Role of microRNAs in myogenesis and their effects on meat quality in pig - A review

  • The demand for food is increasing day by day because of the increasing global population. Therefore, meat, the easiest and largely available source of protein, needs to be produced in large amounts with good quality. The pork industry is a significant shareholder in fulfilling the global meat demands. Notably, myogenesis- development of muscles during embryogenesis- is a complex mechanism which culminates in meat production. But the molecular mechanisms which govern the myogenesis are less known. The involvement of miRNAs in myogenesis and meat quality, which depends on factors such as myofiber composition and intramuscular fat contents which determine the meat color, flavor, juiciness, and water holding capacity, are being extrapolated to increase both the quantity and quality of pork. Various kinds of microRNAs (miRNAs), miR-1, miR-21, miR22, miR-27, miR-34, miR-127, miR-133, miR-143, miR-155, miR-199, miR-206, miR-208, miR-378, and miR-432 play important roles in pig skeletal muscle development. Further, the quality of meat also depends upon myofiber which is developed through the expression of different kinds of miRNAs at different stages. This review will focus on the mechanism of myogenesis, the role of miRNAs in myogenesis, and meat quality with a focus on the pig.

Splitting Algorithms and Recovery Rules for Zero Anaphora Resolution in Korean Complex Sentences (한국어 복합문에서의 제로 대용어 처리를 위한 분해 알고리즘과 복원규칙)

  • Zero anaphora occurs frequently in Korean complex sentences, and it makes the interpretation of sentences difficult. This paper proposes splitting algorithms and zero anaphora recovery rules for the purpose of handling zero anaphora, and also presents a resolution methodology. The paper covers quotations, conjunctive sentences and embedded sentences out of the complex sentences shown in the newspaper articles, with an exclusion of embedded sentences of auxiliary verb. We manage the quotations using the equivalent noun phrase deletion rule according to subject person constraint, the nominalized embedded sentences using the equivalent noun phrase deletion rule, the adnominal embedded sentences using the relative noun phrase deletion rule and the conjunctive sentences using the conjunction reduction rule in reverse. The classified table of the endings which relate to a formation of the complex sentences is used for splitting the complex sentences, and the syntactic rules, applied when being omitted, are used in reverse for recovering zero anaphora. The presented rule showed the result of 83.53% in perfect resolution and 11.52% in partial resolution.

  • Osteoarthritis (OA) is the most common degenerative joint disorder in the elderly population. To identify OA-associated genetic variants and candidate genes, we conducted a genome-wide association study (GWAS). A total 3,793 samples (476 cases: wrist + knee and 3317 controls) from a community-based epidemiological study were genotyped using the Affymetrix SNP 5.0. An intronic SNP (rs4789934) in the TIMP2 (tissue inhibitor of metalloproteinase-2) showed the most significance with OA (odd ratio [OR] = 2.06, 95% confidence interval [CI] = 1.52-2.81, p = $4.01{\times}10^{-6}$). Furthermore, a poly-morphism (rs1352677) in the NKAIN2 ($Na^+/K^+$ transporting ATPase interacting 2) was suggestively associated with OA (OR = 1.43, CI = 1.22-1.66, p = $7.01{\times}10^{-6}$). The present study provides new insights into the identification of genetic predisposing factors for OA.

LINC00562 drives gastric cancer development by regulating miR-4636-AP1S3 axis

  • Dysregulation of certain long non-coding RNAs may facilitate tumor initiation and progression. However, numerous carcinogenesis-related long noncoding RNAs have not been characterized. The goal of this study was to elucidate the role of LINC00562 in gastric cancer (GC). The expression of LINC00562 was analyzed using real-time quantitative PCR and Western blotting. The proliferative capacity of GC cells was determined using Cell Counting Kit-8 and colony-formation assays. The migration of GC cells were evaluated using wound-healing assays. The apoptosis of GC cells was assessed by measuring the expression levels of apoptosis-related proteins (Bax and Bcl-2). Xenograft models in nude mice were constructed for in vivo functional analysis of LINC00562. The binding relationship between miR-4636 and LINC00562 or adaptor protein complex 1 sigma 3 (AP1S3), obtained from public databases, was confirmed using dual-luciferase and RNA-binding protein immunoprecipitation experiments. LINC00562 was expressed in GC cells at high levels. Knockdown of LINC00562 repressed GC cell growth and migration, promoted apoptosis in vitro, and inhibited tumor growth in nude mouse models. LINC00562 directly targeted miR-4636, and miR-4636 depletion restored the GC cell behavior inhibited by LINC00562 absence. AP1S3, an oncogene, binds to miR-4636. MiR-4636 downregulation increased AP1S3 level, restoring GC cell malignant behaviors inhibited by AP1S3 downregulation. Thus, LINC00562 exerts carcinogenic effects on GC development by targeting miR-4636-mediated AP1S3 signaling.

Effect of NaCl/Monosodium Glutamate (MSG) Mixture on the Sensorial Properties and Quality Characteristics of Model Meat Products

  • Sodium chloride is an important ingredient added to most of foods which contributes to flavor enhancement and food preservation but excess intake of sodium chloride may also cause various diseases such as heart diseases, osteoporosis and so on. Therefore, this study was carried out to investigate the effect of monosodium glutamate (MSG) as a salty flavor enhancer on the quality and sensorial properties of the NaCl/MSG complex and actual food system. For characterizing the spray-dried NaCl/MSG complex, surface dimension, morphology, rheology, and saltiness intensity were estimated by increasing MSG (0-2.0%) levels at a fixed NaCl concentration (2.0%). MSG levels had no effect of the characteristics of the NaCl/MSG complex, although the addition of MSG increased the surface dimension of the NaCl/MSG complex significantly (p<0.05). Furthermore, the effect of MSG on enhancing the salty flavor was not observed in the solution of the NaCl/MSG complex. In the case of an actual food system, model meat products (pork patties) were prepared by replacing NaCl with MSG. MSG enhanced the salty flavor, thereby increasing overall acceptability of pork patties. Replacement of NaCl with MSG (<1.0%) did not result in negative sensorial properties of pork patties, although quality deterioration such as high cooking loss was found. Nevertheless, MSG had a potential application in meat product formulation as a salty flavor enhancer or a partial NaCl replacer when meat products were supplemented with binding agents.

Risk Assessment of Airborne Toxic Metals in Thejon Industrial Complex (대전공단지역의 대기중 독성금속에 대한 위해도 평가)

  • The research centers on the concentration profile and risk assessment of toxic metals for ambient air in Taejon industrial complex. Airborne concentrations of each toxic metal for risk assessment were obtained from 2-year sampling by high volume air sampler and analysis by ICP-MS and ICP-AES in the complex. The long-term arithmetic mean of human carcinogen, arsenic, hexavalent chromium and nickel subsulfide was 5.53, 2.16 and $3.46ng/m^3$ while the mean of probable human carcinogen, beryllium, cadmium and lead was 0.08, 2.35, $293.29ng/m^3$, respectively. And the long-term arithmetic mean concentration of non-carcinogenic metal, manganese was $55.91ng/m^3$. The point risk estimate for the inhalation of carcinogenic metals was $3.6{\times}10^{-5}$, which was higher than a risk standard of $10^{-5}$. About 75% of the cancer risk was to the inhalation of human carcinogen, arsenic. Thus, it is necessary to properly manage arsenic risk in Taejon industrial complex. The point hazard index by the inhalation of manganese was 1.1. Therefore, an investigation into Taejon industrial complex is needed to obtain more fine long-term concentration data for airborne non-carcinogenic metals including manganese.

Synthesis and Characterization of Red Electrophosphorescent Polymers Containing Pendant Iridium(III) Complex Moieties

  • A series of fluorene-carbazole copolymers containing the pendant phosphor chromophore $Ir(absn)_2(acac)$ (absn: 2-(1-naphthyl)benzothiazole; acac: acetylacetone) were designed and synthesized via Yamamoto coupling. In the film state, these copolymers exhibited absorption and emission peaks at approximately 389 and 426 nm, respectively, which originated from the fluorene backbone. However, in electroluminescent (EL) devices, a significantly red-shifted emission at approximately 611 nm was observed, which was attributed to the pendant iridium(III) complex. Using these copolymers as a single emission layer, polymer light-emitting devices with ITO/PEDOT:PSS/polymer:DNTPD/TmPyPb/LiF/Al configurations exhibited a saturated red emission at 611 nm. The attached iridium(III) complex had a significant effect on the EL performance. A maximum luminous efficiency of 0.85 cd/A, maximum external quantum efficiency of 0.77, maximum power efficiency of 0.48 lm/W, and maximum luminance of 883 $cd/m^2$ were achieved from a device fabricated with the copolymer containing the iridium(III) complex in a 2% molar ratio.

WORKGLOW: A P2P-based Web Service Orchestration Supporting Complex Workflow Patterns (복잡한 워크플로우 패턴들을 지원하는 P2P 기반 웹 서비스 오케스트레이션)

  • Web services are considered as the critical component in the business plans of corporations as they offers the potential for creating highly dynamic and versatile distributed applications that span across business boundaries. Web Service Orchestration studies composition of already-existing web services to create new value-added services. The composite web services could be executed in a centralized or peer-to-peer(P2P) orchestration model. Compared with centralized-orchestration model, the P2P-based orchestration model provides better scalability, reliability, and performance for the overall services. However, recent P2P-orchestration solutions have limitation in supporting complex workflow patterns. Therefore, they could not effectively handle sophisticated business workflow, which contains complex workflow patterns. In this paper, we propose the WORKGLOW system, which can deal with complex workflow patterns while it is able to perform composite services in P2P orchestration manner. Comparing with centralized orchestration systems, the WORKGLOW brings up more business logic advantages, better performance, and higher flexibility with only a little overhead.

  • Ribose-5-phosphate isomerase A (RpiA) plays an important role in interconverting between ribose-5-phosphate (R5P) and ribulose-5-phosphate in the pentose phosphate pathway and the Calvin cycle. We have determined the crystal structures of the open form RpiA from Vibrio vulnificus YJ106 (VvRpiA) in complex with the R5P and the closed form with arabinose-5-phosphate (A5P) in parallel with the apo VvRpiA at $2.0{\AA}$ resolution. VvRpiA is highly similar to Escherichia coli RpiA, and the VvRpiA-R5P complex strongly resembles the E. coli RpiA-A5P complex. Interestingly, unlike the E. coli RpiA-A5P complex, the position of A5P in the VvRpiA-A5P complex reveals a different position than the R5P binding mode. VvRpiA-A5P has a sugar ring inside the binding pocket and a phosphate group outside the binding pocket: By contrast, the sugar ring of A5P interacts with the Asp4, Lys7, Ser30, Asp118, and Lys121 residues; the phosphate group of A5P interacts with two water molecules, W51 and W82.

Effects of NaCl Replacement with Gamma-Aminobutyric acid (GABA) on the Quality Characteristics and Sensorial Properties of Model Meat Products

  • This study investigated the effects of ${\gamma}$-aminobutylic acid (GABA) on the quality and sensorial properties of both the GABA/NaCl complex and model meat products. GABA/NaCl complex was prepared by spray-drying, and the surface dimensions, morphology, rheology, and saltiness were characterized. For model meat products, pork patties were prepared by replacing NaCl with GABA. For characteristics of the complex, increasing GABA concentration increased the surface dimensions of the complex. However, GABA did not affect the rheological properties of solutions containing the complex. The addition of 2% GABA exhibited significantly higher saltiness than the control (no GABA treatment). In the case of pork patties, sensory testing indicated that the addition of GABA decreased the saltiness intensity. Both the intensity of juiciness and tenderness of patties containing GABA also scored lower than the control, based on the NaCl reduction. These results were consistent with the quality characteristics (cooking loss and texture profile analysis). Nevertheless, overall acceptability of the pork patties showed that up to 1.5%, patties containing GABA did not significantly differ from the control. Consequently, the results indicated that GABA has a potential application in meat products, but also manifested a deterioration of quality by the NaCl reduction, which warrants further exploration.
