New Records on Three Harpacticoid Copepods Associated with Marine Macroalgae in Korea

  • Three harpacticoid copepods, Dactylopodamphiascopsis latifolius (Sars, 1909), Diosaccus ezoensis Ito, 1974 and Ambunguipes rufocincta (Brady, 1880) are recorded as new to the Korean fauna with illustrations, on the basis of the specimens collected mainly from the marine macro algae during the period from June 1990 to September 1997.

  • Endophytic fungi have currently been acknowledged as the most promising source of bioactive compounds for drug discovery, and considerable progress has been made in exploring their diversity, species richness, and bioprospecting. Fungal endophytes from unique environmental settings offer a pool of potentially useful medicinal entities. Owing to the constant stresses imposed on macroalgae by marine environments, it is believed that algae and their associated endophytic symbionts represent a good source of structurally diverse bioactive secondary metabolites. Despite the proven significance of active metabolites of algal endophytes, little have been exploited. This review highlights the latest discoveries in algicolous endophytic research, with particular focus on the bioactive metabolites from algal endophytes. Compounds are classified according to their reported biological activities, like anticancer, antibacterial, antifungal, and antioxidant properties. Present experimental evidence suggests that a majority of the bioactive metabolites were reported from Phaeophyceae followed by Rhodophyceae and Chlorophyceae. An intensive search for newer and more effective bioactive metabolites has generated a treasure trove of publications, and this review partially covers the literature published up to 2016.

Metal Concentrations in some Brown Seaweeds from Kongsfjorden on Spitsbergen, Svalbard Islands (스발바드군도 스피츠베르겐섬 콩스피요르드에 서식하는 갈조류의 중금속 농도)

  • Concentrations of Al, As, Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, Zn were determined in four arctic brown algae (Laminaria saccharina, L. digitata, Alaria esculenta, Desmarestia aculeata) in an attempt to examine for their metal accumulation capacity and also to assess their contamination levels. Macroalgae were collected from shallow subtidal waters (<20m) of Kongsfjorden (Kings Bay) on Spitsbergen during the period of the late July to early August 2003. Metal concentrations highly varied between sampling sites, species and tissue parts. Input of melt-water laden with terrigenous sediment particles seemed to have a large influence on baseline accumulations of some metals (Al, Fe, Mn, Pb etc.) in the macroalgae, causing a significant spatial variation. There were also significant concentration differences between the young and old tissue parts in L. saccharina, L. digitata and A. esculenta. While Al, Fe, Mn, Pb were higher in the perennial parts (stipes and holdfast below meristematic region), Cd and As concentrations were significantly higher in the young blades above the meristematic region. Zn and Cr, on the other hand, showed little differences between the tissue parts. The highest metal concentrations were found in D. aculeata, which seems to be due to its filamentous fine branches leading to high surface/volume ratios. The lowest concentrations were found in the two Laminaria spp., the blades of which are thicker than D. aculeata and A. esculenta. No distinct signs of contamination were detected in the brown algal species analyzed. Added to this, the results of the present studies suggest the potential utility of L. saccharina, L. digitata and A. esculenta as biomonitors for metal pollution monitoring in this area.

Oxidative Stress and Antioxidant Activities of Intertidal Macroalgae in Korea

  • The oxidative stress level and antioxidant activities in two green algae (Ulva pertusa and Ulva linza), two brown algae (Agarum cribrosum and Dictyota dichotoma), and three red algae (Grateloupia lanceolata, Carpopeltis affinis, and Gracilaria verrucosa) collected from intertidal regions of Korea were assessed. In the two green algae, although the total glutathione content was not as high as that of the brown algae, the glutathione pool was extremely reduced, and the glutathione reductase (GRd)/glutathione peroxidase (GPx) activity ratio was high, which apparently plays an important role for protection against oxidative damage, as manifested by low lipid peroxidation. In the brown algae, which exhibited a low lipid peroxidation level that was comparable to the green algal species, the highest glutathione content, together with high GPx activity, appears to be the most important factor in their antioxidant protection. The red algal species exhibited extremely high lipid peroxidation levels. They also contained the lowest and most oxidized glutathione among the species, as well as the lowest GRd activity. In spite of the marked difference in the glutathione content, the significant difference in the activity of ${\gamma}$-glutamylcysteine ligase, the rate limiting enzyme for glutathione synthesis, among the species was not exhibited. Our results suggest that there is a significant difference in the levels of oxidative stress and antioxidant capacity among the algal species, and that the glutathione system, especially the efficiency of glutathione recycling, plays a vital role in antioxidative protection in algal species.

Patterns of Interactions among Neighbor species in a High Intertidal Algal Community

  • Three dominant rocky intertidal macroalgae, the fucoids Fucus gardneri and Pelvetiopsis limitata (Phaeophyta) and the red alga Mazzaella cornucopiae (= Iridaea cornucopiae) on the west coast of Vancouver Island, British Columbia, Canada were used in a series of field experiments to examine interspecific interactions. These experiments showed complex patterns which included an interchange of negative (inhibition) and positive (facilitation) interactions depending on neighbor distance. Less fucoid recruitment occurred in the plots with greater percent cover of a turfforming red alga, M. cornucopiae. However, experimentally removing Mazzaella turf (the turf was considered to be "blocking" fucoid recruits or "shading" growing recruits) did not increase recruitment. This result indicated that there may be another factor(s) involved in the survivorship of juvenile fucoids in the turf-removed plots. Morphological differences in adult plants between Mazzaella and the two fucoids resulted in another type of interaction; these began when fucoids successfully settled and grew nearby or within the red algal turf. By monitoring microhabitat at the individual plant level for two years, I found that survivorship of fucoid recruits showed different species-specific patterns. The patterns also varied as the microhabitat changed from Mazzaella turf edge to open space. For F. gardneri, longevity of P.limitata at all distances tested was similar. A reason for greater longevity of F. gardneri individuals at edge microhabitats may be that these sites have one side open to light and nutrients and another site that buffers them from desiccation and wave impact. In the Mazzaella-Fucus interaction, neighbor distance was a key factor in determining whether the outcome of the interaction would be competition or facilitation (or protection). This study provides experimental evidence that detectable biological interactions occur in this upper intertidal algal community where physical conditions are usually severe, and also indicates the importance of small scale examination in understanding macroalgal interactions in intertidal habitats.

Occurrence of Green Macroalgae (Ulva prolifera) Blooms in the Northern East China Sea in Summer 2008 (2008년 여름철 북부 동중국해에서 대규모 녹조(가시파래) 출현)

  • This study examined extensive patches of floating green macroalgal (Ulva prolifera) mats in the northern East China Sea (ECS) using satellite images from mid May through July 2008 and field observation made during early August 2008 cruise. It was previously reported that the massive macroalgal blooms occurred in the coastal areas of Qingdao in China. During our field survey, researchers noticed widely distributed floating patches of macroalgal mats ranging in size from tens of centimeters to a few hundred meters in diameter. Meteorological data in the northern ECS showed high irradiance, high air-temperature, and predominant southerly winds in summer. In the study area during the survey period, surface waters were characterized by the Changjiang Diluted Water (CDW) mass, which contained high concentrations of nitrate and phosphate. The internal transcribed spacer (ITS) sequence of U. prolifera found in the northern ECS was the same as those of U. prolifera sampled from Qingdao blooms, suggesting a possibility that U. prolifera found in two regions would be derived from the same origin. We suggest that U. prolifera in the nearshore Jiangsu Province drifted into the northern ECS and proliferated under favorable meteorological and oceanographic conditions during the summer of 2008.

Isolation and Characterization of a Novel Agar Degrading Bacterium, Alteromonas macleodii subsp. GNUM08120, from Red Macroalgae (홍조류로부터 신규 한천분해미생물 Alteromonas macleodii subsp. GNUM08120의 분리 및 동정)

  • An agar-hydrolyzing marine bacterium, strain GNUM08120, was isolated from Sargassum fulvellum collected from Yeongil bay of East Sea of Korea. The isolate was Gram-negative, aerobic, motile with single polar flagellum, and grew at 1-10% NaCl, pH 5.0-8.0, and $15-37^{\circ}C$. G+C content and the predominant respiratory quinone were 46.13 mol% and Q-8, respectively. The major cellular fatty acids were Summed feature 3 (24.5%), $C_{16:0}$ (21.7%), and $C_{18:1}{\omega}7c$ (12.5%). Based on 16S rRNA gene sequence similarity and DNA-DNA hybridization analyses, strain GNUM08120 was identified as a novel subspecies of Alteromonas macleodii, designated Alteromonas macleodii subsp. GNUM08120. Production of agarase by strain GNUM08120 was likely repressed by the effect of carbon catabolite repression caused by glucose. The crude agarase prepared from 12-h culture broth of strain GNUM08120 exhibited an optimum pH and temperature for agarase activity at 7.0 and $40^{\circ}C$, respectively. The crude enzyme produced (neo)agarobiose, (neo)agarotetraose, and (neo)agarohexaose as the hydrolyzed product of agarose.

Feeding Habits and Growth of the Sea Urchin, Strongylocentrotus pulcherrimus (A. Agassiz) Reared in the Laboratory (실험실에서 사육한 말똥성게 Strongylocentrotus pulcherrimus (A. Agassiz)의 식성과 성장)

  • Strongylocentrotus pulcherrimus is one of the very important fishery stock in this country since its high demand for raw gonad. Although the demand has been continuously increasing, the natural stock of this species seems to be decreasing. Since fresh macroalgae are not always available, the substitution of terrestrial vegetables for algae as a principal food was examined for the intensive indoor culture of the sea urchin. The results from the studies on selective and non-selective feeding habits and nutritional efficiencies using 13 food stuffs (6 macroalgae, 5 vegetables and 2 animal products) indicate that algae can be substituted with vegetables in sea urchin culture. Although the growth of body weight was the highest when sea urchins were fed Undaria Pinnatifida, the gonad index of the sea urchins fed on spinach, radish leaf and lettuce was higher than that of this brown alga. Considering the different food efficiencies of body and gonad growth, spinach and radish leaf will be adequate during gonad growth season, while lettuce and Chinese cabbage will be suitable for test growth season. In particular, radish leaf, which is usually not eaten by humans, could be highly beneficial and very economical in the sea urchin culture.

A Simple Method for Extraction of High Molecular Weight DNA fromPorphyra Tenera (Rhodophyta) Using Diatomaceous Earth

  • The innate soluble polysaccharides and phenolic compounds of marine macroalgae are serious contaminants which interfere with experimental procedures such as restriction enzyme digestion, polymerase chain reaction (PCR) and other enzymatic reactions using extracted DNA samples. The viscous polysaccharides are co-precipitated with DNA samples by isopropanol or ethanol precipitation in conventional experiment. To overcome the problem, a method for the isolation of high molecular weight DNA from Porphyra tenera is developed with the application of diatomaceous earth column. The isolated DNAs by this method were about 50-100 kb in size and could be digested well with restriction enzymes. The nuclease activity seemed to be minimal, and high reproducibility in the arbitrary primed PCR for RAPD analyses was a distinctive feature. These results suggest that this method is very efficient in isolating nucleic acid from macroalgae including Porphyra.
