두경부 편평상피세포암 세포주의 염색체 이상 분석: 비교유전체보합법과 Array 비교유전체보합법 (Cytogenetic Analysis in Korean Head and Neck Cancer Cell Lines: Comparative Genomic Hybridization(CGH) and Array-CGH)

  • Head and neck squamous cell carcinoma(HNSCC) is notorious for its poor outcome and increasing incidence. But, the studies of cytogenetic analysis in HNSCC are relatively rare, because of difficulties in culturing solid tumor cells and complexity in chromosomal DNA abberations associated with the lesions. The purpose of this study is to evaluate the location of chromosomal aberrations in Korean HNSCC cell lines (SNU-1041, 1066, and 1076) with comparative genomic hybridization(CGH) and array based CGH(array-CGH). Chromosomal gains of 3q23-q27, 5p13-p15.3, 7p21-pter, 8q11.2-q12, 8q21.1-qter, 9q22-q34, 16q22-q24, and 20q11.2-qter, as well as chromosomal losses on 3p10-p14 were found in all 3 SNU cell lines. Losses on 3p15- p23, 4q22-q27, 4q31.3-qter, 6q14-q15, 7q31-q34, 8p12-pter, 18q21-q23, and 21q11.2-q12 were observed in 2 of 3 cell lines. In array-CGH, many genes were altered including gains of PIK3CA, MYC, EVI1, MAD1L1 genes and losses of SERPIN genes. These aberrations of gene and chromosome coincide with other results of study, generally. These data about the patterns of chromosomal aberrations could be a basic step for understanding more detailed genetic events in the carcinogenesis and also provide information for diagosis and treatment in HNSCC.

도로터널 내 환기소 위치별 방재 효율에 관한 실험적 연구 (An experimental study of smoke extraction efficiency along with ventilation building location in the mad tunnel)

  • 본 연구에서는 도로터널 화재 시 지하 및 지상 환기소의 방재 효율을 검토하기 위해 터널 축소모형실험을 실시하였다. 터널 모형은 Froude 상사를 기본으로 도로터널을 1/50로 축소하여 20 m의 모형 터널을 제작하였다. 방재 효율 분석에 적용된 연직갱은 설제 터널 설계시 고려된 두 가지 모형으로서 실터널 560, $280\;m^3/s$의 배연량을 1/50으로 상사하여 적용하였다. 결과로 모두 연직갱을 통한 CO 가스의 전량 배출이 가능한 것으로 분석되었다. 두 환기소의 CO 농도는 지상 환기소가 지하 환기소 보다 2.23~2.73 ppm 작게 배출되었다. 그리고 지상 환기소의 경우 터널 벽면의 냉각효과로 인해 화재 후 온도의 상승이 지하 환기소보다 $0.53{\sim}0.94^{\circ}C$ 낮게 나타났다. 이상의 축소모형 환기소 내 CO농도 및 온도변화의 분석 결과, 방재적인 측면에서 볼 때 도로터널 내 환기소의 위치는 지하보다 지상에 위치하는 것이 유리할 것으로 판단되었다.

다양한 전처리에 따른 중온혐기-고온호기 복합 슬러지 처리공정의 슬러지 처리효율 및 메탄 생성량 변화 (Effects of diverse Pre-treatment methods on the sludge digestion and methane production in combined mesophilic anaerobic and thermophilic aerobic sludge digestion process)

  • 본 연구에서는 유입 슬러지에 다양한 전처리 방법을 적용하여 전처리 방법이 중온혐기-고온호기 복합 슬러지 처리 공정의 슬러지 소화효율과 메탄가스 생성량에 미치는 영향을 비교 검증하였다. 실험실 규모의 슬러지 소화장치를 제작하여 서로 다른 유입 슬러지 전처리방법을 적용하여 4단계로 실험을 진행하였다. 1단계에서는 전처리를 하지 않은 슬러지를 공급하였고, 2, 3, 4단계에서는 각각 열처리, 열-알칼리처리, 장기 알칼리 처리(7일)를 거친 유입 슬러지를 공급하였다. 실험 결과, 1단계에서 4단계까지 진행되는 동안 총COD 제거율은 44%에서 76%까지 증가하였으며, 메탄 생성량 또한 101mL/L/day에서 165, 256mL/L/day까지 크게 증가하였다. 한편, 4단계에서는 7일간의 장기 알칼리 처리를 하였음에도 불구하고 3단계에 비해 총COD 제거율과 메탄 생성량이 증가하지 않았다. 결론적으로, 유입 슬러지의 전처리를 통해 복합 슬러지 처리 공정의 슬러지 제거 효율과 메탄생성량을 크게 증가시킬 수 있었으며, 여러 가지 전처리 방법 중 열-알칼리 처리법이 가장 효율적임을 실험결과를 통해 알 수 있었다.

가상주행과 실차주행의 운전자 주행행태 차이에 관한 연구 (A Study on the Compensation of the Difference of Driving Behavior between the Driving Vehicle and Driving Simulator)

  • PURPOSES : The use of virtual driving tests to determine actual road driving behavior is increasing. However, the results indicate a gap between real and virtual driving under same road conditions road based on ergonomic factors, such as anxiety and speed. In the future, the use of virtual driving tests is expected to increase. For this reason, the purpose of this study is to analyze the gap between real and virtual driving on same road conditions and to use a calibration formula to allow for higher reliability of virtual driving tests. METHODS : An intelligent driving recorder was used to capture real driving. A driving simulator was used to record virtual driving. Additionally, a virtual driving map was made with the UC-Win/Road software. We gathered data including geometric structure information, driving information, driver information, and road operation information for real driving and virtual driving on the same road conditions. In this study we investigated a range of gaps, driving speeds, and lateral positions, and introduced a calibration formula to the virtual record to achieve the same record as the real driving situation by applying the effects of the main causes of discrepancy between the two (driving speed and lateral position) using a linear regression model. RESULTS: In the virtual driving test, driving speed and lateral position were determined to be higher and bigger than in the real Driving test, respectively. Additionally, the virtual driving test reduces the concentration, anxiety, and reality when compared to the real driving test. The formula includes four variables to produce the calibration: tangent driving speed, curve driving speed, tangent lateral position, and curve lateral position. However, the tangent lateral position was excluded because it was not statistically significant. CONCLUSIONS: The results of analyzing the formula from MPB (mean prediction bias), MAD (mean absolute deviation) is after applying the formula to the virtual driving test, similar to the real driving test so that the formula works. Because this study was conducted on a national, two-way road, the road speed limit was 80 km/h, and the lane width was 3.0-3.5 m. It works in the same condition road restrictively.

Structural basis of novel TRP14, thioredoxin-related protein that regulates TNE-$\alpha$ signaling pathways

  • Thioredoxin (Trx) is a small redox protein that is ubiquitously distributed from achaes to human. In diverse organisms, the protein is involved in various physiological roles by acting as electron donor and regulators of transcription and apoptosis as well as antioxidants. Sequences of Trx within various species are 27~69% identical to that of E. coli and all Trx proteins have the same overall fold, which consists of central five β strands surrounded by four α helices. The N-terminal cysteine in WCGPC motif of Trx is redox sensitive and the motif is highly conserved. Compared with general cysteine, the N-terminal cysteine has low pKa value. The result leads to increased reduction activity of protein. Recently, novel thio.edoxin-related protein (TRP14) was found from rat brain. TRP14 acts as disulfide reductase like Trx1, and its redox potential and pKa are similar to those of Trx1. However, TRP14 takes up electrons from cytosolic thioredoxin reductase (TrxR1), not from the mitochondrial thioredoxin reductase (TrxR2). Biological roles of TES14 were reported to be involved in regulating TNF-α induced signaling pathways in different manner with Trx1. In depletion experiments, depletion of TRP14 increased TNF-α induced phosphorylation and degradation of IκBα more than the depletion Trx1 did. It also facilitated activation of JNK and p38 MAP kinase induced by TNF-α. Unlike Trx1, TRP14 shows neither interaction nor interference with ASK1. Here, we determined three-dimensional crystal structure of TRP14 by MAD method at 1.8Å. The structure reveals that the conserved cis-Pro (Pro90) and active site-W-C-X-X-C motif, which may be involved in substrate recognition similar to Trx1 , are located at the beginning position of strand β4 and helix α2, respectively. The TRP14 structure also shows that surface of TRP14 in the vicinity of the active site, which is surrounded by an extended flexible loop and an additional short a helix, is different from that of Trx1. In addition, the structure exhibits that TRP14 interact with a distinct target proteins compared with Trx1 and the binding may depend mainly on hydrophobic and charge interactions. Consequently, the structure supports biological data that the TRP14 is involved in regulating TNF-α induced signaling pathways in different manner with Trx1.

주부들의 식품안전 관련 구매행동 및 정보요구도 조사 (A Survey on the Housewives' Purchasing Behavior and Needs for Food Safety Information)

  • 본 연구는 주부들의 식품안전과 관련한 식품구매행동과 식품안전에 대한 정보요구도를 조사하기 위하여 수행되었다. 연구를 위한 자료는 서울 및 수도권 지역 거주 만 20세 이상의 주부를 대상으로 2009년 3월 9일부터 4월 3일까지 수집되었다. 결과를 요약하면 첫째, 식품안전 관련 구매행동에서 '유통기한을 확인'한다는 응답이 4.59로 가장 높았고, '국내산 구입'(4.08), '가격보다 안전을 고려'(3.66) 등이 다음으로 높게 나타났다. 둘째, 식품안전에 관한 교육 경험이 있다고 응답한 사람은 18.5%에 그쳤는데, 식품안전 위해요소에 관한 항목별 지식수준에 있어 '환경호르몬' 항목은 정답률이 89.1%로 가장 높았고 '광우병' 항목은 25.8%로 가장 낮았으며, 전체 지식수준은 총 13점 만점에 평균 8.56점으로 그다지 높지 않은 것으로 나타났다. 셋째, 식품안전에 관한 정보탐색도는 2.61로 낮게 나타났는데, 식품안전에 관한 정보원천으로는 TV/라디오가 4.04로 가장 높았고 신문/잡지(3.80), 인터넷(3.44), 가족/친지/친구(3.39) 등이 다음으로 높게 나타났다. 넷째, 식품안전에 관한 정보요구도는 3.82로 높게 나타나 평소 식품안전에 관한 정보를 많이 필요로 하고 있는 것으로 나타났다. 따라서 정부, 제조업자, 가공 및 유통업자 등은 식품안전 및 식품위해요소들에 관한 정확하고 충분한 정보를 다양한 정보원을 통해 제공해야 하며 또한 각종 교육프로그램을 개발 및 운영하여야 한다. 이를 통해, 보다 적극적인 소비자들의 정보탐색 활동을 유도하고 식품안전관련 지식수준을 향상시켜 나감으로써, 궁극적으로 소비자들의 안전을 고려한 올바른 식품구매행동을 유도해 나가야 할 것이다.

황제내경(黃帝內徑)에 나타난 맥진법(脈診法)에 관한 연구(硏究) (A study on the method of Macjin(脈診) in The Whang Di Nei Qing(黃帝內徑))

  • This thesis is a study on the diagnostic theory and methods of Macjin(checking of pulse) as described in the Whang Di Nei Qing. There are four methods of Macjin in this text, namely: Dong Macjin(動脈法), Sam Bu Gu Who Macjin(三部九脈診). In Young Gi Gu Macjin(人迎氣口脈診法), Nei Qing's Gi Gu Macjin(內徑의 氣口脈法). Each method developed in it's own way, but they are all closely related and became the basis of today's Macjin. I examined the methods of Macjin in the Whang Di Nei Qing from a perspective of "change". The main questions of this study are: "What brought about the development of the different methods?". "How do the methods differ?", and "how did these methods evoke into today's Macjin?". The method of Macjin generated spontaneously from the observation and treatment of disease. Dong Macjin was the first method developed and became the basis of subsequent forms of Macjin. The accumulation of medical knowledge and the influence of oriental philosophy fueled the evolution of Macjin. Chronologically, Macjin methods developed starting with Dong Macjin and eventually into Sam Bu Gu Who Macjin. In Young Gi Gu Macjin, and Nei Qing's Gi Gu Macjin. The different methods of Macjin vary in how many pulse points are checked, and were established with simplicity and effectiveness in mind. Dong Macjin involves the checking the whole body. Sam Bu Gu Who Macjin involves nine points. In Young Gi Gu Macjin involves four points. and Nei Qing's Gi Gu Macjin involves two. In it's early development, the checking of a patient's pulse was used to diagnose only localized disorders in comparison with symptoms. It evolved with Oh Jang Mad(五臟脈, five main types of pulse) to be able to check internal organs with the introduction of Nei Qing's Gi Gu Macjin. After the division of Chon Kwan Chuck(寸關尺), it evolved further to be able to make a detailed diagnosis by using the result of pules checks. Nowadays, we can make 28 forms of diagnostic indices from these development. In conclusion, Macjin can be used practically and effectively in the diagnosis of disease. In using three methods of Macjin(In Young Gi Gu Macjin, Nei Qing's Gi Gu Macjin, and today's Gi Gu Macjin, which can determine the whole body's Siate of well-being) in conjunction with Dong Macjin(which helps pinpoint the localized disorder), an exact diagnosis can be obtained. I strongly feel that we should acquire objectivity by accommodating Macjin with modern methods of medicine.

Validating the Structural Behavior and Response of Burj Khalifa: Synopsis of the Full Scale Structural Health Monitoring Programs

  • New generation of tall and complex buildings systems are now introduced that are reflective of the latest development in materials, design, sustainability, construction, and IT technologies. While the complexity in design is being overcome by the availability and advances in structural analysis tools and readily advanced software, the design of these buildings are still reliant on minimum code requirements that yet to be validated in full scale. The involvement of the author in the design and construction planning of Burj Khalifa since its inception until its completion prompted the author to conceptually develop an extensive survey and real-time structural health monitoring program to validate all the fundamental assumptions mad for the design and construction planning of the tower. The Burj Khalifa Project is the tallest structure ever built by man; the tower is 828 meters tall and comprises of 162 floors above grade and 3 basement levels. Early integration of aerodynamic shaping and wind engineering played a major role in the architectural massing and design of this multi-use tower, where mitigating and taming the dynamic wind effects was one of the most important design criteria established at the onset of the project design. Understanding the structural and foundation system behaviors of the tower are the key fundamental drivers for the development and execution of a state-of-the-art survey and structural health monitoring (SHM) programs. Therefore, the focus of this paper is to discuss the execution of the survey and real-time structural health monitoring programs to confirm the structural behavioral response of the tower during construction stage and during its service life; the monitoring programs included 1) monitoring the tower's foundation system, 2) monitoring the foundation settlement, 3) measuring the strains of the tower vertical elements, 4) measuring the wall and column vertical shortening due to elastic, shrinkage and creep effects, 5) measuring the lateral displacement of the tower under its own gravity loads (including asymmetrical effects) resulting from immediate elastic and long term creep effects, 6) measuring the building lateral movements and dynamic characteristic in real time during construction, 7) measuring the building displacements, accelerations, dynamic characteristics, and structural behavior in real time under building permanent conditions, 8) and monitoring the Pinnacle dynamic behavior and fatigue characteristics. This extensive SHM program has resulted in extensive insight into the structural response of the tower, allowed control the construction process, allowed for the evaluation of the structural response in effective and immediate manner and it allowed for immediate correlation between the measured and the predicted behavior. The survey and SHM programs developed for Burj Khalifa will with no doubt pioneer the use of new survey techniques and the execution of new SHM program concepts as part of the fundamental design of building structures. Moreover, this survey and SHM programs will be benchmarked as a model for the development of future generation of SHM programs for all critical and essential facilities, however, but with much improved devices and technologies, which are now being considered by the author for another tall and complex building development, that is presently under construction.

근거리 무선통신 기술 기반 차량간통신 시스템 개발 (A development of the Vehicle-To-Vehicle communication system using the Dedicated Short Range Communication technology)

  • 본 논문에서는 5.8 GHz 대역의 근거리전용 무선통신기술(DSRC)을 이용한 차량과 차량간의 무선통신에 관하여 연구하였다 첨단화, 지능화 되고 있는 현재의 도로교통체계에서 주행하는 차량간의 통신은 첨단의 지능형 주행을 위한 중요기술이다. 주행 중에 통신 반경 내에 있는 주변 차량들의 속도, 위치, 제동, 운전 상태에 관한 정보를 교환함으로서 원활하고 안전한 교통흐름을 도모할 수 있다. 따라서 본 연구에서는 고속의 차량 이동 환경에서 1Mbps 급의 대용량 데이터 처리 능력을 갖는 5.8 GHz 대역의 DSRC 기술을 이용하여 LOS (Line Of Sight) 조건에서 사용이 가능한 차량간통신 시스템을 개발하였다. 이를 위하여 완전한 능동형의 차량간통신이 가능하도록 물리계층, 데이터 링크계층과 논리계층 그리고 차량간통신 서비스를 위한 응용계층으로 구분하여 개발하였다. 본 연구를 통하여 ASV (Advanced Safety Vehicle)나 SSVS (Super Smart Vehicle System)의 실현을 위한 차량간 무선통신 기술을 확립하였고 이를 통하여 첨단의 ITS 구현 가능성을 확인할 수 있었다.

장면전환에 효율적인 H.264/AVC 비트율 제어 기법 (An Effective of Rate Control for Scene Change in H.264/AVC)

  • H.264/AVC 표준은 기존의 부호화 기법보다 뛰어난 압축 성능 때문에 비디오 데이터의 압축과 전송에 널리 응용될 것으로 전망된다. 그러나 H.264/AVC 표준도 기존의 비디오 압축표준과 마찬가지로 가변길이부호화 방식을 따르기 때문에, 동영상을 부호화할 때 발생하는 데이터량은 시간의 흐름에 따라 큰 폭으로 변한다. 이 때문에 PSTN이나 ISDN 망과 같이 대역폭이 고정된 네트워크를 통해 비디오 비트 스트림을 전송하고자 할 경우에는 부호기로부터 출력되는 비트량을 제어할 필요가 있다. 본 논문에서는 전송율이 고정된 비디오 전송채널 환경에서 H.264/AVC 부호화 기법으로 압축된 영상 데이터를 전송할 때 장면전환에 효율적인 비트율 제어 방식을 제안한다. 즉, 제안하는 비트율 제어 방식은 장면 전환과 같이 움직임이 많은 부분에서는 이전 프레임과 현재 프레임간의 움직임 변화량을 예측하여 움직임의 양에 따라 2차원 RD 모델 식을 수정하여 프레임 단위로 비트량을 효율적으로 제어하는 방법이다. 또한 장면전환이 많은 영상일 경우에 버퍼의 넘침을 방지하기 위해서 효율적인 프레임 스킵 방법을 제안한다. 실험 결과는 화면간 움직임이 많은 영상의 경우, 기존의 비트량 제어 방법이 채널 전송에 부적합할 정도로 비트량을 초과하지만, 제안한 방법은 채널 전송에 적합하게 효율적으로 부호화한다. 또한 기존 비트율 방법보다 제안한 방법의 프레임 스킵 개수가 적어 화면간의 끊김 현상이 줄었다.
