Antiproliferative Activity of Marrubium persicum Extract in the MCF-7 Human Breast Cancer Cell Line

  • Aim: Developing antitumor drugs from natural products is receiving increasing interest worldwide due to limitations and side effects of therapy strategies for the second leading cause of disease related mortality, cancer. Methods: The antiproliferative activity of a methanolic extract from the aerial parts of Marrubium persicum extract was assessed with the MCF-7 breast cancer cell line using the MTT test for cell viability and cytotoxicity indices. In addition, antioxidant properties of the extract were evaluated by measuring its ability to scavenge free DPPH radicals. Moreover, the total phenolic and flavonoid content of the extract was determined based on Folin-Ciocalteu and colorimetric aluminum chloride methods. Results: The findings of the study for the antiproliferative activity of the methanolic extract of M. persicum showed that growth of MCF-7 cells was inhibited by the extract in a dose and time dependent manner, where a gradual increase of cytotoxicity effect has been achieved setting out on 200 ${\mu}g/mL$ concentration of the plant extract. The antioxidant assay revealed that the extract was a strong scavenger of DPPH radicals with an $RC_{50}$ value of 52 ${\mu}g/mL$. The total phenolic and flavonoids content of the plant extract was 409.3 mg gallic acid equivalent and 168.9 mg quercetin equivalent per 100g of dry plant material. Conclusion: Overall, M. persicum possesses potential antiproliferative and antioxidant activities on the malignant MCF-7 cell line that could be attributed to the high content of phenolics and flavonoids, and therefore warrants further exploration.

  • This laboratory has recently reported the synthesis and in vitro antitumor activity of PT(II) complexes containing ethylenediamine and diphosphine. In view of the reports of others, cisplatin is toxic to the kidney since the kidney's vulnerability to PT(II) complexes may originate in its ability to accumulate and retain platinum to a greater degree than other organs. The in vitro cytotoxicity of these synthetic PT(II) complexes on the primary cultured proximal tubular cells of rabbit kidney and renal cortical cells of human kidney was investigated. Three endpoints for cytotoxicity tests were evaluated:3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), $^3H$-thymidine uptake and the glucose consumption tests. The rank order of sensitivity exhibited $^3H$-thymidine uptake>MTT>glucose consumption test. The agents with diphosphine leaving group were significantly less cytotoxic than cisplatin. Moreover, 1,2-bis(diphenylphosphino)ethane (DPPE) exhibited less cytotoxicity than 1.3-bis (diphenylphosphino)propane (DPPP) against on rabbit and human cultured kidney cells. Based on these results, the decreased nephrotoxicity of these new complexes over cisplatin appeared to be partially attributable to a leaving group of DPPP and DPPE. This novel class of platinum compound represents a valuable lead in the development of a "third-generation" agent.

섬유성이형성증 유래세포의 특성연구 (CHARACTERISTICS OF FIBROUS DYSPLASIA DERIVED CELLS)

  • Purpose: Fibrous dysplasia (FD) is a fibro-osseous disease associated with activating missense mutations of the gene encoding the $\alpha$-subunit of stimulatory G protein. FD may affect a single bone (called monostotic form) or multiple bones (called polyostotic form). The extent of lesions reflects the onset time of mutation. In this study, cells from monostotic FD in maxilla of a patient were isolated and cultured in vitro for characterization. Materials and Methods: The single cells were released from FD lesion which was surgical specimen from 15 years-old boy. These isolated cells were cultured in vitro and tested their proliferation activity with MTT assay. In osteogenic media, these cells underwent differentiation process comparing with its normal counterpart i.e. bone marrow stromal cells. The proliferated FD cells were detached and transplanted into the dordsal pocket of nude mouse and harvested in 6 weeks and 12 weeks. Results and Summary: FD cells have an increased proliferation rate and poor differentiation. As a result, cells isolated from FD lesion decreased differentiation into osteoblast and increased proliferation capacity. MTT assay presented that proliferation rate of FD cells were higher than control. However, the mineral induction capacity of FD was lesser than that of control. Monostotic FD cells make fewer amounts of bone ossicles and most of them are woven bone rather than lamellar bone in vivo transplantation. In transplanted FD cells, hematopoietic marrow were not seen in the marrow space and filled with the organized fibrous tissue. Therefore, they were recapitulated to the original histological features of FD lesion. Collectively, these results indicated that the FD cells were shown that the increased proliferation and decreased differentiation potential. These in vitro and in vivo system can be useful to test FD cell's fate and possible.

Amylocolatosis sp.가 생산하는 항암물질의 정제 및 구강암 모델에 미치는 항암 효과 (Purification of Materials Produced by Amylocolatosis sp. and Anticancer Effect in Oral Cancer Model)

  • 각종 토양시료로부터 M배지와 AMP배지에서 생육하는 500여종의 방선균을 분리하였으며, 그 중 전배양배지(GMY 배지)에서 가장 뛰어난 항암활성능을 소유한 균주를 불리하고 배양하여 세포외 항암성물질을 분리하여 이 물질을 완전정제하고 MTT 정량분석을 실시하여 암세포에 대한 세포독성검사를 실시하였다. 1. 정제는 배양한 균체를 완전히 제거하고 동량의 ethylacetate를 처리하여 배양액중의 항암성분을 추출하고 무수 magnesium sulfate로 건조 후 농축, ethanol로 용해, 10배량의 water을 첨가하여 $4^{\circ}C$에서 overnight 시킨 후 추출액을 원심분리(12,000 rpm, 30분)하여 methylene chloride로 용해시켜 silica gel 60 column(${\phi}35{\times}600mm$, Merck Co.), methylene chloride-ethanol(96:4) 용매로 용출하고 sephadex LH-20 column(${\phi}15{\times}300mm$, Pharmacia LKB)에서 100% methanol로 용출하여 HPLC(Waters, ODS)를 이용하여 최종적으로 완전 정제하였다. 2. Compound는 Gram(+) 세균(6균주), Gram(-) 세균(11균주), 효모(2균주), 곰팡이(1균주)에 대하여 항균 효과를 나타내었다. 3. 완전 정제된 물질에 대한 항암효과를 측정한 결과, 모든 실험 암세포에 대해서 뛰어난 항암효과를 나타내었으며 protein성 물질은 아닌 것으로 추정되었다.

연근 추출물에서 주름개선 소재의 개발 (Development of Anti-Wrinkle Agent from Nelumbo nucifera Root Extract)

  • 본 연구에서는 연근의 추출물에서 기능성화장품 소재를 개발하고자 하였다. 용매는 70~100%의 에탄올을 이용하였으며, 연근의 유효성분으로 nuciferine을 확인하였다. 연근 추출물의 화장품소재로서의 응용가능성을 보기 위해 안전성시험(MTT assay), 주름개선 효과시험(Elastase 활성저해 시험) 및 항산화효과시험(DPPH free radical scavenging assay)을 실시하였다. 100% 에탄올로 연근을 추출할 경우 $100{\mu}g/ml$의 농도에서 세포생존율이 80% 이상이어서 연근 추출물은 화장품소재로서 비교적 안전함을 알 수 있었다. 또한 연근 추출물은 $100{\mu}g/ml$의 농도에서 elastase 활성억제가 40~50%를 보여주어서 비교적 우수한 주름개선 효과를 나타내었다. 50% 항산화 저해능을 보이는 농도($FSC_{50}$)는 $5.0{\sim}38{\mu}g/ml$로 연근 추출물의 항산화효과가 매우 우수하였다. 따라서 연근 에탄올추출물은 주름개선 기능성화장품 소재로서 그 가능성이 높음을 알 수 있었다.

Cell attachment and proliferation of bone marrow-derived osteoblast on zirconia of various surface treatment

  • PURPOSE. This study was performed to characterize the effects of zirconia coated with calcium phosphate and hydroxyapatite compared to smooth zirconia after bone marrow-derived osteoblast culture. MATERIALS AND METHODS. Bone marrow-derived osteoblasts were cultured on (1) smooth zirconia, (2) zirconia coated with calcium phosphate (CaP), and (3) zirconia coated with hydroxyapatite (HA). The tetrazolium-based colorimetric assay (MTT test) was used for cell proliferation evaluation. Scanning electron microscopy (SEM) and alkaline phosphatase (ALP) activity was measured to evaluate the cellular morphology and differentiation rate. X-ray photoelectron spectroscopy (XPS) was employed for the analysis of surface chemistry. The genetic expression of the osteoblasts and dissolution behavior of the coatings were observed. Assessment of the significance level of the differences between the groups was done with analysis of variance (ANOVA). RESULTS. From the MTT assay, no significant difference between smooth and surface coated zirconia was found (P>.05). From the SEM image, cells on all three groups of discs were sporadically triangular or spread out in shape with formation of filopodia. From the ALP activity assay, the optical density of osteoblasts on smooth zirconia discs was higher than that on surface treated zirconia discs (P>.05). Most of the genes related to cell adhesion showed similar expression level between smooth and surface treated zirconia. The dissolution rate was higher with CaP than HA coating. CONCLUSION. The attachment and growth behavior of bone-marrow-derived osteoblasts cultured on smooth surface coated zirconia showed comparable results. However, the HA coating showed more time-dependent stability compared to the CaP coating.

박테리오파지의 살균소독제 응용을 위한 안전성 평가 (Safety evaluation of bacteriophages for application as sanitizers)

  • 본 연구에서는 6종의 E. coli 및 S. aureus 유래 박테리오파지 용액에 존재하는 내독소 함량 조사와 파지의 세포독성 여부에 대해 평가하였다. 대표적인 생물학적 내독소 시험법인 LAL assay을 시행하여 9-10 log PFU/mL 농도의 내독소 함량을 확인한 결과, 파지의 임상적용에는 부적합한 수치이나 식품에 존재하는 병원균의 살균 목적의 사용에는 매우 유해하지 않은 수치임을 확인할 수 있었다. 박테리오파지 용액의 세포독성평가를 확인하기 위해 MTT 분석을 시행하여 세포 생존율을 확인하였다. E. coli 파지 용액과 S. aureus 파지 용액 처리군 모두에서 98% 이상의 생존율이 관찰되어 파지용액에 존재하는 내독소 및 파지가 세포독성을 유발하지 않는다는 것을 확인하였다. 그러므로 일반적인 방법으로 증폭, 농축한 파지 용액은 인체에 대한 유해성이 적으며 식품에 살균소독제로 적용하더라도 문제가 없을 것으로 사료된다.

메탄올로 추출한 여주 분획성분의 항균 및 항발암 효과 (The Effects on Antimicrobial and Anticarcinogenic Activity of Momordica Charantia L.)

  • 중국에서 약용 및 음용식물로 사용되고 있는 여주 (Mo-mordica charantia L.)를 이용하여 추출, 분획한 후 항균, 항암 및 암예방 효과를 살펴보았다 여주의 MCU층을 제외한 모든 층에서 항균력을 보였으며 특히 Staphylococru aureus의 MCMEE층에서 가장 높은 항균활성을 나타내었다 실험에 사용한 각균에 대한 여주 분획물의 암세포 증식 억제 효과를 MTT assay로 실험한 결과, 3종의 인체 암세포 HepG2, HeLa 및 MCF극에서 모두 여주의 ethyleth-er 분획층인 MCMEE와 ethylacetate 분획층인 MCMEA 층에서 아주 높은 암세포 증식억제 효과를 나타내었다. HepG2세포를 이용하여 암예방 QR 유도 활성을 측정한 결과, 다른 분회층에 비해 비극성 용매층인 hexane 분획층 MCMH에서 아주 높은 QR 유도를 활성시키는 것으로 나타났다. 본 실험 결과, 여주에는 식품 보존과 항균제로서의 개발가능성이 보이며. 항발암 효과를 보기위한 암세포 증식 억제 효과는 ethylether층 MCMEE과 ethylacetate층 MCMEA층에서 월등하였고, 비극성 용매층인 hexane 분획층에는 암발생을 예방하는 quinone reductase inducer가 있는 것으로 사료되며. 나아가 단계적인 생리활성 연구의 분리 동정이 계속 이루어져야 할 것으로 사료된다.

Cytocompatibility and cell proliferation evaluation of calcium phosphate-based root canal sealers

  • Objectives: This study aimed to evaluate the cell viability and migration of Endosequence Bioceramic Root Canal Sealer (BC Sealer) compared to MTA Fillapex and AH Plus. Materials and Methods: BC Sealer, MTA Fillapex, and AH Plus were placed in contact with culture medium to obtain sealers extracts in dilution 1:1, 1:2 and 1:4. 3T3 cells were plated and exposed to the extracts. Cell viability and migration were assessed by 3-(4,5-dimethylthiazoyl)-2,5-diphenyl-tetrazolium bromide (MTT) and Scratch assay, respectively. Data were analyzed by Kruskal-Wallis and Dunn's test (p < 0.05). Results: The MTT assay revealed greater cytotoxicity for AH Plus and MTA Fillapex at 1:1 dilution when compared to control (p < 0.05). At 1:2 and 1:4 dilutions, all sealers were similar to control (p > 0.05) and MTA Fillapex was more cytotoxic than BC Sealer (p < 0.05). Scratch assay demonstrated the continuous closure of the wound according to time. At 30 hours, the control group presented closure of the wound (p < 0.05). At 36 hours, only BC Sealer presented the closure when compared to AH Plus and MTA Fillapex (p < 0.05). At 42 hours, AH Plus and MTA Fillapex showed a wound healing (p > 0.05). Conclusions: All tested sealers demonstrated cell viability highlighting BC Sealer, which showed increased cell migration capacity suggesting that this sealer may achieve better tissue repair when compared to other tested sealers.

Chemical Composition and Biological Activities of Essential Oils Extracted from Korean Endemic Citrus Species

  • The aim of this study was to analyze the chemical composition of 14 kinds of citrus oils and to test their biological activities. Citrus essential oils were obtained by steam distillation from immature fruits collected from Jeju Island and were analyzed using gas chromatograph (GC)-flame ionization detectors (FID) and GC-MS. Limonene (55.4% to 91.7%), myrcene (2.1% to 32.1%), ${\alpha}$-pinene (0.6% to 1.6%) and linalool (0.4% to 6.9%) were the major components in most citrus species. To evaluate in vitro antibacterial activity, all essential oils were tested against Propionibacterium acnes and Staphylococcus epidermidis. Nine out of fourteen citrus oils exhibited antibacterial activity against P. acnes, but not against S. epidermidis. The effects of the citrus oils on DPPH radical scavenging, superoxide radical anion scavenging, nitric oxide radical, and cytotoxicity were also assessed. Three essential citrus oils, Joadeung, Dongjunggyul, and Bujiwha, exhibited potent inhibitory effects on nitric oxide production. Two essential oils, Dongjunggyul and Joadeung, showed potent free radical scavenging activities in the DPPH assay. For future applications in cosmetic products, we also performed MTT assays in a human dermal fibroblast cell line. The majority of the essential oils showed no cytotoxicity. The results indicate that citrus essential oils can be useful natural agents for cosmetic application.
