 무기성분 흡수와 과실품질에 미치는 영향 (Bio-Green' Functional Water Supply Influences Mineral Uptake and Fruit Quality In Tsugaru Apples)

  • 바이오 그린 기능수는 지하수 순수화$\longrightarrow$처리 촉매제 첨가$\longrightarrow$에너지 imprinting$\longrightarrow$여과 과정을 거쳐 제조되는 미약 에너지 발생 신소재로서 10년생 사과 쓰가루/M26 품종의 수관하부에 1995년 4월 20일, 5월 20일 및 6월 20일 ‘바이오 그린’ 기능수를 주당 0, 5, l0$\ell$씩 관주 처리하여 얻어진 결과는 다음과 같다. 사과원 토양의 화학적 특성에 있어서 무처리구의 pH 5.73에 비하여 기능수 처리구는 pH 6.31-6.43이었고, 기능수 처리에 의하여 치환성 Ca 및 Mg 함량이 증가되었다. 한편 P$_2$ $O_{5}$, K 및 B 함량은 차이가 인정되지 않았다. 기능수 처리에 의하여 수확기 사과 과실의 당함량과 과피의 안토시아닌 함량이 증가되었고, Ca 함량이 현저히 증가되었다. 그러나 N, K, Mg는 처리간에 차이가 없었다. 기능수가 처리된 사과의 수체특성에 있어서 뿌리활력과 잎의 광합성 능력이 향상되었다. 과실 저장중 (4$^{\circ}C$) 기능수 처리구의 과실은 고두병 발생이 현저히 감소되었고, 호흡과 에틸렌 발생량이 상대적으로 적었으며, 높은 과실 경도를 나타냄으로써 과실 저장력이 향상되었다.

쓰가루 사과즙을 이용한 두부의 품질 및 항산화 특성 (Quality and Antioxidant Properties of Tofu Coagulated with 'Tsugaru' Apple (Malus domestica Borkh) Juice)

  • 본 연구에서는 '쓰가루' 사과즙을 이용하여 제조한 두부의 품질, 항산화 활성 및 관능 특성을 분석하고자 하였다. 두부제조를 위해 사용한 '쓰가루' 사과즙은 60%의 수율, $10.3^{\circ}Brix$, pH 3.89, 0.51%의 총산을 나타내었고, 총폴리페놀 함량은 2.26 mg GAE/g, ABTS 라디칼 소거능은 $20{\mu}g$ AE/g, DPPH 라디칼 소거능은 $75{\mu}g$ AE/g을 나타내었다. '쓰가루' 사과즙을 첨가하여 두부의 제조가 가능하였으며, 사과즙의 첨가량이 증가할수록 제조된 두부의 수율과 pH는 감소하였으나 총산은 증가하는 경향을 나타내었다. 또한 조직감에서는 경도와 씸힘성이 증가하였으나 응집성은 다소 감소하는 경향을 나타내었으며, 색도에서는 L값이 감소하고 a값과 b값은 증가하여 전반적으로 어두운 갈색을 나타내었다. 이러한 갈색화 반응의 주요 원인은 사과즙의 효소적 갈색화 반응에 기인하므로 비타민 C 등의 환원제, 산미제 및 무기염을 사용하는 방법으로 갈변 방지를 최소화할 수 있을 것으로 생각된다. 그리고 항산화 활성에서는 총폴리페놀 함량과 DPPH 라디칼 소거능이 증가한 반면 ABTS 라디칼 소거능은 일정한 범위를 유지하였다. '쓰가루' 사과즙 첨가량에 따라 제조된 두부의 맛, 색, 풍미, 전체적인 선호도에서 30%(w/v) '쓰가루' 사과즙으로 제조한 두부의 선호도가 가장 높게 나타났다. 따라서 본 연구에서는 '쓰가루' 사과즙으로 제조한 두부의 품질, 항산화 특성 및 관능적 특성을 확인함으로써 '쓰가루' 사과 품종을 이용한 다양한 가공식품의 제조 가능성을 제시하였다고 생각된다.
