Antiallergic Action of Magnolia Officinalis on Immediate Hypersensitivity Reaction

  • We studied the effect of aqueous extract of Magnolia officinalis bark (Magnoliaceae) (MOAE) on the immediate hypersensitivity reaction. MOAE (0.01 to 1g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in rats. MOAE (0.1 and 1g/kg) also significantly inhibited local immunoglobulin E (lgE)-mediated passive cutaneous anaphylactic reaction. When MOAE was pretreated at concentrations ranging from 0.01 to 1g/kg, the levels of plasma histamine were reduced in a dose-dependent manner. MOAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) Igl. The level of cyclic AMP (CAMP) in RPMC, when MOAE was added, significantly increased compared with that of the normal control. Moreover, MOAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP Igl-induced tumor necrosis factor-$\alpha$ production from RPMC. These results indicate that MOAE inhibits immediate hypersensitivity reaction in vivo and in vitro.

Inhibitory Principles from Magnolia officinalis on Tumor Necrosis $Factor-{\alpha}$ Production in Lipopolysaccharide-Activated RAW264.7 cells

  • In the course of a search for tumor necrosis factor $(TNF)-{\alpha}$ inhibitory compounds from medicinal plants, we identified neolignans, honokiol and magnolol, from the alcoholic extract of Magnolia offcinalis as active inhibitory principles. These compounds dose-dependently inhibited $TNF-{\alpha}$ production without displaying cytotoxicity and their inhibitory activities measured by $IC_{50}$ values were 53.7 and $61.4\;{\mu}M$, respectively.

중국 및 일본산 후박 잎의 형태 및 성분 비교 (The Comparison of Morphological and Constituents of the Leaves of Magnolia officinalis, M. biloba and M. obovata)

  • In this study, we analyzed and quantified the contents of magnolol (1), honokiol (2), and obovatol (3) in the leaves of Magnolia species by HPLC. HPLC methods were developed and validated by ICH guide lines comparing the linearity, intraday precision, inter-day precision. The optimal conditions were as follows; wavelength: 288 nm, column: Zorbax Eclipse XDBC18 $(2.1\times150\;mm,\;5{\mu}m)$, mobile phase: Acetonitril - 0.1 % $H_3PO_4$ (45:55, v/v) buffer, flow rate: 0.5 ml/min. The contents of magnolol, honokiol and obovatol were 1.08%, 2.37%, and 0.57% in the leaves of M. officinalis, 0.50%, 0.03%, and 1.54% in the leaves of M. biloba, 0.37%, 0.03%, and 0.76% in the leaves of M. obovata. The contents of effective ingredients in the leaves of M. officinalis and M biloba were higher than that of M. obovata. Especially, the content of obovatol in the leaves of M. biloba was remarkably high. This result showed that the shapes of leaves have a certain relationship with the content of obovatol.

Inhibitory Effect of Oriental Herbal Medicines on Tumor Necrosis $Factor-{\alpha}$ Production in Lipopolysaccharide-stimulated RAW264.7 cells

  • Eleven out of 118 herbal medicines which are frequently used in Korean traditional prescriptions for inflammatory diseases exhibited more than 50% of inhibition on $TNF-{\alpha}$ production in LPS-stimulated RAW264.7 cells by their total ethanol extracts with 0.1 mg/ml as a final concentration. The active 11 total extracts were prepared from Angelica koreana, Coptis japonica, Cynanchum paniculatum, Magnolia frgesii, Magnolia officinalis, Panax ginseng, Patrinia scabiosaefolia, Pterocarpus santalius, Rhapontica uniflora, Saussurea lappa. Of them, Coptis japonica, Magnolia fargesii and Saussurea lappa also significantly inhibited $TNF-{\alpha}$ production in vivo. These total extracts were sequentially fractionated with n-hexane, methylene chloride, ethyl acetate, n-butanol and water. Among the solvent-fractionated extracts with 0.05 mg/ml as a final concentration, inhibitory effects of Angelica koreana, Magnolia fargesii, Magnolia officinalis, Pterocarpus santalinus, Rhapontica uniflora and Saussurea lappa markedly showed in one or two solvent fractions suggesting that the principles may be concentrated by subfractionation as the main compounds.

Effect of Macleaya cordata and Magnolia officinalis plant extracts on oxidative stress control in lambs fed a high-concentrate diet

  • Objective: The objective of this experiment was to compare conventional antioxidants and plant extracts for oxidative stress control in lambs fed a high-concentrate diet. Methods: Forty-eight male Dorper×Santa Ines lambs with an initial weight of 20±1.49 kg and 60 days of age, were used to evaluate the effects of feeding a combination of Macleaya cordata and Magnolia officinalis plant extracts (0 vs 320 mg/kg dry matter [DM]) in combination with selenium+vitamin E (0 vs 100 IU/kg DM of vitamin E and 0.1 mg/kg DM of selenium) in a completely randomized block design in a 2×2 factorial arrangement. The animals were housed in individual pens and received a high-concentrate diet consisting of 80% whole corn and 20% protein pellet for 60 days. The animals were weighed at the beginning of the experiment and every 14 days for performance monitoring. Three blood samplings were performed during the experimental period for the evaluation of oxidative and protein parameters. Results: The treatments with vitamin E and selenium as additives had a positive influence on final weight, daily weight gain, carcass weight, and selenium content in longissimus muscle (p = 0.01). Plant extracts tended to improve final weight (p = 0.064) and daily weight gain (p = 0.059), showing similar effect as selenium and vitamin E. There was no effect of treatment on blood proteins, indicating that the animals were healthy throughout the experiment. Conclusion: The use of plant extracts had a similar effect as the addition of selenium and vitamin E, with dietary inclusion of additives resulting in better performance of lambs but both supplements did not have strong influence on oxidative stress.

고지방식이유도 실험용 Rat에서 Statin 및 한약 2종(후박, 하수오) 투여에 따른 혈중 지질, 조직 변화 비교 연구 (Dyslipidemic Effect of Pravastatin and Herbal Medicine in Rats)

  • Objective: The goal of this preclinical study was to compare the dyslipidemic effect of pravastatin with that of herbal medicine in rats. Methods: In total, 40 rats were divided into 4 groups: Normal (10 rats), Control (10 rats), Statin alone (10 rats), and the MO-PM-S group (10 rats), which was given the powder of the cortex of Magnolia officinalis Rehd. et Wils., the root of Polygonum multiflorum Thunb, and pravastatin. The Control group, the Statin alone group, and the MO-PM-S group were all given a high-fat (45%) diet that made them obese. After 2 weeks of drug administration, the dyslipidemic effect of pravastatin was compared with that of herbal medicine in rats by analyzing the lipid profiles, measuring the body weights, and taking biopsies (liver, aorta). Results: The herbal medicine and the statin complex group got a much lower TG level and a slightly higher HDL-cholesterol level than the other groups. However, it got a higher total cholesterol and LDL-cholesterol level than the other groups. In biopsies, 30% of the Statin alone group and 10% of the MO-PM-S group showed mild histopathologic findings in the liver. Conclusion: The cortex of the Magnolia officinalis Rehd. et Wils. and the root of Polygonum multiflorum Thunb have dyslipidemic effects from the perspective of a TG level and HDL-cholesterol. However, the herbal mixture has a raising effect on both the LDL-cholesterol and the total cholesterol levels. Therefore, we cannot conclude that the herbal mixture helps to prevent dyslipidemia. In liver biopsies, the group administered with both the herbal mixture and the statin showed less histopathologic findings than the group administered with statin alone. This means that the herbal mixture helps to prevent fatty degeneration of the liver.

요엽후박나무 추출물의 입술 주름 개선에 대한 연구 (Effects of Magnolia Officinalis Bark Extract on Improvement of Lip Wrinkles)

  • 입술은 각질층이 매우 얇아 수분증발에 취약하며, 노화 과정에서 주름이 증가하고, 붉은색을 잃으며, 볼륨이 감소하게 된다. 매력적인 입술을 가지기 위해 시행되는 지방이식, 필러 주입을 대체하기 위한 성분에 대한 연구는 아직 보고된 바가 거의 없으며, 최근 새로운 지방세포의 수를 증가시키는 것이 인체 내 지방을 늘릴 수 있는 방법으로 제안되고 있다. 우리는 선행연구에서 지방줄기세포를 지방세포로 분화 유도하는 천연물질로써 요엽후박나무 추출물(Magnolia officinalis bark extract)의 우수한 효능을 확인하였다. 본 연구에서는 요엽후박나무 추출물이 바이오 프린팅으로 제작한 지방 유사 구조체에서 지방(lipid droplet)의 양을 증가시키면서 분화를 촉진시킴을 3D 수준에서 확인하였다. 다음으로 입술 주름에 미치는 영향을 확인하기 위해 주름 사진으로 부터 명암값의 표준편차(SDGV)를 J 이미지 소프트 웨어를 사용하여 측정함으로써 객관적 측정 방법을 확립하였고, 주름 정도에 따른 입술 주름 그레이드를 도출하여 정량화하였다. 결과적으로 요엽후 박나무 추출물 1%를 함유한 제품을 12주간 사용했을 때, 입술 주름을 유의하게 개선시킬 수 있음을 확인하였다. 본 연구 결과는 요엽후박나무 추출물이 지방줄기세포를 지방세포로 분화 유도하는 효능을 가지며, 이러한 효능이 입술 주름을 개선하는데 도움을 줄 수 있다는 점을 시사하고, 따라서 요엽후박나무 추출물은 입술 주름과 볼륨을 개선하는 화장품 후보 소재로 적용 가능하다는 것을 보여준다.

후박(厚朴)의 외.내부형태 및 이화학패턴연구 (A Study of Morphology and Pattern Analysis in Magnoliae Cortex)

  • Objectives: To determine the standards for discrimination of Magnoliae Cortex, the experiment of specific external-internal characters and the physicochemical pattern analysis were performed. Methods: External characteristics was observed using a stereoscope. Paraffin-mediated sectioned materials were stained by Ju's method. Physicochemical patterns of materials were analyzed using HPLC. Results: 1. Botanical characteristics: Magnolia officinalis had one seed and a white flower, while M. obovata had two seeds and a white flower. Machilus thunbergii had berry and spherical fruits and yellowish green panicles. 2. External characteristics: M. officinalis and M obovata were dark and thick. M. officinalis was gray brown and greasy while M. obovata was light-gray, less oily and smoothly sectioned. Machilus thunbergii was thin and relatively light or yellow-brown, coarsely sectioned and faintly specific scents. 3. Internal characteristics: The bast parts of M. officinalis and M. obovata were commonly wider than Machilus thunbergii The cork cortex of M. officinalis was $10{\sim}mg/L$ cell layers with many oil cells, while that of M. obovata was $4{\sim}7$ cell layers with less oil cells. Machilus thunbergii's xylem which consisted of ring-shaped cambium at 1st and 2nd part was occupied in large portion. 4. Physicochemical pattern: Both M. officinalis and M. obovata involved honokiol and magnolol. All kinds of M. officinalis involved Magnatriol B but one kind of M. ovobata and all of Machilus thunbergii didn't. Machilus thunbergii showed different pattern of chromatogram from that of 2 species above. Conclusions: These results could be used as standards for discrimination of Magnoliae Cortex and as the method of objectification in medicinal herbs giving the basic resource for bioactivity research.
