A Rare Case of Canine Pericardial Malignant Mesothelioma: Clinicopathologic Findings, Diagnostic Investigations, and Clinical Course with Epirubicin Treatment

  • An adult male dog was presented for hemorrhagic pericardial effusion. Echocardiography and computed tomography revealed nodule-like lesions on the pericardium. Cytology of pericardial effusion and excisional pericardial lesions indicated neoplastic effusion. Histopathology indicated an inflamed neoplasm with a primary differential diagnosis of hemangiosarcoma and malignant mesothelioma (MM). Immunohistochemistry showed that atypical cells were positive for cytokeratin and vimentin, but negative for CD31, strongly favoring pericardial MM. Postoperative NT-proBNP level remained increased, which led to the administration of epirubicin to minimize potential cardiotoxicity. During the 4 cycles of epirubicin treatment, a total cumulative dose of 108 mg/m2 was administrated and no effusion recurrence was observed. After a month post-completion of chemotherapy, however, pleural effusion was detected with cardiac masses. The owner requested no further diagnostic investigations and chemotherapy. Due to deteriorating conditions, the dog died 132 days after the first presentation. Our case is the first notable attempt to treat canine malignant mesothelioma with epirubicin, providing the clinicopathologic, diagnostic routine, and clinical course of the affected dog.

Antitumor and Free Radical-Scavenging Activities of Various Extract Fractions of Fruits and Leaves from Prunus mume (금매와 매화 잎 추출물의 프리라디칼 억제 활성 및 항암 효과)

  • This study investigated the free radical-scavenging and antitumor activities of hot water, water, acetone, ethanol, ethyl acetate, chloroform, and hexane extracts of fruits and leaves from Prunus mume. The various extracts were evaluated for their total polyphenol, flavonoid, and tannin contents, scavenging activities by DPPH and ABTS analyses, reducing power, protective effects against oxidative stress in L-132 cells, and antitumor activities against A549, HeLa, and U87 cancer cells. Ethanol extracts of fruits and leaves showed the highest total polyphenol content (336.41 and 523 mg GAE/100 g, respectively). DPPH and ABTS radical-scavenging activities increased according to concentration of fruit. DPPH radical-scavenging activity of ethanol extracts from leaves was 65.48% at $200{\mu}g/mL$. All extract fractions of leaves showed high ABTS radical-scavenging activities. The reducing power activities increased according to increasing concentration of fruits and leaves. All extracts of leaves performed better than extracts of fruits in terms of protective effects against oxidative stress in L-132 cells. Ethyl acetate, chloroform, hexane, ethanol extracts of fruits and leaves showed anticancer activities against A549, HeLa, and U87 cancer cells. However, ethanol extracts of fruits and leaves showed no toxicity in normal cells (BNLCL2). This study suggests that antioxidant activities of fruits and leaves from P. mume depend on polyphenol contents. Thus, fruits and leaves from P. mume can be useful as natural antioxidant compounds.

Preparation and Characterization of Jochung, a Grain Syrup, with Apple (사과 첨가 조청의 제조 및 특성)

  • This study was performed to investigate the effect of apple and maltitol as ingredients on the quality of Jochung, a grain syrup. Four kinds of Jochung products were prepared from steamed-rice, apple juice, heated-apple sarcocarp (at $70^{\circ}C$, 60 min), and a mixture (sarcocarp : maltitol=5:1, w/w) by saccharifying (at $55^{\circ}C$, 8 hrs) with a malt (100 g/500 g rice), mixing the ingredients (steamed-rice : ingredient=5:5, w/w), filtering, and heating the filtrate (at $95^{\circ}C$, 2 hrs): product (A) with apple juice added before saccharified, product (B) with apple juice added after saccharified, product (C) with heated-apple sarcocarp added after saccharified, and product (D) with the mixture added after saccharified. The product (D) had the lowest pH value ($4.60\pm0.01$) of any other products. The contents of reducing sugar and total phenolic compound were the highest in the product (A) among all the products, which comprised $68.10\pm6.71$% and $7.36\pm0.85$ mg/g, respectively, resulting in good quality. The solidity and the dextrose equivalence had the highest value in the product (B) and the product (C), respectively. The malic acid content ($4.10\pm0.02$%) of the product (D) was the highest of any other organic acids identified by HPLC. Hunter L, a, and b values of the product (D) were the highest compared to other products. In sensory evaluation, the product (A) had generally higher score in all sensory attributes. It was concluded from the chemical and sensory evaluation that adding the apple juice before saccharified might be an effective method for manufacturing good quality rice-Jochung.

Fundamental Study on Recycling as Construction Material of Dredged Harbor Soil Mixed with Clay and Glass Frit (점토와 유리프리트를 혼합한 항만준설토의 건축자재 재활용에 관한 기초연구)

  • This research was performed to evaluate the recycling feasibility as a construction material of dredged harbor soil mixed with clay and glass frit. Concentration of heavy metals of the dredged soil from D harbor was severly high, showing Zn of 526.0~13,150.1 mg/kg. The dredged soil was maily composed of 48.30 wt% $SiO_2$, 16.60 wt% $Al_2O_3$, 10.10 wt% CaO, 7.75 wt% $Fe_2O_3$. The clay and the glass frit contained 70.82 wt% $SiO_2$ and $Al_2O_3$ 18.78 wt%, and 71.75 wt% $SiO_2$, 13.99 wt% CaO, 8.51 wt% $Na_2O$, respectively. After adding 10~40 wt% to the clay and sintering them at $1,000^{\circ}C$ or $1,100^{\circ}C$, the compressive strength of the sintered specimens showed $132.6{\sim}178.5kgf/cm^2$ or $581.2{\sim}793.7kgf/cm^2$, respectively. In case of SC46 with the addition 40 wt% of the dredged soil to the clay, the compressive strength ($793.7kgf/cm^2$) of specimen sintered at $1,100^{\circ}C$ was over 5 times higher than that at $1,000^{\circ}C$. The specimen mixed with 40 wt% of dredged soil, 60 wt% of clay and 1 wt% of glass frit satisfied the 1st grade standard for clay brick by KS L 4201. The results of all specimens by Korean Standard Leaching Test also satisfied the standard criteria.

The Decreased Expression of Fbxw7 E3 Ligase Mediated by Cancer Upregulated Gene 2 Confers Cancer Stem Cell-like Phenotypes (CUG2 유전자에 의하여 감소된 FBXW7 E3 ligase 발현이 유사-종양줄기세포 표현형을 유도)

  • The detailed mechanism by which cancer upregulated gene 2 (CUG2) overexpression induces cancer stem cell-like phenotypes is not fully understood. The downregulation of FBXW7 E3 ligase, a tumor suppressor known for its proteolytic regulation of oncogenic proteins such as cyclin E, c-Myc, Notch, and Yap1, has been frequently reported in several types of tumor tissues, including those in the large intestine, cervix, and stomach. Therefore, we investigated whether FBXW7 is involved in CUG2-induced oncogenesis. In this study, the decreased expression of FBXW7 was examined in human lung adenocarcinoma A549 (A549-CUG2) and human bronchial BEAS-2B cells (BEAS-CUG2) overexpressing CUG2 and compared with control cells stably expressing an empty vector (A549-Vec or BEAS-Vec). Treatment with MG132 (a proteosome inhibitor) prevented the degradation of FBXW7 and Yap1 proteins, which are substrates of the FBXW7 E3 ligase. To address the role of Fbxw7 in the development of cancer stem cell (CSC) phenotypes, we suppressed Fbxw7 protein levels using its siRNA. We observed that decreased levels of FBXW7 enhanced cell migration, invasion, and spheroid size and number in A549-Vec and BEAS-Vec cells. The enforced expression of FBXW7 produced the opposite results in A549-CUG2 and BEAS-CUG2 cells. Furthermore, the downregulation of FBXW7 elevated the activities of EGFR, Akt, and ERK1/2 and upregulated β-catenin, Yap1, and NEK2, while the enforced expression of FBXW7 generated the opposite results. We thus propose that FBXW7 downregulation induced by CUG2 confers CSC-like phenotypes through the upregulation of both the EGFR-ERK1/2 and β-catenin-Yap1-NEK2 signaling pathways.

Effect of Polluted Irrigation Water on the Rice Growth and the Grain Quality (오염(汚染) 관개수(灌漑水)가 벼 생육(生育) 및 미질(米質)에 미치는 영향(影響))

  • This study was conducted to establish a safe cultivation technique of rice in the area irrigated with polluted water. The changes of water and grain quality were investigated in the two paddy soils which are located in the adjacent of Geumho river in 1991. The results obtained in this study are as follows : 1. The qualities of irrigation water were shown to be in the range of 6.7~7.4 in pH, 21.3~52.8ppm in COD. 3.2~5.3ppm in $NH_4-N$ and 1.6~6.0ppm in $PO_4$, respectively. Concentration of COD and $NH_4-N$ were over the standard levels. Therefore, the water pollution was mainly caused by organic waste matters. 2. Ranges of the Soil pH of Gyeongsan and Gyuam series were 5.6~6.0, 6.1~6.3 respectively. The contents of avaiable $P_2O_5$ and $SiO_2$ were high in the silicate treatment plots among other plots. 3. Degree of irrigation water pollution of Geumho river showed the highest peak in June. 4. Plant height, number of panicle and yield of rice grain were not decreased by the diminution of nitrogen fertilizer application. 5. The $Mg/(K{\times}N)$ ratio and Gel consistency of milled rice were increased in the nitrogen decreasing plot, but the alkali digestability value and protein content were decreased. Therefore, it was desirable to decrease the amount of nitrogen fertilizer application.

Characteristics of Heavy Metals (Ba, Cr) Distribution in Soil (토양 중 중금속(Ba, Cr)의 분포특성 평가)

  • This study was performed to provide fundamental data to establish the new soil pollution standards and the soil contamination management plans in a rational manner. The distribution characteristics of new soil contaminants such as barium (Ba) and chromium (Cr) in soils (n=140) were investigated in relation to land-use classification and geological features. Also, the sequential extraction test was conducted to evaluate fate and mobility of new soil contaminants. The soil samples taken from 140 sites were analyzed to survey distribution levels of selected new soil contaminants. The average concentration and range for hazardous metals (Ba, Cr) were Ba 128.946 (26.757~489.587) mg/kg, Cr 30.121 (2.579~132.783) mg/kg. Based on land use classification, the highest Ba concentration was found in factory soils, followed by dry field and park soils, while Cr concentration was highest in rice paddy soils, followed by dry field and factory soils. Within 10 geological units investigated the highest Ba and Cr concentrations were observed in the soils from Okcheon group and metamorphic rocks, respectively. The BCR (European Community Bureau of Reference) sequential extraction was conducted to identify chemical distributional existence of 2 elements of soils from each geological unit. Ba in soils is mainly assumed to exists as reducible form (such as BaSO4, BaCO3) and Cr in soils mainly is assumed to exist as residual form (such as Cr2O3, CrxFe1-x(OH)3(x < 1)).

Rainfastness of 5 Fungicides on the Leaf Surface of Hot Pepper (고추잎 표면에서 5종 살균제의 내우성)

  • In order to elucidate the relationship between the rainfastness of fungicides and their water solubilities, the fungicide residues on the leaf surface of hot pepper was assessed and compared after the drop-wise applications of fungicide solutions on leaf surface followed by artificial raining. As the raining was progressed after application of aqueous acetone solution of fungicides, the residue levels of fungicides were drop rapid at the early stage of raining, but the decreasing rates of residue level were slowed down thereafter. The initial rainfastness was reversely proportional to the water solubilities of the fungicides. Whole amount of dimethomorph residue, which water solubility is 18 mg/L, was washed off by 2.5 mm of raining. Although WP formulations of fungicides showed remarkable decreases of rainfastness compared to the aqueous acetone formulations, the fungicides having low water solubility showed better rainfastness. Chlorothalonil and mepanipyrim suspension concentrates was better in rainfastness than their WP formulation, and the rainfastness of mepanipyrim suspension was reversely proportional to the median diameter of suspension particles in the range of 1 to 4 ${\mu}m$. In brief, the rainfastness of 5 fungicides tested on the pepper leaf was, in the early stage of raining, closely related to water solubility. But, as the raining is progressed, the effect of the unknown factor, which is related with the particle size of fungicides, becomes serious.

Phytoremediation of Soils Contaminated with Heavy Metal by Hosta longipes in Urban Shade (도시 내 음지 중금속 오염지에 대한 비비추의 식물정화 효과)

  • Hosta longipes is one of the most popular ornamental perennials in use in Korea today, and is mainly used as a groundcover plant in urban shaded places. In this study, the pytoremediation effect of Hosta longipes was tested using four concentrations (Control, 100, 250 and $500mg{\cdot}kg^{-1}$) of Cd, Pb and Zn in soil. The plants were planted in $300mm{\times}200mm{\times}250mm$ drainless-containers, which were filled with a artificial amended soil for 7 months. The results showed that the contents of heavy metals cadmium and lead in the shoot of Hosta longipes increased with increasing heavy metal concentration levels exception of zinc. The amount of zinc, cadmium, and lead accumulated in roots were increased with heavy metal concentration levels up in soil. The shoot/root ratios(TF; translocation factor) values were found to be more than 80% of total Zn, Cd, and Pb take up by Hosta longipes. These results indicated that root is the major part for accumulation of heavy metal. The removal contents of zinc, cadmium, and lead increased significantly with the increasing heavy metal concentration in the soil, which was planted with Hosta longipes. The heavy metal concentration accumulation in plant/soil ratios (BF; bioaccumulation factor) values for three metals were found to be more than 30% in cadmium and lead, but lower in zinc treatments. The different responses of Hosta longipes suggest that in heavy metal contaminated soils the plant adsorbs available metals depending on the concentration soils in which they are present. Therefore, this species can be an efficient phytoremediator for soils contaminated with cadmium and lead in urban shaded places.

Clinical Experience of Continuous Epidural Analgesia Using Baxter $Infusor^{(R)}$ (Baxter $Infusor^{(R)}$를 이용한 경막외 진통제 지속 주입)

  • Recently a non-electronic, disposable and portable infusor(Baxter infusor with patient control module, Baxter health care Co., Deerfield IL 60015 USA: BI $\bar{c}$ PCM) has been developed that will deliver both a continuous drug infusion as well as allow the patient to deliver extra doses of medication on a demand basis under predetermined limitation of analgesics. Patients may also not require as high analgesic dose rate to control pain when the acceptable and tolerable level of pain relief can be maintained by this device. From April l99l, we have used a total l93 units of BI $\bar{c}$ PCM. These units consisting of two components which one made by a balloon reservoir(capacity 65 ml, flow rate 0.5 ml/hr) to store medication and to regulate the pump power(490 torr), and another two PCMs to regulate additional analgesic administration by patients demand at intervals of 1S minutes and 60 minutes. The dose administered to the patient can be varied by changing the concentration of the infusate within the balloon reservoir. These devices were utilized for the pain control of 44 patients. These patients were divided into two groups. Twenty seven cases had cancer pain and 17 cases had non-cancer pain. The Touhy needle(No. l8 G.) tip was inserted into the epidural space and was used to guide the catheter to the spinal nerve level corresponding to the most painful area. The device was connected to the opposite site of the catheter tip and was filled with 60 ml of mixture solution such as 0.5% bupivacaine 15 ml, morphine HCl 10 mg, trazodone 10 ml, Tridol 3 ml and normal saline 31 ml were administed as the initial dose. When the initial dose was less effective, the next dose could be varied by increasing the concentration of bupivacaine, by adding more morphine (5~10 mg), and by reducing the volume of normal saline. Using these modules of drug self administration, we experienced the following: 1) Improvement of patient's self titration of analgesic requirement was provided. 2) The patients anxiety with pain recurrence resulting from delays in administering pain control medication was decreased significantly. 3) The working load accompanying with the single bolus injection as the usual method was reduced remarkably. 4) There was urinary retention in 5 cases and pruritus in 4 eases which developed as side effects but respiratory depression and vomiting was not encountered in a single case.
