Quantification of Vibrio parahaemolyticus Using a Most Probable Number-Polymerase Chain Reaction Assay Targeting the H-NS gene (MPN 및 H-NS 유전자를 표적으로 하는 PCR assay를 병용한 장염비브리오(Vibrio parahaemolyticus)의 정량)

  • We applied a combination of most probable number-polymerase chain reaction (MPN-PCR) methods using a PCR procedure targeting the H-NS (VP1133) gene to detect Vibrio parahaemolyticus presence and density in seawater as well as within short-necked clam Ruditapes philippinarum tissues collected from Gomso Bay, Korea. In 30 seawater samples, V. parahaemolyticus levels ranged from less than 1.8 to $1.1{\times}10^3MPN/100mL$, and samples from August showed higher than those from other months. Furthermore, the levels of V. parahaemolyticus in six short-necked clam samples ranged from $7.8{\times}10^2$ to $2.1{\times}10^3MPN/100g$, approximately 2.5 times higher than in seawater samples from the corresponding month. Our results provide data on V. parahaemolyticus contamination in seawater and short-necked clam tissues, and help to improve quantitative methods of assessing V. parahaemolytcius levels.

Survey on the microbiological quality of meat in Seoul (소.돼지 도체표면의 미생물학적 고찰)

  • This survey was conducted to evaluate the microbiological quality of raw beef and pork products from January to December in 1999. A total of 107 beef and 157 hog carcasses were collected from two abattoirs located in Seoul. The result showed that beef carcasses had an average bacterial loading around 139,000 bacteria/$\textrm{cm}^2$ of carcass surface, indicating a little bit higher count than the results reported in USA and Australian meat. However, overall hygienic status was found to be acceptable for all examined carcasses because 84.4% of product rated excellent, good or acceptable comparable to USA of 91.6% and Australia of 88%. The analysis of data on overnight-chilled to weekend-chilled carcasses indicated that the microbiological growth occurred in the chiller during the weekend chill with increases in total viable count from 130,000cfu/$\textrm{cm}^2$ to 400,000cfu/$\textrm{cm}^2$. Qualitative testing for escherichia coli, EC + MUG was used as a most probable number (MPN) method along with the petrifilm method. The average of MPN/$\textrm{cm}^2$ of E coli biotype 1 was 29MPN/$\textrm{cm}^2$ for beef carcasses and 1,100 MPN/$\textrm{cm}^2$ for hog carcasses, respectively. However, 41% of beef and 16.3% of hog carcasses were shown to be less than < 3 MPN/$\textrm{cm}^2$ in E coli biotype 1 examination. Although salmonella enteritis, S typhimurium and E coli O157:H7 were all negative, listeria monocytogenes was recovered from only one hog surface samples of the 89 carcasses tested.

  • Sediments collected from the Jungnang-cheon and its tributaries were used to enumerate anaerobic bacteria by most probable number (MPN) methods. A simple method was developed to detect ferrous ion in the culture fluid in order to count the number of iron ion reducers, and sulfate-reducing bacteria (SRB) and methanogens were detected by the presence of FeS precipitate in the culture or methane in the head space, respectively. The numbers of iron reducer was in the range of 10$^{7}$ - 10$^{8}$ /g in the sediment of the stream containing higher organic content than the tributaries. The sediments of tributaries were analyzed to contain iron reducers less than 10$^{7}$ cells/g. With one exception the numbers of SRB and methanogens were less than 10$^{3}$ cells/g in the sediment. From these results it is concluded that organics in the sediment support the growth of iron reducers, which out-compete SRB and methanogens.

Distribution of Pathogenic Vibrio spp. in Seawater of the Geum River Estuary Area, West Coast of Korea (금강 하구 해역의 해수에 병원성 비브리오균(Vibrio spp.)의 분포)

  • The pathogenic Vibrio genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. V. cholerae, V. vulnificus, and V. parahaemolyticus are the main species that can be potent human pathogens and the leading cause of septicemia, wound infections, and seafood borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrios in seawater. We obtained a total of 80 seawater samples from the Geum River estuary area in the west coast of Korea from April to December 2021. Pathogenic Vibrios was determined using a combination of the most probable number-polymerase chain reaction (MPN-PCR) methods. The detection levels of V. cholerae, V. parahaemolyticus, and V. vulnificus in the seawater samples were 7.5%, 68.8%, and 30.0%, respectively. The quantitative results were as follows: 3.6-3.6 MPN/100 mL in V. cholerae, 3.6-3,400 MPN/100 mL in V. parahaemolyticus, and 3.6-4,300 MPN/100 mL in V. vulnificus. Overall, these results provide novel insight into the necessity for seawater sanitation in the Geum River estuary area, and could help reduce the risk of seafood-borne outbreaks caused by pathogenic Vibrios.

Evaluation of the Bacteriological Quality of a Shellfish-growing Area in Kamak Bay, Korea

  • The sanitary quality of a shellfish-growing area in Kamak Bay, Korea, was assessed through a bacteriological examination of seawater and oysters from January 2004 to December 2006. From seawater samples collected at 28 stations, the range of geometric mean (GM) and the estimated 90th percentile for most-probable-number (MPN) values of fecal coliforms were <1.8-2.4 MPN/100 mL and 1.8-6.0 MPN/100 mL, respectively. The observed fecal coliform GM and the estimated 90th percentile did not exceed the fecal coliform water quality standards of 14 MPN/100 mL and 43 MPN/100 mL. Therefore, the bacteriological quality of seawater at this shellfish-growing area met the Korean Shellfish Sanitation Program (KSSP) criteria for a growing area used for export. The range of the fecal coliform GM and the estimated 90th percentile MPN values of oyster samples were 19.2-160 MPN/100 g and 20.2-166.9 MPN/100 g, respectively, and the range of the viable cell count was 30-1900 CFU/g. Thus, the fecal coliform value for the oysters and the viable cell count were less than the current shellfish quality standards of 230 MPN/100 g and 50,000 CFU/g, respectively. The bacteriological quality of the oysters complied with the criteria for domestic use and export of shellfish.

Distribution of Pathogenic Vibrio Species in Seawater in Gomso Bay and Byeonsan, West Coast of Korea (곰소만 및 변산 해역 해수에 병원성 비브리오균(Vibrios spp.)의 분포)

  • The pathogenic Vibrio genus contains halophilic bacteria that are distributed in marine and freshwater environments. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are potent human pathogens and leading causes of septicemia, wound infection, and seafood-borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrio species in seawater off the west coast of Korea. Sixty-four seawater samples were obtained from different sites in Gomso Bay and Byeonsan from April 2018 to November 2018. Pathogenic Vibrio species were detected using a combination of most probable number (MPN)-polymerase chain reaction methods. V. cholerae, V. vulnificus, and V. parahaemolyticus were found in 0.0%, 20.3%, and 65.6% of seawater samples, respectively. Quantitative results revealed 3.6-23 MPN/100 mL of V. vulnificus, and 3.6-930 MPN/100 mL of V. parahaemolyticus in the samples. Overall, these results provide new insight into the necessity for seawater sanitation in Gomso Bay and Byunsan; they also provide evidence that will help reduce outbreaks of seafood-borne illness caused by pathogenic Vibrio species.

Evaluation of Bacteriological Safety for the Shellfish growing Sea waters in Seocheon Area, Korea (충남 서천 패류생산해역에서의 세균학적 위생안전성 평가)

  • A Sanitary survey in an Important shellfish-growing area of Biin Bay in Seocheon-gun Korea, was conducted to evaluate bay conditions and compliance with the bacteriological criteria for areas designated for the production of shellfish for export. Seawater samples were collected monthly at 55 sampling stations established in the survey area from January 2006 to December 2008. Bacteriological water quality did not change in response to a small rainfall (10.0 mm), but it increased abruptly in response to rainfall to 62.5 mm The most probable number (MPN) value for total coliforms and fecal coliforms in 1, 980 seawater samples ranged from <1.8 to >1, 600 MPN/100mL and from <1.8 to 330 MPN/100mL, respectively. The geometric mean and estimated $90^{th}$ percentile value for total coliforms ranged from 2.0 to 10.3 MPN/100mL and from 7.0 to 42.6 MPN/100mL respectively. The geometric mean and estimated percentile value for fecal coliforms ranged from 1.8 to 4.0 MPN/100mL and from 1.9 to 18.3 MPN/100mL, respectively. Accordingly, the bacteriological water quality of Biin Bay met the National Shellfish Sanitation Program (NSSP) and Korea Shel1fish Sanitation Program(KSSP) criteria for areas designated for shellfish production for export in Korea.

유류오염토양 복원을 위한 설계인자 선정에 관한 연구

  • The objective of this study was to decide the designing factor for remediaton of the contaminated site. The soil and ground-water samples were analyzed and hydro- geological characteristics was assayed for the survey of pollution level. Also air-permeability test and MPN(most probable number) test were conducted for selecting the designing factor. The contaminants were mainly found in north-west part of the site and were expected to move toward the south. Ex-situ technology was expected more useful than in-situ one with the results of air-permeability test saying that air permeability was relatively low. Additional microbes were expected for remediation efficiency because residual microbes were loosely populated. The choosing of the designing factor was requisite for remediation of contaminated site.

Prevalence and Quantification of Vibrio parahaemolyticus in Raw Salad Vegetables at Retail Level

  • The purpose of this study was to investigate the biosafety of Vibrio parahaemolyticus in raw salad vegetables at wet markets and supermarkets in Malaysia. A combination of the most probable number-polymerase chain reaction (MPN-PCR) method was applied to detect the presence of V. parahaemolyticus and to enumerate their density in the food samples. The study analyzed 276 samples of common vegetables eaten raw in Malaysia (Wild cosmos=8; Japanese parsley=21; Cabbage=30; Lettuce=16; Indian pennywort=17; Carrot=31; Sweet potato=29; Tomato=38; Cucumber=28; Four-winged bean=26; Long bean=32). The samples were purchased from two supermarkets (A and B) and two wet markets (C and D). The occurrence of V. parahaemolyticus detected was 20.65%, with a higher frequency of V. parahaemolyticus in vegetables obtained from wet markets (Wet market C=27.27%; Wet Market D=32.05%) compared with supermarkets (Supermarket A=1.64%; Supermarket B=16.67%). V. parahaemolyticus was most prevalent in Indian pennywort (41.18%). The density of V. parahaemolyticus in all the samples ranged from <3 up to >2,400 MPN/g, mostly <3 MPN/g concentration. Raw vegetables from wet markets contained higher levels of V. parahaemolyticus compared with supermarkets. Although V. parahaemolyticus was present in raw vegetables, its numbers were low. The results suggest that raw vegetables act as a transmission route for V. parahaemolyticus. This study will be the first biosafety assessment of V. parahaemolyticus in raw vegetables in Malaysia.

The Detection and a Quantitative Evaluation of Entomopathogenic Nematodes in Cultivated Rhizosphere Soil (경작지 근권 토양내 곤충병원성 선충의 검출 및 정량적 평가)

  • The direct count and MPN (Most Probable Number) methods were used to measure the number of nematodes in soils collected from cultivated and non-cultivated fields. As a result, the number of nematodes from cultivated soils was higher than the non-cultivated soils (NC -1, NC -2). On the other hand, upon measuring the value from the organo farming cultivated soils (OC, OR) and conventional cultivated soils (CC, CR), the former showed 16 times higher than the latter. These results indicate that nematode population which can multiply in the organic compounds abundantly exist in the organo farming cultivated soil. Isolated entomopathogenie nematodes are composed of two orders, which were Rhabditida and Diplogasterida. To determine the pathogene-city of them using the 5th larvae and pupae silkworm, and the following mean $LD_{90}$ values were found: 24 to 30 hours in Rhabditida and 36 to 48 hours in Diplogasterida nematode, respectively. This study indicates that nematodes are sensitive to this kind of environmental disturbance. Isolated entomopathogenic nematodes were suggested that aye quite within the realms of possibility for biological control agents.
