A liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI/MS/MS) method was developed for the determination and confirmation of clenbuterol in bovine muscle and milk. Clenbuterol and clenbuterol-D9 using as an internal standard in samples were extracted with ethyl acetate after hydrolysis and evaporated to dryness. The extracts were dissolved in 20% methanol and cleaned using HLB solid-phase extraction cartridge. The analytes were detected by LC-ESI/MS/MS on a $C_{18}$ column. Mass spectral acquisition was done in selected reaction monitoring (SRM) in positive ion mode to provide a high degree of sensitivity. Using MS/MS with SRM mode, the transitions (precursor to product) monitored were m/z 277${\rightarrow}$203 for clenbuterol, and m/z 286${\rightarrow}$204 for internal standard. The limits of quantitation (LOQ) and mean recoveries of clenbuterol in bovine muscle were $0.2{\mu}g/kg$ and 84.3~91.1%, respectively. The LOQ and mean recoveries in milk were $0.05{\mu}g/kg$ and 87.7~98.3%, respectively.
This article was conducted to induce the transgenic hairy roots and determine the effect of culture conditions on optimum growth of hairy roots by Agrobacterium rhizogenes strain, 15834 in Lycium chinense Miller. Hairy roots of L. chinense Miller. were induced from leaf segments by co-cultivation with A. rhizogenes. When the hairy roots were cultured in various MS medium strength and sucrose concentrations, the highest growth of hairy roots was observed in half-strength MS media supplemented with 3% sucrose, respectively. In air lift bioreactor cultures, the liquid medium contained with 1/2 MS and 3% sucrose was also the best for optimum growth of hairy roots.
This study is performed to estimate the prevalence of metabolic syndrome among male workers and to identify the relationships with many related factors including anthropometry, hematological index, serum lipid level, dietary-related behaviors and health-related behaviors. According to the age groups, the 20s are significantly higher in normal and risk groups than in the metabolic syndrome (MS) group, the 30s are significantly higher in MS group than the other groups. The levels of AST and ${\gamma}$-GTP both show significant differences in the order of MS group ($30.3{\pm}8.8U/l$, $91.1{\pm}40.2U/l$) > risk group ($25.7{\pm}8.1U/l$, $41.8{\pm}20.2U/l$) > normal group ($22.8{\pm}6.0U/l$, $26.6{\pm}10.7U/l$). For the frequency of breakfast consumption, the response of 'Every day' is significantly higher in MS group than normal and risk groups, but the response of 'Not at all' is significantly higher in normal group than MS and risk groups. The drinking amount is positively correlated with ${\gamma}$-GTP in normal group, and it is negatively correlated with the hematocrit level, but it is positively correlated with the systolic blood pressure in MS group. AST is positively correlated with glucose concentrations of the MS group. The ALT is positively correlated with waist circumferences and systolic blood pressure in the risk group. The results of this study show that breakfast frequency, education level, drinking amount, drinking frequency, exercise frequency, AST, ALT and ${\gamma}$-GTP levels are all important risk factors of MS. Therefore, it is very important to maintain a healthy life style for the prevention of MS incidence.
Background: Ginsenosides are the characteristic and principal components which manifest a variety of the biological and pharmacological activities of the roots and rhizomes of Panax ginseng (GRR). This study was carried out to qualitatively and quantitatively determine the ginsenosides in the cultivated and forest GRR. Methods: A rapid and sensitive ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time of flight tandem mass spectrometry (UPLC-DAD-QTOF-MS/MS) was applied to the qualitative analysis of ginsenosides and a 4000 QTRAP triple quadrupole tandem mass spectrometer (HPLC-ESI-MS) was applied to quantitative analysis of 19 ginsenosides. Results: In the qualitative analysis, all ingredients were separated in 10 min. A total of 131 ginsenosides were detected in cultivated and forest GRR. The method for the quantitative determination was validated for linearity, precision, and limits of detection and quantification. 19 representative ginsenosides were quantitated. The total content of all 19 ginsenosides in the forest GRR were much higher than those in the cultivated GRR, and were increased with the growing ages. Conclusion: This newly developed analysis method could be applied to the quality assessment of GRR as well as the distinction between cultivated and forest GRR.

Background: Polycystic ovarian syndrome (PCOS) is one of the most common endocrinopathies among reproductive-age women. Its metabolic features often overlap with those associated with metabolic syndrome (MS) and insulin resistance syndrome (IRS). The objective of this study was to determine the prevalence and predictors of MS and IRS in infertile Vietnamese women with PCOS. Methods: A cross-sectional study was conducted at a tertiary fertility centre at Hue University Hospital from June 2016 to November 2017. A total of 441 infertile women diagnosed with PCOS based on the revised 2003 Rotterdam consensus criteria were enrolled. MS and IRS were defined based on the National Heart, Lung, and Blood Institute/American Heart Association Adult Treatment Panel III 2005 and American College of Endocrinology IRS 2003 criteria, respectively. Complete clinical and biochemical measurements of 318 women were available for analysis. Independent predictors of MS and IRS were identified using multivariate logistic regression. Results: The overall prevalence of MS and IRS in women with PCOS was 10.4% and 27.0%, respectively. We identified older age (>30 years) and obesity as independent predictors of MS and IRS. Elevated anti-$M{\ddot{u}}llerian$ hormone levels increased the risk of IRS, but not that of MS. Conclusion: MS and IRS are prevalent disorders among infertile Vietnamese women with PCOS. PCOS is not solely a reproductive problem. Screening and early intervention for MS and/or IRS based on anthropometric, metabolic, and reproductive hormone risk factors should be an integral part of fertility care.
A simple, specific, and economical LC-MS/MS method was investigated for the screening of 43 prescribed antihypertensive and related drugs in human urine. The urine samples were simply prepared by diluting and mixing with internal standard before directly introduced to the LC-MS/MS system, which is fast, straightforward, and cost-effective. Fractional factorial, Box-Behnken, and I-optimal design were applied to screen and optimize the mass spectrometric and chromatographic factors. The analysis was carried out on a triple quadrupole mass spectrometer system utilizing multiple reaction monitoring with positive and negative electrospray ionization method. Chromatographic separation was performed on a Thermo Scientific Accucore RP-MS column (50 × 3.0 mm ID., 2.6 ㎛) using two separate gradient elution programs established with the same mobile phases. Chromatographic separation was performed within 12 min. The optimal method was validated based on FDA guideline. The results indicated that the assay was specific, reproducible, and sensitive with the limit of detection from 0.1 to 50.0 ㎍/L. The method was linear for all analytes with coefficient of determination ranging from 0.9870 to 0.9981. The intra-assay precision was from 1.44 to 19.87% and the inter-assay precision was between 2.69 and 18.54% with the recovery rate ranges from 84.54 to 119.78% for all drugs measured. All analytes in urine samples were stable for 24 h at 25℃, and for 2 weeks at -60℃. The developed method improves on currently existing methods by including larger number of cardiovascular medications and better sensitivity of 12 analytes.
The aim of this study is to evaluate the reference value for electrocardiogram in healthy captive raccoon dogs. Forty-one free-ranging adult raccoon dogs rescued from Wildlife rescue centre, Kangwon National University were enrolled in this study. The 6-lead electrocardiogram was obtained in all raccoon dogs without any chemical restraints. The mean heart rate was $146.10{\pm}43.31$ beats/min (95% confidence interval 132.84~159.36 beats/min). The mean respiration rate was $35.73{\pm}11.56$ breaths/min (95% confidence interval 32.19~39.27 breaths/min). The mean systolic blood pressure was $136{\pm}29.26$ mmHg (95% confidence interval 127.99~145.91 mmHg). Electrocardiographical features were also evaluated in all raccoon dogs. The mean duration and amplitude of P-wave were $38.2{\pm}4.0$ ms (range 28-40 ms) and $0.128{\pm}0.039$ mV (range 0.09~0.20). The mean duration and amplitude of QRS complexes were $48.5{\pm}7.2ms$ (range 36-60 ms) and $1.330{\pm}0.650$ mV (range 0.15~2.30). The range of the mean electrical (QRS) axis was $-91^{\circ}{\sim}+96^{\circ}$ ($10^{\circ}{\sim}60^{\circ}$; 95% of confidence interval). The mean corrected QT (QTc) interval was $273.7{\pm}32.7ms$ (range 212-333 ms), while the mean PR interval was $76.1{\pm}10.0ms$ (range 50-82 ms). To the authors' knowledge, this is the first study to provide references in electrocardiogram (ECG) in healthy captive raccoon dogs.
In order to micropropagate uniform plantlets of Philodendron cannifolium in vitro, the shoot tips were cultured on MS media supplemented with $0.5{\sim}10.0$ mg/L BA or $0.05{\sim}0.1$ mg/L thidiazuron(TDZ). The adventitious multi-bud clusters from basal part of shoots were formed on MS media containing $2.0{\sim}5.0$ mg/L BA or $0.05{\sim}0.1$ mg/L TDZ. But the shoots grown on MS media with TDZ showed necrosis by the lack of chlorophyll. The adventitious multi-bus clusters were cut into $5{\sim}7$ mm sections and cultured on MS media containing BA and TDZ for shoot proliferation. Shoots were proliferated vigorously on MS medium supplemented with $1.0{\sim}3.0$ mg/L BA with up to 30 shoots. But abnormally swollen hard calli were formed from basal parts of shoots on MS media with TDZ and high concentration of BA(10.0 mg/L). The proliferated shoots on same media also showed necrosis by the lack of chlorophyll. The shoot growth and rooting were favorable on MS media containing $0.5{\sim}2.0$ mg/L IBA. The rooted plantlets were acclimatizated effectively in soil mixed with perlite 1:vermiculite 1 or vermiculite alone. Fifteen mL of liquid medium containing 10 g/L activated charcoal and 30 g/L sucrose were added in same vessels after small shoots were proliferated to stimulate shoot growth and rooting. After 8 weeks in culture, the shoots were dipped into high concentration of IBA solution. and planted in soil mexed with perlite 1:vermiculite 1. The shoot growth and rooting were favorable in dipping treatments of $500{\sim}2,000$ ppm IBA solutions for 10 sec.
The efficacy of the alginate microsphere (Alginate MS) oral vaccine against Miamiensis avidus in olive flounder (Paralichthys olivaceus) was confirmed through challenge infections by both immersion and injection routes. In trial 1, the formalin-inactivated M. avidus coated with alginate, designated as 'IMa+Alginate MS' group, and the IMa group were administered with vaccines mixed with feed, with a total antigen dose of 3.75 × 106 cells/fish. When challenged with immersion infection at five weeks post vaccination, the relative percent survival (RPS) in the IMa+Alginate MS group was 50% (immersed in 50% seawater) and 37.5% (immersed in 100% seawater). The group that received only IMa showed a low survival rate. In trial 2, the antigen was fed mixed with feed at a total dose of 2.38 × 106 cells/fish for 5 days. Two weeks after oral vaccination, fish were intraperitoneally injected for infection. The RPS in the IMa+Alginate MS group was 30.8%, while the IMa-only group showed no vaccine efficacy. At five weeks post vaccination, when subjected to challenge infection by immersion in 50% seawater, the IMa+Alginate MS group recorded a RPS of 42.9%, whereas the IMa group had a RPS of 14.3%. The results of this study indicate that coating M. avidus antigen with alginate can provide higher protection in olive flounder compared to administering the antigen alone.

Xanthium sibiricum is used as a traditional folk medicine for the treatment of cancer, fever, headache, nasal sinusitis, and skin pruritus. This study aimed to identify components from Xanthium sibiricum extracts using an SPE-800MHz NMR-MS hyphenated system. The simultaneous acquisition of MS and NMR spectra from the same chromatographic peaks significantly increases the depth of information acquired for the compound and allows the elucidation of structures that would not be possible using MS or NMR data alone. LC -NMR analysis was conducted using a HPLC separation system coupled to 800 MHz spectrometer equipped with a cryoprobe, and a SPE unit was used to automatically trap chromatographic peaks using a HPLC pump. LC-MS analysis was conducted with a Q-TOF MS instrument using ESI ionization in the negative ion mode. Using the hyphenated analysis, several secondary metabolites were identified, such as 3',5'-O-dicaffeoylquinic acid, 1',5'-O-dicaffeoyl- quinic acid, and ethyl caffeate. These results demonstrate that the SPE-800MHz NMR-MS hyphenated system can be used to identify metabolites within natural products that have complex mixtures.
