In this study, we tried to separate the photosensitizer that induces apoptosis of leukemia cells (U937) from perilla leaves. Perilla leaves (Perilla frutescens Britt var. japonica Hara) are a popular vegetable in Korea, being rich in vitamins (A and E), GABA, and minerals. Dried perilla leaves were extracted with methanol to separate the photosensitizer by various chromatographic techniques. The structure of the isolated compound (PL9443) was identified by 1D-NMR, 2D-NMR, and FAB-mass spectroscopy. Absorbance of the UV-Vis spectrum was highest at 410 nm and was confirmed by the 330, 410, and 668 nm. PL9443 compound was determined to be pheophorbide, an ethyl ester having a molecular weight of 620. It was identified as a derivative compound of pheophorbide structure when magnesium comes away from a porphyrin ring. Observation of morphological changes in U937 cells following cell death induced by treated PL9443 compound revealed representative phenomena of apoptosis only in light irradiation conditions (apoptotic body, vesicle formation). Results from examining the cytotoxicity of PL9443 substance against U937 cells showed that inhibition rates of the cell growth were 99.9% with the concentration of 0.32 nM PL9443. Also, the caspase-3/7 activity was 99% against U937 cells with the concentration of 0.08 nM of PL9443 substance. The result of the electrophoresis was that a DNA ladder was formed by the PL9443. The PL9443 compound is a promising lead compound as a photosensitizer for photodynamic therapy of cancer.
This article purports to clarify the doctrinal characteristics of the $Yog{\bar{a}}c{\bar{a}}ra$ school's hermeneutic interpretations of the "theory of the three turns of the Dharma Cakra" in the Saṃdbinirmocana-sūtra through early Indian $Yog{\bar{a}}c{\bar{a}}ra$ treatises such as the $Yog{\bar{a}}c{\bar{a}}rabb{\bar{u}}mi-vy{\bar{a}}kby{\bar{a}}$ and the. $Vy{\bar{a}}khy{\bar{a}}yukti$. It will probe how these interpretations apply co the theory of two truths or that of three natures($trisvabh{\bar{a}}va$) among the main doctrines of the $Yog{\bar{a}}c{\bar{a}}ra$ school. Especially, the peculiar characteristic of the "theory of the three turns of the Dharma Cakra" is such chat the thought of ${\acute{s}}{\bar{u}}nyat{\bar{a}}$ in the lineage of $Praj{\bar{n}}{\bar{a}}p{\bar{a}}ramita-s{\bar{u}}tras$ is regarded as incomplete, as the early school of Madhyamaka represented by $N{\bar{a}}g{\bar{a}}rjuna$ is conceived of as belonging to the second period of turn. Speaking of the further details of the "theory of the three turns of the Dharma Cakra", the $Yog{\bar{a}}c{\bar{a}}ra$ school subdivides the realm of saṃvṛti satya in $N{\bar{a}}g{\bar{a}}rjuna^{\prime}s$ theory of two truths; that is, it divides the saṃvṛti into merely linguistic existence and actual existence, and the thus-created structure of the theory of three natures on the basis of ocher-dependent nature(paratantra-$svabh{\bar{a}}va$) makes it possible to establish the doctrinal system of the thought of ${\acute{s}}{\bar{u}}nyat{\bar{a}}$ that is not subject to "nihilism or ${\acute{s}}{\bar{u}}nyat{\bar{a}}$ attached to evil." In effect, the above hermeneutic interpretation of the "theory of the three turns of the Dharma Cakra" is inherited into the structure of the $abh{\bar{u}}taparikalpa$ in the $Madhy{\bar{a}}nta-vibh{\bar{a}}ga$ so that, as seen in the commentary of Sthiramati, it is ascertained to apply to later doctrines through its secure establishment. To summarize its characteristics succinctly, firstly the $abh{\bar{u}}taparikalpa$ newly established as a saṃvṛti-satya is set up as the other-dependent nature, which is seen to have been set up particularly in order to sublate both the $Sarv{\bar{a}}stiv{\bar{a}}da^{\prime}s$ realist "view of being" and the Madhyamaka's "view of ${\acute{s}}{\bar{u}}nyat{\bar{a}}$" that impairs the ocher-dependent nature as a samvṛti-satya. In other words, according to the five kinds of views suggested in Sthiramati's commentary, the three natures are seen to be presented as the fundamental truth in order to unify all the doctrinal systems available ever since the beginning of Buddhism. Then, the theory of three natures is established principally on the basis of the $abh{\bar{u}}taparikalpa$, while the two truths of the $Yog{\bar{a}}c{\bar{a}}ra$ school are clearly ascertained to have been embedded in the structure of the $abh{\bar{u}}taparikalpa$. In fact, this might be understood to reflect the unique ontological view of reality or truth in the $Yog{\bar{a}}c{\bar{a}}ra$ School.

Traditionally, aqueous extracts of Puerariae radix had been used for the treatment of alcohol-related problems. In the present study, the effect of Puerariae radix on cell proliferation and expression of nitric oxide synthase (NOS) in the dentate gyrus of alcohol-intoxicated rats were investigated via 5-bromo-2' -deoxyuridine (BrdU) immunohistochemistry and nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) histochemistry, respectively. Sprague-Dawley rats weighing 150 ± 10 g were divided into four groups: the control group, the Puerarias radix-treated group, the alcohol-treated group, and the alcohol- and Puerariae radix-treated group. The numbers of both BrdU-positive and NADPH-d-positive cells in the dentate gyrus were inhibited significantly by alcohol administration, while Puerariae radix treatment was shown to increase those numbers. In this study, it was revealed that Puerariae radix possesses protective effect against alcohol-induced suppressed new cell formation and NOS expression in the dentate gyrus. Based on the results, it is possible that NO, which might play an important role in the regulation of cell proliferation, is a major target of the toxic effects of alcohol.
A dextransucrase (LcDS) gene from Leuconostoc citreum HJ-P4 has been amplified and cloned in E. coli. The LcDS gene consists of 4,431 nucleotides encoding 1,477 amino acid residues sharing 63-98% of amino acid sequence identities with other known dextransucrases from Leuc. mesenteroides. Interestingly, 0.1 mM of IPTG induction at $15^{\circ}C$ remarkably increased the LcDS productivity to 19,187 U/I culture broth, which was over 330-fold higher than that induced at $37^{\circ}C$. Optimal reaction temperature and pH of LcDS were determined as $35^{\circ}C$ and pH 5.5 in 20 mM sodium acetate buffer, respectively. Meanwhile, 0.1 mM $CaCl_2$ increased its activity to the maximum of 686 U/mg, which was 2.1-fold higher than that in the absence of calcium ion. Similar to the native Leuconostoc dextransucrase, recombinant LcDS could successfully produce a series of isomaltooligosaccharides from sucrose and maltose, on the basis of its transglycosylation activity.
Purpose. The purpose of this study were to investigate the effects of a 12-week brisk walking program on triglycerides (TG) and apolipoprotein B (Apo B) and to compare these effects to those of a brisk walking plus diet program in middle-aged overweight/obese ($BMI{\geq}23$) Korean women with hypertriglyceridemia. Method. This analysis was done with nineteen middle-aged overweight/obese Korean women who completed either the brisk walking program (9 women) or the brisk walking plus diet program (10 women) for 12 weeks. The brisk walking consisted of walking for 20 to 50 minutes/day at an intensity of 40 to 70% of heart rate reserve (HRR) for 3 to 6 days/week. The diet consisted of 60 minutes of group education and 20 to 30 minutes of individual counseling every week. TG and Apo B were assessed pre- and post-intervention. Results. TG and Apo B decreased significantly after the brisk walking program (Z = -2.31, p = 0.021; Z = -2.59, p = 0.009). TG and Apo B lowering effects of the brisk walking program were not significantly different from those of the brisk walking plus diet program (U = 37.0, P = 0.549; U = 42.0, P = 0.842). Conclusion. Brisk walking can be an effective intervention for overweight/obese middle-aged women with hypertriglyceridemia in reducing cardiovascular risk by lowering TG and Apo B levels. Adding diet to brisk walking may have no additional significant effects on changes in TG and Apo B.
Recently, the new technology for the recycling of waste plastics as an alternative he1 of blast furnace ha been developed. In this shldy, the test of plastics injection into a tuyere af the foundry blast furnace were carried out. The injection rate of plastics far this tcst facility war expressed as follows, ${W}_{s}=0.265\frac{{delta}PA}{U}_{g}$, where. Ws, AP, A and Ug are plastic conveying ratc (kgisec), pressure drop between feed hopper and blaqt pressure (alm), cross sectional area of conveying pipe (mi) and superficial velocity of transport air (mhzc) respcctiuely. From the results of semi-continuous test operation during 96 hours, the replacement raho mned out to be 1.38 according to the injection rate of 6. 4 kg-plasticsit-p. With increasing the rate of plaslics injcchon, the content of hydrogen in top gas became increased and the brick temperahlre at bzlly was also increased due to Lhe changes ot the combustion zone shape.
The purpose of this study was to investigate the effects of dietary habits and nutrient intakes on skin condition of female university students. The subjects were 95 female university students in Daegu. This study examined anthropometric measurements, dietary intake, food intake habits and skin condition (moisture, elasticity, sebum (U-zone, T-zone), pores, evenness and melanin pigmentation. The subjects in this study had an average age of 21.4, an average height and weight of 161.2 cm and 52.9 kg, respectively, and the average body mass index (BMI) was 20.3 $kg/m^2$. The means of skin elasticity, pores, evenness, and melanin pigmentation were within the normal range of women in their 20s and the skin moisture state was slightly lower than the normal range, which showed a dry tendency. The sebum in the U-zone and the T-zone was lower than the standard values, which showed less secretion in both areas. The subjects with high nut intake showed a better skin pore state (p < 0.05). Those with a high intake of instant foods showed a sig-nificantly low skin moisture and a significantly higher sebum rate (p < 0.05). The group with high skin elasticity and the group with less melanin pigmentation showed a significantly low animal protein intake (p < 0.05). The group with high skin elasticity also showed a significantly higher vitamin A and niacin intakes as well (p < 0.05). The group with high skin evenness showed a significantly lower animal lipid intake and a significantly higher vitamin E intake (p < 0.05). In addition, those with high skin elasticity and evenness as well as low melanin pigmentation showed a tendency of high intake of antioxidant vitamins, such as vitamin A, carotene, vitamin C or vitamin E. In summary, a moderate intake of plant-based products and also the antioxidant vitamins within such products seems to have a beneficial effect on maintaining a healthy and good skin condition.

Seaweed biomass was estimated in the rocky subtidal zone of Daejin on the mid-east coast of Korea using a destructive sampling method from February to November 2007. Monthly sampling was conducted at depths of 5, 10 and 15 m using 50 $\times$ 50 cm quadrats. A total of 73 species, including 5 chlorophytes, 25 phaeophytes, 43 rhodophytes and one seagrass were identified. Average seaweed biomass was 2,852.3 g wet wt. $m^{-2}$ and biomass values varied monthly from 1,189.8 g to 7,523.7 g. The species dominant in biomass were Laminaria japonica (481.7 g), Sargassum confusum (470.85 g), Undaria pinnatifida (422.57 g), Costaria costata (282.16 g), Odonthalia corymbifera (174.46 g), S. sagamianum (163.22 g) and S. horneri (122.18 g). The vertical distribution of algae was characterized by S. confusum and U. pinnatifida at 5 m, L. japonica and U. pinnatifida at 10 m, and O. corymbifera and L. japonica at 15 m depth. The C/P, R/P and (R+C)/P values were 0.20, 1.72 and 1.92, respectively.
Low density lipoproteins (LDL) play important roles in the pathogenesis of atherosclerosis. Diabetes is associated with accelerated atherosclerosis leading to cardiovascular disease in diabetic patients. Although LDL stimulates the proliferation of arterial smooth muscle cells (SMC), the mechanisms are not fully understood. We examined the effects of native LDL and glycated LDL on the extracellular signal-regulated kinase (ERK) pathway. Addition of native and glycated LDL to rat aorta SMCs (RASMCs) stimulated ERK phosphorylation. ERK phosphorylation was not affected by exposure to the $Ca^{2+}$ chelator BAPTA-AM but inhibition of protein kinase C (PKC) with GF109203X, inhibition of Src kinase with PP1 ($5{\mu}M$) and inhibition of phospholipase C (PLC) with U73122/U73343 ($5{\mu}M$) all reduced ERK phosphorylation in response to glycated LDL. In addition, pretreatment of the RASMCs with a cell-permeable mitogen-activated protein kinase kinase (MEK) inhibitor (PD98059, $5{\mu}M$) markedly decreased ERK phosphorylation in response to native and glycated LDL. These findings indicate that ERK phosphorylation in response to glycated LDL involves the activation of PKC, PLC, and MEK, but is independent of intracellular $Ca^{2+}$.
