Effect of Acer tegmentosum M. Extracts on Hepatocarcinoma Cell (산겨릅나무 추출물의 간암세포의 증식억제 효과)

  • The objective of this study was to investigate the anticancer effects of Acer tegmentosum M. extracts. HepG2 hepatocarcinoma cells were treated with ethanol, chloroform, ethylacetate, butanol, aqueous fraction and hot water extract. The antiproliferative effect was evaluated by trypan blue exclusion, MTT-based viability assay and morphology. The trypan blue test showed that anticancer effect of the A. tegmentosum M. extracts on HepG2 cells increased gradually in proportion to the increasing concentration of the fractions. The butanol fraction showed the highest anticancer activity against HepG2 cells (p<0.05). The MTT assay indicated that the growth inhibition by the butanol fraction was dose-dependent. These results suggest that A. tegmentosum M. has the potential to inhibit the growth of hepatocarcinoma cells.

Anti-tumor effect of new compound, 127, through the induction of apoptosis (새로운 화합물 c-127의 세포고사 유도에 의한 항암효과)

  • Purpose : We screened more than 350 compounds with an endoperoxide ring structure in search of an anti-leukemic drug and found that compound 127 (c-127) could induce significant cytotoxicity in HL-60 cells. In this study, we investigated the molecular mechanisms of compound 127-induced antitumor activity on HL-60 cells. Methods : HL-60 cells were cultured in Rosewell Park Memorial Institute 1640 and cell viability was measured by MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide], a tetrazole assay. Apoptosis was assessed by a DNA fragmentation test. Apoptotic machineries were determined by Western blot analysis. Results : C-127 could induce a cytotoxic effect at 24 h and apoptosis at 6 h, which was demonstrated with MTT assay and DNA fragmentation test, respectively. The apoptotic effect of this drug was caused by the activation of the intracellular caspase-8,3 activation, the cleavage of pro-apoptotic Bid, and the increase of c-Jun expression accompanied with JNK (Jun N-terminal kinases) phosphorylation. On the contrary, it increased the expression of anti-apoptotic Bcl-2 levels, leading to the induction of the induction of anti-apoptotic effect. Taken together, the present study demonstrated that c-127 was a potent inducer of cytotoxicity on HL-60 cells through apoptotic mechanisms, which included the activation of caspase family, the regulation of Bcl-2 family, and the activation of JNK signaling pathway. Conclusion : Our results suggest that c-127 has a strong antitumor activity through the regulation of various apoptotic machineries on HL-60 cells. The compound may be utilized as an effective and potentially therapeutic drug in leukemia.

The Effects of Pulsatilla Koreana for Anti - Inflammatory and Cellular Activity of Periodontal Tissue (백두옹 추출물의 치주조직 세포에 활성도 및 항염 효과에 관한 연구)

  • This study was performed to define the cytotoxicity and the anti-inflammatory action of Pulsatilla koreana extracts. To analyze cytotoxic effects, gingival and periodontal ligament fibroblasts were used, and anti-inflammatory actions related to reduction of $IL-1{\beta}$ and $PGE_2$ production were performed in vitro, for the suggestion of efficacy and safety on periodontal therapeutic use of Pulsatilla koreana extracts. We extracted ethylacetate and butylalcohol from well-dried and ground Pulsatilla koreana throughout multiple processing, then used different concentration solution(0.1 %, 0.2 %, 0.4 %, 0.01 %, 0.02 %, 0.04 %, 1 %, 2 %) of ethylacetate and butylalcohol extracts to examine eytotoxic effects and anti-inflammatory actions Cytotoxic effects were examined by ELISA reader using MTT(Methyl Thiazol-2-YL-2, 5-diphenyl Tetrazolium bromide)solution following culture of human gingival and periodontal ligament fibroblasts. Synthesis of $IL-1{\beta}$was examined by $IL-1{\beta}$ enzyme-immunoassay(EIA)system after separation and culture of monocyte, and $PGE_2$ was examined by $PGE_2EIA$ system after culture of gingival fibroblasts. The results were as follows: 1. In the MTT test of gingival fibroblasts, the change of optical density was decreased significantly at 2 % of butylalcohol extracts and 0.04 %, 0.1 %, 0.2 %, 0.4 %, 1 %, 2 % of ethylacetate extracts.(p<0.05) 2. In the MTT test of periodontal ligament cells, the change of optical density were not differ significantly. but butylalcohol and ethylacetate extracts except from butylalcohol 0.01 % showed high cell cytotoxity. 3. Both ethylacetate and butylalcohol extracts from Pulsatilla koreana inhibited the synthesis of $IL-1{\beta}$and inhibition effect of ethylacetate extracts were higher than butylalcohol extracts. 4. Both ethylacetate and butylalcohol extracts from Pulsatilla koreana inhibited the synthesis of $PGE_2$, and ethylacetate extracts were higher than butylalcohol extracts. In conclusion, ethylacetate and butylalcohol extracts from Pulsatilla koreana showed little cell cytotoxity for gingival and periodontal ligament fibroblasts, and the inhibition of $IL-1{\beta}$ and $PGE_2$ sysnthesis, therefore it is considered that these extracts can be developed as the therapeutics of the periodontal disease.

Development of the Functional Beverage Containing the Prunus mume Extracts (매실 추출물을 함유한 기능성 음료 개발)

  • This study was performed to investigate the cytotoxic effect of the Prunus mume extracts containing beverages on the growth of SNU-16 gastric cancer cell, SNU-C2A colon cancer cell, SNU-449 liver cancer cell and HeLa cervical cancer cell. The inhibitory effect on the growth of the cancer cell lines was examined by MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay, cytotoxicity test and microscopy. Also this study aimed to compare the changes of blood electrolytes and osmolarity during submaximal exercise for the intake of the Prunus mume extracts containing beverages. 20% Prunus mume extracts containing beverage exhibited the greatest inhibitory effect on the growth of SNU-16 and significantly inhibited at the concentration of $1000\;{\mu}g/mL$ in the MTT assay. Morphological changes in SNU-16 which treated with the same beverage were observed under inverted microscope. The change of blood electrolytes and osmolarity during submaximal exercise showed no significant differences between before and after intake of the beverage in both groups.

Screening Test of Selaginella tarmariscina for Liver Protective Effects in Primary Cultured Hepatocytes (일차 배양한 간세포에서의 권백의 간보호효과의 Screening Test)

  • Selaginellae Herba has been used as folk medicine for antineoplastics, coagulants, antidotes and invigorants. To find an in vitro screening method for liver protective effect of Selaginellae Herba in benzo(a)pyrene intoxicated injury were examined in primary cultured rat hepatocytes. Using MTT assay, herba concentration showed dose dependently viability. The lowest concentration of benzo(a)pyrene giving cytotoxicity revealed around 50gM. The hepatoprotective effect of Selaginellae Herba in both water and chloroform extracts was also increased dose dependently.

Functional Cosmetic Effect of Porcine Placeta (Porcine Placenta의 기능성 화장품소재 특성)

  • Porcine placenta was treated with alkali, acid and enzyme treatment to obtain extracts. Heavy metal contents such as Pb, As, and Hg were low enough to satisfy cosmetic agent standard. As a result of safety test(MTT assay), porcine placenta extracts showed over 80% of cell viability at $50{\mu}g/ml$, and cell toxicity was relatively lower. From antioxidation test using DPPH free radical scavenging assay, antioxidation effect was highest as 63% at $50{\mu}g/ml$ when porcine placenta was treated with alkali in pH 9. From whitening effect test using tyrosinase inhibition assay, tyrosinase inhibition effect was 30% at $50{\mu}g/ml$ concentration in alkali treated procine placenta, however, the efficiency was lower compared with arbutin or vitamin C. In anti-wrinkle effect test from elastase inhibition assay, elastase inhibition effects were 20~30% at $50{\mu}g/ml$ for 5 kinds of porcine placenta treatments, which was superior to standard, and especially, protease treated extracts showed best results. Skin formulation including 1% porcine placenta was made and the formulation was very stable for temperature and storage period. From this research, porcine placenta extract showed high potential for anti-wrinkle functional cosmetic agent.

Efficacy and Safety of Micro Current Tape on Chronic Low Back Pain: One Group Pre-Post Test Design, Multi Center Pilot Study (만성 요통 통증 완화에 대한 미세전류 테이프의 안전성 및 유효성 평가)

  • Objectives The aim of this study was to evaluate the efficacy and safety of micro current taping therapy (MTT) on patients with chronic low back pain (CLBP). Methods We included 50 participants who met the inclusion criteria and 5 participants dropped out during the sessions due to the following reasons: 3 participants were personal reasons, 1 participant was taking medication, 1 participant was fore arm fracture. We attached "I" shaped 40 cm, 2 tapes along the erector muscles of the spine starting from both iliac crest and another "I" shaped 30 cm, 1 tape on the painful site horizontally. This procedure was done 8 times and participants visited a total of 9 times including a final visit for evaluation. We measured visual analog scale (VAS), range of motion (ROM) and schober's test on every visit. Participants completed a questionnaire of oswestry disability index (ODI) and Beck's depression inventory (BDI) on the first and last visits. Results In VAS for pain intensity and bothersomeness, there were significant decreases after 1st, 6th treatments. In range of extension, there was significant increase after 3rd treatment at first. In range of flexion, there was significant increase after 2nd treatment at first. In range of left flexion, there was significant increase after 4th treatment at first. In range of right flexion, there was significant increase after 3rd treatment at first. in schober's test, there was significant increase after 2nd treatment at first. In VAS, ROM, schober's test, BDI, ODI, there were significant improvement after all treatments had done. Conclusions There was a significant effect of MTT on CLBP. And there was no adverse effect.

An Anti-Tumor Effects of Dioscorea bulbifera L. on Stomach Cancer Cells (황약자(黃藥子)의 위암세포(胃癌細胞)에 대(對)한 항암(抗癌) 효능(效能) 검색(檢索))

  • Background : Nowadays a lot of research is based on natural substances or materials world wide since many kinds of side effects are accompanied by anti tumor chemotherapy. In Chinese medicine, Dioscorea bulbifera L is widely used to treat many kinds of cancer, but in Korea it is rarely used. Therefore, we need to scientifically identify anti tumor effects of Dioscorea bulbifera L. Objective : We aimed to identify anti tumor effects of Dioscorea bulbifera L on the stomach cancer cells through molecular biological methods. Materials & Methods : We used AGS, a stomach cancer cell from American Type Culture Collection. We injected the boiled extract of Dioscorea bulbifera L 5 ul(sample 1), 10ul(sample 2) to cultured media(ml) for 0,6,12,18,24 hours. We measured the killing effect on stomach cancer cells through Tryphan blue exclusion test and suppressive effect on viability of stomach cancer cells via MTT assay. Results : Tryphan blue exclusion test showed that each test group killed more stomach cancer cells than the controlled group with a dosage-dependent, but not significantly. MTT assay showed that each test group had a more suppressive effect on viability of stomach cancer cells than the controlled group without a dosage-independent, but not significantly. The cell cycle analysis via flow cytometry showed that the test group extended cell cycle, and there was no peak in M phase, the number of sub G1, G0, G1 phase cells increased a little, but not significantly. Conclusion : This experiment showed that Dioscorea bulbifera L. has an anti-tumor effect, but not significantly. This is in vitro experiment and basic experiment on Dioscorea bulbifera L. We hope more progressive researches on Dioscorea bulbifera L. will be conducted and its anti tumor will be more accurately identified.

산삼의 배양 및 그 응용에 관한 연구

  • Korea mountain ginseng known as oriental miracle drug is an important medicinal plant. The effect of mountain ginseng adventitious roots extract has been described. The valuable root of mountain ginseng contained several kinds of ginsenosides that have been confirmed to have many active functions for the human body. However, the study of mountain ginseng has a limit because the price of wild ginseng is very expensive and rare. The mountain ginseng adventitious roots were derived from mountain ginseng callus that were induced from mountain ginseng roots. Adventitious roots were separated from callus and grown in solid media(Murachige and stoog media). It was cultured in a 20L bioreactor. After culturing for 40days, adventitious roots were harvested. Afterwards the harvested mountain ginseng adventitious roots were dryed and extracted. We examined the effect on melanogenesis of mountain ginseng adventitious roots extract. Here, we report the inhibitory effect of melanin biosynthesis on the adventitious roots extract of In vitro test. Also, we assessed the safety of adventitious roots extract. In vitro, cytotoxicity of adventitious roots extract was assessed in mouse fibroblast using two method: The neutral red uptake assay and the MTT assay. In vivo, the allergic and irritant were Patch teated in 30 patients. Consequently, extract of mountain ginseng adventitious roots have inhibitory effect on melanin biosynthesis in B-16 melanoma cell test, tyrosinase inhibitory test and DOPA auto-oxidation test. There were decreased 86%(0.5% concentration), 45%(1% concentration) and 61%(1% concentration), respectively.

A study of mesenchymal stem cell proliferation and surface characteristics of the titanium discs coated with MS275/PLGA by an electrospray (Electrospray법을 이용한 MS275/PLGA submicron 코팅 티타늄에서의 표면변화와 간엽줄기세포증식에 관한 연구)

  • Purpose: This study was conducted to identify the surface characteristics of titanium discs coated with MS275/PLGA by electrospray and which is effective to mesenchymal stem cell proliferation. Materials and methods: We used anodized surface coated with PLGA as a control group and anodized surface coated with MS275 $0.5{\mu}M$, $1{\mu}M$, $1.5{\mu}M$ as test groups. To examine that the coating particles are nanometer sized, FE-SEM was used and AFM was utilized to determine the difference of coating surface roughness. We checked the mesenchymal stem cell proliferation by using MTT assay on $1^{st}$, $4^{th}$, $7^{th}$ days. Results: There was no significant difference between control groups and test groups in AFM results (P>.05). In MTT assay results, mesenchymal stem cell proliferation was increased with time, at $7^{th}$ day, cell viability on discs coated with $1.5{\mu}M$ MS275 was significantly higher than control group (P<.05). As SEM showed, the number of cells on all discs was increased and the morphology of cell attachment was also wider and closer with time. Conclusion: Titanium surface coated with MS275/PLGA showed significantly higher cell proliferation and the more density of MS275 was dispersed on titanium discs, the faster cells grew.
