TPA로 분화된 U937 세포에서 사람 세포거대바이러스에 의한 c-jun Promoter 활성도의 변화 (The Change of c-jun Promoter Activity in TPA-Induced U937 Cells Infected with Human Cytomegalovirus (HCMV))

  • Transient transfection assay has been done to evaluate whether the c-jun activation would be prerequisite to the induction of permissiveness against human cytomegalovirus using in vitro cell model in which U937 has been induced to express CD11b and CD14 to become potential monocyte/macrophage cells by TPA treatment. U937 cells were treated with $10\;{\mu}M$, $50\;{\mu}M$ or $100\;{\mu}M$ of TPA. The cell morphology change was observed and the expression of the CD11b and CD14 was confirmed by FACS. Differentiated cells were transfected with pJLuc reporter vector which contained the wild type murine c-jun promoter spanning the SP1, CTF, ATF/CREB and MEF-2 binding sites upstream of the firefly luciferase gene. After 48 hrs of transfection, the cells were infected with HCMV Towne strain and the luciferase activity was assessed at 1 hand 4 h pi. The transfection assay showed no activation of the c-jun promoter at 1 h pi, instead, it showed 2 times increase of the its activity at 4 h pi. There was no difference of the c-jun promoter activation between TPA treated and untreated U937 cells, implying that c-jun activation might not be prerequisite for allowing cells to be premissive to HCMV, although HCMV infection itself could activate c-jun promoter.

Genome-Wide Association Study of Bone Mineral Density in Korean Men

  • Osteoporosis is a medical condition of global concern, with increasing incidence in both sexes. Bone mineral density (BMD), a highly heritable trait, has been proven a useful diagnostic factor in predicting fracture. Because medical information is lacking about male osteoporotic genetics, we conducted a genome-wide association study of BMD in Korean men. With 1,176 participants, we analyzed 4,414,664 single nucleotide polymorphisms (SNPs) after genomic imputation, and identified five SNPs and three loci correlated with bone density and strength. Multivariate linear regression models were applied to adjust for age and body mass index interference. Rs17124500 ($p=6.42{\times}10^{-7}$), rs34594869 ($p=6.53{\times}10^{-7}$) and rs17124504 ($p=6.53{\times}10^{-7}$) in 14q31.3 and rs140155614 ($p=8.64{\times}10^{-7}$) in 15q25.1 were significantly associated with lumbar spine BMD (LS-BMD), while rs111822233 ($p=6.35{\times}10^{-7}$) was linked with the femur total BMD (FT-BMD). Additionally, we analyzed the relationship between BMD and five genes previously identified in Korean men. Rs61382873 (p = 0.0009) in LRP5, rs9567003 (p = 0.0033) in TNFSF11 and rs9935828 (p = 0.0248) in FOXL1 were observed for LS-BMD. Furthermore, rs33997547 (p = 0.0057) in ZBTB and rs1664496 (p = 0.0012) in MEF2C were found to influence FT-BMD and rs61769193 (p = 0.0114) in ZBTB to influence femur neck BMD. We identified five SNPs and three genomic regions, associated with BMD. The significance of our results lies in the discovery of new loci, while also affirming a previously significant locus, as potential osteoporotic factors in the Korean male population.

제브라피쉬 근육성장에서의 carnosic acid의 효과 (Effects of Carnosic Acid on Muscle Growth in Zebrafish (Danio rerio))

  • 로즈마리의 주요 성분인 carnosic acid는 carnosol, rosmarinic acid, ursolic acid 등과 같은 폴리페놀의 한 성분으로 다양한 생리활성 기능이 보고되어 있다. 본 연구에서는 로즈마리 유래 폴리페놀인 carnosic acid가 제브라피쉬 근육성장에 미치는 영향을 근육 내 주사와 사료를 통해서 확인해 보았다. 근육 내 주사 실험을 통해서 CA는 제브라 피쉬의 근육 내 단백질 함량을 증가시키고 중성지방의 함량을 감소시켰다. 또한 조직학적 분석 결과 근섬유의 평균 면적이 커지는 근섬유의 과비대 효과를 나타내었다. 사료 실험 결과 근육 내 단백질 및 중성지방의 함량에는 영향을 미치지 않았으며 조직학적 분석 결과 근육 내 주사 실험에서와 마찬가지로 근 섬유의 과비대를 유도하였다.

돼지 웅성 생식선 줄기세포의 체외배양기법 개발 (Development of In Vitro Culture System for Male Germline Stem Cells in Porcine)

  • Spermatogonial stem cells(SSCs) only are responsible for the generation of progeny and for the transmission of genetic information to the next generation in male. Other in vitro studies have cultured SSCs for proliferation, differentiation, and genetic modification in mouse and rat. Currently, information regarding in vitro culture of porcine Germline Stem Cell(GSC) such as gonocyte or SSC is limited and is in need of further studies. Therefore, in this study, we report development of a successful culture system for gonocytes of neonatal porcine testes. Testis cells were extracted from $10{\sim}14$-day-old pigs. These cells were harvested using enzymatic digestion, and the harvested cells were purified with combination of percoll, laminin, and gelatin selection techniques. The most effective culture system of porcine gonocytes was established through trial experiments which made a comparison between different feeder cells, medium, serum concentrations, temperatures, and $O_2$ tensions. Taken together, the optimal condition was established using C166 or Mouse Embryonic Fibroblast(MEF) feeder cell, Rat Serum Free Medium(RSFM), 0% serum concentration, $37^{\circ}C$ temperature, and $O_2$ 20% tension. Although we discovered the optimal culture condition for proliferation of porcine gonocytes, the gonocyte colonies ceased to expand after one month. These results suggest inadequate acquirement of ingredients essential for long term culture of porcine GSCs. Consequently, further study should be conducted to establish a successful long-term culture system for porcine GSCs by introducing various growth factors or nutrients.

미소안면침의 얼굴 축소 및 피부상태 개선 효과 (The Effect of Miso Facial Acupuncture on Facial Reduction and Improvement of Skin Condition)

  • Objectives : The purpose of this study is to evaluate the effects of Miso facial acupuncture on facial reduction and improvement of skin condition. Methods : From March to May 2012, 20 women in their twenties to forties with no facial diseases were recruited. Miso facial acupuncture was performed on the both side of their face twice a week, total seven times. The 3D face photographs of each participant were checked 5 times ; before treatment, after 1st treatment, 4th treatment, 7th treatment and 10 days from the last treatment using RS-400FL(Morpheus Co. Korea) and REAL FACE. The surface distances of the 4 lines on the face were measured using Renai MEF program. The improvement of skin condition was evaluated by water content, trans epidermal water loss(TEWL), erythema index and melanin index using MPA 5. They were checked 4 times ; before treatment, 4th treatment, 7th treatment and after 10 days from the last treatment. We analyzed data using Paired t-test(p<0.05) and Wilcoxon signed rank test(p<0.05). Results : One case was dropped out due to pain and edema after 4th treatment. The surface distances of the 4 lines on face were significantly reduced and maintained after 10 days from last treatment. Water content was significantly increased and maintained after 10 days from last treatment. TEWL was significantly decreased. Erythema index was significantly decreased from the 3rd evaluations. Melanin index was slight decreased with no significance at the 2nd evaluations. Conclusions : These results show that Miso facial acupuncture can reduce sizes of the face and improve skin condition such as water content, TEWL and erythema index except melanin index.
