삼백초(三白草)와 어성초(魚腥草)의 암세포(癌細胞)에 대한 독성억제(毒性抑制) 효과(效果) (Cytotoxic Effects of Methanol Extracts from Saururus Chinensis Bail and Herba Houttuyniae on Cancer Cell Lines)

  • This study was performed to determine the cytotoxic effect of methanol extract from medicinal plants. 1. The cell viability was determined by MTT method. Their cytotoxic activities against three cancer cell lines such as A549, MDA-MB-231 and SNU-C4 cell line were tested. 2. Among them, The methanol extract of Saururus Ohinensis Bail showed the strongest cytotoxic effect against SNU-C4 cells. These results suggest that the methanol extract of Saururus Ohinensis Bail possessed a potential antitumorous agent. 3. The free radical scavenging activity using DPPH method was the strongest of Saururus Chinensis Bail extract.

조각자(皂角刺) 추출물이 인간 유래 유방암 세포의 유전자 발현에 미치는 영향 (Effects of Gleditsiae Spina(GS) on Gene Expression of Human Breast Cancer Cells)

  • Purpose: Gleditsiae spina (GS) has been used to treat patients with several diseases such as carbuncle, swelling and parasites. Recently GS is known to have anticancer activity in abdominal solid tumor, but the effects of GS on breast cancers is not clarified. For these reasons, we investigated effects of Gleditsiae spina (GS) on gene expression of human breast cancer cells. Methods: We investigated the effects of GS on proliferation of breast cancer cell line, MDA-MB-231. In addition, the genetic profile for the effect of GS on breast cancer cells was measured using microarray technique, and the functional analysis on these genes was conducted. Results: Total 1,434 genes were up-regulated and 2,483 genes down-regulated in the cells treated with GS. Genes induced or suppressed by GS were all mainly concerned with metabolic process, regulation of biological process and protein binding. The network of total protein interactions was measured using cytoscape program, and some key molecules that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conclusion: These results suggest possibility of GS as anti-cancer drug for breast cancer, and also suggest that related mechanisms are involved in regulation of intra-cellular metabolism in breast cancer cells.

Ethanol but not Aqueous Extracts of Tubers of Sauromatum Giganteum(Engl.) Cusimano and Hett Inhibit Cancer Cell Proliferation

  • Background: Both alcohol and aqueous extracts of Sauromatum giganteum(Engl.) Cusimano and Hett, the dried root tuber of which is named Baifuzi in Chinese, have been used for folklore treatment of cancer in Northeast of China. However, little is known about which is most suitable to the cancer therapy. Materials and Methods: Serum pharmacology and MTT assays were adopted to detect the effects of ethanol and aqueous extracts of Sauromatum giganteum(Engl.) Cusimano and Hett, prepared by heat reflux methods, on proliferation of different cancer cells. Results: Cancer cells treated with medium supplemented with 10%, 20%, 40% serum(v/v) containing ethanol extract had a decline in viability, with inhibition rates of 7.69%, 21.8%, 41.9% in MCF-7 cells, 42.8%, 48.1%, 51.8% in SGC-7901 cells, 44.1%, 49.2%, 53.7% in SMMC-7721 cells, 6.8%, 15.2%, 39.8% in HepG2 cells, 7.57%, 16.3%, 36.2% in HeLa cells, 6.24%, 12.5%, 27.4% in A549 cells, and 7.20%, 17.5%, 31.3% in MDA-MB-231 cells, respectively. Viability in the aqueous extract groups was no different with that of controls. Conclusions: An ethanol extract of Sauromatum giganteum(Engl.) Cusimano and Hett inhibited the proliferation of SMMC-7721, SGC-7901 and MCF-7 cells, which supports the use of alcoholic but not aqueous extracts for control of sensive cancers, which might include hepatocarcinoma, gastric cancer and breast cancer.

Iris Nertschinskia Ethanol Extract Differentially Induces Cytotoxicity in Human Breast Cancer Cells Depending on AKT1/2 Activity

  • Recently, we reported that an ethanol extract of Iris nertschinskia induces p53-dependent apoptosis in the MCF7 human breast cancer cell line. However, the detailed mechanisms were not fully explored. Here, we demonstrate another aspect of the activity of I. nertschinskia in breast cancer cells. We compared the response to an ethanol extract of I. nertschinskia in two different human breast cancer cell lines, Hs578Tand MDA-MB231, respectively with relatively low and high AKT1/2 activity by trypan blue exclusion assay and FACS analysis. Knockdown of endogenous AKT1 or AKT2 in breast cancer cells by RNA interference determined the sensitivity to I. nertschinskia ethanol extract compared to control cells. The I. nertschinskia ethanol extract induced cell death in a manner that depended on the level of phosphorylated AKT1/2 protein and was associated with a significant increase in the sub-G1 cell population, indicative of apoptosis. Our results indicate that an ethanol extract of I. nertschinskia differentially induces cell death in breast cancer cells depending on their level of phosphorylated AKT1/2.

삼백초 추출물의 항암활성과 카드뮴에 대한 독성억제효과 (Effects of Saururus Chinensis Baill Extracts on Anticancer Activity and Cadmium Induced Cytotoxicity)

These results suggest that the methanol extract of Saururus Chinensis Baill. possessed a potential antitumorous agent. 2. In vitro the antitoxic activity of ethanol extract of Saururus Chinensis Bail on NIH 3T3 fibroblasts was evaluate by the MTT (3-(4,5-dimethyl- thiazol-2-yl)-2,5-diphenyl-2H-tetra캐lium bromide) and SRB (sulforhodamine B protein) assays. The number of NIH 3T3 fibroblasts were increased and tend to regenerate. These results suggest that Saururus Chinensis Bail extract retains a potential antitoxic activity. 3. Complexation of Cd (II) ion with ligands such as quercetin has been determined by UV-vis spectrophotometric method in tris buffer solution at various pH. It was found that only 1 : 1 Cd-complex is formed by the interaction between catechol moiety in ring B and cadmium [Cd(II)] in aqueous solution. The spectral parameters for Cd-complex were determined by Beer's law at various pH. It has shown that 1 : 1 Cd-complex has a maximum absorbance and red shift by the alkaline pH.
