랫드에서의 UTI의 약물동태학 및 조직 분포 (Pharmacokinetics and Tissue Distribution of UTI in the Rat)

  • The purpose of this study was to determine pharmacokinetic parameters and tissue distribution patters of urinary trypsin inhibitor(UTI) in Sprague-Dawley rats. $Na^{125}$I was conjugated to UTI to make $^{125}I-UTI$ and the concentrations were determined by $\gamma$-counter. With the aid of nonlinear least-square regression analysis for i.v bolus injection of 1,000 unit UTI including $^{125}I-UTI$, the temporal concentration curves were best fitted by 2-compartment open model. The distribution phase half-life was 0.39$\pm$0.02 hours whereas the elimination half-life was 12.99$\pm$1.05 hours in male rats. The volume of distribution and total body clearance in male rats were 0.28$\pm$0.01 1/kg and 83.16$\pm$1.15 ml/kg/h, respectively. We could not find any difference of pharmacokinetic parameters of UTI between male and female rats. UTI were distributed widely in rat organs. In both male and female rats, the kidney was the highest distributed organ. Amount of UTI in 24 hour cumulative urine in male rats was 36.22$\pm$8.74% and that in 48 hours was 43.32$\pm$10.55%. Excretion via feces was very scanty, with the 24 hours cumulative amount being only 2.76$\pm$0.97%. This data suggest the main excretion route of UTI is urine.

  • 본 연구는 골다공증유발 rat에 녹용추출물의 투여가 예방 및 치료효과에 미치는 영향을 구명하고자, 양측 난소를 적출한 후 골다공증이 유발된 rat에 암, 수녹용 추출물 525, 1,250 mg/kg을 투여했을 때 체중, 대퇴골의 중량, 장기중량, 골회분량의 측정 및 조직학적, 전자현미경적 검사 등을 실시하였다. 1. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 체중은 정상대조군에 비하여 유의한 증가를 나타냈다 (p<0.05). 2. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 우측, 좌측의 femur 중량은 정상대조군에 비하여 감소하였다. 3. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 골회분량은 정상대조군에 비하여 약간 감소된 치를 나타냈으나 유의성은 인정되지 않았다. 4. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 간과 비장 및 신장의 각각 정상대조군에 비하여 감소된 치를 나타냈다. 5. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 조직학적 및 전자현미경적 소견은 정상대조군에서는 소주골간의 연결이 비교적 잘 유지되어 있고 골소실을 발견할 수 없었으나 난소적출군은 정상군에 비하여 소주골이 가늘어졌거나 소주골간 연결부분의 대다수가 끊어져 골양이 많이 소실되었음이 관찰되었으며, 녹용처리군에서는 소주골의 형태가 굵고 연결부분이 잘 유지되어 정상군과 유사한 소견이 관찰되었다. 위의 결과들에서 암, 수녹용 추출액은 난소제거후의 골소설을 억제하므로 골다공증의 예방과 치료에 효과가 있는 것으로 판단된다.

  • Chronic adminstraction of morphine to adult male rats has long been known to lower hepatic cytochrome p-450 content and its dependent mixed-function oxidase activity. Following the treatment of adult male rats with morphine, pethidine pentazocine and codeine and also by concomitant adminstration of naloxone activities of microsomal electron transfer in the adult male rats were examined. In present study, the acute treatment of mature male rats with a dose of narcotic drugs higher than that used chronically also reduces their hepatic cytochrome p-450.

마늘첨가식이(添加食餌)가 흰쥐의 혈청(血淸) 콜레스테롤, 글루코오스의 함량(含量) 및 혈액응고시간(血液凝固時間)에 미치는 영향(影響) (Effect of Garlic Diets on the Changes of Serum Cholesterol Glucose Level and Coagulation Time in Rats)

  • Effects of garlic on hypocholesterolemia, anticoagulation and hypoglycemia were studied in the present experiments using male rats. The results were summarized as follows. 1. The supplementation of $2{\sim}4%$ garlic to 2% cholesterol diets did not affect food intake and weight gain in male rats. 2. Rat's groups fed the diets supplemented with $2{\sim}3%$ garlic (C.D.E.F) to 2% cholesterol diet (B) decreased serum total cholesterol levels by $16{\sim}32%$, triglyceride levels by $18.6{\sim}26.8%$ and ${\beta}/{\alpha}-lipoportein$ rations by $42{\sim}58%$, but increased HDL-cholesterol levels by $29{\sim}65%$ as compared to B group, and so the author assumes that garlic supplementation exerts hypocholesterolemic effect on cholesterol - fed rats because of the increase of HDL and HDL - cholesterol levels. 3. Rat's groups fed the diets supplemented with $2{\sim}4%$ garlic (C.D.E.F) to 2% cholesterol plus 0.25% bile salt diet (B) increased whole blood coagulation times, prothrombin times and fibrinolytic activities, but decreased plasma fibrinogen levels as compared to B group, and so the author assumes that garlic supplementation exerts anticoagulative effect because of the inhibition of fibrinogen synthesis in male rat's liver. 4. Rat's groups fed the diets supplemented with $2{\sim}4%$ garlic (B.C.D.E) to standard diet (A) decreased serum glucose levels by $1{\sim}24%$, but increased serum insulin concentrations by $0.5{\sim}3.0$ times as compared to A group, and so the author assumes that garlic supplementation exerts hypoglycemic effect because of the increase of serum insulin levels by stimulation pancreatic secretion of insulin from ${\beta}-cells$ in the islets of Langerhans.

향사양위탕(香砂養胃湯)이 Propylthiouracil로 유발된 Rat 갑상샘기능저하성 웅성 생식기 손상에 미치는 영향 (Favorable Effects of Hyangsayangyi-tang on the Hypothyroidism related Rat Male Reproductive Organ Damages induced by Propylthiouracil)

  • Objectives : Hyangsayangyi-tang (香砂養胃湯) is a polyherbal formula, has been used as one of the representative So-eumin (少陰人) prescriptions in Sasang-medicine (四象醫學), a famous Korean medicinal theory. The aqueous extract of Hyangsayangyi-tang (HSYYT) was evaluated for its possible ameliorative effect in the regulation of hypothyroidism related reproductive organ damages in propylthiouracil (PTU)-induced rat model. Methods : HSYYT aqueous extracts were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 500, 250 and 125 mg/kg (bodyweight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28days. Results : PTU induced hypothyroidism and related male reproductive organ (testis, epididymis and prostate) damages were favorably and dose-dependently inhibited by treatment of HSYYT 500 and 250 mg/kg, and they also effectively regulated the PTU-induced abnormal antioxidant defense system changes in the testis. No effective or any harmful changes on the PTU induced hypothyroidism and related male reproductive organ damages were observed in HSYYT 125 mg/kg treated rats as compared with PTU control in this experiment. Conclusions : The results obtained in this study suggest that oral administration of 500 and 250 mg/kg of HSYYT showed favorable effects on the hypothyroidism and related reproductive organ damages through augmentation of antioxidant defense system in the testis, and it is considered that HSYYT may be help to ameliorate the hypothyroidism and related organ damages in clinics.

Evaluation of immunocontraceptive vaccine composed of gonadotropin-releasing hormone conjugated with granulocyte-macrophage colony-stimulating factor in male rats

  • Surgical castration performed to reduce male-associated problems results in pain and microbial infections in male animals. Therefore, immunocontraception, which is mediated by the animal's own antibodies against reproductive hormones, has been recommended as an alternative to surgical castration when considering the animal's welfare. In this study, a new immunocontraceptive vaccine composed of six tandem copies of gonadotropin-releasing hormone (GnRH) fused to rat granulocyte-macrophage colony-stimulating factor (GM-CSF) was developed, and its efficacy was evaluated in male rats. Three different doses (10, 50, and $100{\mu}g$) of recombinant GM-CSF-GnRH protein were injected three times at intervals of two weeks into male rats. The rats vaccinated with three doses of GM-CSF-GnRH produced a significantly higher level of antibodies against GnRH than that in the negative control rats. Severe atrophy of gonads was observed in rats vaccinated with three doses of GM-CSF-GnRH but not in the negative control rats. The results reveal that the new GnRH vaccine conjugated with rat GM-CSF induces efficient immunocontraception in male rats. This formulation of the immunocontraceptive vaccine would be applicable to both domestic and pet male animals.

Spontaneous Peripheral Ameloblastic Odontoma in a Male Sprague-Dawley Rat

  • Peripheral ameloblastic odontoma is a rare variant of odontogenic tumor occurring in the extraosseous region. The present report describes a spontaneous tumor in male Sprague-Dawley (SD) rats. The clinically confirmed nodule in the right mandibular region was first observed when the rat was 42 weeks and remained until the terminal sacrifice date when the animal was 48 weeks of age. At necropsy, a well demarcated nodule, approximately $2.5{\times}2.0{\times}2.0cm$, protruded from the ventral area of the right mandible. The nodule was not attached to mandibular bone and was not continuous with the normal teeth. Histopathologically, the tumor was characterized by the simultaneous occurrence of an ameloblastomatous component and composite odontoma-like elements within the same tumor. The epithelial portion formed islands or cords resembling the follicle or plexiform pattern typical of ameloblastoma and was surrounded by mesenchymal tissue. Formation of eosinophilic and basophilic hard tissue matrix (dentin and enamel) resembling odontoma was observed in the center of the tumor. Mitotic figures were rare, and areas of cystic degeneration were present. Immunohistochemically, the epithelial component was positive for cytokeratin AE1/AE3 (CK AE1/AE3), and the mesenchymal component and odontoblast-like cells were positive for vimentin, in the same manner as in normal teeth. On the basis of these findings, the tumor was diagnosed as a peripheral ameloblastic odontoma in an extraosseous mandibular region in a SD rat. In the present study, we report the uncommon spontaneous peripheral ameloblastic odontoma in the SD rat. We also discuss here the morphological characteristics, origin, histochemical, and immunohistochemical features for the diagnosis of this tumor.

피부조직 콜라겐의 DSC 특성 (Differential Scanning Calorimetry of Skin Collagen)

  • 피부조직 콜라겐의 열안정성에 대한 기초자료를 얻고자 DSC를 이용하여 Tm(transition temperature)와 ${\Delta}H(enthalpy)$를 측정하였다. 수화시간과 수화율에 따른 불용성 콜라겐의 DSC 특성은 수화시간에 비하여 수화율이 콜라겐 구조안정성에 미치는 영향이 큰 것으로 나타났으며, 수화율이 증가할수록 Tm은 낮아지고 ${\Delta}H$는 증가하였다. 주령 및 성별에 따른 acetone dried skin의 Tm 및 ΔH는 암컷의 경우보다 수컷의 경우가 다소 높았으며, 주령이 증가할수록 Tm과 ${\Delta}H$가 서서히 증가하고 있어 열안정성은 나이와 관계가 있음을 보여주었다. Acetone dried skin의 경우와는 달리 염가용성 콜라겐의 DSC 특성값은 암컷의 경우가 수컷의 경우보다 다소 높았으며, 암컷의 Tm과 ${\Delta}H$는 주령의 증가에 따라 유의적으로 감소하는 경향을 보였다. 이러한 결과는 피부조직내에서의 콜라겐 구조는 성별 또는 주령에 따라 달리 변화되고 있음을 간접적으로 시사한 것으로 생각되었다.

Examination of the Presence and Differential Expression of Connexin Isoforms in the Urinary Bladder of the Male Rat During Postnatal Period

  • The coordinated action of the detrusor muscle cells in the urinary bladder is governed by cell-cell communication through gap junction, consisted of connexin (Cx) molecules. Even though a number of researches have been mostly focused on expressional changes of a few Cx isoforms in clinically dysfunctional condition of the bladder, less attention has been paid for investigation of Cx isoforms present in the bladder. Using real-time PCR analysis, the present study examined Cx isoforms expressing in the male rat bladder during postnatal period. Also, expressional patterns of Cx isoforms were evaluated in the bladder at different postnatal ages. Of a total of 13 Cx isoforms tested in the present study, we were able to detect mRNAs of 6 Cx isoforms in the rat urinary bladder, including Cxs 31, 31.1, 32, 37, 40, and 45. The transcript levels of Cxs 31, 31.1, 37, 40, and 45 were gradually increased from 1 week of age until 25 days of age, followed by transient decreases at 45 days of age. However, abundance of Cx32 transcript was drastically increased at 15 days of age, followed by a sharp drop at 45 days of age. These results indicate that differential expression of Cx isoforms in the bladder during postnatal development would be necessary for maintaining proper function of the bladder. A question remains to be answered if significant decreases of transcript levels of some Cx isoforms at the elderly are associated with age-dependent dysfunction of the bladder.

복합한방처방 SH21-B의 랫드와 Beagle 견에 대한 단회 경구투여 독성시험 (Acute Oral Toxicity Test of Oriental Medical Prescription SH21-B)

  • This study was performed to evaluate the acute oral toxicity of an oriental medical prescription for obesity treatment, SH21-B, in Sprague-Dawley rats and Beagle dogs. SH21-B was administered in rats at does of 0mg/kg, 2,000mg/kg, and 5,000mg/kg. And also SH21-B was administered in Beagle dogs at does of 150mg/kg, 300mg/kg, and 600mg/kg. The rats and dogs of both sexes were observed daily for 14 days after single oral administration. Two female rats, one administered at 2,000mg/kg and the other administered at 5,000mg/kg, died, but no dead animal was observed among male rats. Therefore LD50 in the female rat is observed to be 8,710mg/kg, and MLD(Minimum Lethal Dose) of the male rat is observed to be more than 5,000mg/kg. Among dogs, no dead animal was observed up to 600mg/kg and MLD is observed to be more than 600mg/kg.
