  • iPod Touch provided so various application software beyond the early music playback function, and enhanced its UI to the level of UX. Because of these, it was evaluated as innovative. In addition, even though various images were provided through the Internet, some people still classify MP3P as audio device in its product category. As for how audio device became video or image device and which factor contributed to such happening, there may be various factors, such as social, cultural, and technical change, other than fundamental change of medium itself. This study is regarding medium characteristics of MP3Player. In other words, this study classified genealogical changes, which can be categorized into 4 points. This study also analyzed the property of each point according to this classification. The result of analysis showed that there is a relation with technological as well as social context in such genealogical change in MP3player. Even though there are various elements in the change of a device, it can be suggested that the factors of genealogical change in MP3player are influenced by a totality of social needs and contexts and technical changes.

  • Commercially used disposable cups undergo fragmentation in the environment and become microplastics (MPs). These MPs can be ingested by aquatic organisms and cause a range of adverse effects. We assessed the acute and chronic toxicity of disposable cup-derived MP fragments in Daphnia magna. MP fragments were identified as a polyethylene terephthalate (PET) fragment with a size of 33.18 ± 7.76 ㎛. The presence of three additives including 1- Propanone. 1-phenyl-3-[2-(phenylmethoxy)phenyl]-, p-Xylene and ethylbenzene was analyzed from MP fragments. The 48 h acute toxicity revealed that 20 % of immobilization and mortality were found at the highest concentration of PET MP (200 mg L-1). The 21 d chronic toxicity revealed that PET MP fragments significantly (p < 0.05) more reduced survival rate (31 %), total offspring (52 %) in D. magna compared with control group. The developmental abnormality of offspring (3.5%) by PET MP fragments was significantly (p < 0.05) higher than control groups (0.3%). These results are possibly induced by gut blocking by ingestion of MP fragments and their longer retention time. These findings indicate that the fragmentation of disposable cups (PET polymers) into small-sized MP fragments pose a significant ecological risk to aquatic organisms. Further studies are required to elucidate the underlying toxicity mechanisms.

  • Di- and tri-methylation of lysine 4 on histone H3 (H3K4me2 and H3K4me3, respectively) are epigenetic markers of active genes. Complex associated with Set1 (COMPASS) mediates these H3K4 methylations. The involvement of COMPASS activity in secondary metabolite (SM) biosynthesis was first demonstrated with an Aspergillus nidulans cclA knockout mutant. The cclA knockout induced the transcription of two cryptic SM biosynthetic gene clusters, leading to the production of the cognate SM. Monascus spp. are filamentous fungi that have been used for food fermentation in eastern Asia, and the pigment Monascus azaphione (MAz) is their main SM. Monascus highly produces MAz, implying that the cognate biosynthetic genes are highly active in transcription. In the present study, we examined how COMPASS activity modulates MAz biosynthesis by inactivating Monascus purpureus cclA (Mp-cclA) and swd1 (Mp-swd1). For both ${\Delta}Mp-cclA$ and ${\Delta}Mp-swd1$, a reduction in MAz production, accompanied by an abated cell growth, was observed. Suppression of MAz production was more effective in an agar culture than in the submerged liquid culture. The fidelity of the ${\Delta}Mp-swd1$ phenotypes was verified by restoring the WT-like phenotypes in a reversion recombinant mutant, namely, trpCp: Mp-swd1, that was generated from the ${\Delta}Mp-swd1$ mutant. Real-time quantitative Polymerase chain reaction analysis indicated that the transcription of MAz biosynthetic genes was repressed in the ${\Delta}Mp-swd1$ mutant. This study demonstrated that MAz biosynthesis is under the control of COMPASS activity and that the extent of this regulation is dependent on growth conditions.

  • The Systems Engineering method is increasingly recognized as a central in the Acquisition of Weapon Systems in South Korea. Because of these trends, issues concerned with improving weapon system design are increasingly raised. This paper presents a development of Operational Mode Summary/Mission Profile (OMS/MP) in terms of Weapon Systems Acquisition for application to Systems Engineering method. For a systematic approach, a precise concept of OMS/MP is derived by analyzing the concept of conventional OMS/MP. This paper reviews the results of a series of previously reported examples aiming to develop the OMS/MP of a weapon system. Based on this, the limitations of the conventional OSM/MP applications are investigated. And these limitations can be resolved by the Fundamentals of Systems Engineering. Finally, the OMS/MP framework based on the Fundamentals of Systems Engineering has been proposed and is successfully implemented for the Harbor Underwater Surveillance System.

  • Barley yellow dwarf virus (BYDV) belongs to Luteovirus and is limited only at phloem related tissues. An open reading frame (ORF) 4 of BYDV codes for the movement protein (MP) of BYDV gating plasmodesmata (PD) to facilitate virus movement. Like other Luteoviruses, ORF 4 of BYDV is embedded in the ORF3 but expressed from the different reading frame in leaky scanning manner. Although MP is a very important protein for systemic infection of BYDV, there was a little information. In this study, MP was characterized in terms of subcellular localization and programmed cell death (PCD). Gene of MP or its mutant (ΔMP) was expressed by Agroinfiltration method. MP was clearly localized at the nucleus and the PD, but ΔMP which was deleted distal N-terminus of MP showed no localization to PD exhibited the different target with original MP. In addition to PD localization, MP appeared associated with small granules in cytoplasm whereas ΔMP did not. MP associated with PD and small granules induced PCD, but ΔMP showed no association with PD and small granules did not exhibit PCD. Based on this study, the distal N-terminal region within MP is seemingly responsible for the localization of PD and the induction small granules and PCD induction. These results suggest that subcellular localization of BYDV MP may modulate the PCD in Nicotiana benthamiana.

  • Conflicting evidence has been obtained regarding whether transient receptor potential cation channels (TRPC) are store-operated channels (SOCs) or receptor-operated channels (ROCs). Moreover, the Ca/Na permeability ratio differs depending on whether the current-voltage (I-V) curve has a doubly rectifying shape or inward rectifying shape. To investigate the calcium permeability of TRPC4 channels, we attached GCaMP6s to TRPC4 and simultaneously measured the current and calcium signals. A TRPC4 specific activator, (-)-englerin A, induced both current and calcium fluorescence with the similar time course. Muscarinic receptor stimulator, carbachol, also induced both current and calcium fluorescence with the similar time course. By forming heteromers with TRPC4, TRPC1 significantly reduced the inward current with outward rectifying I-V curve, which also caused the decrease of calcium fluorescence intensity. These results suggest that GCaMP6s attached to TRPC4 can detect slight calcium changes near TRPC4 channels. Consequently, TRPC4-GCaMP6s can be a useful tool for testing the calcium permeability of TRPC4 channels.
