Study of Takliikgi-tang on the Anti-Cancer Action in Mice (탁리익기탕(托裏益氣湯)이 항암(抗癌) 및 면역기능(免疫機能)에 미치는 작용(作用) 연구(硏究))

  • Objective : This Study was to investigate effects of Takliikgi-tang on the anti-cancer and proliferation of immunocytes, nitric oxide (NO) production of peritoneal macrophages. Methods : We used Takliikgi-tang extract (TLT) with freeze-dried, 8weeks-old male mice and cancer cell lines (L1210, Sarcoma-180) for this Study. The cytotoxicity and proliferation of cells were tested using a colorimetric tetrazoliun assay (MTT assay). Results : 1. TLT was significantly showed cytotoxicity on the L1210, 5-180 cell lines. 2. TLT was significantly increased proliferation of thymocytes in vitro. 3. TLT was significantly increased proliferation of thymocytes and splenocytes in normal mice. 4. TLT was significantly increased NO production from peritoneal macrophages in normal mice. 5. TLT was significantly decreased proliferation of L1210 cells in L1210 cells transplanted mice. 6. TLT was significantly increased proliferation of thymocytes and splenocytes by all-dosage in L1210 cells transplanted mice. 7. TLT was significantly increased NO production from peritoneal macrophages in L1210 cells transplanted mice. Conclusions : The present author thought that TLT had action of anti-cancer by becoming immunocytes activity (NO production, proliferation of thymocytes).

Comparative Study of Ginseng Radix and Insamhodo-tang on Immune Reaction (인삼과 인삼호도탕(人參胡挑湯)의 면역반응에 대한 비교 연구)

  • Immunological activities of Ginseng Radix water extract (GE) were compared with theirs of Insamhodo-tang water extract (IHT). GE and IHT (500 mg/kg) were administered p.o. twice a day for 5 days to C57BL/6 mice, respectively. IHT decreased the cell viability of murine thymocytes and splenocytes, but increased the population of $CD4^+$ cells in thymocytes, and the population of $Thy1^+$ cells and $CD4^+$ cells in splenocytes in contrast to GE. In addition, IHT decreased the phagocytic activity of peritoneal macrophages, but increased the production of nitric oxide from peritoneal macrophages in contrast to GE. These results suggest that IHT decrease a specific- and nonspecific immune reaction in comparison with GE.

  • Out of twenty rabbits which died of rabbit hemorrhagic disease spontaneously occurring in Korea, five animals had a concurrent infection with Encephalitozoon cuniculi in the cerebrum. The lesions were composed of granulomas, leptomeningitis and perivascular cuffing with mononuclear cells. The granulomas consisted of a central necrotic focus surrounded by an infiltration with plasma cells, lymphocytes and macrophages. Gliosis was associated with the granulomas. Gram-positive organisms were detected in the cerebrum from two rabbits. They were oval to rod-shaped with blunt round ends. The distribution of the pathogens was investigated by the direct avidin-biotin peroxidase complex method. They were present in pseudocysts and macrophages. Pseudocysts were found in the granulomas as well as the neuropil without cellular reactions. Some organisms were present within reticulo-endothelial cells of blood capillaries and macrophages in the subarachnoid spaces. These organisms had ultrastructural characteristics consistent with Encephalitozoon cuniculi.

Effects of Okbyungpoongsan Administration on Innate and Specific Immune Response in the Mouse (옥병풍산(玉屛風散)이 생쥐의 선천성 및 특이적 면역반응에 미치는 영향)

  • Okbyungpoongsan(OBPS) has long been known to have anti-allergic effect. In order to evaluate the influence on innate and specific immune response, the effects of OBPS on vascular permeability. hypersensitivities and phagocytic functions were measured. As the results, OBPS increased phagocytic activity of peritoneal macrophages in vitro and in vivo. But OBPS depressed formation of reactive oxygen intermediates(ROI) in vitro and in vivo, while the drug enhanced generation macrophages. Foot pad swelling in the mouse and contact hypersensitivity against dinitroflouorobenzene were decreased. OBPS had no effect on NK cells. But OBPS decreased vascular permeability induced by histamine without statistical significance. These results demonstrate that OBPS suppresses hypersensitivity reactions without affecting phagocytic functions and formation of ROI from macrophages. It also means that OBPS acts as a effective inducer to synthesis of nitric oxide which is effective for the infectious disease while it does damage to tissue less as it suppresses ROI, So we can conclude that OBPS could be used for the treatment of the disease related with immune function.

Effect of Scutellariae Radix Water Extract on Hydrogen Peroxide Production in LPS-induced RAW 264.7 Mouse Macrophages (황금(黃芩) 물추출물이 LPS로 유발된 마우스 대식세포의 hydrogen peroxide 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate effects of Scutellariae Radix water extract on hydrogen peroxide production in LPS-induced RAW 264.7 mouse macrophages. Methods : Scutellariae Radix produced from South Korea (SK) and Scutellariae Radix produced from China (SC) were extracted by hot water. Effects of SK and SC on hydrogen peroxide production in LPS-induced RAW 264.7 were measured by dihydrorhodamine 123 assay after 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL. Results : SK significantly increased hydrogen peroxide production in LPS-induced RAW 264.7 cells for 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL (P < 0.05). But SC did not represent any significant effect on hydrogen peroxide production in LPS-induced RAW 264.7 cells. Conclusions : These results suggest that Scutellariae Radix, especially produced from South Korea, has the immune-enhancing property related with its increasement of bacteriocidal hydrogen peroxide production in LPS-induced macrophages.

Inhibitory Effect of the Leaves of Rumex crispus L. on LPS-induced Nitric Oxide Production and the Expression of iNOS and COX-2 in Macrophages

  • In this study, the anti-inflammatory effects of ethanol extract of Rumex crispus L. and its fractions were investigated in RAW 264.7 macrophages. To evaluate the anti-inflammatory effects of extract, we studied nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-alpha ($TNF-{\alpha}$) levels in RAW 264.7 cells. The ethanol extract of R. crispus L. significantly decreased NO production and the levels of other inflammatory factors, such as PGE2 and $TNF-{\alpha}$, in lipopolysaccharide (LPS)-stimulated macrophages in a dose-dependent manner. We also assessed the inducible nitric oxide synthase (iNOS) and the cyclooxygenase 2 (COX-2) protein expression by western blot. Ethyl acetate fraction of R. crispus L. had the strongest anti-inflammatory effect. These results suggest that ethyl acetate extract of R. crispus L. might be beneficial in the treatment of chronic inflammatory diseases.

Epigenetic Control of Oxidative Stresses by Histone Acetyltransferases in Candida albicans

  • Candida albicans is a major pathogenic fungus in humans, and meets at first the innate immune cells, such as macrophages, in its host. One important strategy of the host cell to kill C. albicans is to produce reactive oxygen species (ROS) by the macrophages. In response to ROS produced by the macrophages, C. albicans operates its defense mechanisms against them by expressing its oxidative stress response genes. Although there have been many research studies explaining the specific transcription factors and the expression of the oxidative stress genes in C. albicans, the regulation of the oxidative stress genes by chromatin structure is little known. Epigenetic regulation by the chromatin structure is very important for the regulation of eukaryotic gene expression, including the chromatin structure dynamics by histone modifications. Among various histone modifications, histone acetylation is reported for its direct relationship to the regulation of gene expression. Recent studies reported that histone acetyltransferases regulate genes to respond to the oxidative stress in C. albicans. In this review, we introduce all histone acetyltransferases that C. albicans contains and some papers that explain how histone acetyltransferases participate in the oxidative stress response in C. albicans.

Inhibitory effects of Coptidis Rhizoma on the LPS-induced production of nitric oxide and $TNF-{\alpha}$ in mouse macrophage cells (황련의 쥐 대식세포로부터 LPS에 의해 유도되는 nitric oxide 및 $TNF-{\alpha}$의 생성억제효과)

  • Objectives : Coptidis Rhizoma has been known traditional medicine with antimicrobial activities. We investigated inhibitory effects of Coptidis Rhizoma extract on lipopolysaccharide(LPS)-induced nitric oxide production from mouse macrophages. Methods : After Coptidis Rhizoma extract was pretreated in BV2, mouse brain macrophages and RAW264.7 mouse macrophages, cells were activated with LPS. To investigate cytotoxicity Coptidis Rhizoma extract, cell viability was measured by MTT assay. The production of nitric oxide(NO) and inducible nitric oxide synthase(iNOS) was determined in each culture supernatant and mRNA by Griess reaction and RT-PCR. The production of $TNF-{\alpha}$ from cells was measured by ELISA. Results : Coptidis Rhizoma extract significantly inhibited LPS-induced NO production in BV2 and RAW264.7 cells. Coptidis Rhizoma extract also greatly suppressed mRNA expression of iNOS in BV2 and RAW264.7 cells activated by LPS. Conclusion : These data suggests that Coptidis Rhizoma extract may have an anti-inflammatory effect through the inhibition of NO production.

OK-432 Suppresses Proliferation and Metastasis by Tumor Associated Macrophages in Bladder Cancer

  • OK-432, a Streptococcus-derived anticancer immunotherapeutic agent, has been applied in clinic for many years and achieved great progress in various cancers. In the present study, we investigated its anticancer effect on bladder cancer through tumor associated macrophages (TAMs). MTS assay validated OK-432 could inhibit proliferation in both T24 and EJ bladder cell lines. OK-432 also induced apoptosis of bladder cancer cells in vitro. Consequently, we demonstrated that OK-432 could suppress the bladder cancer cells migration and invasion by altering the EMT-related factors. Furthermore, using SD rat model, we revealed that OK-432 inhibited tumor growth, suppressed PCNA expression and inhibited metastasis in vivo. Taken together, these findings strongly suggest that OK-432 inhibits cell proliferation and metastasis through inducing macrophages to secret cytokines in bladder cancer.
