Immunogold Labellings and Expression of Metallothionein in Regenerating Rat Liver (재생중인 흰쥐 간의 메탈로사이오닌에 대한 면역-금 표지 및 발현에 관한 연구)

  • Metallothionein (MT) is a low molecular weight, cysteine-rich, metal-binding, and non-enzymatic protein. The present study was carried out to investigate the expression of MT gene as well as the localization of MT in regenerating rat liver. In partial hepatectomized rats, MT mRNA was detected as early as 1 hr and reached a maximal level by 8 hr after the operation. Thereafter, this level decreased gradually until 24 hr, and it became similar to that of sham control. Meanwhile, time course of MT immunoreactivity using immunogold-labelling revealed that the number of gold particles in hepatocytes increased significantly by 12 hr, but decreased at 48 hr after partial hepatectomy. Ultrastructurally, immunogold particles indicating the presence of MT were distributed in both the cytoplasm and the nucleus of the rat hepatocytes, particularly the particles were distributed at rough endoplasmic reticulum and nucleolus and did not seem to adhere to mitochondria or lysosomes in proliferating hepatocytes. Briefly, high level of MT mRNA expression and the intense immunoreactivity and/or the specific localization of MT was observed during liver regeneration. These results suggest that MT possibly involves hepatocyte proliferation via the storage or the supply of various metal ions in the regenerating rat liver.

Is Mitochondrial DNA Copy Number Associated with Clinical Characteristics and Prognosis in Gastric Cancer?

  • Alterations in mitochondrial DNA (mtDNA) have been studied in various cancers. However, the clinical value of mtDNA copy number (mtCN) alterations in gastric cancer (GC) is poorly understood. In the present study, we investigated whether alterations in mtCNs might be associated with clinicopathological parameters in GC cases. mtCN was measured in 109 patients with GC by quantitative real-time PCR. Then, correlations with clinicopathological characteristics were analyzed. mtCN was elevated in 64.2% of GC tissues compared with paired, adjacent, non-cancerous tissue. However, the observed alterations in mtCN were not associated with any clinicopathological characteristics, including age, gender, TN stage, Lauren classification, lymph node metastasis, and depth of invasion. Moreover, Kaplan-Meier survival curves revealed that mtCN was not significantly associated with the survival of GC patients. In this study, we demonstrated that mtCN was not a significant marker for predicting clinical characteristics or prognosis in GC.

  • The purpose of this study was to investigate the metallothionein of acute cadmium poisoning mice as a Cadmium index. Forty male ICR mice were injected with cadmium chloride solution from 1/8LD$\sub$50/ to 1/2 LD$\sub$50/ dose. At 24 hours after exposed Cd, I examined Cd and metallothionein (MT) in tissues (liver and kidney) and fluids (whole blood and urine) and also measured low molecular proteins, N-acety1-${\beta}$-D-glucosaminidase (NAG) and ${\beta}$$\sub$2/- microglobuline (${\beta}$$\sub$2 /-MG) in urine. The concentration of Cd and MT of liver, kidney whole blood and urine were increased with dose dependent manner. Urinary Cd and urinary MT had very good significance (p<0.01) and urinary MT had good significance with kidney Cd and NAG but not ${\beta}$$\sub$2/-MG. Conclusionally MT in urine was very correlated with kidney Cd and urine Cd. So MT maybe useful as a Cd poisoning index.

Stromelysin-1 and Membrane type-MMP-1 Expressions in Human Chronic Periodontitis with Type 2 Diabetes Mellitus (단순 만성 치주염 환자와 제 2형 당뇨병을 동반한 만성 치주염 환자에서 Stromelysin-1와 Membrane type-MMP-1 Expressions)

  • Purpose: The purposes of this study were to compare and quantify the expression of Stromelysin-1 and MT-MMP-1 in the gingival tissues of patients with type 2 diabetes mellitus(DM) and healthy adults with chronic periodontitis. Materials and Methods: Gingival tissue samples were obtained during periodontal surgery or tooth extraction. According to the patient's systemic condition & clinical criteria of gingiva, each gingival sample was devided into three groups. Group 1 (n=8) is clinically healthy gingiva without bleeding and no evidence of bone resorption or periodontal pockets, obtained from systemically healthy 8 patients. Group 2 (n=8) is inflammed gingiva from patients with chronic periodontitis. Group 3 (n=8) is inflammed gingiva from patients with chronic periodontitis associated with type 2 DM. Tissue samples were prepared and analyzed by Western blotting. The quantification of Stromelysin-1 and MT-MMP-1 were performed using a densitometer and statistically analyzed by one-way ANOVA followed by Tukey test. Results: In the analysis of expression levels, Stromelysin-1 and MT-MMP-1 expressions were similar in group 1 and 2. Stromelysin-1 and MT-MMP-1 expressions was more increased in group 3 than group 1, 2. The difference between group 3 and group 1, 2 was statistically significant. Also, in the interrelationship of Stromelysin-1 and MT-MMP-1 expressions, expressions of Stromelysin-1 and MT-MMP-1 showed increasing tendency in chronic periodontitis associated with type 2 DM and it seems that the MT-MMP-1 expressions were increasing in proportion to Stromelysin-1 expressions. Conclusion: It is suggested that Stromelysin-1 and MT-MMP-1 may be partly involved in the progression of periodontal inflammation associated with type 2 DM, as related to a metabolism of other factors, such as AGE, plasmin and other inflammatory mediators. Therefore, the expression levels of Stromelysin-1 and MT-MMP-1 can be inflammatory markers of periodontal inflammed tissue with type 2 DM.

Effects of Seeding Rate on Forage Yield and Chemical Composition of Echinochloa crusglli Var. Frumentacea(Roxb) Wight In Jeju Region (제주지역에서 파종량 차이에 따른 청예피의 사료수량 및 조성분 변화)

  • In order to identify the optimum seeding rate of Jeju barnyard grass (Echinochloa crusgalli var. fiurnentacea(Roxb) Wight), 15, 20, 25, 30, 35 and 40 kilograms per a hectare were planted in April 16, 2000, respectively. The growth characterization, the yield and chemical composition were then investigated in June 28(1st cutting) and August 30(2nd cutting), 2000. According to increasing the seeding rate, plant height(155.5 to 162 cm), the yield of fresh forage(61 to 73 MT/ha), dry matter forage(11.9 to 16.9 MT/ha), crude protein(0.9 to 1.6 MT/ha), and TDN(6.1 to 9.7 MT/ha) were gradually increased until the seeding furrow, 35 kg/ha. In 40 kg/ha of seeding furrow, however, those results were decreased than that of the former seeding furrow. The contents of crude protein(7.4 to 10.4%), ether extract(3.3 to 5.2%), nitrogen free extract(46.4 to 47.8%), and TDN(52.1 to 60.4%) were also similar to those of the plant heights and the each yield excepted the seeding furrow, 40 kg/ha. Namely, the each results about the contents increased in 40kg/ha of seeding furrow. According to increasing the seeding rate, the stem diameter, number of leaves, number of withering leaves, and fresh weight were gradually decreased. The contents of crude fiber(30.3 to 27.5%) and crude ash(12.7 to 9.0%) were also decreased These results showed that the optimum seeding rate for the best of the feed production was the seeding furrow, 35 kg/ha.

Effects of Seeding Rate on Growth Characters, Yield Potential and Feeding Value in Jeiu Italian Millet (제주조의 파종량 차이에 따른 생육반응, 수량성 및 사료가치 변화)

  • A Jeju local variety of Italian millet (Setaria italica Beauvois) was grown at five seeding rates (6, 9, 12, 15 and 15 kg/ha) from May 1 to August 25, 2003 at Jeju to determine influence of seeding rate on agronomic characters, forage yield and quality. Days to flowering increased from 89 to 96 days as seeding rate increased from 6 to 18 kg/ha. The plant height increased from 128 to 148cm as seeding rate was increased from 6 to 12 kg/ha and then decreased to 131cm at a rate of 18 kg/ha. With increasing seeding rate, stem diameter, number of leaves, number of nodes, leaf width and weight per plant decreased but leaf length had the same trend with plant height. As seeding rate increased from 6 to 12 kg/ha, fresh furage, dry matter, crude protein, and total digestible nutrients (TDN) yield increased from 35.93 to 54.83 MT/ha, 10.49 to 17.43 MT/ha, 0.94 to 1.79 MT/ha and 5.33 to 9.27 MT/ha, respectively, and then decreased with further increased seeding rates. Crude protein, either extract, nitrogen free extract, and TDN content increased 9.0 to 12.01%, 1.3 to 1.8%, 45.6 to 47.8% and 50.8 to 55.0%, respectively, but crude fiber content decreased 35.2 to 30.0% and crude ash declined 5.8 to 8.4% as seeding rate increased from f to 18 kg/ha. The optimum seeding rate to obtain the highest forage yield was estimated to be 12 kg/ha.

Distribution of positive cells by two monoclonal antibodies (MT1, MB2) in rat spleens (Rat 비장에서 MT1 과 MB2 항체의 양성반응세포 분포)

  • This study was designed to investigate the distributions of the positive cells in rat spleens by two monoclonal antibodies of MT1 and MB2. The spleens of immature 10 rats (Sprague Duwely, approximately 200gm) were collected and paraffin-embedded sections of spleens were stained with immunohistochemical methods. Higher proportions of MT1-positive cell number in spleens were ordered as marginal zone(8.5~18.1%), red pulp(2.1~8.8%) and periarterial lymphoid sheath(0~1.6%) in white pulp, and those of MB2-positive cell number are ordered as the central area of the periarterial lymphoid sheath(100%), red pulp(29.1~45.0%), marginal zone(15.2~30.4%), and peripheral area of periarterial lymphoid sheath(2.3~3.5%). The positive cells by MB2 are more numerous in number than by MT1. The above results were concluded that the positive cells by above two monoclonal antibodies were scattered throughout the red pulp and marginal zone, but in the central area of the periarterial lymphoid sheath, the MB2-positive cells only were present.

  • We isolated Actinomycetes strain GMT 1155 and purified the active compound, MT 1155, on the morphological reversion of ts/NRK cell from the isolate. MT 1155 was identified as toyocamycin having antifungal and antitumor activities from physico-chemical properties and UV, IR, $^1H$-NMR, $^13C$-NMR and mass spectrum. MT 1155 showed the morphologically reversional activity on ts/NRK cell and the cytotoxicity on CTLL cell at the final concentrations of 1.7 JlM and 0.2 11M, respectively and its $IC_{50}$ value on protein kinase A enzyme was 2.3 $\mu$M. Also it had strong antifungal activity against several pathogenic fungi but not antibacterial activity. And it did not inhibit both protein kinase C activity and the bleb-formation of K562 cell induced by phorbol esters.

  • Recently, due to the development of network environment, the internet market has been expanding, so it is necessary to standardize the data format and API to exchange information among objects. Therefore, MPEG (Moving Picture Expert Group), an international standardization organization, is establishing the MPEG-IoMT (ISO/IEC 23093) project to standardize the Internet of Things. MPEG-IoMT establishes Core Experiment (CE) and discusses overall data exchange such as data exchange procedure, markup language and communication method. In this paper, core experiments 1, 2, 4, and 5 of the core experiments of MPEG-IoMT will be discussed. The performance information of the sensor, the sensor data, the performance information of the driver, and the exchange procedure of the control command are explained and the exchange of the media additional data is discussed. We compare the markup language and communication method through experiment.

Role of Chemical Fertilizer and Change of Agriculture in Korea (우리나라 농업의 변천과 비료의 역할)

  • The self-supply rate of Korea in 2006 was approximately 27.3 % by importing 13.99Mt for 19.79Mt of demanded amount. Among the imported crops, wheat, corn, and soybean consumed 95 % for the total imported amount, and wheat, corn, and soybean were 3.5Mt(Table use : 0.22Mt; Feed stuff : 0.13Mt), 8.7Mt(Table use : 0.19Mt; Feed stuff : 0.68Mt), and 1.2Mt (Table use : 0.03Mt; Feed stuff : 0.09Mt), respectively. On the other hand, our government has prepared the strategies for a great fear of food according to sharp price rise of the international crops by maintaining the self-supply rate of 5 % excluding 5.23Mt of rice in Korea. Also concern for recycled energy known as future energy for era of high oil price and global warming due to green house gas is rapidly growing. Therefore, our country which has relied on import of the whole oil needed in Korea and has to keep Kyoto Agent to request reduction of green house gas fully support research and practical use for agricultural products as resource of alternate energy. At first, we have to develop the mass production technology in order to secure a program of self-supply of food for bioenergy production utilizing agricultural product in Korea. But we assume that this matter is difficult to achieve under the current agriculture system that more emphasizes the environment conservation such as environmentally-friendly agriculture than production of food.
