Proteases and Protease Inhibitors Produced in Streptomycetes and Their Roles in Morphological Differentiation

  • Streptomycetes are Gram-positive microorganisms producing secondary metabolites through unique physiological differentiation [4]. The microbes show unusual morphological differentiation to form substrate mycelia, aerial mycelia, and arthrospores on solid medium [19]. Substrate mycelium growth is sustaining with sufficient nutrients in the culture medium. The concentration of a specific individual substrate in the culture environment is the most important extracellular factor allowing vegetative mycelia growth, where extracellular hydrolytic enzymes participate in the utilization of waterinsoluble substrates. However, with starvation of nutrients in the culture medium, the vegetative mycelia differentiate to aerial mycelia and spores. It has been considered that shiftdown of essential nutrients for mycelia growth is the most important factor triggering morphological and physiological differentiation in Streptomyces spp. Since proteineous macromolecule compounds are the major cellular components, these are faced to endogenously metabolize following a severe depletion of nitrogen source in culture nutrients (Fig. 1). Various proteases were identified of which production was specifically related with the phase of mycelium growth and also morphological differentiation. The involvement of proteases and protease inhibitor is reviewed as a factor explaining the mycelium differentiation in Streptomyces spp.

새로운 Hyperbranchedpolyimidesandpolyamides: 합성, 말단기 변형, 경화 연구, 그리고 물리적 성질 (New Hyperbranched Polyimides and Polyamides: Synthesis, Chain-End Functionalizations, Curing Studies, and Some Physical Properties)

  • While aromatic polyimides and polyamides have found widespread use as high performance polymers, the present work addressed the need for organosoluble materials through the use of a hyperbranching scheme. The $AB_2$ monomers were prepared. The $AB_2$ monomers were then polymerized via aromatic fluoride-displacement and Yamazaki reactions to afford the corresponding hydroxyl-terminated hyperbranched polyimides (HT-PAEKI) and amine-terminated hyperbranched polyamides, respectively. HT-FAEKI was then functionalized with allyl and propargyl bromides as well as epichlorohydrin to afford allyl-terminated AT-PAEKI, propargyl-terminated PT-PAEKI, and epoxy (glycidyl)-terminated ET-PAEKI, in that order. All hyperbranched poly(ether-ketone-imide)s were soluble in common organic solvents. AT-PAEKI was blended with a bisphenol-A-based bismaleimide (BFA-BMI) in various weight ratios. Thermal, rheological, and mechanical properties of these blend systems were evaluated. Two characteristic hyperbranched polyamides, which the one has para-electron donating groups to the surface amine groups and the other has para-electron withdrawing groups to the surface amine groups, were selected to compare BMI curing behaviors. The electron rich polymer displayed ordinary Michael addition type exothermic reaction, while electron deficient polymer did display unusual curing behaviors. Based on analytical data, the later system provided the strong evidences to support room temperature curing of BMI by reactive intermediates instead of reactive primary amine groups on the macromolecule surface.

  • Intercellular signaling is a crucial biological process for the coordination of cell differentiation, organ development and whole plant physiology. The intercellular movement of macromolecule signals such as proteins and RNAs has emerged as a novel mechanism of cell-to-cell communication in plant. Plasmodesmata, which are intercellular symplasmic channels, provide a key pathway for cell-to-cell trafficking of regulatory proteins / RNAs. This review specifically focuses on integrating the recent understanding on non-cell autonomous macromolecules, their function and regulatory mechanisms of intercellular trafficking through plasmodesmata.

Electrophoretic Tissue Clearing and Labeling Methods for Volume Imaging of Whole Organs

  • Detailed structural and molecular imaging of intact organs has incurred academic interest because the associated technique is expected to provide innovative information for biological investigation and pathological diagnosis. The conventional methods for volume imaging include reconstruction of images obtained from serially sectioned tissues. This approach requires intense manual work which involves inevitable uncertainty and much time to assemble the whole image of a target organ. Recently, effective tissue clearing techniques including CLARITY and ACT-PRESTO have been reported that enables visualization of molecularly labeled structures within intact organs in three dimensions. The central principle of the methods is transformation of intact tissue into an optically transpicuous and macromolecule permeable state without loss of intrinsic structural integrity. The rapidly evolving protocols enable morphological analysis and molecular labeling of normal and pathological characteristics in large assembled biological systems with single-cell resolution. The deep tissue volume imaging will provide fundamental information about mutual interaction among adjacent structures such as connectivity of neural circuits; meso-connectome and clinically significant structural alterations according to pathologic mechanisms or treatment procedures.

Using Harmonic Analysis and Optimization to Study Macromolecular Dynamics

  • Mechanical system dynamics plays an important role in the area of computational structural biology. Elastic network models (ENMs) for macromolecules (e.g., polymers, proteins, and nucleic acids such as DNA and RNA) have been developed to understand the relationship between their structure and biological function. For example. a protein, which is basically a folded polypeptide chain, can be simply modeled as a mass-spring system from the mechanical viewpoint. Since the conformational flexibility of a protein is dominantly subject to its chemical bond interactions (e.g., covalent bonds, salt bridges, and hydrogen bonds), these constraints can be modeled as linear spring connections between spatially proximal representatives in a variety of coarse-grained ENMs. Coarse-graining approaches enable one to simulate harmonic and anharmonic motions of large macromolecules in a PC, while all-atom based molecular dynamics (MD) simulation has been conventionally performed with an aid of supercomputer. A harmonic analysis of a macroscopic mechanical system, called normal mode analysis, has been adopted to analyze thermal fluctuations of a microscopic biological system around its equilibrium state. Furthermore, a structure-based system optimization, called elastic network interpolation, has been developed to predict nonlinear transition (or folding) pathways between two different functional states of a same macromolecule. The good agreement of simulation and experiment allows the employment of coarse-grained ENMs as a versatile tool for the study of macromolecular dynamics.

5-Fluorouracil 유도체 합성 및 항암작용 (Synthesis and Antitumor Activity of $N^1$-derivatives of 5-Fluorouracil)

  • In order to find out a proper connecting bridge between 5-fluorouracil(5-FU) and a macromolecule such as a polypeptide, potentially hydrolytic N$^{1}$-derivartives of 5-FU have been systhesized and evaluated for their biological activity. When tested with in vitro leukemic L$_{1210}$ cells all the obtained derivartives exhibited slightly higher antitumor activity than the parent 5FU. Among them the N$^{1}$ -carbamoyl analogue 2 and N$^{1}$-acetamido analogue 6b showed 50% inhibition of the L$_{1210}$ cell growth at the concentrations of 5.01$\times$10$^{-8}$M and 1.03$\times$10$^{-7}$M, respectively. When tested against sarcoma 180 tumor cells inoculated into mice, the compounds 2 and 6b exhibited, respectively, 62% and 54% inhibition of the solid tumor growth at the 5-time doses of 100 mg/kg/day. Both compounds, N$^{1}$-carbamoyl analogue 2 and N$^{1}$-acetamido analogue 6b, realeased the parent 5-FU when incubated in the L$_{1210}$ cell cultural media for 5 hrs.

  • 십자류 흐름 평판형 cell에서 비대칭 cellulose acetate 막으로 PEG #6000 및 dextran 70T 용액의 한외여과를 압력차와 도입용액 농도 변화에 따라 실험하여 막투과량 및 용질 배제도를 측정하고, 막투과량에 미치는 농도분극층 저항을 고찰하였다. 이 결과 농도분극층 저항 $R_{b1}$을 농도분극층내의 고분자 용액의 평균 농도 $C_{b1}$과 상관시켜 다음의 무차원 관계식으로 나타낼 수 있었다. $\frac{R_{b1}}{{R_m}}=\alpha[\frac{\rho_{b1}}{C_{b1}}]^\beta$

  • The focus of this paper is to develop a mathematical model for investigating the dynamic performance of a polymer electrolyte membrane fuel cell. The model in this work is based on physical laws having clear significance in replicating the fuel cell system and can easily be used to set up different operational strategies. Simulation results display the transient behavior of the voltage within each single cell, and also within a number of such single cells combined into a fuel cell stack system. A linear as well as a nonlinear analysis of the polymer electrolyte membrane fuel cell system(PEMFC) has been discussed in order to present a complete and comprehensive view of this kind of modeling. Also, a comparison of the two kinds of analysis has been performed. Finally, the various characteristics of the fuel cell system are plotted in order to help us understand its dynamic behavior. Results indicate that there is a considerable amount of error in the modeling process if we use a linear model of the fuel cell. Thus, the nonlinearities present in the fuel cell system should be taken into account in order to obtain a better understanding of the dynamic behavior of the fuel cell system.

Recent progress in the synthesis of luminescent copper clusters

  • Luminescent metallic clusters have attracted great interest due to their unique optical, electronic and chemical features. Comparing with intensively studied Au and Ag Clusters, Cu clusters are superior in the aspects of cost and wide industrial demanding. However, tiny copper clusters are extremely prone to aggregate and undergo susceptibility of oxidation, thereby the synthesis of fluorescent zero valent copper clusters is rather challenging. In this review, synthetic strategies towards luminescent copper clusters, including macromolecule-protection and micro molecule-capping, have been systematically surveyed. Both "bottom-up" and "top-down" synthetic routes are found to be effective in fabricating luminescent copper clusters, some of which are quite stable and possess decent luminescence quantum yields. In general, the synthesis of fluorescent copper clusters remains at its infant stage. A great deal of effort on developing novel and economic synthetic routes to produce bright and stable copper clusters is highly expected in future.

Co-immunomodulatory Activities of Anionic Macromolecules Extracted from Codium fragile with Red Ginseng Extract on Peritoneal Macrophage of Immune-Suppressed Mice

  • In this study we investigated the immune effects of oral administration of anionic macromolecules extracted from Codium fragile (CFAM) and red ginseng extract mixture on the peritoneal macrophage cells in immune-suppressed mice. Cyclophosphamide (CY) induces the immune-suppressed condition. CY-treated mice were orally fed with different concentrations of CFAM supplemented with red ginseng extract and the peritoneal macrophages collected. CY treatment significantly decreased the immune activities of peritoneal macrophages, compared to the normal mice. The administration of CFAM mixed with red ginseng extract significantly boosted the viability of macrophage cells and nitric oxide production of peritoneal macrophages. Further, the oral administration of CFAM mixed with red ginseng extract up-regulated the expression of iNOS, COX-2, and TLR-4 as well as cytokines such as IL-1β, IL-6, TNF-α, and IFN-γ more than the red ginseng-treated group. This study showed that CFAM enhanced the immune activity of red ginseng extract in the peritoneal macrophage cells of immune-suppressed mice. Furthermore, CFAM might be used as a co-stimulant of red ginseng extract through the regulation of macrophage cells for the enhancement of human health and immunity.
