Analytical Determination of Cyanide in Maesil (Prunus mume) Extracts (매실추출제품의 시안화합물 분석법에 관한 연구)

  • Picrate, enzyme-picrate and instrumental analysis methods using IC (Ion Chromatography) and HPLC (High Performance Liquid Chromatography) were compared for their effectiveness in determining cyanide in extracts of Maesil, which is classified as a harmful substance. First, the picrate method showed the shortest analysis time (about 5 hr). The color of picrate paper changed at 0.01 mg/$200\;mL\;CN^-$. However, it was difficult to detect cyanide from amygdalin of glucosides. Second, we performed a qualitative analysis for total cyanide (free cyanide and cyanide from amygdalin) by the enzyme-picrate method using $\beta$-glucosidase and a quantitative analysis by spectrophotometry. Finally, analysis of cyanide by IC and HPLC required the longest determining time (about 17 hr) as well as pretreatment for each free cyanide and amygdalin. These results suggest that enzyme-picrate is the most effective analysis method for the detection of cyanide in Maesil extracts.

Effect of Maesil(Prunus mume) Juice on Antimicrobial Activity and Shelf-Life of Wet Noodle (매실(Prunus mume) 착즙액이 항균성과 생면의 저장성에 미치는 영향)

  • The effect of addition with maesil(Prunus mume) juice for extending the shelf-life of wet noodle was investigated by measuring quality changes such as total microbial count and pH. The Prunus mume juice showed antimicrobial activities against Salmonella enteritidis, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, and Escherichia coli. When the wet noodles containing Prunus mume juice were kept at $4^{\circ}C$ for 20 days, it was showed that the drop of pH was not significantly occurred during the storage. Total microbial counts for control exceeded the initial putrefactive criterion level of $1.0{\times}10^6\;cfu/g$ at 16 days of storage at $4^{\circ}C$. However, total microbial count of wet noodles with the addition of 10%, 20% and 30% Prunus mume juice were $3.0{\times}10^2\;cfu/g,\;3.0{\times}10^2\;cfu/g,\;and\;1.5{\times}10^2\;cfu/g$, respectively, and these bacterial counts were still less than the criterion level ever at 20 days of storage. The addition of Prunus mume juice extended the shelf-life of wet noodle appreciably.

Induction of Apoptosis by Treatment of Human Prostate Cancer LNCaP Cells with Methanol Fractions from Prunus mume (매실(Prunus mume) 메탄올 분획물의 처리에 따른 인체 전립선암세포 LNCaP의 apoptosis 유도 효과)

  • This study examined the growth inhibitory effect of the methanol fraction of maesil (Prunus mume) extract (MMF) on LNCaP, PC-3, and RC-58T human prostate cancer cell lines. Among these cell lines, LNCaP was the most sensitive to the inhibitory effects of MMF. Observation of the morphology and apoptotic body formation in the LNCaP cells revealed morphological changes, nuclear damage, and condensation in response to MMF treatment. The suppressive effect of MMF was related to the intrinsic apoptosis pathway, as indicated by increased expression of the pro-apoptotic proteins Bax, capase-3, capase-9, and PARP and decreased expression of the anti-apoptotic protein Bcl-2. Combined treatment with MMF and the AIF inhibitor N-phenylmalemide (N-PM) indicated that MMF treatment alone had a significant growth suppression effect. The involvement of the extrinsic apoptosis pathway was also confirmed by increased expression of AIF and Endo G. The growth suppression effect of MMF was also significant when compared to the effects of a combination of the PI3K inhibitor LY294002 and MMF. The reduced expression of PI3K, p-Akt, and p-mTOR confirmed the involvement of the PI3K/Akt/ mTOR signaling pathway in regulating the anti-proliferative properties of MMF. In conclusion, the growth suppression effect of MMF in the LNCaP human prostate cancer cell line shows the possibility of using this natural product in functional foods.

Effect of Natural Plant Components on the Nitrite-scavenging (천연식물성분이 아질산염 소거에 미치는 영향)

  • The purpose of this presents is to investigate, using natural food extracts(Green tea; Camellia sinesis, Du'chung; Eucommia ulmoides Oliver, Eu sung cho; Houttuynia cordate Thunb, Sam back cho; Saurus Chinensis, Baek hwa sa seal oho; Oldenladia diffusa Roxb., Laver; Porphyra tenera, Sea mustard; Undaria pinnatifda and Sea staghorn; Condium fragile) and juices(Sweet pepper; Capsicum annuum var. angulosum, Kale; Brassia oleracea var. acephala, Cucumber; Cucumis sativus, Onion; Allium cepa, Tomato; Lycopericon esculentum, Maesil; Prunus mume, Plum; Prunus salicina and Grape; Vitis spp.), the effect of natural plant components on the nitrite-scavenging under the different levels of pH. From the above mentioned extracts and juices, the content of vitamin C was detected, containing 65.1∼77.1 mg/100g, at the highest level in the Green tea, and followed by vegetables and fruits in order. The nitrite scavenging effect of teas, medicinal plants, and seaweed extracts, in the reaction system under the condition of pH 1.2, were 57.0∼100%, 50.0∼100%, and 18.0∼99.0%, respectively. Effect of Maesil(Prunus mume) byproduct Obtained from Maesil Liqueur Manufacture on Kimchi Fermentation (매실 리큐르 제조 부산물인 매실의 첨가가 김치 발효에 미치는 영향)

  • Quality changes of kimchi added with 10 or 20% Prunus mume liqueur byproduct (PLB), obtained after producing Prunus mume liqueur. during fermentation at $10^{\circ}C$ for 25 days were investigated. The pH and titrtable acidity in 20% PLB added kimchi were changed more gradually during fermentation for 25 days compared to control. Total bacteria and lactic acid bacteria counts in kimchi added with 20% PLB were lower than those of control during fermentation for 15 days. Kimchi fermentation was delayed about 10 days with 20% PLB. L and a values of kimchi added with PLB decreased but b value increased with increasing the concentration of PLB. In the sensory evaluation of kimchi fermented for 10 days, the texture score of PLB added kimchi was higher than that of control, and increased with increasing the concentration of PLB. There were no significant differences (p < 0.05) in overall acceptability among control kimchi and PLB added kimchi.

Effects of Herbal Sports Drinks with Omija, Maesil and Molasses on the Endurance and Energy Metabolites of Experimental Animals (오미자, 매실 및 당밀을 함유하는 한방스포츠음료가 실험동물의 지구력과 에너지 대사 성분에 미치는 영향)

  • The antifatigue and endurance promoting properties of two Korean medicinal herb extracts and molasses with various mineral components were studied by evaluating forced-swimming capacity and biochemical parameters in ICR mice. The treatment groups were orally administered mineral beverages which were contained 6% sugar with the mixture of Maesil (Prunus mume fruit) extracts, Omija (Schisandra chinensis fruit) extracts and molasses for 4 weeks. The exercised forced-swimming tests were conducted after 28 days of beverage supplementation. The swimming times to exhaustion were longer 1.5${\sim}$2 times in group 6 and group 10 than control goup (Control: 93.2${\pm}$10.4 sec; Beverage 6; 190.8${\pm}$25.6 sec, Beverage 10; 173.6${\pm}$21.8 sec; p<0.05). Moreover, the activity of hexokinase (Control: 5.23${\pm}$0.38 ${\mu}mol$l/g tissue; Beverage 6: 5.99${\pm}$0.18 ${\mu}mol$/g tissue, Beverage 10: 6.13${\pm}$0.25 ${\mu}mol$/g tissue, p<0.05) and citrate synthase (control: 42.9${\pm}$1.87 ${\mu}mol$/g tissue; Beverage 6: 56.8${\pm}$3.98 ${\mu}mol$/g tissue, Beverage 10; 59.5${\pm}$3.09 ${\mu}mol$/g tissue, p<0.05) were also significantly higher than those of control group. Even if the treatment groups had long swimming than control group, there is no significant difference in the glycogen contents of gastrocnemus muscle or liver between the control group and each treatment group. This demonstrated an improvement in endurance. These results suggest that reported herbal beverage is very effective to combat fatigue, improve endurance and increase overall physical activity.

Relative Abundance of Stink Bugs on Four Stone Fruits (Prunus spp.) in Korea (핵과류 4종에 발생하는 노린재의 종류와 상대적인 풍부도)

  • Stink bugs (Hemiptera: Pentatomidae) are economically important pests of fruit trees in Korea. The aim of the present study was to survey the relative abundance of stink bugs on four stone fruits, maesil (Prunus mume Sieb. et Zucc), cherry (Prunus avium L.), plum (Prunus salicina Lindl), and peach (Prunus persica (L.) Batsch), from 2017 to 2019 in the field. Four stink bug species were observed, including Carbula putoni (Jakovlev), Dolycoris baccarum (L.), Halyomorpha halys (Stål), Plautia stali Scott. H. halys was the most abundant (65%), followed by P. stali (26%), C. putoni (7%), and D. baccarum (2%). H. halys was the dominant species in maesil, cherry, and peach fruits, whereas P. stali was the dominant species in plum fruits. Most (81%) of the stink bugs observed were adults, with nymphs accounting for only 19% of the observed specimens. More stink bugs were observed during the late season than during the early season. These finding suggest that control strategies should be developed for the management of H. halys and P. stali at harvest in stone fruit tree orchards in Korea.
