Biological Activity Review of Mume Fructus and Processed Mume Fructus (오매(烏梅)와 증숙 오매(蒸熟 烏梅)의 생리활성 연구)

  • Objectives: Presently Mume Fructus (MF) undergoes fumigation, which produces benzo[a]pyrene. As a primary analysis with the aims to minimize the production of benzo[a]pyrene and to suggest standards for processing the MF, the steaming method was chosen among the various processing methods, and reviewed through a series of experiments.Methods: Methods:Pitted and un-pitted MF were steamed and processed into samples. After testing level of benzo[a]pyrene, the samples were analyzed for amount of polyphenol and flavonoids. Scavenging activities of the samples for the DPPH and ABTS radicals were tested. In order to measure anti-inflammatory effects of the samples, cell survival rate was investigated using CCK-8 Assay. Also, water extracts of dried and steamed MF were administered to the RAW 264.7 cells to compare expressions of NO, PGE2, IL-1β, and TNF-α. In addition, anti-diarrhea effects of the herbal medicine were tested on animal models with diarrhea induced by MgSO4 and Castor oil.Results: Regardless of pitting, processed MF contained no benzo[a]pyrene. Anti-oxidation effect increased in relation to the frequency of steaming process. However, extracts of dried and steamed MF suppressed different kinds of inflammation factors, and extract of dried MF showed superior anti-diarrhea effect than extract of steamed MF.Conclusions: It is suggested that steaming method of MF is recommended for processing the herbal medicine without the production of benzo[a]pyrene. But regarding that dried and steamed MF showed differences in their anti-oxidative, anti-inflammatory, and anti-diarrhea effects, it is recommended to perform further researches on different efficacies of MF according to their processing methods.

Phase 4, Post-Marketing Safety Surveillance of the MF59-Adjuvanted Influenza Vaccines FLUAD® and VANTAFLU® in South Korean Subjects Aged ≥65 Years

  • Backgroud: Influenza vaccination is recommended for adults aged ${\geq}65$ years as they are at high risk of significant morbidity and mortality. This open-label, multicenter, post-marketing surveillance study assessed the safety of the MF59-adjuvanted trivalent inactivated subunit influenza vaccine, which is marketed as $FLUAD^{(R)}$ and $VANTAFLU^{(R)}$, in South Korean subjects aged ${\geq}65$ years. Materials and Methods: Solicited local and systemic adverse events (AEs) were collected from day 1 to 4 of the study. All unsolicited AEs and serious AEs (SAEs) were recorded from day 1 until study termination (day 29). Results: Of the 770 subjects enrolled ($FLUAD^{(R)}$, n = 389; $VANTAFLU^{(R)}$, n = 381), 39% overall experienced any solicited AE. Local AEs were reported by 33% of subjects overall; with the most common events being injection-site pain (30%) and tenderness (27%). Systemic AEs were reported by 19% of subjects overall with the most common events being myalgia (11%) and fatigue (8%). Conclusion: These results show that the MF59-adjuvanted influenza vaccine known as $FLUAD^{(R)}$ or $VANTAFLU^{(R)}$ had acceptable safety profiles in older adults (aged ${\geq}65$ years) in South Korea.

Toxicity of Hematoporphyrin-Coated Magnetic Ferrofluid in Rats

  • The purpose of this study was to investigate the toxicity of hematoporphyrin-coated magnetic ferrofluid (HP-MF) through intravenous administration in Sprague-Dawley rats. Each group was treated with either saline, or the HP-MF at 0.5, 1, 1.5, 2 and 4 ml/kg body weight (b.w.) for the observation of survival rate, clinical symptoms, laboratory values and histopathological findings. In this study, HP-MF was evaluated for the survival rates, symptoms, laboratory values and histopathological examination after treatments. The result revealed that the animals in the group of HP-MF at 2 and 4 ml/kg b.w. showed some lethality. In serum biochemistry, the levels of AST, ALT and ALP were increased in the MF and HP-MF treated groups. However, histopathological examination for the suspected organs showed no evidence of hepatotoxicity and nephrotoxicity of typical iron poisoning. Though the toxicity of HP-MF was higher than that of HP, long retention of hematoporphyrin via HP-MF provides additional benefit over conventional hematoporphyrin. HP-MF could be utilized as a potential photodynamic agent in cancer therapy. It is suggested to develop an efficient external magnetic device to attract hematoporphyrin in the target site, thereby enabling to administering a small amount of HP-MF.

Antioxidant Activities of Various Solvent Extracts from Canola Meal (추출용매별 canola meal의 항산화 활성)

  • This study was performed to investigate antioxidant activity of various solvent extracts (methanol, ethanol, acetone and water) from canola meal. As the concentration of all extracts increased, antioxidant activities increased gradually. Among solvent extracts from canola meal, methanol extract showed the highest antioxidant activities with $EC_{50}$ values of 0.9, 3.5, 1.3, and 8.5 mg/mL for DPPH radical scavenging ability, ABTS radical scavenging ability, reducing power, and chelating ability, respectively, and it had the highest total phenolics and total flavonoids contents. Furthermore, methanol extract was fractionated into three fractions (MF 1, MF 2, and MF 3) using Sephadex LH-20 column. The highest antioxidant activity was found on MF 2 for DPPH radical scavenging ability, on MF 3 for ABTS radical scavenging ability and reducing power, and on MF 1 for chelating ability at the same concentration of 1.5 mg/mL. The results indicated that each fraction containing various antioxidant materials seemed to have different influence at each antioxidant activity.

Effect of Neck Position on Muscle Fatigue during Shaker Exercise

  • Objective: The aim of this study is to find the influence on muscle fatigue by changing an angle of neck while Shaker exercise. Background: Shaker exercise is commonly applied to dysphagia patients for strengthen suprahyoid muscle(SHM) and relaxing upper esophageal sphincter. Method: Experiments were conducted by measuring muscle fatigue with surface electromyography(SEMG) in case of neck $15^{\circ}$ and $50^{\circ}$ flexion, in addition to original method. For the study 30 undergraduate students were participated as subjects. Muscle fatigue was measured with variations of median frequency(MF) and median frequency slope(MFS) by attaching SEMG to 3 muscles. At the same time, perceived exertion was measured by using Borg's rating of perceived exertion(RPE). Results: A MF was increased when low angle in SHM, both male and female. It means that muscle fatigue was significantly decreased(p<.05). RPE was significantly increased when low angle(p<.01). Perceived exertion explained 15.2% of the variation of MF in SHM. And regression equation was, MF in SHM = 59.918 + (4,910 * RPE). Conclusion: The outcome shows that it is possible to use the efficient method which makes muscle fatigue decrease by reducing the angle of neck flexion. Application: The results might contribute to develop the effective Shaker exercise method.

Clinical Efficacy of Bunny Multifocal Intraocular Lens after Cataract Surgery (버니 다초점인공수정체의 임상적 효용성)

  • Purpose: To compare the postoperative clinical outcomes after cataract surgery and implantation using the BunnyLens $MF^{(R)}$ and $ReSTOR^{(R)}$ multifocal intraocular lenses. Methods: Sixty-five eyes implanted with multifocal intraocular lenses were divided into two groups involving either $ReSTOR^{(R)}$ (39 eyes) or BunnyLens $MF^{(R)}$ (26 eyes) lenses. In these two groups, the distant and near visual acuity, astigmatism, spherical equivalent, and contrast sensitivity test were examined at preoperative and postoperative 1 week, 8 weeks, and 24 weeks. We compared the clinical efficacy between the two groups before and after cataract surgery using statistical analysis. Results: The mean value of distant and near visual acuity, and spherical equivalent of both groups after intraocular lens implantation were significantly improved, compared with the preoperative values (p < 0.05), while there was no significant differences between the two groups (p > 0.05). The means of postoperative distant and near visual acuity, astigmatism, and contrast sensitivity test were not significantly different between $ReSTOR^{(R)}$ and BunnyLens $MF^{(R)}$ lenses (p > 0.05). The mean numeric error of spherical equivalent at the final postoperative 24 weeks was $-0.17{\pm}0.50$ diopters (D) for the $ReSTOR^{(R)}$ lenses and $-0.34{\pm}0.52D$ for the BunnyLens $MF^{(R)}$ lenses (p > 0.05). Conclusions: BunnyLens $MF^{(R)}$ and $ReSTOR^{(R)}$ lenses showed no significant difference in clinical efficacy, including distance and near vision, spherical equivalent error, and contrast sensitivity test after cataract surgery. However, it should be noted that BunnyLens $MF^{(R)}$ lenses had a tendency toward myopic shift compared with $ReSTOR^{(R)}$ lenses.

Effect of $N_2$-backflushing Time in Carbon Ceramic UF & MF System for Paper Wastewater Treatment

  • The wastewater discharged from a paper plant was filtrated by 3 kinds of tubular carbon ceramic UF and MF membranes with $N_2$-backflushing. The filtration time (FT) was fixed at 8 min or 16 min, and $N_2$-backflushing time (BT) was changed in 0${\~}$60 sec. The optimal condition was discussed in the viewpoints of total permeate volume ($V_T$), dimensionless permeate flux (J/Jo) and resistance of membrane fouling ($R_f$). In the viewpoints of $V_T$, J/Jo and $R_f$, the optimal $N_2$-BT was 40 sec at both FT for M9 (MWCO: 300,000 Daltons) and C005 ($0.05{\mu}m$) membranes. However, for C010 ($0.1{\mu}m$) it was 10 sec at FT=8 min, and 20 sec at FT=16 min in the viewpoints of J/Jo and $R_f$, and 5 sec at both FT in the viewpoints of $V_T$. It means that the short $N_2$-BT could reduce the membrane fouling and recover the permeate flux sufficiently for MF membrane having a large pore size as C010. Average rejection rates of pollutants were higher than $99.0\%$ for turbidity and $22.8{\~}59.6\%$ for $COD_{cr}$, but rejection rates of total dissolved solid (TDS) were lower than $8.9\%$. Therefore, the low turbidity water purified in our system could be reused for paper process.

Production of Fructose Corn Syrup by Glucose Isomerase (Glucose isomerase 효소를 이용한 이성화당(과당) 생산에 관한 연구)

  • Two strains S-P and S-P-2, both Streptomyces sp., have been isolated and were found to have relatively high specific enzyme activity compared to other organisms reported. The specific activity of the enzyme produced from these two strains were 0.25 and 0.2 international units respectively. The productivity of the enzyme achieved was about 50 IU/l/hr. Glucose isomerase form these strains was found to be stable under the temperature of heat treatment (at $65^{\circ}C$) for fixation of enzyme inside the dell. This organism has an advantage in that it did not require toxic metalic ion for enzyme activity and could utilize xylan in leu of xylose as an inducer. The optimal temperature and pH of enzymatic reaction purpose of using these data for the optimal operation and designing of enzyme reactor system. The reaction mechanism was found to follow the single substrate reversible reaction kinetics. The kinetic constants determined experimentally are : $K_{mf}=0.33M,\;K_{mb}=1.0M,\;V_{mf}=0.88{\mu}mole\;per\;min.,\;V_{mb}= 2.96{\mu}mole\;per\;min.\;and\;K_{eq}=0.74.
