CD133, a pentaspan transmembrane glycoprotein, is generally used as a cancer stem cell marker in various human malignancies, but its biological function in cancer cells, especially in glioma cells, is largely unknown. Here, we demonstrated that forced expression of CD133 increases the expression of IL-$1{\beta}$ and its downstream chemokines, namely, CCL3, CXCL3 and CXCL5, in U87MG glioma cells. Although there were no apparent changes in cell growth and sphere formation in vitro and tumor growth in vivo, in vitro trans-well studies and in vivo tumor xenograft assays showed that neutrophil recruitment was markedly increased by the ectopic expression of CD133. In addition, the clinical relevance between CD133 expression and IL-$1{\beta}$ gene signature was established in patients with malignant gliomas. Thus, these results imply that glioma cells expressing CD133 are capable of modulating tumor microenvironment through the IL-$1{\beta}$ signaling pathway.

Gliomas are the most common malignant brain tumor and cause the most deaths. Manual brain tumor segmentation is expensive, time-consuming, error-prone, and dependent on the radiologist's expertise and experience. Manual brain tumor segmentation outcomes by different radiologists for the same patient may differ. Thus, more robust, and dependable methods are needed. Medical imaging researchers produced numerous semi-automatic and fully automatic brain tumor segmentation algorithms using ML pipelines and accurate (handcrafted feature-based, etc.) or data-driven strategies. Current methods use CNN or handmade features such symmetry analysis, alignment-based features analysis, or textural qualities. CNN approaches provide unsupervised features, while manual features model domain knowledge. Cascaded algorithms may outperform feature-based or data-driven like CNN methods. A revolutionary cascaded strategy is presented that intelligently supplies CNN with past information from handmade feature-based ML algorithms. Each patient receives manual ground truth and four MRI modalities (T1, T1c, T2, and FLAIR). Handcrafted characteristics and deep learning are used to segment brain tumors in a Global Convolutional Neural Network (GCNN). The proposed GCNN architecture with two parallel CNNs, CSPathways CNN (CSPCNN) and MRI Pathways CNN (MRIPCNN), segmented BraTS brain tumors with high accuracy. The proposed model achieved a Dice score of 87% higher than the state of the art. This research could improve brain tumor segmentation, helping clinicians diagnose and treat patients.
목 적: WHO 등급 3/4의 신경아교종 환자에서 수술 후 방사선 치료의 결과와 예후인자를 분석하였다. 대상 및 방법: 1988년부터 2007년까지 수술 후 고악성도 신경아교종으로 확진된 환자들 중 방사선 치료를 시행한 99명을 대상으로 하였다. 나이는 18~77세(중앙값, 51세), 남, 여가 각각 55, 44명이었다. 수술은 완전절제, 아전절제(50% 이상 절제), 부분절제(50% 미만 절제), 조직검사만 시행한 예가 각각 16, 38, 22, 23명이었고, WHO 등급 3, 4, 기타(unspecified high grade glioma)가 각각 32, 63, 4명이었다. 방사선 치료는 1.8 또는 2 Gy 통상 분할치료 혹은 1.2 또는 1.5 Gy 하루 2회 치료 방법으로 하였고 중앙 생물학적 등가선량은 $72.2\;Gy_{10}$ ($18.6{\sim}83.3\;Gy_{10}$)였다. 동시항암화학방사선치료나 연속화학방사선요법을 적용한 환자가 49명이었다. 환자들의 생존율 및 실패양상, 예후인자, 부작용에 대해 후향적으로 분석하였다. 결 과: 대상 환자의 중앙 추적 관찰기간은 11개월이었다. 국소치료 실패한 환자가 54명(54.5%)이었고, 이중 12명(22.2%)은 방사선치료부위 밖의 병변 진행을 동반하였다. 전체 생존율은 1년 56.6%, 2년 29.3%였고, 중앙생존기간은 13개월이었다. 무진행생존율은 1년, 2년이 각각 31.3%, 18.2%였으며, 중앙생존기간은 7개월이었다. 전체생존율에 영향을 미치는 인자로는 나이(p=0.0001), 수술 절제 정도(완전절제를 기준으로, 아전절제 p=0.023, 부분절제 p=0.009, 조직생검 p=0.002), 수술 후 영상검사에서 조영증가 여부(p=0.049)였다. 무진행생존율에 영향을 미치는 인자는 나이(p=0.036)와 수술 후 영상검사에서 조영증가 여부(p=0.006)였다. 방사선치료중과 후에 3등급 이상의 부작용이 각각 3명과 1명에서 발생하였다. 결 론: 나이, 수술절제 정도, 수술 후 영상검사에서 조영증가 여부가 생존율에 유의한 요인으로 분석되었다. 국소치료실패가 대부분을 차지하여, 국소 제어율 향상을 위한 추가적인 연구가 필요할 것이다.
목적: 신경고종의 자기공명영상 소견과 표피성장인자수용체의 발현 사이의 상관관계를 알아보고자 본 실험을 시행하였다. 대상 및 방법: 수술적 제거 혹은 생검으로 확진된 41예의 신경교종(저등급 성상세포종 8예, 역형성 성상세포종 12예, 다형성 교아세포종 21예) 환자에서 시행한 자기공명영상을 종양의 경계, 괴사, 균질도, 출혈, 조영증강, 그리고 종괴주위의 부종에 대하여 분석하여 각 소장인자수용체의 염색을 시행한 후 그 염색 분포 및 강도에 대한 등급값을 정하였다. 각 환자에서 얻어진 종양 조직을 면역조직화학법으로 표피성 장인자수용체의 염색을 시행한 후 그 염색 분포 및 강도에 대해 등급값을 정했다. 각 자기공명소견과 표피성장인자수용체 발현의 상관관계를 통계적으로 알아보았다. 결과: 자기공명영상에서 종괴주위 부종만이 표피성장인자수용체의 염색 분포(r=0.71, p=0.00) 및 염색 강도(r=0.69, p=0.00)와 유의한 상관관계를 보이는 소견이었다. 나머지 소견은 표피성장인자수용체의 발현과 통게적으로 유의한 상관관계를 보이지 않았다. 결론: 자기공명영상은 신경교종의 진단에 있어 유용한 방법이며, 신경교종에서 종괴주위 부종은 그 조직병리학적 악성도의 예측 뿐만 아니라, 표피성장인자수용체의 발현을 예측하는 데에도 도움이 되는 소견이다.
Objectives : It has been known that vascular endothelial growth factor(VEGF), as an endothelial cell-specific mitogen, induces angiogenesis, and possesses vascular permeability and procoagulant properties. Peritumoral brain edema(PTBE) is a common accompaniment of malignant gliomas. It results from microvascular extravasation of plasma and proteins through the interendothelial spaces. The correlation between pathological grading, PTBE, neovascularization, and the expression of VEGF were analyzed in 31 patients with astrocytic tumors. Methods : Astrocytic tumor samples(8 astrocytomas, 14 anaplastic astrocytomas, and 9 glioblastomas) from 31 patients( 21 males and 10 females : average age $37{\pm}24$ years) who underwent surgery were examined retrospectively for the expression of VEGF and CD31(microvasculature) immunohistochemically. The extent of PTBE was examined by using preoperative CT or MRI as an edema index(EI). In addition to VEGF and CD31, several causative factors including tumor size, histologic type were compared with EI. Results : Only one of 8 astrocytomas, and majority of high grade(21 of 23 anaplastic astrocytomas and glioblastomas) tumors demonstrated PTBE(p<0.05). The majority of high grade tumors showed higher expression of VEGF (p<0.01). High grade tumors showed even higher CD31 expression(p<0.05), however, there was no close correlation between expression of VEGF and CD31. The EI was increased significantly, just as VEGF(p<0.01), but CD31 expression was not correlated with high EI. Conclusion : These data suggest that VEGF expression is closely correlated with PTBE and histological grading in astrocytic tumors. Microvasculature(CD31) in tumors is highly correlated with histological grading, however, shows no correlation with the expression of VEGF and PTBE.
Objective : To determine the presentation, incidence, and risk factors of seizures in patients treated for brain tumors. Methods : One hundred patients who consecutively underwent a craniotomy for the treatment of supratentorial brain tumors were assessed. The pathologies of the patients enrolled in the study included glioma [n=56], meningioma [n=31], metastatic brain tumor [n=7], primary central nervous system lymphoma [n=4], and central neurocytoma [n=2]. Anti-epileptic drugs [AEDs] were administered to all patients for up to six months after the surgery. Pre-defined variables for outcome analysis included tumor grade and location, extent of tumor resection, number of seizures, age at tumor diagnosis, adjuvant therapy, medication and radiological abnormalities. Results : Thirty patients [30%] presented at least a single episode of seizure at the time of admission. Five of these patients [16.7%] developed the seizure during the follow-up period. Newly developed seizure was noticed in six out of seventy patients [8.6%] without prior seizure. Histopathology was malignant gliomas in 10 and supratentorial meningioma in one. Early seizure developed only in two patients. Conclusion : Compared with patients without seizure, patients with seizure at the time of admission showed younger age [p=0.003], a higher portion of low-grade glioma [p=0.001], tumor location in the frontal and temporal lobes [p=0.003] and cortical involvement [p=0.017]. Our study suggestes that tumor progression is considered a significant risk factor for seizure development in glioma patients.

Objective : The choice of tumor antigen for dendritic cell[DC]-loading has still been an unresolved problem in the DC-based vaccine strategies against malignant gliomas that has not been found well-characterized tumor specific antigens. In this study, we compare tumor-specific T cell response induced by glioma apoptotic body[GAB]-pulsed DCs to response induced by glioma cell lysate-pulsed ones quantitatively. Methods : DCs generated in the presence of granulocyte macrophage-colony stimulating factor and interleukin[IL]-4 from peripheral blood mononuclear cells[PBMCs] of HLA-A2 positive healthy donors were cultured. Each GABs and glioma cell lysate generated from HLA-A2 positive T98G glioblastoma cells were co-incubated with DCs. $CD8^+$ T lymphocytes isolated from PBMCs of same donors were cultured in media containing IL-2 and either stimulated by GAB- or lysate-pulsed DCs three times at a weekly interval. The interferon[IFN]-${\gamma}$ concentrations of each cell culture supernate were measured by enzyme immunoassay technique. Cytolytic activity of the generated cytotoxic $CD8^+$ T cells either stimulated with GAB- or lysate-pulsed DCs was determined by a standard 4-h $^{51}Cr$-release assay. Results : IFN-${\gamma}$ production and cytolytic activity of effector T cells stimulated by GAB-pulsed DCs were significantly higher than those of T cells stimulated by lysate-pulsed ones. Conclusion : These results indicate the choice of antigen is a critical determinant in the induction of antitumor immunity against malignant glioma. Antigen preparations from GABs represent a promising alternative to glioma cell lysate in DC-based glioma vaccine strategies.
UHRF2 is a member of the ubiquitin plant homeo domain RING finger family, which has been proven to be frequently up-regulated in colorectal cancer cells and play a role as an oncogene in breast cancer cells. However, the role of UHRF2 in glioma cells remains unclear. In this study, we performed real-time quantitative PCR on 32 pathologically confirmed glioma samples (grade I, 4 cases; grade II, 11 cases; grade III, 10 cases; and grade IV, 7 cases; according to the 2007 WHO classification system) and four glioma cell lines (A172, U251, U373, and U87). The expression of UHRF2 mRNA was significantly lower in the grade III and grade IV groups compared with the noncancerous brain tissue group, whereas its expression was high in A172, U251, and U373 glioma cell lines. An in vitro assay was performed to investigate the functions of UHRF2. Using a lentivirus-based RNA interference (RNAi) approach, we down-regulated UHRF2 expression in the U251 glioma cell line. This down-regulation led to the inhibition of cell proliferation, an increase in cell apoptosis, and a change of cell cycle distribution, in which S stage cells decreased and G2/M stage cells increased. Our results suggest that UHRF2 may be closely related to tumorigenesis and the development of gliomas.
혈관분포도(vascularity) 및 세포조밀도(cellularity)와 같은 종양의 생물학적 특성을 고려한 임상표적체적을 결정하기 위하여, 국부혈류용적영상(regional cerebral blood volume map, rCBV map)과 겉보기확산계수영상(apparent diffusion coefficient map, ADC map)의 종양 체적을 해부학적 영상 위에 맵핑 할 수 있는 소프트웨어를 개발하였다. 개발한 프로그램은 해부학적 영상 및 기능 영상 간 mutual information, affine transform, non-rigid registration을 이용한 영상 정합 기능을 제공한다. 영상 정합 후 기준 영상과 정합된 영상에서 획득한 각 segmented bone의 겹치는 체적 비율 및 contour 간 평균 거리를 이용하여 정합도 평가도 가능하다. 잔여 종양이 있는 악성신경아교종 환자의 영상을 이용하여 소프트웨어의 기능을 평가하였을 때, bone segmentation과 contour 간 평균 거리 차이를 이용한 정합도는 각각 약 74%와 2.3 mm였으며, 수동정합을 이용하여 2~5% 정도의 정합도를 향상 시킬 수 있었다. 종양의 생물학적 특성을 치료 계획에 반영할 수 있도록, color map을 이용하여 rCBV map을 분석하였으며, ADC map에서 설정한 관심 영역의 평균 확산 계수와 표준 편차 등을 계산하여 종양의 예후 인자 및 악성도를 평가하였다. 두 기능 영상이 공통적으로 나타내는 종양 체적에서 얻은 생물학적 인자를 평면 위에 맵핑하여 종양의 특성을 쉽게 파악할 수 있는 multi-functional parametric map을 구성하였다. 또한 각기능 인자에 대응되는 악성 종양의 임계값을 적용하여 주변 종양 세포에 비하여 혈관 분포도는 높으면서 확산 계수는 낮아 악성 종양 세포일 확률이 높은 영역을 구분할 수 있었다. 각 기능 영상 위에서 설정한 생물학적 종양 체적 및 악성도가 높은 국소 체적은 해부학적 영상 위에 표시하여 dicom 파일로 출력할 수 있었다. 개발한 소프트웨어는 기능적 다중영상을 이용하여 생물학적 종양 체적을 해부학적 영상 위에 맵핑하는데 적용할 수 있으며, 해부학적 영상에서 파악하기 어려운 종양의 특성 변화들을 치료 계획에 활용할 수 있다. 나아가 개발한 소프트웨어를 이용하여, 한 종류의 영상을 참고하여 종양 체적을 결정했을 때 발생할 수 있는 오류를 줄이고, 치료 전이나 치료 과정에서 나타나는 종양의 조직학적, 생리학적 특성을 치료 계획에 접목하는데 활용할 수 있다.
목적: all-trans retinoic acid (ATRA)는 뇌종양 세포의 증식억제효과가 있으며, ATRA와 방사선의 병용은 악성 뇌종양의 치료 효과를 증진시키는 방법이 될 수 있다. 그러나 ATRA에 의해 항산화효소가 증가되며 이로 인해 방사선에 의해 생성된 reactive oxygen species (ROS)가 제거된다면 방사선의 효과는 낮아질 수 있다. 본 연구에서는 ATRA에 의해 유도되는 카탈라제(catalase)에 의한 방사선감수성의 변화를 보고자하였다. 대상 및 방법: 백서 교종세포(36B10)을 대상으로 ATRA 및 ATRA의 화학적 억제제인 3-amino-1, 2, 4-triazole (ATZ) 와 병용하여 카탈라제 활성도, 방사선감수성 및 ROS의 변화를 측정하였다. 카탈라제 활성도는 $H_2O_2$의 소멸을 자외선 분광광도계로 측정하는 방법을 이용해 정량하였으며, 방사선감수성은 단일집락군형성능력으로, ROS 는 2, 7-dichlorofluorescein diacetate 를 분광광도계로 측정하였다. 결과: 카탈라제 활성도는 ATRA의 농도(10, 25, $50{\mu}M$)에 따라 증가하였다. ATRA ($10{\mu}M$)와 방사선(4 Gy)의 병용에 의해 생존분획은 상승적(supra-auditive)으로 감소하였으며, 이 감소된 생존분획은 ATZ 동시 투여에 의해 증가하였다. ATRA $10{\mu}M$ 또는 $25{\mu}M$을 48시간 처리 후 ROS는 대조군에 비해 각각 1.5배, 2배 증가하였고, 4 Gy와 ATRA의 병용군에서는 2.5배 증가하였다. ATRA와 방사선의 병용에 의해 증가된 ROS는 ATZ에 의해 감소되었다. 결론: ATRA에 의해 유도되는 카탈라제는 방사선감수성을 감소시키지 않으며, 오히려 ROS의 증가에 의해 방사선감수성을 상승시켰다. 따라서 ATRA와 방사선의 병용은 뇌종양의 치료에 유용한 방법이 될 수 있을 것으로 보인다.

① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
