Effect of Faba Bean Isolate and Microbial Transglutaminase on Rheological Properties of Pork Myofibrillar Protein Gel and Physicochemical and Textural Properties of Reduced-Salt, Low-Fat Pork Model Sausages

  • The study was performed to determine the effect of faba bean protein isolate (FBPI) alone or in combination with microbial transglutaminase (MTG) on the rheological properties of pork myofibrillar protein gel (MPG), and physiochemical and textural properties of reduced-salt, low-fat pork model sausages (LFMSs). The cooking yields of MPGs with MTG or FBPI alone decreased and increased, respectively. However, the combination of FBPI and MTG was similar to the control (CTL) without FBPI or MTG. Gel strength values of MPG added with both FBPI and MTG were higher than treatments with FBPI or MTG alone. The hydrophobicity values of CTL were lower than those of MPG with FBPI alone, whereas the addition of MTG decreased the hydrophobicity of MPGs. The incorporation of FBPI alone or in combination with MTG decreased sulfhydryl groups (p<0.05). Shear stress values of MPGs with MTG tended to be higher than those of non-MTG treatments at all shear rates, and the addition of FBPI into MPGs increased shear stress values. Reduced-salt (1.0%) LFMSs with FBPI alone or combined with MTG had both lower cooking loss and expressible moisture values than those of CTL and similar values to the reference sample (REF, 1.5% salt). Textural properties of reduced-salt LFMSs with FBPI or MTG were similar to those of REF. These results demonstrated that the combination of FBPI and MTG could improve the water binding capacity and textural properties of pork MPGs and LFMSs and might be suitable for application in the development of healthier meat products.

Production of Microbial-Transglutaminase [MTG] from Streptoverticillium mobaraense

  • Mineral salts in medium usually profoundly influence microorganism growth and protein synthesis. In order to produce microbial transglutaminase (MTG) with a high yield from Streptoverticillium mobaraense, we screened the minerals $CaCl_2,\;CoCl_2,\;FeSO_4,\;ZnSO_4,\;MnSO_4\;and\;CuSO_4$ for MTG fermentation. The results indicated that appropriate $FeSO_4$ concentrations could significantly promote cell growth and stimulate the production of MTG. With 15 mg/L of $FeSO_4$ added to medium, 58% improvements were noted in MTG productivity (2.24 U/mL). NaCl, $CaCl_2,\;and\;CoCl_2$ enhanced MTG productivity by less than 15%, and the optimal concentrations were determined as 1 g/L, 2 g/L, and 30 mg/L respectively. Furthermore, it was determined that 7.5 mg/L of $ZnSO_4$ in medium could augment MTG productivity by 20% and induce the stationary phase for MTG production to a period 24 hr earlier. This basic and novel discovery should result in the development of a good complement to the previously defined culture media for MTG fermentation.

Heterologous Expression of Recombinant Transglutaminase in Bacillus subtilis SCK6 with Optimized Signal Peptide and Codon, and Its Impact on Gelatin Properties

  • Microbial transglutaminases (MTGs) are widely used in the food industry. In this study, the MTG gene of Streptomyces sp. TYQ1024 was cloned and expressed in a food-grade bacterial strain, Bacillus subtilis SCK6. Extracellular activity of the MTG after codon and signal peptide (SP Ync M) optimization was 20 times that of the pre-optimized enzyme. After purification, the molecular weight of the MTG was 38 kDa and the specific activity was 63.75 U/mg. The optimal temperature and pH for the recombinant MTG activity were 50℃ and 8.0, respectively. MTG activity increased 1.42-fold in the presence of β-ME and 1.6-fold in the presence of DTT. Moreover, 18% sodium chloride still resulted in 83% enzyme activity, which showed good salt tolerance. Cross-linking gelatin with the MTG increased the strength of gelatin 1.67 times and increased the thermal denaturation temperature from 61.8 to 75.8℃. The MTG also significantly increased the strength and thermal stability of gelatin. These characteristics demonstrated the huge commercial potential of MTG, such as for applications in salted protein foods.

Liver PET/MRI 검사 시 MR 기반 호흡 움직임 보정 방법의 유용성 평가 (Evaluation of MR Based Respiratory Motion Correction Technique in Liver PET/MRI Study)

  • PET/MRI 검사 시 호흡에 의한 움직임은 영상의 질 저하는 물론 종양의 크기, 표준섭취계수의 오차를 발생시키는 원인이 된다. 본 연구에서는 일체형 PET/MRI 장비에서 MR기반 호흡 움직임 보정 방법의 적용에 따른 종양의 표준섭취계수, 크기와 영상 품질 변화를 평가하고자 하였다. Biograph mMR 3.0T (Siemens, Germany)장비에서 2016년 3월부터 7월까지 $^{18}F-FDG$ liver PET/MRI 검사를 시행한 30명 ($62.5{\pm}10.2$세)의 데이터를 분석하였다. 7분의 PET listmode 데이터를 획득하는 동안 MR 기반 호흡 움직임 보정 방법인 MAG, MTG와 NG T1 weighted MR 영상을 획득하였다. Gated PET 영상의 재구성은 35% efficiency window가 적용된 MAG와 MTG로부터 획득된 감쇄보정영상을 이용하여 시행하였다. Non-gate, MAG, MTG 영상에서 측정된 종양의 표준섭취계수와 Z축 방향의 크기 그리고 반치폭을 분석하였다 평균 $SUV_{max}$$SUV_{peak}$는 NG 대비 MAG 13.15%(P<0.0001), 8.66%(P<0.0001), MTG 13.27%(P<0.0001), 8.80%(P<0.0001) 증가하였으며 Z-축에서 평균 종양의 크기와 반치폭은 MAG 14.47%(P<0.0001), 15.49%(P=0.0004), MTG 14.89%(P<0.0001), 15.79%(P=0.0003) 감소하였으며 통계적으로 유의한 차이를 보였다. MAG와 MTG 비교 평가에서 MTG의 $SUV_{max}$$SUV_{peak}$는 MAG 대비 0.07%(P=0.8802), 0.13%(P=0.7766) 증가하였으며 Z-축에서 평균 종양의 크기와 반치폭은 0.49%(P=0.2786), 0.36%(P=0.2488) 감소하였다. 약 7분과 2분의 추가 검사시간이 필요한 MAG와 MTG에서 표준섭취계수와 종양의 크기, 반치폭에서 통계적으로 유의한 차이가 없었다. 간 PET/MRI 검사 시 MR 기반 호흡 움직임 보정 방법을 적용하였을 때 NG 대비 MAG, MTG 모두에서 표준섭취계수와 종양의 크기 및 공간분해능이 개선되었으며 MAG와 MTG의 결과 값은 통계적으로 유의한 차이가 없었다. 호흡에 의한 움직임에 영향을 받는 다양한 상 복부 검사에 MBRMCT를 적용 시 추가적인 장비의 설치 없이 약 2분의 추가 검사시간이 필요한 MTG 방법 적용하여 NG 대비 보다 정확한 정보를 제공할 수 있을 것으로 사료된다.

$2{\mu$}$ CMOS 공정을 이용한 BLC, MTG 가산기의 전기적 특성 (Electrical Characteristics of BLC, MTG Adders Using $2{\mu}m$ CMOS Process)

  • In this paper, BLC adder/subtractor and MTG adder which can be used as a fundamental operation block in VLSI processors are designed, and their structural and electrical characteristics are analyzed and compared. Also, two circuits are fabricated usign 2\ulcorner CMOS process and their time delays for critical paths are measured. For 8 bit binary addition, the measured critical delays for MSB sum of the BLC adder/subtractor are 26 nsec for rising delay and 32nsec for falling. Those for MSB carry out of the MTG adder are 28nsed and 38nsec, respectively. The BLC adder/subtractor has a layout area which is 4 times larger than the MTG adder, and a fast operation speed. On the contrary, the MTG adder has a small layout area and a large time delay.

마이크로 스케일 난류에 의한 화염안정성 및 NOx 생성 (Flame Stability and NOx Formation by Micro scale Turbulence)

  • The effect of micro scale turbulence on flame structure and stability were experimentally investigated by changing the area of micro turbulence generator(MTG) and air velocity in terms of low NOx and high efficiency combustion. NOx and CO concentration were also measured for different MTG areas to investigate whether a vane swirler having MTG has a possibility of using as part for low NOx combustor. From the obtained results, it is shown that flame stability region increases and flame size becomes small as MTG area increases since MTG in itself makes small scale recirculation flow and swirler does large scale recirculation one. It is also shown that low NOx concentration(about 20${\sim}$30ppm@$O_2$ 11%) is achieved for all MTG areas without any increase in CO concentration regardless of air velocity range tested in this study when the equivalence ratio is 0.7. The results obtained in this study can give basic guideline for the design of compact low NOx high efficiency combustor using a vane swirler having MTG.

Inconsistency in the Improvements of Gel Strength in Chicken and Pork Sausages Induced by Microbial Transglutaminase

  • This research investigated variation in the improvement of the texture of chicken and pork sausages induced by microbial transglutaminase (MTG). The extractability of myofibrillar proteins from these sausages as well as the ${\varepsilon}-({\gamma}-glutamyl)$lysine (G-L) content were also investigated. MTG treatment of sausages significantly increased the breaking strength values in both meat types, especially for samples incubated at $40^{\circ}C$. However, values of the breaking strength in both meat types were increased differently. The variation in protein extractability of samples incubated at $40^{\circ}C$ for both meat types could lead to some consideration of the mechanisms and the high accessions of myosin heavy chain (MHC) to MTG. SDS-PAGE analysis showed significant changes in the density of the bands after adding MTG, especially for the pork samples in which the bands disappeared totally. The G-L content in the presence of MTG was double that in control samples of both meat types. This study suggests that the binding ability of myofibrillar proteins with MTG is strong. This leads us to suggest that MTG functions positively with different improvements in the texture of chicken and pork products that are treated mechanically, such as sausages. Variability in gel improvement level between chicken and pork sausages was observed; this resulted from the variation in meat proteins in response to MTG, as well as to the original glutamyl and lysine content.

Monothiolglycerol이 동결 융해 후 미니돼지 정자의 활성산소 억제, 미토콘드리아 활성 그리고 DNA Integrity에 미치는 영향 (Effect of Monothioglycerol on ROS Inhibition, Mitochondrial Activity, and DNA Integrity in Frozen-thawed Miniature Pig Sperm)

  • Cryopreservation and in vitro fertilization (IVF) protocols are important in genetic studies and applications to transgenic animals. Various studies about boar sperm cryopreservation have been studied for a long time. Those were about the use of extenders, the choice of sugars, the cooling and warming rates. The factors that influence the boar sperm are the dramatic changes in temperatures, osmotic and toxic stresses, and reactive oxygen species (ROS) generation. Among these factors, ROS generation is the main damage to DNA which is a principal genetic material and the most important for the practical applications. So we wondered whether ROS generation could be reduced. In previous study, monothioglycerol (MTG) was essential for the culture of embryo stem cells. Therefore we added MTG in the freezing extender based on lactose-egg yolk (LEY) with trehalose. For the assessment of the frozen-thawed sperm, we focused onmotility, membrane integrity and DNA damage. First, we used a computer-aided sperm analysis system for overall conditions of sperm such as motility and viability. Then we performed the sperm chromatin structure assay for DNA integrity and hypo-osmotic swelling test for membrane integrity. And our result showed the existence of MTG in the freezing extender caused less damage to DNA and higher motility in frozen-thawed boar sperm. Also we checked a relative antioxidant activity of MTG in modified Modena B extender. We concluded that this reagent can activate sperm mitochondria at MTG $0.2{\mu}M$, contribute to sperm motility and DNA integrity but there was no significant difference on membrane integrity. Also antioxidant activity of MTG in modified Modena B extender was proved.

Evaluation of Acid-treated Fish Sarcoplasmic Proteins on Physicochemical and Rheological Characteristics of Pork Myofibrillar Protein Gel Mediated by Microbial Transglutaminase

  • Fish sarcoplasmic protein (SP) is currently dumped as waste from surimi industry and its recovery by practical method for being the non-meat ingredient in meat industry would be a strategy to utilize effectively the fish resource. This study was aimed to apply pH treatment for fish SP recovery and evaluated its effect on pork myofibrillar protein (MP) gel. The pH values of fish SP were changed to 3 and 12, and neutralized to pH 7 before lyophilizing the precipitated protein after centrifugation. Acid-treated fish SP (AFSP) showed about 4-fold higher recovery yield than that of alkaline-treated SP and water absorption capacity was also about 1.2-fold greater. Because of the high recovery yield and water absorption capacity, AFSP was selected to incorporate into MP with/without microbial transglutaminase (MTG). The effects of AFSP and MTG on the physicochemical and rheological characteristics of MP and MP gel were evaluated. MTG induced an increase shear stress of the MP mixture and increase the breaking force of MP gels. MP gel lightness was decreased by adding AFSP. MP gel with MTG showed higher cooking loss than that without MTG. A reduction of cooking loss was observed when the AFSP was added along with MTG, where the insoluble particles were found. Therefore, AFSP could be contributed as a water holding agent in meat protein gel.

Effects of Multiple-target Anti-microRNA Antisense Oligodeoxyribonucleotides on Proliferation and Migration of Gastric Cancer Cells

  • Backgrounds: To investigate the inhibiting effects of multi-target anti-microRNA antisense oligonucleotide (MTg-AMOs) on proliferation and migration of human gastric cancer cells. Methods: Single anti-microRNA antisense oligonucleotides (AMOs) and MTg-AMOs for miR-221, 21, and 106a were designed and transfected into SGC7901, a gastric cancer cell line, to target the activity of these miRNAs. Their expression was analyzed using stem-loop RT-PCR and effects of MTg-AMOs on human gastric cancer cells were determined using the following two assay methods: CCK8 for cell proliferation and transwells for migration. Results: In the CCK-8 cell proliferation assay, $0.6{\mu}mol/L$ was selected as the preferred concentration of MTg-AMOs and incubation time was 72 hours. Under these experimental conditions, MTg-AMOs demonstrated better suppression of the expression of miR-221, miR-106a, miR-21 in gastric cancer cells than that of single AMOs (P = 0.014, 0.024; 0.038, respectively). Migration activity was also clearly decreased as compared to those in randomized and blank control groups ($28{\pm}4$ Vs $54{\pm}3$, P <0.01; $28{\pm}4$ Vs $59{\pm}4$, P < 0.01). Conclusions: MTg-AMOs can specifically inhibit the expression of multiple miRNAs, and effectively antagonize proliferation and migration of gastric cancer cells promoted by oncomirs.
