In vitro Effect of Aloe on the Respiratory Burst Activity of Olive Flounder (Paralichthys olivaceus) Leucocytes (알로에가 in vitro에서 넙치 백혈구의 호흡폭발에 미치는 영향)

  • The immunostimulating effect of aloe on respiratory burst activity was investigated by measurements of the chemiluminescent responses (CL) of olive flounder (Paralichthys olivaceus) kidney phagocytes in vitro. The phagocytes incubated with 0.5 and $1{\mu}g/ml$ of aloe for 24 hours showed significantly increased respiratory burst activities compared to control. The result of this study suggests that aloe can be used as an immunostimulant for olive flounder culture.

  • In this work, the solvent permeation and separation performance of organic solvent nanofiltration (OSN) membranes were evaluated. Particularly, the PuraMem (PM) series developed for nonpolar solvents were analyzed and tested in dead-end filtration system. PM membranes exhibited higher permeance for nonpolar solvents compared to polar solvents, and their rejection data did not follow conventional trends with respect to solute size. The data showed that simple solution-diffusion model is not suitable to describe the OSN membrane behavior, and a better solvent-solute-membrane interaction parameter must be developed.

Developmental Competence of Intrafollicular Oocytes Derived from Preantral Follicle Culture with Different Protocols after Parthenogenetic Activation

  • This study was conducted to improve efficiency of a follicle culture system without reducing developmental competence of intrafollicular oocytes. Preantral follicles (100 to $125{\mu}m$ in diameter) of F1 hybrid (B6CBAF1) mice were cultured singly for 216 h in modified ${\alpha}$-MEM-glutamax medium, to which 2.5 IU/ml hCG and epidermal growth factor was added 16 h prior to the end of culture. Medium change was either performed three times (54 h interval), twice (72 h interval), once (108 h interval), or not at all (216 h interval). Maturation (progression to the metaphase II stage) of intrafollicular oocytes was detected from 4 days after culture in the three-times change treatment, while all treatments yielded mature oocytes from day 5 of culture. Compared with the three-times change, decreasing the change frequency to once did not reduce the capacity to begin maturation (germinal vesicle breakdown of 82 to 86%), to mature (78 to 79%) and to develop into blastocysts after parthenogenetic activation (29 to 32%). Morphological parameters were similar among these treatments. Except for the no medium change treatment, similar colony-forming activity of inner cell mass cells after culturing of blastocysts in leukemia inhibitory factor-containing medium was detected, while the morphology of the colony-forming cells deteriorated in the change-once treatment compared with the change twice or three-times. In conclusion, the efficiency of the preantral follicle culture system could be improved by reducing frequency of medium change up to a 72 h interval (three times in total 216 h culture) without decreasing developmental competence of oocytes.

Quality of Life and Related Factors in Patients with Chronic Hepatitis B* (만성 B형 간염환자의 삶의 질과 제요인)

  • Purpose: This study aimed to investigate LDQOL (Liver Disease Quality of Life) and its related factors of patients with Hepatitis B. Method: A cross-sectional descriptive study. The LDQOL was formally translated to Korean and reliability was examined. One hundred thirty eight patients following gastroenterology outpatient clinic of S. hospital in Seoul, Korea participated. Results: The mean age of the patients was 45.43 years and 87.7% were men. The mean score of LDQOL was 77.34, and mean scores of subscale were followed; symptom of liver disease (SxLD) (82.12), effect of liver disease (ELD) (25.50), concentration (Conc) (84.47), memory (Mem) (83.24), health discomfort, (HD) (75.18), sexual function (SFun) (75.71), sexual problem (SProb) (84.70), Loneliness (85.50), Hopeless (67.43), and stigma of liver disease (SLD) (91.64). Women had a lower LDQOL score for Loneliness (p=.034), and over 45 year-old patients had a lower LDQOL overall score (p=.000). Patients who were HBV carriers, or who had Chronic Hepatitis B or Liver Cirrhosis reported lower QOL respectively(p=.032). Conclusion: Although the liver disease itself seemed to be stable, patients with HBV experienced poor QOL in ELD, SProb, SFun, and Hopeless. Therefore nursing interventions in these aspects are needed.

In Vitro Culture of the Isolated Mouse Preantral Follicles: Effect of Different Types of FSH and Vitrification (생쥐 Preantral 난포의 체외배양: FSH의 종류와 농도 및 초자화 냉동보존의 영향)

  • Objectives: 1) To compare the efficacy of urofollitropin (Follimon) to that of recombinant human FSH (rhFSH) on the growth and maturation of mouse early preantral follicles in vitro, and 2) effect of vitrification on the growth and maturation of preantral follicles and oocytes . Methods: Isolated early preantra1 follicles (100-130 ${\mu}m$ diameter) were cultured for 12 days in 20 ${\mu}l$ ${\alpha}$-MEM media drop under the mineral oil. Follimon or rhFSH was added to the culture medium at various concentrations (0, 10, 100, and 1000 mIU/ml). Results: With Follimon, the dose of 10 mIU/ml showed the best follicle survival, growth, and MIl rate of oocyte than the other concentrations. Whereas the optimal dose of rhFSH was 100 mIU/ml. Despite the different optimal doses, the efficacy of two different FSHs on the follicle growth and maturation was similar. Isolated mouse preantral follicles were cryopreserved by vitrification and cultured in vitro for 12 days with 100 mIU/ml rhFSH. Despite the decreased follicular survival rate after thawing, the follicular growth and maturation rate of its oocyte were comparable to those of the fresh follicle. Conclusion: Results from the present study revealed that 1) the optimal doses of Follimon and rhFSH for in-vitro culture of mouse follicles are different, and 2) the frozen-thawed follicles develop normally after vitrification.

Anti-inflammatory Effect of Yongseollan on the LPS-activated RAW 264.7 Cells

  • This study was conducted to evaluate the inhibitory effects of Yongseollan(YSL) on the production of nitric oxide (NO) and the expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-activated RAW264.7 cells. YSL is tropical plant originating from Mexico. The biological activity of this plant is not yet evaluated systematically. The aim of the present work is to investigate a potential anti-inflammatory activity of YSL. The RAW264.7 cells were cultured in D MEM/F12 medium for 24 hrs. After serum starvation, cells were treated with YSL for 1 hr, followed by stimulating NO production with a LPS. We found that YSL has an inhibitory effect on the production of NO, iNOS expression and $phospho-I{\kappa}B$ expression. YSL also inhibited tumor necrosis factor $(TNF)-{\alpha}$, interleukin (IL)-6, and $IL-1{\beta}$. Moreover, YSL inhibited cyclooxygenase (COX)-2 expression and prostanglandin E2 (PGE2). These findings showed that YSL could have some anti-inflammatory effects which might play a role in therapy in Gram-negative bacterial infections.

A method using artificial neural networks to morphologically assess mouse blastocyst quality

  • Background: Morphologically classifying embryos is important for numerous laboratory techniques, which range from basic methods to methods for assisted reproduction. However, the standard method currently used for classification is subjective and depends on an embryologist's prior training. Thus, our work was aimed at developing software to classify morphological quality for blastocysts based on digital images. Methods: The developed methodology is suitable for the assistance of the embryologist on the task of analyzing blastocysts. The software uses artificial neural network techniques as a machine learning technique. These networks analyze both visual variables extracted from an image and biological features for an embryo. Results: After the training process the final accuracy of the system using this method was 95%. To aid the end-users in operating this system, we developed a graphical user interface that can be used to produce a quality assessment based on a previously trained artificial neural network. Conclusions: This process has a high potential for applicability because it can be adapted to additional species with greater economic appeal (human beings and cattle). Based on an objective assessment (without personal bias from the embryologist) and with high reproducibility between samples or different clinics and laboratories, this method will facilitate such classification in the future as an alternative practice for assessing embryo morphologies.

Effect of Musical Rhythm on the CPR education of Adolescent (청소년 대상 심폐소생술 교육에서 음악적 리듬 적용의 교육효과)

  • We conducted this study to verify effect of applying musical rhythm during the cardiopulmonary resuscitation(CPR) education. From June to October, 2013, we recruited participants of CPR education among mem's high school students. We divided total 57 participants to 2 groups. For experimental group we use musical rhythm during cardiac compression practice to assist adequate cardiac compression rate and for control group do not apply musical rhythm. The group with musical rhythm show more adequate compression rate(16.4sec) than non-rhythmic group(14.4sec). 90% of rhythmic group keep adequate compression rate, but only 44% of non-rhythmic group keep adequate rate. In conclusion, applying musical rhythm to CPR education is more effective to keep correct cardiac compression rate.

BcI-2 Over-expression Reduced the Serum Dependency and Improved the Nutrient Metabolism in a NS0 Cells Culture

  • The over-expression of Bcl-2 has greatly improved the culture period, specific growth rate, and maximum viable cell density of NS0 cells culture under low serum condition. Further analysis of these data suggests that a saturation model of the Monod type can be used to represent the relationships of specific growth rate and initial serum concentration. The ${\mu}_{max}$ and $K_s$ for the Bcl-2 cell line is $0.927day^{-1}\;and\;0.947\%(v/v)$ respectively, which are $21\%$ greate and $7\%$ lower respectively than its control counterpart. Study on the amino acid supplementation revealed that Bcl-2 cell lines possess greater improvement in the specific growth rate and maximum viable cell density compared to the control cell lines. A further increase in the amino acid supplementation has resulted a $17\%$ decrease in specific growth rate and no improvement in maximum viable cell density in the control culture. However, the Bcl-2 cell line exhibited a better growth characteristic in this culture condition compared to that of control cell lines. The higher specific growth rate and maximum viable cell density of the Bcl-2 cell line in medium fortified with serum and MEM EM suggested a more efficient nutrient metabolism compared to that in the control cell line. The low serum and amino acid utilisation rate and the higher cell yield may prove to be important in the development of serum/protein free culture.

  • Si wafer direct bonding(SDB) technology is very attractive for both Si-on-insulator(SOI) electronic devices and MEMS applications. This paper presents on pre-bonding according to HF pre-treatment conditions in Si wafer direct bonding. The characteristics of bonded sample were measured under different bonding conditions of HF concentration and applied pressure. The bonding strength was evaluated by tensile strength method. A bond characteristic on the interface was analyzed by using FT-IR, and surface roughness according to HF concentration was analyzed by AFM. Si-F bonds on Si surface after HF pre-treatment are replaced by Si-OH during a DI water rinse. Consequently, hydrophobic wafer was bonded by hydrogen bonding of Si-OH$\cdots$(HOH$\cdots$HOH$\cdots$HOH)$\cdots$OH-Si. The pre-bonding strength depends on the HF pre-treatment condition before pre-bonding. (Min : $2.4kgf/cm^2{\sim}$Max : $14.9kgf/cm^2$)
