Most studies on transgenic bioreactors have focused on expression levels of interest genes. In this study we examined whether transgenic bioreactors would inherit expression level of the Oansgene to long-term generations independently of transgene sources. We employed three transgenic mice, which were separately reported, carrying different transgenes and copy numbers, 27 kb of hLF and 22 kb of hIL-10 genomic sequences, and 1.3 kb of hTPO cDNA, respectively. Three females of the transgenic lineages crossbred with a wild-type male up to 20 generations to test transgenic frequencies of their progenies and to determine expression levels of the transgenes. Ultimately, transmission rates of kLF, hIL-10, and hTPO were $64.3{\pm}7.0$, $59.3{\pm}9.8$, and $56.1{\pm}9.7$, respectively, appeared following Mendelian pattern of inheritance. Notably, we found that levels of expressions of hLF, hIL-10, and hTPO in milk were sustained to high numbers of generations. No transgene silencing of expression was observed in every generations of all transgenic mice. In conclusion, we suggest that once established animal bioreactors could consistently transmit the transgene to continual generations, without loss of expressional activity, independently of transgene sources.
We investigated the effects of maternal and paternal parentage, and ovule position in a pod on seed development, weight, and progeny performance, using an annual crop commonly called cowpea (Vigna unguiculata L.). The experiment was conducted in a greenhouse. Nine maternal parents were randomly selected, and 3 individuals were chosen as paternal parents based on the male fitness components. Five types of hand-pollination were attempted on each flower of the maternal plants to evaluate the influence of paternal identify. Independent variables such as parentage and ovule position affected the pattern of seed development. Ovules in the middle position of the pod exhibited a much higher probability of seed maturation than those at the stylar and basal positions, confirming the position effect. These independent variables also appeared to exert a significant effect on seed weight, although it was not possible to evaluate the relative importance of those variables due to a significant interaction between parents. Mixed pollination tended to yield heavier seeds than other paternal parents. Ovules in the middle and basal end positions produced heavier seeds than those at the stylar end regardless of maternal and paternal parentage. Seed weight was independent of all measurements of progeny performance. At week 2 after germination significant paternal effects were found to be mediated through the ovule position in a pod. In particular, the progeny derived from self-pollination performed worse than the progeny from other paternal parentage. The progeny performance at week 4 after germination was affected by maternal parents independently of seed weight. These results show that paternal and maternal effects were quite consistent from the seed development to early growth of the progeny. Consequently, maternal and paternal effects on seed development and weight, and progeny performance may reflect, at least to some extent, genetic contribution of both parents. If so, it is possible for sexual selection on maternal and paternal function to generate the evolution of reproductive traits related to seed production.
Biometric evaluation and reproductive biology of Oreochromis niloticus were studied from Geray Reservoir. Between November 2021 and August 2022, 316 O. niloticus specimens were collected from this reservoir. The observed male to female sex ratio was 1:1.14, which was statistically insignificant compared with the hypothetical 1:1 sex ratio (X2 = 1.27, p > 0.05). The length-weight relationship was curvilinear with allometric growth (b < 3) and statistically significant from the expected isometric growth pattern (b = 3). The regression equation for females was TW = 0.0264TL2.72 (r2 = 0.90, n = 168), males (TW = 0.0183TL2.79, r2 = 0.93, n = 148), and combined sexes (TW = 0.0229TL2.76, r2 = 0.91, n = 316). Gonadosomatic index (GSI) ranged from 0.8 to 1.4 (males) and 1.8 to 4.9 (females). The breeding season lasted from March to June, with a GSI peak in April. Fecundity ranged from 279 to 1,528 eggs./fish, with a mean of 1,025 eggs./fish. The correlation between absolute fecundity and length was more significant (F = 0.76TL2.8, R2 = 0.94, p < 0.05) than with weight (F = 17.683TW0.71, R2 = 0.92, p < 0.05). This study provides basic information on the biology of O. niloticus in the Geray Reservoir for future fishery management tools for the conservation and utilization of this valuable freshwater fish.
Monthly changes in the gonadosomatic index (GSI) and hepatosomatic index (HSI) of wild river puffer Takifugu obscurus, and water quality environment in spawning area during breeding season were investigated from March 1995 to February 1996. Monthly changes in GSI and HSI of T. obscurus, that was cultured in low salinity, were calculated. The external morphology of the gonads, germ cell differentiation during gametogenesis and the reproductive cycle with the gonad developmental phases were investigated by histological analysis. The optimum water quality environment in Ganggyung, Choongcheongnam-do, where is spawning ground of wild T. obscurus, was $15-20^{\circ}C$ (water temperature) and 0 psu (salinity). Monthly changes in the GSI in females and males reached a maximum in May, and then rapidly decreased. Therefore, it is assumed that in the natural condition the spawning period of wild T. obscurus is May to June. In females and males, it showed a negative correlationship between the GSI and HSI. The external morphology of the gonads in female and male T. obscurus, that was cultured in low salinity, is composed of a pair of saccular structure. Based on monthly changes in the GSI, it is assumed that in female T. obscurus, that was cultured in low salinity, spawn from March through May. Therefore, it showed a negative correlationship between changes in the GSI and HSI. On the whole, in females and males, it showed a similar pattern between wild and cultured T. obscurus. The reproductive cycle with the gonad developmental phases can be classified into successive five stages in females: the early growing stage, late growing stage, mature stage, ripe and spent stage, and recovery and resting stage. In males, that can be divided into successive four stages: the growing stage, mature stage, ripe and spent stage, and recovery and resting stage. In case of wild T. obscurus, the spawning period has once a year, however, those cultured in the high water temperature ($20-27^{\circ}C$) - low salinity (under 3.3 psu) condition have reproductive characteristics having possibilities of discharge of eggs and sperms year-round as a multiple spawner.
Gonadal development, gametogenesis, reproductive cycle, gonad index, flesh weight rate, and first sexual maturity of the turban shell, Lunella coronata coreensis were investigated by histological observation. The materials used were collected monthly from the rocky intertidal zone of Daehang-ri, Buan-gun, Jeollabuk-do, on the west coast of Korea, from July 1998 to June 1999. Sex of L coronata coreensis was separate. The gonad was widely located in the spirals of the visceral mass buried in the digestive gland. The ovary and testis were composed of a number of oogenic follicles and speymatogenic follicles, respectively. Monthly variations in the gonad index increased from March ($23.86{\pm}3.73$) when the water temperature increased and reached the maximun in July ($49.76{\pm}6.47$). And then, the gonad index sharply decreased in September ($15.58{\pm}2.33$). The flesh weight rate ranged from $25.2{\%}$ to $32.3{\%}$, and its variation showed a similar pattern to the gonad index. Individuals $<5.9 mm$ in shell height could not take part in reproduction in both sexes. Percentages of first sexual maturity of female and male specimens ranging from $7.0{\~}7.9 mm$ in shell heights were $84.6{\%}\;and\;91.7{\%}$, respectively, and $100{\%}$ in those over 8.0 mm in shell height in both sexes took part in reproduction. By studying the monthly changes of the morphological features and sizes of germ cells during gametogenesis in the gonad, the reproductive cycle of this species could be devided into five successive stages: early active (December to April), late active (January to July), ripe (May to August), spawning (July to September), and recovery (September to March). The spawning period of this species was once a year between July and September, and the main spawning occurred in July when the seawater temperature reached above $24.8^{\circ}C$. The fully ripe eggs were $150{\~}160\;{\mu}m$ in diameter.
Procymidone is a fungicide with anti-androgenic properties widely used to protect fruits from fungal infection, which induces an excessive reactive oxygen species production in male reproductive organs. In this study, to clarify whether procymidone affect the cellular antioxidant system of prostate at onset of puberty, gene expression patterns of the representative antioxidant enzymes such as cytoplasmic glutathione peroxidase (GPx1), phospholipid hydroperoxide GPx (PHGPx), selenoprotein P (SePP), cytoplasmic copper/zinc superoxide dismutase (SOD1), and manganese SOD (SOD2) were investigated in the rat ventral prostates exposed to procymidone using real-time RT-PCR analyses. Seven-week-old Sprague-Dawley rats castrated at 6 weeks old were treated with procymidone (25, 50, or 100 mg/kg per day) orally for 7 consecutive days after testosterone propionate (0.4 mg/kg per day) administration by subcutaneous injection. As compared to normal control animals, GPx1 mRNA expression in prostates significantly increased by the administration with TP and/or procymidone. However, PHGPx and SOD1 mRNA levels significanatly decreased by over 25 mg/kg of procymidone treatment and SePP and SOD2 mRNA levels was significanatly reduced by over 50 mg/kg of procymidone treatment. These findings indicate that procymidone may affect the antioxidant system of prostatic cells in up-regulation mode of GPx1, but in down-regulation modes of PHGPx, SePP, SOD1, and SOD2, suggesting that procymidone may affect differently the cellular antioxidant system of prostate according to the exposure doses.
This study was carried out to investigate the morphological changes of gonadotropes in pituitary gland and spermatogenic cells in testis, obtained from 150 of 3-year-old immature and mature male rainbow trout (Oncorhynchus mykiss) during the reproductive cycles from March to February in the following year. In the maturation cycle of the pituitary gonadotropes of cultured rainbow trout, three periods can be distinguished i.e. a period of resting(March-August), a period of full spermatogenesis (September-November), and a period of breeding (December-February). The ultrastructures of the gonadotropes largely parallel the cyclical changes in the tests. The seminiferous tubules contain all spermatogenetic stages and sperm cells in a period of early maturation. At first, the size of the nucleus and cytoplasm decrease gradually at every stages from spermatogonia to spermatids. In the secondary spermatocytes, the small mitochondria are located over the outer cytoplam. In spermatids, the cytoplasmic masses move toward the posterior part of the nucleus. Some of the information concerning sexual function in the male diabetes has been focused upon the problems of endocrine or semen parameters. However, the characteristics of acrosome reaction and spermatozoa concentration at the epididymis and vas deferens have scarcely been studied, and the causes of the infertility has not been critically identified. So, we designed to inspect the spermatozoa concentration and the characteristics of acrosome reaction at epididymis and vas deferens of diabetic Wistar rat induced by streptozotocin (STZ, 70 mg/kg, ip). Experimental animal was sacrificed at 3 days and 14 days after the STZ injection. In the diabetes-induced rat, the levels of insulin and glucose had a pattern of inverse proportion. The spermatozoa concentrations in caput and corpus epididymis were significantly decreased in all diabetic condition. In cauda epididymis, however, there was significant decrease in sperm concentration at 14 days onward. In diabetic rat, the spontaneous reaction rate of spermatozoa of cauda and vas deferens were significantly higher than the control group. The ARIC (acrosome reaction to ionophore challenge) value of caudal sperm was 28.7 at control, 22.1 at 3 days, and 8.3 at 14 days. In the present study the spermatozoa concentration was decreased and the spontaneous reaction rate was increased by diabetes. In ARIC-test, it is revealed that the fertility of spermatozoa of 14 days group was lower than control or 3 days group. Diabetes mellitus may be provoke the decreased fertilization rate and subsequent infertility and subsequent infertility.
