On Improvement of Garlic Productivity by Inactivation of Virus in Garlics (마늘 Virus 불활성화에 의한 생산성 향상에 관하여)

  • The effect of heat or chemotherapeutant treatment on the mosaic virus infected garlic(Allium sativum L.) scales and that of chemotherapeutant added to the culture medium were summarized as following. The treatment of the virus infected garlic scales at $37\~57^{\circ}C$ for 35 days to one hour in water or in air shelved no effort of inactivating the virus. Although treatment of the garlic scales at $62\~72^{\circ}C$ for 90 to five minutes reduced the mosaic symptom on the leaves of the garlic plants grown after the heat treatment, it reduced the growth vigor of the plants so greatly that complete inactivation of the virus in garlics was not feasible. The mosaic symptom on the leaves of garlic plant was reduced when the infected garlic scales were grown after 24 hours soaking in $10\~50\;ppm$ Malachite Green, 2,4-Dichlorophnoxy Acetic Acid, or in $20\~100\;ppm$ Quinhydron. These chemotherapeutants, however, inhibited the growth of garlic plant at the high concentration. Garlic scales soaked in $10\~50\;ppm$ Naphthyl Acetic Acid showed the least mosaic symptom without its complete extiction on the garlic leaves. When incorporated into the modified Murashige-Skoog's medium, $0.5\~l.5\;ppm$ Naphthyl Acetic Acid could inactivate the mosaic virus in newy developed garlic plants showing no mosaic symptom on the leaves, no inclusion bodies and intact nuclei in the leaf tissue cells.

Effects of pH and Temperature on the Adsorption of Cationic Dyes from Aqueous Suspension by Maghnia Montmorillonite (수용액으로부터 양이온 염료 흡수에 대한 pH 및 온도 효과)

  • The effects of pH and temperature on the removal of two dyes (neutral red; NR and malachite green oxalates; MG) from aqueous effluents using Maghnia montmorillonite clay in a batch adsorption process were investigated. The results showed the stability of the optical properties of MG in aqueous solution and adsorbed onto clay under wide range of pH 3-9. However, the interaction of NR dye with clay is accompanied by a red shift of the main absorption bands of monomer cations under pH range of 3-5, whereas, those of neutral form remains nearly constant over the pH range of 8-12. The optimal pH for favorable adsorption of the dyes, i.e. ${\geq}$90% has been achieved in aqueous solutions at 6 and 7 for NR and VM respectively. The most suitable adsorption temperatures were 298 and 318 K with maximum adsorption capacities of 465.13mg/g for NR and 459.89 mg/g for MG. The adsorption equilibrium results for both dyes follow Langmuir, Freundlich isotherms. The numerical values of the mean free energy $E_a$ of 4.472-5.559 kj/mol and 2.000-2.886 kj/mol for NR and MG respectively indicated physical adsorption. Various thermodynamic parameters, such as ${\Delta}H^{\circ}$, ${\Delta}S^{\circ}$, ${\Delta}G^{\circ}$ and Ea have been calculated. The data showed that the adsorption process is spontaneous and endothermic. The sticking probability model was further used to assess the potential feasibility of the clay mineral as an alternative adsorbent for organic ion pollutants in aqueous solution.

Decolorization of Azo, Triphenylmethane and Heterocyclic Dyes by Irpex zonatus BN2 (송곳니구름버섯(Irpex zonatus) BN2에 의한 아조계, 트리페닐메탄계 및 헤테로싸이클릭계 염료의 탈색)

  • The present research was undertaken to investigate the activity of ligninolytic enzymes and the decolorization capability of some dyes with Irpex zonatus BN2, isolated from nature and identified. For the assay of enzyme activities, the isolate did not produce lignin peroxidase (LiP) and veratryl alcohol oxidase (VAO), but laccase and manganese dependent peroxidase (MnP). While the activity for MnP was low $(61.6\;nmol/mg{\cdot}protein)$, its laccase activity was very high $(1185.9\;nmol/mg{\cdot}protein)$. Moreover, laccase had appeared earlier than MnP. When the isolate was incubated with each dye for 10 days, the decolorization rates of azo dyes, such as orange II, orange G, tropaeolin O and congo red were 98.0%, 97.4%, 99.0% and 95.3%, respectively. In case of heterocyclic dyes, eosin Y, toludine blue, methyl blue and azur B were 97.4 %, 98.7%, 99.9% and 94.0% respectively. Finally the results of triphenylmethane dye such as basic fuchsin, malachite green and crystal violet were 98.5%, 95.7% and 99.4%, respectively. The results suggest that laccase of Irpex zonatus BN2 should be played an important role in the decolorization of the dyes.

Photocurrent and Its Stability Enhancement of Dye-sensitized Nanoparticle $TiO_2$ Solar Cells (염료감응 나노입자 $TiO_2$ 태양전지의 광전류와 그 안정성 향상)

  • A solar cell based on dye-sensitized photoelectric conversion was studied by electrochemical and spec-trofluorometric methods for the purposes of enhancing its efficiency and stability of $TiO_2$ solar cells. Nanocrystalline $TiO_2$ was used to prepare photoelectrodes, and photosensitizing dyes such as malachite green oxalate, basic blue3, rhodamine B, and bromocresol purple were chosen as sensitizers. Electrochemical oxidation potentials and absorption and emission wavelengths of dyes were used to determine energy levels of the dyes. By comparing excited energy levels of the dyes with the conduction band edge potential $(E_{c,s})\;of\;TiO_2$ calculated by using the flat-band potential $(E_{fb})\;of\;TiO_2$, properties of a dye required to fabricate a high efficient photosensitizing solar cell with high short-circuit current $(J_{sc})$ were suggested. Enhanced stability of photocurrent was obtained by coating a $TiO_2|ITO$ electrode with Polypyrrole that Possibly Prevented the recombination between the conduction band electrons and oxidized dyes and suppressed the direct electrode redox reactions of dyes on ITO.

Comparison of Pigments and Estimation of Production Period in Old and New Celestial Charts Folding Screens (신구법천문도 채색 안료 비교 및 제작시기 추정)

  • The pigments of three old and new celestial charts folding screens(『Celestial Chart(Folding Screen)』 and 『Old and New Celestial Charts, Eight-Panel Folding Screen』 of National Folk Museum of Korea and 『Koudou-Nanboku-Ryousouseizu』 of National Diet Library of Japan) were analyzed to estimate their dating. It was estimated that the 『Celestial Charts(Folding Screen)』 was painted using traditional pigments from the Joseon dynasty such as azurite, indigo lake, malachite, atacamite, vermilion, iron oxide, cochineal, gamboge, orpiment, lead white, talc and soot. The green and blue colors of the 『Old and New Celestial Charts, Eight-Panel Folding Screen』 and 『Koudou-Nanboku-Ryousouseizu』 were painted using artificial inorganic pigments such as emerald green and ultramarine blue. These pigments were imported from Europe post the mid-19th century. In the 『Old and New Celestial Chart, Eight-Panel Folding Screen』, only artificial inorganic pigments were used for green and blue colors. However in the 『Koudou-Nanboku-Ryousouseizu』, emerald green and atacamite in green color, and ultramarine blue and indigo lake in blue color were used together. Based on both the results of pigment analysis and the study of star charts and inscriptions, the 『Celestial Charts(Folding Screen)』 was painted post mid-18th century. The 『Koudou-Nanboku-Ryousouseizu』 and 『Old and New Celestial Charts, Eight-Panel Folding Screen』 were painted after green and blue artificial pigments were imported in the mid-19th century. The 『Koudou-Nanboku-Ryousouseizu』 in which both traditional and western artificial pigments were used, can be dated earlier than the 『Old and New Celestial Chart, Eight-Panel Folding Screen』.

A Study on a Morphological Identification of Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora and Codonopsis pilosula (사삼(沙參), 양유근(羊乳根), 제니 및 당삼(黨參)의 형태(形態) 감별에 관한 연구)

  • Objectives : A morphological classification among Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora and Codonopsis pilosula was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Codonopsis pilosula is like a long cone, with amole-like projected parts at the beginning of the roots. 2. Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora, and Codonopsis pilosula are all similar as following: 1) Cork layers are composed of a few to over 10 subsidiary cells, with individual or a group of stone cells among them. 2) Phloem is wide and has a milk tube. Conclusion : Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora, and Codonopsis pilosula are all similar and difficult to be distinguished through microscopic observation, so that chemical experiments should be conducted for their identification.

  • Objectives : A morphological classification among Sinomenium acutum, Cocculus trilobus and Aristolochia fangchi was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Fiber bundles of pericycle in S. acutum are like a crescent moon, stone cells were found among fiber bundles, and starch grain and raphides of calcium oxalate were included in parenchyma. 2. C. trilobus has the fiber bundle of pericycle resembling a crescent moon, but does not have the raphidesof calcium oxalate. 3. A. fangchi has the raphides of calcium oxalate, but does not have the fiber bundles of pericycle. 4. Stems and roots of S. acutum arc mostly similar, but only stems have the pith. Conclusion :' C. trilobus and A. fangchi can be distinguished from each other by the fiber bundles of pericycle and the raphides of calcium oxalate.

Comparative Efficacy of Antifungal Agents for Aquaculture Fish and their Eggs (양식 어류와 이들 난에 대한 항곰팡이성 약물들의 효과 비교)

  • In fresh water fish hatcheries and farms, Saprolegniales often cause serious mortality to the fish and their eggs. Malachite green is an effective antifungal agent, but is carcinogenic to fish and humans. Alternative antifungal agents are needed. Presently, we tested various concentrations of MBT-01108 (Opuntia ficus-indica extracts) alone and in combination with bronopol, formalin and sodium chloride (MBT-01108 mixture) on in vitro mycelial growth and in vivo remediation of adult eel, Anguilla japonica, infected with Saprolegnia sp. and fertilized eggs of chum salmon, Oncorhynchus keta, to evaluate the compounds' antifungal efficacy on eyed-egg and hatching rates. MBT-Oll08 mixtures incorporating bronopol and formalin at respective concentrations of 50 and 30 parts per million (ppm), and 100 and 20 ppm were most effective in controlling Saprolegnia in vitro and in vivo (P<0.05). Repeated daily exposures to 50 ppm and 100 ppm MBT-01108 were more effective than exposure every 2-3 days post-fertilization for the inhibition of Saprolegnia infection of rainbow trout, Oncorhynchus mykiss eggs as compared with control (0 ppm).

  • Objectives : In order to distinguish Akebiae Caulis and Aristoiochiae Manshuriensis Caulis, their external and internal states were examined by microscope. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results : 1. Akebiae Caulis has stone cells and fiber bundles in its pericycle part, which include square crystal of calcium oxalate. Outside of neogenesis has a lot of cells with square crystal of calcium oxalate. 2. Aristoiochiae Manshuriensis Caulis has a little lignified fiber bundles in its pericycle part, and parenchymatous cell has delicate starch grains and druse of calcium oxalate. Conclusions : In conclusion, internal forms of Akebiae Caulis and of Aristoiochiae Manshuriensis Caulis are different in that each includes square crystal of calcium oxalate and druse of calcium oxalate respectively, so that it may be possible to distinguish them with optical microscope.

  • Objectives : Because Ligusticum sinense Root as well as those of Angelica tenuissima Root are sole as Gobon(藁本) in domestic markets, morphological characteristics of them are attempted to be distinguished. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results : 1. Angelica tenuissima Root has considerable number of thick roots, but Chinese Ligusticum sinense Root and Ligusticum jeholense Root are composed of rhizomes mostly with small roots. 2. Intersection of rhizomes of Angelica tenuissima Root shows a 'V'-like arrangement of xylem vessel. 3. Chinese Ligusticum sinense Root and Ligusticum jeholense Root did not show the 'V'-like arrangement, and sliced fiber bundles were found in xylem. 4. There was no difference between Chinese Ligusticum sinense Root and Ligusticum jeholense Root. Conclusions : In conclusion, Angelica tenuissima Root, Chinese Ligusticum sinense Root, and Ligusticum jeholense Root show characteristic differences in internal forms, so that it is possible to distinguish them with optical microscope.
