Antitumor Activity of Chitosan Oligosaccharides Produced in Ultrafiltration Membrane Reactor System

  • Chitosan oligosaccharides (COSs) were prepared and fractionated into three groups of COS [a high molecular weight COS (HMWCOS), medium molecular weight COS (LMWCOS), and low molecular weight COS (LMWCOS)] according to their molecular weight, using an ultrafiltration membrane enzymatic bioreactor designed earlier [8]. Antitumor activity of these COSs was then examined against Sarcoma 180 solid (S180) or Uterine cervix carcinoma No. 14 (Ul4) tumor cell-bearing mice. Among these COSs, MMWCOS with molecular weight range from 1.5 to 5.5 kDa effectively inhibited the growth of both tumor cells in the mice. In addition, the administration of MMWCOS resulted in increased thymus weight among lymphoid organs. The mice treated with MMWCOS showed improved survival rate and larger number of survivors after 40 days of feeding. The most effective of MMWCOS far antitumor activity in the S180- or U14-bearing mice was 20 mg/kg/day or more.

미생물 합성에 의한 poly(3-hydroxybutyrate)의 분자량과 말단관능기 제어 (Control of Molecular Weight and Terminal Groups of Poly(3-hydroxybutyrate) in Bio-synthesis)

  • In the bio-synthesis of poly(3-hydroxybutyrate)(PHB), which is a kind of poly(3-hydroxyalkanoate)(PHA), aimed to control the low molecular weight of PHB and obtain a telechelic PHB. As a result of incubation of R. eutropha at $30^{\circ}C$ with ethylene glycol added as a chain transfer agent, PHB content on the dry cell weight increased up to 24h, however, it decreased after that, and the molecular weight of PHB increased from 9h to 12h, and then, decreased up to 72h. The decrease of the content and the molecular weight of PHB indicates that PHB was decomposed as an energy source in bacterial cells and was incorporated into metabolic pathways. $^1H-NMR$ of the obtained PHB after incubation for 72h was measured to determine the terminal groups of the PHB during incubation. As the results of $^1H-NMR$ measurement, the peaks derived from ethylene glycol in both terminals of PHB were observed. Which indicate that the terminal reaction was caused by the addition of ethylene glycol, and that telechelic PHB having hydroxyl group at the both terminals where molecular weight was controlled was successfully synthesized.

High Molecular Weight Conjugated Polymer Thin Films with Enhanced Molecular Ordering, Obtained via a Dipping Method

  • The fabrication of polymer field-effect transistors with good electrical properties requires the minimization of molecular defects caused by low molecular weight (MW) fractions of a conjugated polymer. Here we report that the electrical properties of a narrow bandgap conjugated polymer could be dramatically improved as a result of dipping a thin film into a poor solvent. The dipping time in hexanes was controlled to efficiently eliminate the low molecular weight fractions and concomitantly improve the molecular ordering of the conjugated polymer. The correlation between the structural order and the electrical properties was used to optimize the dipping time and investigate the effects of the low MW fraction on the electrical properties of the resulting thin film.

ABS 중에 첨가된 저분자 화합물의 파단 거동에 미치는 영향 (Fracture Behaviour of Lubricants in ABS Terpolymer)

  • ABS 수지의 파단 거동을 살펴 보기 위해 rubber content, molecular weight, lubricant의 종류 및 양과 파단 speed, 주위 환경온도를 변수로 측정한 후 전자현미경 사진으로 그 파단면을 살펴 보았다. Tension하에서 rubber content와 lubricant content는 감소할수록, molecular weight와 tension speed는 증가할수록 strength는 증가하였으며, 파단면은 rubber content, speed, molecular weight는 감소할수록, liquid lubricant를 사용할수록 파단부위 주변의 변형이 일어나지 않았으며, 파단면도 수지가 서로 미끄러져 파단되는 상태가 아닌 상분리와 같은 형상으로 분리파단되었다. Impact하에서 rubber content, molecular weight, 주위 환경온도, lubricant content가 증가할수록 strength도 증가하였으며, 파단면은 사진상의 파단표면적이 작은 것이 높은 strength를 나타내고 있다.

실크의 가용화 조건이 효소분해 실크 펩타이드의 분자량 분포에 미치는 영향 (Effect of Solubilization Conditions on Molecular Weight Distribution of Enzymatically-Hydrolyzed Silk Peptides)

  • 실크의 피브로인 단백짐의 가수분해에 의한 실크 펠타이드의 제조를 위하여 설크의 가용화 조건이 분자량 분포에 미치는 영 향을 고찰하였다 염 화칼숨, ethylenediamine 및 황산에 의해 가용화된 실크의 평균 분자량은 각각 41600, 3308과 1268 dalton 으로서 염화칼숨을 제외한 나머지 두 가용화 방법에서 평균 분 자량이 수천 dalton인 실크 용액을 얻을 수 있었으며 분산계수 는 3-4의 범위뜰 보였다. 가용화 및 분말화한 후 6시간 동안 단백질 분해효소 처리에 의하여 600-1200 dalton의 중량평균 분 자량을 갖는 살크 접타이드틀 얻을 수 있었다. 24시간 염산에 의한 산분해 처리에 의해 얻은 실크 웹타이드의 평균 분자량은 145 dalton으로 80%의 높은 유리 아미노산 함량을 나타내었다. 여러가지 가용화 방법 및 가수분해 방법 중 낮은 농도의 황산용 액으로 우선 실크를 가용화한 후 중화 및 효소처리에 의해 웹타 이드로 가수분해하는 방법이 과도한 아미노산의 유리를 억제하 면서 분자량의 조절도 용이할 것으로 판단되었다.

금강 수계 자연유기물 특성 분석 (Analysis of Natural Organic Matter (NOM) Characteristics in the Geum River)

  • Natural organic matter(NOM) is defined as the complex matrix of organic material and abundant in natural waters. It affects the performance of unit operations for water purification. Several kinds of analytical indicators such as DOC, specific ultraviolet absorbance(SUVA), apparent molecular weight (AMW), fractionation and high performance size exclusive chromatography(HPSEC) have been used to understand characteristics and variations of NOM. This study aims to evaluate the characteristics of NOM in the Geum River system comprising with stream flows and reservoirs. It was identified that SUVA denoting the portion of humic substance in water ranged within 1.60~3.36. Using resin adsorbents, dissolved organic carbon(DOC) was fractionated into three classes: hydrophobic bases(HOB), hydrophobic acids(HOA) and hydrophilic substances(HI). HI dominates in all samples, collectively accounting for more than 62% of the DOC. HOA was the second dominated fraction and it varied considerably but accounted for about 30% of the DOC. The distribution of high molecular weight(HMW) measured by HPSEC being used to determine the molecular weight distribution of aquatic humic substances was 40.1% and 38.7% in reservoir and stream flow, respectively. The distribution of low molecular weight(LMW) in stream flow was 13.2% higher than that in reservoir. And apparent molecular weight less than 1KDa, which include the molecular weight of hydrophilic organic matter, occupied with 69.2% and 68.2% in stream flow and reservoir, respectively. While the molecular weight of 1 to 100 KDa including humic substances ranged with 18.6% and 21.6% in stream flow and reservoir, respectively. Seasonal variation of refractory dissolved organic carbon was similar to that of SUVA.

토하젓의 숙성과정중 Chitin Oligosaccharides 생성 (Formation of Chitin Oligosaccharides during Fermentation of Toha-jeot(Salt-fermented Toha Shrimp))

  • 우리나라 전남의 전통 발효식품인 토하젓(salt-fer-mented Toha shrimp)은 주 원료인 토하(Toha: Caridina denticulata denticulata $D_{E}$ $H_{AAN}$)를 살아 있는 생토하의 껍질(갑각)이 붙어 있는 채로 소금에 절여서 제조하였다. 토하 껍질을 구성하는 토하 chitin이 토하젓의 숙성과정 중 발효에 의해 저분자량의 chitin oligosaccharides 생성이 예상되므로 본 연구에서는 토하젓 숙성과정의 기간별로 토하 chitin 분자량의 감소와 점도 변화 그리고 chitin 분자량 분포를 측정하였다. 토하젓 숙성과정 중 chitin의 점도와 평균 분자량은 숙성기간이 길어짐에 따라 뚜렷히 감소하였다. $10^3의$ 저분자량인 chitin oligosaccharides는 숙성 60일까지는 거의 나타나지 않았으나 숙성 75일 이후부터 6.85% 이상의 많은 양을 보였으며 $10^2의$ 분자량은 숙성 75일 이후부터 생성되기 시작하였다. 이 결과로부터 토하젓의 숙성과정 중 $10^{7}$ 이상 고분자량인 chitin이 가수분해됨에 따라 $10^2과$ $10^3의$ chitin oligosaccharides가 생성됨이 확인되었다.다.

효소를 이용한 저분자 토종 닭발 콜라겐의 제조 및 품질 특성 (Preparation and quality characteristics of low molecular weight collagen treated with hydrolytic enzymes from Korean native chicken feet)

  • 본 연구에서는 단백질 가수분해효소 0.1%와 1% Protamex를 사용하여 저분자 콜라겐을 제조하였다. 토종 닭발의 조단백질과 콜라겐의 함량은 일반 육계에 비해 높은 함량이 나타났다. 단백질 가수분해 효소농도와 반응시간이 증가할수록 낮은 분자량의 콜라겐을 얻을 수 있는 것으로 나타났다. 특히 1% Protamex로 7시간 처리한 시료가 1,000-5,000 Da의 저분자 콜라겐 함량이 55.6%로 나타났으며, 평균 분자량은 5,390 Da로 가장 낮은 분자량이 나타났다. 이는 단백질 가수분해효소 Protamex가 고분자 펩타이드 결합을 저분자 펩타이드로 분해했기 때문이다. 효소처리 콜라겐의 조직감은 고분자 펩타이드의 콜라겐이 저분자 펩타이드로 분해되어 gel을 형성하지 못하고 sol의 형태를 유지하였다. 효소농도와 효소반응시간이 증가할수록 콜라겐의 평균분자량은 작아지나 효소반응 5시간부터 평균분자량의 감소가 미미해지는 경향이 나타났다. 따라서 저분자 콜라겐 효소반응시간은 경제적으로 볼 때 5시간에서 7시간 사이가 적합하다고 할 수 있다. 이 연구결과는 향후 산업적 효소를 이용한 저분자 콜라겐 제조 및 식품 소재 활용의 기초자료로 이용할 수 있을 것이다.

Cellulase의 분자량과 섬유소의 표면공극 상이성이 여수도 및 종이의 강도적 특성에 미치는 영향 (The efect of freeness and paper physical properties treated with high and low molecular weight cellulase in the different surface pore sized fibers)

  • To examine how the difference of molecular weight distribution of cellulase influenced the beating process according to surface pore size of the fiber, high molecular weight enzyme and low one were applied to soft wood pulp, hard wood pulp, cotton linter pulp. Some enzymes with the distribution of low molecular weight penetrated into cellulose in the proportion of surface pore size and the results were negative as like : the low viscosity, decrease of refining Yield, decrease of fine fibers content and so on. But in cotton linter pulp in the small surface area, the fiber softness was increased and it had a positive result that the paper intensity was high. Other enzymes with the distribution of high molecular weight had an enzyme reaction on the most surfaces of cellulose. They were effective in eliminating the fuzz of hydrophile fine fibers and the freeness was increased.

  • The molecular weight of poly-hydroxybutyric acid (PHB) produced by Alcaligenes sp. K-912 is an important parameter characterizing the physical properties of the polymer. The effects of temperature and the levels of glucose, ammonium, phosphate and amino acids on the molecular weight of PHB were investigated. Molecular weight of PHB by temperature varied in the range of 380,000 to 550,000, 400,000 to 600,000 by glucose, 300,000 to 380,000 by phosphate, 400,000 to 1,000,000 by amino acids, respectively under the experimental conditions.
