Effects of Mannyeon-hwan on Acetic Acid-induced Colitis in Rats (Acetic-acid로 대장염을 유발한 생쥐에 투여한 만년환의 치료효과)

  • Objectives: This study is designed to verify the effects of Mannyeon-hwan (MNH) on acetic acid-induced colitis in rats. Methods: Colitis was induced in male Sprague-Dawley rats weighing approximately 250 g by injecting acetic acid through the anus. The rats were classified into four groups: normal group, acetic acid (AA) group, AA+MNH (L) (low concentration) group, and AA+MNH (H) (high concentration) group. The body weight, visual evaluation of the colonic mucosa, anatomical histological changes, and changes in the expression of cytokines in the colon tissue were compared and analyzed. Results: Compared with the normal group, weight loss was observed in mice induced with colitis. Compared with the AA group, weight loss recovery occurred in the AA+MNH (L) and AA+MNH (H) groups, and significant changes were observed after the sixth day. In the visual evaluation of the colonic mucosa, a significant decrease in damage indicators was observed in the AA+MNH (L) and AA+MNH (H) groups compared with the AA group. In terms of anatomical histological changes and changes in the expression of tumor necrosis factor-α and interleukin-6 in colon tissue, a significant decrease was observed in the AA+MNH (L) and AA+MNH (H) groups compared with the AA group. A more pronounced decrease was observed in the AA+MNH (H) group compared with the AA+MNH (L) group. Conclusion: The effects of MNH on colitis were confirmed through research. MNH can be used as a first-line treatment for patients complaining of colitis who visit oriental medicine clinics.

  • To confirm the increase effect of $MNH_4$ which has been compounded as a result of two years' potted rice experiments, we have established simple experiment areas in the paddy field of Kyung-Puk, Kyung-Nam and Chon-Puk Provinces and have done the completely randomized design experiments with the following results. 1. Comparison of the gross weight of the harvested rice (Straw included) : The $MNH_4$ 1-area has produced 10-28% more harvest than that of control-area. 2. Comparison of the weight of the Unpolished Rice: The $MNH_4$ 1-area has shown 10-23% increase compared with the control-area. 3. In the comparison of the nutrients of the rice from the $MNH_4-area$ with those of the rice from the control area, $MNH_4-area$ showed increase of $N_2$, $P_{2}O_5$ and $SiO_2$ but a slight decrease of $K_2O$.

Evaluation of Mutation Rate by Differences of Isozyme Band Patterns on $M_2$ Seedling Treated with Chemical Mutagen in Barley (보리 화학돌연변이제 처리 $M_2$ 유묘의 Isozyme band pattern 차이에 의한 돌연변이율 검정)

  • Mutation rate of M$_2$ plants that were treated with three types of double treatments of chemical mutagens(1.5mol Na$N_2$ + 0.75mol MNH, 0.75mol MNH + 0.75mol MNH and 0.5mol MNH + 0.5mol MNH) were estimated on the rate of chlorophyll mutant, changes of isozyme loci ; esterase (Est), glutamate oxaloacetate transaminase(GOT ; AAT) and leucyl aminopeptydase(LAP ; AMP). Rate of chlorophyll mutants (3.3% =no. of seedling carrying mutant / all number of M$_2$ seedlings $\times$ 100) and rate of esterase isozyme loci mutants(3.5% =no. of plant carrying mutant / all number of M$_2$ plant) in Dema were higher than one of Sacheon 6, but no significant differences in GOT, LAP. Among isozymes, most of mutants in M$_2$ plant of two varieties were found in esterase (73% of total mutants were occurred in esterase loci). Although many of null bands were found in GOT 3, these were not repeatable and no real mutants. It might be due to qualities of starch, amount of extract buffer and degradation of isozyme during electrophoresis and staining.

Effects of rotary inertia shear deformation and non-homogeneity on frequencies of beam

  • In the present study, separate and combined effects of rotary inertia, shear deformation and material non-homogeneity (MNH) on the values of natural frequencies of the simply supported beam are examined. MNH is characterized considering the parabolic variations of the Young's modulus and density along the thickness direction of the beam, while the value of Poisson's ratio is assumed to remain constant. At first, the equation of the motion including the effects of the rotary inertia, shear deformation and MNH is provided. Then the solutions including frequencies of the first three modes for various combinations of the parameters of the MNH, depth to length ratios, and shear corrections factors are reported. To show the accuracy of the present results, two comparisons are carried out and good agreements are found.

Improvement of Mutation Rate and Reduction of Somatic Effects by Double Treatment of Chemical Mutagens in Barley (화학 돌연변이제 이중처리에 의한 돌연변이율 향상 및 생장저해 경감)

  • Mutation tachniques inducing more useful mutations and reducing somatic effects need to be improved for crop breeding. Seeds of barley varieties ; Dema, Grosso were treated with two types of mutagens ; 1) chemical treatment: single treatment or double treatment of two mutagens (N-nitroso-N-methylurea ; MNH, Sodium Azide; NaN$_3$) 2) gamma ray irradiation treatment. After treatment, half of seeds were used for germination test and half of seeds were sown to the field. With the higher dose of mutagen both chemical and gamma ray were plants treated, the higher rate of growth reduction rate was in M$_1$ seedling. In chemical treatment, germination rate of seeds, growth rate of coleoptile and root in double treatment of chemical mutagens were better than single treatments, especially in same dose. Growth inhibition rate of plant in double treatment of 1.0mM MNH(0.5mM MNH + 0.5mM MNH), for example, were less than one of plants of single treatment of 1.0mM MNH in pot and petri dish test. Growth reduction rate of culm and fertility rate in M$_1$ plants double treated in same dose of single treatment were also less than single one. With the higher dose of mutagen both chemical and gamma ray were plants treated, the higher frequency of chlorophyll mutants was in M$_2$ seedling. The rate of chlorophyll mutants in double treatment of chemical mutagens were higher than single treatment. Double treatment methods can be a improved method for induction of new good mutants, which were induced more useful mutations and reduced harmful somatic effects.

  • Positive ion mass spectra of some corticosteroids were obtained by using liquid chromatography-mass spectrometry(LC-MS). The base peak of each compound showed the protonated molecular ion [$MH^+$], ammonium adduct ion [${MNH_4}^+$] or [$MH^+-60$] ion according to its chemical structure and other characteristic mass ions were [$MH^+-18$], [${MNH_4}^+-18$] and so on. Several rat urinary metabolites of methylprednisolone acetate after the oral administration were detected by the thermospray LC-MS. The identified major metabolites were 20-hydroxymethylprednisolone(20-HMP), methylprednisolone(MP) and methylprednisone(11-KMP), which were supposed to be formed by deacetylation at the position of C-21, reduction at C-20, oxidation at C-11, or due to the bond cleavage between C-17 and C-20.

  • The thermally labile compounds with hydroxyl group generate the [${MNH_4}^+$] ion, [$MH^+$] and [$MH^+-OH$] ion by ion-molecule reaction in LC/TSP/MS. But these ions create the trouble in the estimation of molecular weight of an unknown compound because the margin of [${MNH_4}^+$] ion and [$MH^+$] ion is same to that of [$MH^+$] ion and [$MH^+-OH$] ion. If it is compensated for the results by using of the $CF_3COOD+NH_4OH$ LC eluent, the molecular weight of analyte will be able to be confirmed. And this study was tried to recognize whether the fragment ions of thermally labile compound are generated by electron impact or by thermal degradation in ion source.

Nutritional Value of Cottonseeds and It's Derived Products : I. Physical Fractionations and Proximate Composition

  • The study was conducted to determine physicochemical parameters in various physical fractions (linter, hull, kernel, oil and meal) of cottonseed of different varieties (MNH 147, CIM 240, NIAB 78, FH 87, CIM 109, MNH 93, FH 682, GOHAR 87, SLS 1 and B 557). Average components of linter, hull, and kernel in different varieties of cotton were 12.21, 28.24 and 70.42%, respectively. Average percentage of meal and oil was 48.97 and 22.09% in seed, and 69.28 and 30.72% in kernel, respectively. Maximum percentage of meal was recovered from variety CIM 240 and lowest in variety CIM 109. Statistical analysls revealed variety differences (p<0.05) in seed and it's components. Average contents of crude protein, crude fiber and ash was 22.31, 17.74 and 4.27% in seed, 2.85, 56.50 and 2.61% in hull; 32.62, 3.45 and 4.01 % in kernel; 47.15, 5.00 and 5.78% in meal, respectively. Average contents of Ca, p, Mg, K, Na and Cl were 0.09, 0.22, 0.26, 0.65, 0.009 and 0.035% in seed; 0.12, 0.07, 0.09, 0.51, 0.020 and 0.034% in hull and 0.16, 0.59, 0.32, 1.01, 0.03 and 0.07% in meal of different varieties of cotton, respectively. Fe, Zn, Cu and Mn were 141.35, 24.55, 186.50 and 27.12 mg/kg in seed; 158.48, 2.06, 74.60, and 22.17 mg/kg in hulls; and 167.62, 20.30, 185.83 and 20.67 mg/kg in meal, respectively. Significant varietal differences were observed in proximate composition and mineral contents of cottonseeds and derived products. Cottonseeds and their products of varieties FH 87, CIM 109 and MNH 93 showed higher nutrient density while lower was observed in varieties CIM 240, SLS I and FH 682.

  • The study was conducted to evaluate the nutritional value of seeds and various seed fractions of different varieties of cotton (MNH 147, CIM 240, NIAB 78, FH 87, CIM 109, MNH 93, FH 682, GOHAR 87, SLS I and B 557). Linter, hull, kernel and meal were obtained from cottonseed by physical and chemical methods. Free gossypol and available lysine contents of seed and it's fractions were determined. In vitro protein digestibility of cottonseed meal was also determined. Free gossypol and available lysine contents ranged between 0.22-2.26% and 0.64-1.32% in seed, and 0.03-0.29% and 1.38-2.36% in meal, respectively. FH 87 was highest both in free gossypol and available lysine content, and NIAB 78 was lowest in free gossypol content and FH 862 was lowest in available lysine content. In vitro protein digestibility of cottonseed meal ranged between 66,02-79,96%. Statistical analysis revealed significant (p<0.05) varietal differences in free gossypol, available lyslne and in vitro protein digestibility of cottonseed and derived products.

Raman Spectroscopic Studies of $YBa_2Cu_3O_7$ Coated Conductors ($YBa_2Cu_3O_7$ Coated Conductors의 Raman 분광학 연구)

  • We present results of Raman spectroscopic studies of superconducting $YBa_2Cu_3O_7$ (YBCO) coated conductors. Raman scattering is used to characterize optical phonon modes, oxygen content, c-axis misalignment, and second phases of the YBCO coated conductors at a micro scale. A two-dimensional mapping of Raman spectra with transport properties has been performed to elucidate the effect of local propertied on current path and superconducting phase. The information taken from the local measurement will be useful for optimizing the process condition.
