The N-methylpurine-DNA glycosylase (MPG), a ubiquitous DNA repair enzyme, removes N-methylpurine and other damaged purines induced in DNA. Tissue-specific mRNA levels of the N-methylpurine-DNA glycosylase (MPG) were investigated in Balb/c mice of four different growing stages; newborn, 1, 4 and 8-weeks postpartum. MPG expressions in the newborn and the 8-week-old mice were the highest in thymus and testis, respectively. The tested tissues of the newborn mice had consistently higher MPG mRNA level than 8-week-old adults except in testis and thymus. The MPG mRNA level in testis was the lowest in the newborn mice, but it attained the highest in the 8-week-old mice. The levels of MPG mRNA among the different tissues in the newborn and the 8-week-old mice were more than 9.0 and 19.0-fold respectively. These results suggest that the of MPG expression was dependent on the growing stage and had tissue-specificity.
N-Methylpurine-DNA glycosylase (MPG) removes N-methylpurine and other damaged purines in DNA. RT-PCR analysis revealed MPG mRNA expression at various tissues of fetal development from day 8 to day 18 fetus and day 400 mature adult. The MPG transcripts were abundant during fetal development in mice. In placenta, the MPG mRNA was continuously decreased from day 8 post coitum (p.c) to day 18 p.c. fetus. The high level of mRNA in fetal brain and liver was drastically declined in day 400 mature adult. The expression of MPG, originally characterized by its highest level of expression in the epididymis of adult mouse, was detected with high level in several other reproductive organ, including the ovary, oviduct, testis, vas deference, uterus, and seminal vesicles. These results demonstrate developmental stage- and tissue-specific variation of MPG gene expression.
This study was conducted to investigate changes in posture and muscle characteristics by applying mat pilates and kinesio taping to college students with round shoulder posture and to investigate effective interventions. Subjects were 28 men and women with a round shoulder posture, and they were randomly assigned to a mat pilates group (MPG, n=14) and a Kinesio taping group (KTG, n=14). Each group performed the intervention for 4 weeks, and the distance from the ground to the acromion(DGA) and muscle characteristics (F: Frequency; S: Stiffness; D: Decrement) were measured before and after the intervention. Independent T-test was performed for the difference in the amount of change between groups, and the paired-sample T-test was performed for the difference between pre-post values within the group. Statistical significance level was set as 𝛼=.05. As a result of measuring the change in DGA in both groups, it was significantly decreased in MPG (p<.05). As a result of measuring the change in muscle characteristics of the two groups, the change in F in TL was significantly increased in MPG (p<.05), there was no change in TG, and the amount of change in MPG was larger in the amount of change between the two groups. appeared (p<.05). The change in S was significantly increased in MPG (p<.05). Therefore, the mat pilates conducted in this study is considered to be an effective intervention for improving the round shoulder posture and changing the muscle characteristics than kinesio taping.

Methoxy polyethylene glycol-epoetin beta (MPG-EPO), a continuous erythropoietin receptor activator, is a new erythropoiesis-stimulating agent with a long half-life. The purpose of this prospective study is to assess the effects of once-monthly subcutaneous MPG-EPO on hematological responses and nutritional status in peritoneal dialysis patients. Forty four patients undergoing stable peritoneal dialysis were enrolled into the study. Darbepoetin alfa therapy, in peritoneal dialysis patients, was converted to the monthly administration of subcutaneous MPG-EPO for 6 months. The starting dose of MPG-EPO was based on the previous weekly dose of darbepoetin alfa. The dose adjustments were performed to maintain the hemoglobin (Hb) levels in a target range of 10.5-11.0 g/dL. If the Hb levels exceeded 11.0 g/dL, MPG-EPO was temporarily interrupted for 1 month. The mean Hb levels were stable with the values of $9.5{\pm}1.1$ g/dL at baseline, and $10.4{\pm}0.9$ g/dL at the 6th month after conversion. The mean differences in the changes of Hb levels between the baseline and the 6th month were $0.9{\pm}1.4$ g/dL, which was statistically significant. However, the mean differences of iron, transferrin saturation and ferritin concentrations were not significant. It did not show significant differences in the changes of the nutritional parameters. These results suggest that the once-monthly subcutaneous administration of MPG-EPO for 6 months effectively maintains the Hb levels and nutritional status in peritoneal dialysis patients. Taken together, the once-monthly subcutaneous administration of MPG-EPO was practical and might improve the clinical compliance for the management of renal anemia in peritoneal dialysis patients.

Objectives: This was to evaluate the current usage of three erythropoietin stimulating agents (ESA) and their efficacy for management of anemia in peritoneal dialysis (PD) patients with chronic kidney disease. Methods: It was a retrospective comparative study through review of electronic medical records of chronic kidney disease patients undergoing PD at a tertiary teaching hospital from January 1998 to June 2013. Results: Average administration frequency was 1.66 times/week in EPO group, 0.75 times/week in DA group, and 0.19 times/week in MPG-EPO group. At the first 4 weeks, there were significant differences in mean hemoglobin levels between EPO and DA groups ($9.25{\pm}1.28g/dL$, $10.02{\pm}0.95g/dL$ each, p = 0.018) and also in hemoglobin response rates (10.0%, 45.2% each, p = 0.008), but since after 4 week, there had been no significant differences. There also showed no significant differences in achievement of hemoglobin target between the two groups. When converted to MPG-EPO in EPO/DA groups, there showed a slight increase in hemoglobin levels of both groups. MPG-EPO was the highest compared with two other drugs by the average cost based on the average weekly dose. Conclusion: EPO, DA, and MPG-EPO showed similar effects in treatment of anemia of PD patients based on hemoglobin target range (11.0~12.0 g/dL) which NFK-K/DOQI guidelines suggest. Though the average cost of MPG-EPO was higher than the other two drugs, the number of PD patients using MPG-EPO has increased and it is thought that long half-life and low administration frequency of MPG-EPO have improved the compliance of PD patients who have to self-administrate.

With different concentration of Evodiae Fructus, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice divided into 3 groups for each experiment. We chose the caspase-3 for cell apoptosis, MAPK and MPG genes for cell viability and DNA repair. Results: In case of 4, 8, 12 day of Evodiae Fructus, we were examined the mean number of total ovulated oocytes and the number of morphologically normal oocytes. We were also examined the embryonic developmental competence in vitro. In addition we were also examined the differential expression of cell viability related genes, caspase-3, MAPK and MPG according to concentration and duration of Evodiae Fructus administration. MPG gene expressions for cell viability and DNA repaie were increased in dose dependent manner than that of control group in 4-day administration group. Conclusion: It is suggested that the medication of Evodiae Fructus has beneficial effect on reproductive functions of female mice via promotion of cell proliferation.

Purpose: These experiments were undertaken to evaluate the effect of Onpoeum on ovarian functions and differential gone expressions related with cell viabilities caspase-3, MAPK and MPG in female mice. Methods: We administered the Onpoeum to 6-week-old female ICR mice for 4, 8, or 12 days. With different concentration of Onpoeum, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice divided into 3 different groups for each experiment. We chose the Caspase-3 for cell apoptosis, MAPK and MPG genes for cell viability and DNA repair. Results: In case of 4, 8, 12 day of Onpoeum, we were examined the mean number of total ovulated oocytes and the number of morphologically normal oocytes. We were also examined the embryonic developmental competence in vitro. In audition we were examined the differential expression of cell apoptosis, viability and DNA repair related genes, Caspase-3, MAPK and MPG according to concentration and duration of Onpoeum. From these results showed that the administration of Onpoeum played a role of prevention of cell apoptosis and DNA damages and also increased cell proliferation resulted in ovarian functions. Conclusion: It is suggested that the medication of Onpoeum may have beneficial effect on reproductive functions of female mice via prevention of cell apoptosis and DNA damaging and promotion of cell proliferation.

Photo-plethysmography (PPG), which measures changes in the peripheral blood flow of a human body using difference in absorption rate of light, is a measurement method that is studied and used in clinical and various applications due to its simple circuit configuration and measurement convenience. Magneto-plethysmography (MPG), which is newly developed by our team, is a method of measuring changes in the conductivity of biological tissues by using a eddy current induced by a time-varying magnetic field, and is not subject to optical interference. In this study, we investigated the detection characteristics of MPG according to the change of the conductivity of the object and fluid to be measured by simultaneously measuring PPG and MPG. In order to control the speed of fluid known in advance, a blood flow simulator was implemented and used. The fluid used in the experiment was general mineral water and physiological saline (0.9% NaCl) solution. Experimental results show that the amplitude change of the measured PPG was 0.3% in normal water and saline solution, and that of MPG was 77.3%. Therefore, it is considered that the magneto-plethysmography (MPG) has a strong correlation with the conductivity of the fluid.

Methods: We administered the Foeniculi Fructus to 6-week-old female CF-1 mice for 4, 8, 12 days. After administration of Foeniculi Fructus with 0.1, 1, 10, $100\;mg/m{\ell}$ concentration in the comparison of control group with $0\;mg/m{\ell}$, we observed the mean number of total ovulated oocytes and the number of morphologically normal oocytes. After entosomatic fertilization, we observed the rate of fertilized 2-cell embryos to blastocyst stage in vitro. Also we chose the caspase-3 for cell apoptosis, MAPK and MPG genes for cell viability and DNA repair by RT-PCR. Results: 1. In case of 4, 8, 12day administration of Foeniculi Fructus with 0.1, 1, 10, $100\;mg/m{\ell}$, the mean number of total ovulated oocytes and the number of morphologically normal oocytes were increased in the comparison of control group. 2. In case of 4, 8, 12day administration of Foeniculi Fructus with 0.1, 1, 10, $100\;mg/m{\ell}$, the rates of blastocyst formation from 2-cell stages were increased in the comparison of control group. 3. In case of 4, 8, 12day administration of Foeniculi Fructus with 0.1, 1, 10, $100\;mg/m{\ell}$, the gene expression of caspase-3, MAPK, MPG didn't show significant result in the comparison of control group. Conclusion: This study shows that Foeniculi Fructus has significant effects on the increase of the function on ovulation and embryonic development of female mice. But this results have nothing to do with caspase-3, MAPK and MPG genes. So we need a further study for which genes are related to the activation of reproductive functions of Foeniculi Fructus.
Major pelvic ganglia (MPG) in rats are an unique autonomic ganglia, containing both sympathetic and parasympathetic neurons related with the function of bladder, penis and bowel. It has been widely known that ionotropic $GABA_A$ receptors are the molecular target of $\gamma$-aminobutric acid (GABA), a major inhibitory neurotransmitter in central nervous system. However, their functions and regulations of $GABA_A$ receptors expressed in autonomic ganglia have been poorly understood. 1 examined the modulatory role of adenylyl cyclase (AC) and protein kinase A(PKA) on $GABA_A$-induced inward currents in the neurons of rat MPG. $GABA_A$ receptors were identified using immunofluorescent labeling in the rat major pelvic ganglion. Electrophysiological experiments were performed to record the activities of $GABA_A$ receptors. $GABA_A$ receptors were expressed only in sympathetic neurons. GABA induced marked inward currents in a concentration-dependent manner. Mucimol ($5{\mu}M$), a $GABA_A$ receptor agonist induced inward currents were significantly reduced in the presence of SQ 225361 $20{\mu}M$, a AC inhibitor and myristoylated PKA inhibitor 100 nM. In addition, forskolin ($1{\mu}M$), AC activator, augmented the GABA induced currents. The activation of AC/PKA-dependent pathway could involve in the regulation $GABA_A$ receptors, expressed only in sympathetic neurons of rat MPG. These findings are helpful for the better understanding the function of various pelvic organs innervated by MPG.
