A 3-year-old female, 5kg, Shih-tzu developed an acute onset of depression, disorientation, hypersalivation, nystagmus after falling down 2 meter height place. In plain skull radiography, there was fracture line in the frontal and parietal bones and next day magnetic resonance imaging examination was performed. Magnetic resonance imaging of the brain was performed with 3.0 Tesla unit. Under general anesthesia, the dog was placed in prone with its head positioned in a birdcage coil. Transverse, sagittal and coronal fast spin echo images of the brain were obtained with the following pulse sequences: T1 weighted images (TR = 560 ms and TE = 18.6 ms) and T2 weighted images (TR = 3500 ms and TE = 80 ms). Magnetic resonance imaging showed epidural hematoma in the left frontal area resulting in compression of the adjacent brain parenchyma. Left lateral ventricle was compressed secondarily and the longitudinal fissure shifted to the right, representing mass effect. The lesion was iso-to slightly hyperintense on T1 weighted image and iso-slightly hypointense signal on T2 weighted image. At necropsy, there was a skull fracture and epidural hematoma in the left frontal area. Magnetic resonance imaging of epidural hematoma is reviewed.
To compare the accuracy of breath-hold magnetic resonance imaging sequences to establish the most effective superparamagnetic iron oxide-enhanced sequence for detection of hepatic metastases. A total of 100 patients(50men and 50women, mean age: 60years) with liver disease(including malignant and benign liver lesions) were investigated at 3.0T machine (GE, General Electric Medical System, Excite HD) with 8Ch body coil. Pulse sequence for MR imaging decided to the FS-T2-FSE-RT(TR/TE/Thick./Freq./Phase=12857ms/100ms/7mm/512/384), MGRE(TR/TE/Thick./Freq./Phase=100ms/9.7ms/7mm/384/288), in-out of phase echo(TR/$TE_1$, $TE_2$/Thick./Freq./Phase=140ms/2.4, 5.8ms/7mm/352/300), Images obtained before the injection of SPIO. Six sequences were optimized for lesion detection: FS-T2-FSE-RT, multigradient recalled echo data image(MGRE), T2-weighted MGRE with an 9.7msec echo time. Images were reviewed independently by five blinded observers. The accuracy of each sequence was measured by using picture archiving communication system analysis. All results were correlated with findings at multidectator computed tomography examination. Differences between the mean results of the six observers were measured by using paired student t-test analysis. Postcontrast T2-weighted MGRE sequences were the most accurate and were significantly superior to postcontrast FS-T2-FSE-RT, T2-weighted MGRE, in-out of phase MR sequences(p < .05). For all lesions that were malignant or smaller than 1 cm, respectively, contrast to noise ratio of pre and postcontrast sequences were -1and -0.3 for T2-weighted FSE, 0.53 and 4.5 in-out of phase, 7, 7.08, 5.08, 3.32, 1.7, 1.16, 0.79, 0.68 for GRE with 2.9, 7.5, 12.1, 16.6, 21.2, 25.8, 30.4, 35.0 TE values. Breath-hold various TE precontrast sequences offer improvement in sensitivity compared with fixed multigradient recalled echo sequences alone.
Matrix assisted laser desorption ionization (MALDI) mass spectrometry is commonly used to analyze biological molecules such as proteins, peptides and lipids from cells or tissue. Recently MALDI Imaging mass spectrometry (IMS) has been widely applied for the identification of different drugs and their metabolites in tissue. This special feature has made MALDI-MS a common choice for investigation of the molecular histology of pathological samples as well as an important alternative to other conventional imaging methods. The basic advantages of MALDI-IMS are its simple technique, rapid acquisition, increased sensitivity and most prominently, its capacity for direct tissue analysis without prior sample preparation. Moreover, with ms/ms analysis, it is possible to acquire structural information of known or unknown analytes directly from tissue sections. In recent years, MALDI-IMS has made enormous advances in the pathological field. Indeed, it is now possible to identify various changes in biological components due to disease states directly on tissue as well as to analyze the effect of treated drugs. In this review, we focus on the advantages of MALDI tissue imaging over traditional methods and highlight some motivating findings that are significant in pathological studies.

BACKGROUND: Cardiac magnetic resonance fingerprinting (cMRF) enables simultaneous mapping of myocardial T1 and T2 with very short acquisition times. Breathing maneuvers have been utilized as a vasoactive stress test to dynamically characterize myocardial tissue in vivo. We tested the feasibility of sequential, rapid cMRF acquisitions during breathing maneuvers to quantify myocardial T1 and T2 changes. METHODS: We measured T1 and T2 values using conventional T1 and T2-mapping techniques (modified look locker inversion [MOLLI] and T2-prepared balanced-steady state free precession), and a 15 heartbeat (15-hb) and rapid 5-hb cMRF sequence in a phantom and in 9 healthy volunteers. The cMRF5-hb sequence was also used to dynamically assess T1 and T2 changes over the course of a vasoactive combined breathing maneuver. RESULTS: In healthy volunteers, the mean myocardial T1 of the different mapping methodologies were: MOLLI 1,224 ± 81 ms, cMRF15-hb 1,359 ± 97 ms, and cMRF5-hb 1,357 ± 76 ms. The mean myocardial T2 measured with the conventional mapping technique was 41.7 ± 6.7 ms, while for cMRF15-hb 29.6 ± 5.8 ms and cMRF5-hb 30.5 ± 5.8 ms. T2 was reduced with vasoconstriction (post-hyperventilation compared to a baseline resting state) (30.15 ± 1.53 ms vs. 27.99 ± 2.07 ms, p = 0.02), while T1 did not change with hyperventilation. During the vasodilatory breath-hold, no significant change of myocardial T1 and T2 was observed. CONCLUSIONS: cMRF5-hb enables simultaneous mapping of myocardial T1 and T2, and may be used to track dynamic changes of myocardial T1 and T2 during vasoactive combined breathing maneuvers.

Mass spectrometry (MS) is an ideal tool for analyzing multiple types of (bio)molecular information simultaneously in complex biological systems. In addition, MS provides structural information on targets, and can easily discriminate between true analytes and background. Therefore, imaging mass spectrometry (IMS) enables not only visualization of tissues to give positional information on targets but also allows for molecular analysis of targets by affording the molecular weights. Matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) MS is particularly effective and is generally used for IMS. However, the requirement for an organic matrix raises several limitations that get in the way of accurate and reliable images and hampers imaging of small molecules such as drugs and their metabolites. To overcome these problems, various organic matrix-free LDI IMS systems have been developed, mostly utilizing nanostructured surfaces and inorganic nanoparticles as an alternative to the organic matrix. This minireview highlights and focuses on the progress in organic matrix-free LDI IMS and briefly discusses the use of other IMS techniques such as desorption electrospray ionization, laser ablation electrospray ionization, and secondary ion mass spectrometry.

The binding of MS2 bacteriophage coat protein (MCP) to MS2 binding site (MBS) RNA stem-loop sequences has been widely used to label mRNA for live-cell imaging at single-molecule resolution. However, concerns have been raised recently from studies with budding yeast showing aberrant mRNA metabolism following the MS2-GFP labeling. To investigate the degradation pattern of MS2-GFP-labeled mRNA in mammalian cells and tissues, we used Northern blot analysis of ${\beta}$-actin mRNA extracted from the Actb-MBS knock-in and $MBS{\times}MCP$ hybrid mouse models. In the immortalized mouse embryonic cell lines and various organ tissues derived from the mouse models, we found no noticeable accumulation of decay products of ${\beta}$-actin mRNA compared with the wild-type mice. Our results suggest that accumulation of MBS RNA decay fragments does not always happen depending on the mRNA species and the model organisms used.
Chemoresistance to cancer therapy is a major obstacle to the effective treatment of human cancers with cisplatin (DDP), but the mechanisms of cisplatin-resistance are not clear. In this study, we established a cisplatin-resistant human ovarian cancer cell line (COC1/DDP) and identified differentially expressed proteins related to cisplatin resistance. The proteomic expression profiles in COC1 before and after DDP treatment were examined using 2-dimensional electrophoresis technology. Differentially expressed proteins were identified using matrix-assisted laser desorption/ ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and high performance liquid chromatography-electrospray tandem MS (NanoUPLC-ESI-MS/MS). 5 protein spots, for cytokeratin 9, keratin 1, deoxyuridine triphosphatase (dUTPase), aarF domain containing kinase 4 (ADCK 4) and cofilin1, were identified to be significantly changed in COC1/DDP compared with its parental cells. The expression of these five proteins was further validated by quantitative PCR and Western blotting, confirming the results of proteomic analysis. Further research on these proteins may help to identify novel resistant biomarkers or reveal the mechanism of cisplatin-resistance in human ovarian cancers.
Purpose: Fluid-attenuated inversion recovery (FLAIR) imaging can be obtained faster with shorter repletion time (TR), but it gets noisier. We hypothesized that shorter-TR FLAIR obtained at 3 tesla (3T) with a 32-channel coil may be comparable to conventional FLAIR. The aim of this study was to compare the diagnostic value between conventional FLAIR (TR = 9000 ms, FLAIR9000) and shorter-TR FLAIR (TR = 6000 ms, FLAIR6000) at 3T in terms of diffusion-weighted imaging-FLAIR mismatch. Materials and Methods: We recruited 184 patients with acute ischemic stroke (28 patients < 4.5 hours) who had undergone 5-mm diffusion-weighted imaging (DWI) and two successive 5-mm FLAIR images (no gap; in-plane resolution, $0.9{\times}0.9mm$) at 3T with a 32-channel coil. The acquisition times for FLAIR9000 and FLAIR6000 were 108 seconds (generalized autocalibrating partially parallel acquisitions [GRAPPA] = 2) and 60 seconds (GRAPPA = 3), respectively. Two radiologists independently assessed the paired imaging sets (DWI-FLAIR9000 and DWI-FLAIR6000) for the presence of matched hyperintense lesions on each FLAIR imaging. The signal intensity ratios (area of DWI lesion to contralateral normal-appearing region) on both FLAIR imaging sets were compared. Results: DWI-FLAIR9000 mismatch was present in 39 of 184 (21.2%) patients, which was perfectly the same on FLAIR6000. Three of 145 patients (2%) with DWI-matched lesions on FLAIR9000 had discrepancy on FLAIR6000, showing no significant difference (P > 0.05). Interobserver agreement was excellent for both DWI-FLAIR9000 and DWI-FLAIR6000 (k = 0.904 and 0.883, respectively). Between the two FLAIR imaging sets, there was no significant difference of signal intensity ratio (mean, standard deviation; $1.25{\pm}0.20$; $1.24{\pm}0.20$, respectively) (P > 0.05). Conclusion: For the determination of mismatch or match between DWI and FLAIR imaging, there is no significant difference between FLAIR9000 and FLAIR6000 at 3T with a 32-channel coil.
This study attempts to examine the clinical usefulness of High b-value DWI (diffusion weighted imaging) for brain tumors with an edema. Subjects were seven patients selected from 65 patients who received an MRI scan for suspected encephalopathy and confirmed diagnosis at our hospital from February to July 2015 (male: 7, average age : 66 years old). As test equipment, 3.0T MR System (ACHIEVA Release, Philips, Best, The Netherlands) and 8Channel SENSE Head Coill were used. As for the experiment, in b-value 1,000 and 2,000 images, SNR and the lesion at the lesion site and CNR in the normal site opposite to the lesion are measured. WW(window width) and WL(window level) are made equal in MRICro software, and the volume of the lesion is measured from each of b-value and MPR TRA CE image. Using SPSS ver. 1.8.0.0 Mann Whitney-test was analyzed for SNR and CNR, while Kruskal-Wallis test was analyzed for volume.
슬관절 퇴행성 질환을 진단하기 위하여 자기공명영상(magnetic resonance imaging: MRI)이 많이 사용되지만 간혹 슬관절 병후 및 예후를 잘못 진단하는 경우가 종종 발생한다. 본 연구에서는 슬관절 질환의 진단에 도움을 주기 위하여 자화전이(magnetization transfer: MT) 영상법을 소개하고자 한다. 슬관절 환자 7명으로부터 스핀 에코(SE) T2 강조 영상(3,400-3,500/90-100 ms)과 슬관절 환자 7명으로부터 FSE T2 강조 영상(4,500-5,000/100-108 ms)과 또한 슬관절 환자 3명으로부터 gradient echo (GRE) T2 강조 영상들(9/4.56 ms, 50 flip angle, NEX 1)을 획득하였다. 6명의 슬관절 환자에서 지방 억제가능 T2 강조 STIR 펄스시퀀스(TR/TE=2894-3215 ms/70 ms, NEX 3, ETL 9)를 사용하였다. 지방 포화도에 있어서 위상감도 방법은 Larmor frequency 차이에 따른 위상 차이를 이용하므로, 각각의 픽셀에 대한 자화전이율(magnetization transfer ratio: MTR)의 측정은 포화된 영상과 포화되지 않은 영상의 비율에 따라 산출하였다. 따라서 각 입력된 영상들은 동차원성을 가지고, 시각적으로 신호강도 정도가 회색의 명암도만으로 평가될 수 있기 때문에 생리학적, 정량적 진단을 위하여 3차원 등방성 체적영상과 자기공명 삼원색을 매핑하였고 정량적 특정은 자화전이율 지도로서 표현하였다. 자화전이율 영상은 병변 부위에서 높은 대조도를 나타내어 환자의 병태를 추적하는데 도움을 주었고 정량화하였다. 자화전이율 영상들과 기존의 MRI 사이에 명암도 차이는 회색상으로 표현되며, 자화전이율 영상화의 효과에 대해 프로파일 그래프는 자화전이 펄스로 인하여 신호강도에 있어서 정량적 측정값이 상대적으로 감소하였다. 슬관절의 정확한 병리상태를 진단하기 위해 프로파일 그래프의 측정값을 영상과 함께 표현하였다. 본 연구에서 수행한 예비적 데이터들을 통하여 슬관절의 자화전이율 영상들이 매우 임상학적으로 유용함을 확인하였다. 자화전이율 영상에 대한 물리적 변화를 관찰함으로써 자화전이율 영상에 대한 물리적, 기술적 기반에 대한 더 많은 통찰력을 제공할 수 있다. 무릎 질환 환자의 자화전이율 영상들을 이용하여 매우 높은 대조도를 확보할 수 있으므로 슬관절 질환의 정밀 진단에 매우 도움을 줄 수 있다.
