Genome-Wide Association Studies Associated with Backfat Thickness in Landrace and Yorkshire Pigs

  • Although pork quality traits are important commercially, genome-wide association studies (GWASs) have not well considered Landrace and Yorkshire pigs worldwide. Landrace and Yorkshire pigs are important pork-providing breeds. Although quantitative trait loci of pigs are well-developed, significant genes in GWASs of pigs in Korea must be studied. Through a GWAS using the PLINK program, study of the significant genes in Korean pigs was performed. We conducted a GWAS and surveyed the gene ontology (GO) terms associated with the backfat thickness (BF) trait of these pigs. We included the breed information (Yorkshire and Landrace pigs) as a covariate. The significant genes after false discovery rate (<0.01) correction were AFG1L, SCAI, RIMS1, and SPDEF. The major GO terms for the top 5% of genes were related to neuronal genes, cell morphogenesis and actin cytoskeleton organization. The neuronal genes were previously reported as being associated with backfat thickness. However, the genes in our results were novel, and they included ZNF280D, BAIAP2, LRTM2, GABRA5, PCDH15, HERC1, DTNBP1, SLIT2, TRAPPC9, NGFR, APBB2, RBPJ, and ABL2. These novel genes might have roles in important cellular and physiological functions related to BF accumulation. The genes related to cell morphogenesis were NOX4, MKLN1, ZNF280D, BAIAP2, DNAAF1, LRTM2, PCDH15, NGFR, RBPJ, MYH9, APBB2, DTNBP1, TRIM62, and SLIT2. The genes that belonged to actin cytoskeleton organization were MKLN1, BAIAP2, PCDH15, BCAS3, MYH9, DTNBP1, ABL2, ADD2, and SLIT2.

보이차 열수 추출물의 근아세포 근분화에 미치는 영향 (Effect of Pu'er tea extract on C2C12 myoblast differentiation)

  • 현재 노화와 관련된 퇴행성 근육 질환은 심각한 문제로 간주되고 있으나 근육 감소증의 치료 및 예방에 대한 약물 효과는 충분히 연구되지 않다. 이에 중국 전통차인 보이차의 추출물을 근육 감소증의 증상을 완화하기 위한 치료제로서 가치를 평가하고자 하였다. 본 실험에서는 이를 평가하기 위하여 ABTS 분석, MTS 분석, 면역 블롯 분석, 면역 형광 현미경법을 수행하였다. 보이차 추출물은 우수한 라디칼 소거 능을 나타냈으며, 또한 근관을 형성하는 Myh3의 발현이 촉진시켰고, 근관 형성을 증진시켰다. 따라서 보이차는 근육생성을 촉진하는 천연 물질이며 근감소증을 포함한 다양한 근육 질환의 예방 및 치료에 대한 제약 연구의 재료로 가치가 있음을 시사하며, 보이차의 구체적인 지표물질을 확인하고 이에 대한 추가연구가 필요할 것으로 보인다.

Laminin as a Key Extracellular Matrix for Proliferation, Differentiation, and Maturation of Porcine Muscle Stem Cell Cultivation

  • Extracellular matrix (ECM) proteins play a crucial role in culturing muscle stem cells (MuSCs). However, there is a lack of extensive research on how each of these proteins influences proliferation and differentiation of MuSCs from livestock animals. Therefore, we investigated the effects of various ECM coatings-collagen, fibronectin, gelatin, and laminin-on the proliferation, differentiation, and maturation of porcine MuSCs. Porcine MuSCs, isolated from 14-day-old Berkshire piglets, were cultured on ECM-coated plates, undergoing three days of proliferation followed by three days of differentiation. MuSCs on laminin showed higher proliferation rate than others (p<0.05). There was no significant difference in the mRNA expression levels of PAX7, MYF5, and MYOD among MuSCs on laminin, collagen, and fibronectin (p>0.05). During the differentiation period, MuSCs cultured on laminin exhibited a significantly higher differentiation rate, resulting in thicker myotubes compared to those on other ECMs (p<0.05). Also, MuSCs on laminin showed higher expression of mRNA related with maturated muscle fiber such as MYH1 and MYH4 corresponding to muscle fiber type IIx and muscle fiber type IIb, respectively, compared with MuSCs on other ECM coatings (p<0.05). In summary, our comparison of ECMs revealed that laminin significantly enhances MuSC proliferation and differentiation, outperforming other ECMs. Specifically, muscle fibers cultured on laminin exhibited a more mature phenotype. These findings underscore laminin's potential to advance in vitro muscle research and cultured meat production, highlighting its role in supporting rapid cell proliferation, higher differentiation rates, and the development of mature muscle fibers.

길경에서 추출한 polygalacin D가 근원세포 분화 및 근위축에 미치는 영향 (Effects of polygalacin D extracted from Platycodon grandiflorum on myoblast differentiation and muscle atrophy)

  • 본 연구는 근생성 및 근위축 완화효능을 가진 유효소재 발굴의 필요성에 의해 polygalacin D가 근원세포 분화 및 미토콘드리아에 미치는 영향과 항암제 유도 근위축에 대한 완화효과를 각각 세포 및 동물실험을 통해 확인하고자 하였다. 그 결과, polygalacin D는 다핵을 지닌 근관세포의 수와 분화 종결인자인 MHC isoforms의 발현량을 증가시켰고 근육 내 단백질 분해 인자인 MuRF1, Smad2/3의 발현량은 유의적으로 감소시켰다. 또한 미토콘드리아 생합성 조절인자인 Pgc1α의 발현은 증가시키고 미토콘드리아 분열인자인 Drp1과 Fis1의 발현은 감소시켰다. 한편 zebrafish 동물모델을 통해 항암제 유도 근위축에 대한 개선효과를 확인한 결과, polygalacin D는 항암제에 의해 유도된 근위축과 미토콘드리아 손상을 완화시켰다. 이상의 결과들은 polygalacin D가 미토콘드리아 기능 증진을 매개로 근원세포 분화 촉진 및 근육 단백질 분해 저하 효과를 지닐 뿐만 아니라, 미토콘드리아 손상을 개선하여 항암제로 유도된 근위축에 대한 완화 효과를 나타냄을 시사한다. 따라서 본 연구를 통해 polygalacin D가 근생성 및 근위축 예방과 치료를 위한 잠재적인 유효소재로서의 가능성을 제시하였다.

Gynostemma pentaphyllum extract and Gypenoside L enhance skeletal muscle differentiation and mitochondrial metabolism by activating the PGC-1α pathway in C2C12 myotubes

  • BACKGROUND/OBJECTIVES: Peroxisome proliferator-activated receptor-gamma co-activator-1α (PGC-1α) has a central role in regulating muscle differentiation and mitochondrial metabolism. PGC-1α stimulates muscle growth and muscle fiber remodeling, concomitantly regulating lactate and lipid metabolism and promoting oxidative metabolism. Gynostemma pentaphyllum (Thumb.) has been widely employed as a traditional herbal medicine and possesses antioxidant, anti-obesity, anti-inflammatory, hypolipemic, hypoglycemic, and anticancer properties. We investigated whether G. pentaphyllum extract (GPE) and its active compound, gypenoside L (GL), affect muscle differentiation and mitochondrial metabolism via activation of the PGC-1α pathway in murine C2C12 myoblast cells. MATERIALS/METHODS: C2C12 cells were treated with GPE and GL, and quantitative reverse transcription polymerase chain reaction and western blot were used to analyze the mRNA and protein expression levels. Myh1 was determined using immunocytochemistry. Mitochondrial reactive oxygen species generation was measured using the 2'7'-dichlorofluorescein diacetate assay. RESULTS: GPE and GL promoted the differentiation of myoblasts into myotubes and elevated mRNA and protein expression levels of Myh1 (type IIx). GPE and GL also significantly increased the mRNA expression levels of the PGC-1α gene (Ppargc1a), lactate metabolism-regulatory genes (Esrra and Mct1), adipocyte-browning gene fibronectin type III domain-containing 5 gene (Fndc5), glycogen synthase gene (Gys), and lipid metabolism gene carnitine palmitoyltransferase 1b gene (Cpt1b). Moreover, GPE and GL induced the phosphorylation of AMP-activated protein kinase, p38, sirtuin1, and deacetylated PGC-1α. We also observed that treatment with GPE and GL significantly stimulated the expression of genes associated with the anti-oxidative stress response, such as Ucp2, Ucp3, Nrf2, and Sod2. CONCLUSIONS: The results indicated that GPE and GL enhance exercise performance by promoting myotube differentiation and mitochondrial metabolism through the upregulation of PGC-1α in C2C12 skeletal muscle.

Genetic classification and confirmation of inherited platelet disorders: current status in Korea

  • Inherited platelet disorders (IPDs), which manifest as primary hemostasis defects, often underlie abnormal bleeding and a family history of thrombocytopenia, bone marrow failure, hematologic malignancies, undefined mucocutaneous bleeding disorder, or congenital bony defects. Wide heterogeneity in IPD types with regard to the presence or absence of thrombocytopenia, platelet dysfunction, bone marrow failure, and dysmegakaryopoiesis is observed in patients. The individual processes involved in platelet production and hemostasis are genetically controlled; to date, mutations of more than 50 genes involved in various platelet biogenesis steps have been implicated in IPDs. Representative IPDs resulting from defects in specific pathways, such as thrombopoietin/MPL signaling; transcriptional regulation; granule formation, trafficking, and secretion; proplatelet formation; cytoskeleton regulation; and transmembrane glycoprotein signaling are reviewed, and the underlying gene mutations are discussed based on the National Center for Biotechnology Information database and Online Mendelian Inheritance in Man accession number. Further, the status and prevalence of genetically confirmed IPDs in Korea are explored based on searches of the PubMed and KoreaMed databases. IPDs are congenital bleeding disorders that can be dangerous due to unexpected bleeding and require genetic counseling for family members and descendants. Therefore, the pediatrician should be suspicious and aware of IPDs and perform the appropriate tests if the patient has unexpected bleeding. However, all IPDs are extremely rare; thus, the domestic incidences of IPDs are unclear and their diagnosis is difficult. Diagnostic confirmation or differential diagnoses of IPDs are challenging, time-consuming, and expensive, and patients are frequently misdiagnosed. Comprehensive molecular characterization and classification of these disorders should enable accurate and precise diagnosis and facilitate improved patient management.

cDNA Microarray를 이용한 치주인대세포와 치은섬유아세포의 유전자 발현에 대한 연구 (A Comparative Study of Gene Expression Patterns of Periodontal Ligament Cells and Gingival Fibroblasts using the cDNA Microarray)

  • Periodontal ligament(PDL) cells have been known as playing an important roles in periodontal regeneration and gingival fibroblasts are also important to periodontal regeneration by forming connective tissue attachment. There were rare studies about the gene expression patterns of PDL cells and gingival fibroblasts, therefore in this study, we tried cDNA microarray-based gene expression monitoring to explain the functional differences of PDL cells and gingival fibroblasts in vivo and to confirm the characteristics of PDL cells. Total RNA were extracted from PDL cells and gingival fibroblasts of same person and same passages, and mRNA were isolated from the total RNA using Oligotex mRNA midi kit(Qiagen) and then fluorescent cDNA probe were prepared. And microarray hybridization were performed. The gene expression patterns of PDL cells and gingival fibroblasts were quite different. About 400 genes were expressed more highly in the PDL cells than gingival fibroblasts and about 300 genes were more highly expressed in the gingival fibroblasts than PDL cells. Compared growth factor- and growth factor receptor-related gene expression patterns of PDL cells with gingival fibroblasts, IGF-2, IGF-2 associated protein, nerve growth factor, placental bone morphogenic protein, neuron-specific growth- associated protein, FGF receptor, EGF receptor-related gene and PDGF receptor were more highly expressed in the PDL cells than gingival fibroblasts. The results of collagen gene expression patterns showed that collagen type I, type III, type VI and type VII were more highly expressed in the PDL cells than gingival fibroblasts, and in the gingival fibroblasts collagen type V, XII were more highly expressed than PDL cells. The results of osteoblast-related gene expression patterns showed that osteoblast specific cysteine-rich protein were more highly expressed in the PDL cells than gingival fibroblasts. The results of cytoskeletal proteins gene expression patterns showed that a-smooth muscle actin, actin binding protein, smooth muscle myosin heavy chain homolog and myosin light chain were more highly expressed in the PDL cells than gingival fibrobalsts, and ${\beta}-actin$, actin-capping protein(${\beta}$ subunit), actin- related protein Arp3(ARP) and myosin class I(myh-1c) were more highly expressed in the gingival fibroblasts than PDL cells. Osteoprotegerin/osteoclastogenesis inhibitory factor(OPG/OCIF) was more highly expressed in the PDL cells than gingival fibroblasts. According to the results of this study, PDL cells and gingival fibroblasts were quite different gene expression patterns though they are the fibroblast which have similar shape. Therefore PDL cells & gingival fibroblasts are heterogeneous populations which represent distinct characteristics. If more studies about genes that were differently expressed in each PDL cells & gingival fibroblasts would be performed in the future, it would be expected that the characteristics of PDL cells would be more clear.

백지에서 추출한 oxypeucedanin hydrate의 미토콘드리아 기능 관련 근생성 효과 (Effects of oxypeucedanin hydrate isolated from Angelica dahurica on myoblast differentiation in association with mitochondrial function)

  • 본 연구는 근생성 효능을 가진 천연화합물을 발굴하고자 oxypeucedanin hydrate가 근생성과 미토콘드리아에 미치는 영향 및 항암제 유도 미토콘드리아 손상에 대한 완화효과를 C2C12 근원세포와 zebrafish 모델을 통해 각각 확인하였다. 그 결과, oxypeucedanin hydrate는 다핵의 근관세포의 수와 분화말기 표지자인 Myh4의 발현량을 증가시켰고 근육단백질 분해 인자인 MuRF1과 MAFbx의 발현량은 감소시켰다. 또한 미토콘드리아 생합성 조절인자인 Pgc1α, Tfam과 전자전달계 구성인자인 Sdha, Cox1의 발현은 증가시키고, 미토콘드리아 융합인자인 Opa1의 발현 또한 증가시킴과 동시에 미토콘드리아 분열을 표지하는 Drp1의 발현은 감소시켰다. 한편 zebrafish 모델을 통해 항암제 유도 미토콘드리아 손상에 대한 개선효과를 확인한 결과, oxypeucedanin hydrate는 항암제에 의해 유도된 미토콘드리아 손상을 완화시켰다. 이상의 결과들은 oxypeucedanin hydrate가 미토콘드리아 기능 증진을 매개로 근원세포 분화 촉진 및 근육 단백질 분해 저하 효과를 나타냄을 시사한다. 따라서 본 연구를 통해 oxypeucedanin hydrate가 근생성 효과를 나타낼 수 있는 잠재적인 유효소재로서의 가능성을 제시하였다.
