Comparison of Isoflavone Contents and Antioxidant Effect in Cheonggukjang with Black Soybean Cultivars by Bacillus subtilis CSY191

  • BACKGROUND: Soybeans are the rich sources of isoflavones. To date, the changes of isoflavone contents in various black soybeans cheonggukjang during fermentation by Bacillus subtilis CSY191 has not been investigated.METHODS AND RESULTS: This study investigated the changes of total phenolic and isoflavone contents and antioxidant effects during cheonggukjang fermentation made with four black soybean (BS) cultivars including Cheongja, Cheongja#3, Geomjeong#5, and Ilpumgeomjeong with a potential probiotic Bacillus subtilis CSY191. The total phenolic contents, isoflavone-malonylglycoside and -aglycone contents, and antioxidant activity were increased in cheonggukjang at 48 h fermentation, while the content of isoflavone-glycosides was decreased during cheonggukjang fermentation. In particular, the Cheongja#3 soybean fermented at 37℃ for 48 h displayed the highest antioxidant activities, compared to those of the other BS cultivars tested. Also, the highest levels of total phenolic, daidzein, glycitein, and genistein were present at concentrations of 17.28 mg/g, 283.7 g/g, 39.9 g/g, and 13.2 g/g at the end of Cheongja#3 soybean fermentation.CONCLUSION: The results from this study suggested that the enhanced antioxidant activity of cheonggukjang of BS might be related to increased levels of total phenolic, isoflavon-aglycone, and malonyl-glycoside contents achieved during fermentation. Furthermore, fermented Cheongja#3 soybean showed the highest levels of enhanced antioxidant activities than the other BS cultivars.

Comparison of Surface and Air Temperature depending on Cover Materials in Playground (운동장의 피복 유형별 표면 및 대기온도 비교)

  • Playground is frequently used for physical and sports activity by students as well as by common people, which is constructed with various cover materials on the ground. This research surveyed the surface temperature in Kyungpook National University Sangju campus playground which is covered with various cover materials [bare field, zoysiagrass (Zoysia japonica) turf field, urethane track, concrete field, epoxy field and artificial turf field] in Sangju, Gyeongsangbukdo, Korea. Temperature was measured 4 times per day at 09:00, 12:00, 15:00, and 18:00 from May to October 2014 in surface and 1 m height above the ground. Surface temperature was different, depending on cover materials and survey time. Bare field and zoysiagrass turf field was lower surface temperature than other sites. Higher surface temperature site was different depending on survey time. Urethane track and artificial turf field was hotter than other sites at 12 and 15 hours, however concrete and epoxy field was hot at 18 hours. One meter above ground temperature was the highest in artificial turf field except at 18 hours. So natural turf, zoysiagrass playground will increase the athletic performance by reduce the surface and above ground temperature.

Expression of Heat Shock Protein and Antioxidant Genes in Rice Leaf Under Heat Stress

  • We have previously investigated the proteome changes of rice leaves under heat stress (Lee et al. in Proteomics 2007a, 7:3369-3383), wherein a group of antioxidant proteins and heat shock proteins (HSPs) were found to be regulated differently. The present study focuses on the biochemical changes and gene expression profiles of heat shock protein and antioxidant genes in rice leaves in response to heat stress ($42^{\circ}C$) during a wide range of exposure times. The results show that hydrogen peroxide and proline contents increased significantly, suggesting an oxidative burst and osmotic imbalance under heat stress. The mRNA levels of chaperone 60, HSP70, HSP100, chloroplastic HSP26, and mitochondrial small HSP responded rapidly and showed maximum expression after 0.5 or 2 h under heat stress. Transcript levels of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and Cu-Zn superoxide dismutase (Cu-Zn SOD) showed a rapid and marked accumulation upon heat stress. While prolonged exposure to heat stress resulted in increased transcript levels of monodehydroascorbate reductase, peroxidase, glyoxalase 1, glutathione reductase, thioredoxin peroxidase, 2-Cysteine peroxiredoxin, and nucleoside diphosphate kinase 1, while the transcription of catalase was suppressed. Consistent with their changes in gene expression, the enzyme activities of APX and DHAR also increased significantly following exposure to heat stress. These results suggest that oxidative stress is usually caused by heat stress, and plants apply complex HSP- and antioxidant-mediated defense mechanisms to cope with heat stress.

  • e-seal은 RFID(Radio Frequency IDentification) 기술을 사용하여 원격에서 자동으로 봉인상태를 확인할 수 있는 컨테이너 봉인 장치를 말한다. RFID의 특징상 반도체 칩에 기록된 정보를 제 삼자가 쉽게 판독 및 변조할 수 있다는 취약점으로 인하여 활성화되지 못하고 있는 실정이다. ISO에서는 RFID의 취약점을 보안하기 위한 표준작업(ISO 18185)을 진행 중이다. 이 중, ISO 18185-4는 e-seal에 저장되는 자료나 리더와의 RF통신에서 데이터 보호를 위한 표준이다. 이와 관련된 연구로는 인증 프로토콜과 ISO 18185-4를 위한 보고서로 제출된 보안 프로토콜이 있다. 제안된 e-seal 보안 프로토콜을 적용하기 위해서는 e-seal과 리더 간의 데이터를 암/복호화할 키가 필요하지만, 키 서버를 통해 전달받은 마스터 키를 데이터 암/복호화 키로 바로 사용하는 것은 보안 상의 문제점을 야기할 수 있기 때문에 PRF(Pseudo Random Function)을 이용하여 마스터 키로부터 MTK(Mutual Transient Key)를 유도하고, MTK를 암/복호화 키로 사용해야 한다. 기존의 PRF는 일방향 해시 함수(MD5, SHA 등)를 기반으로 하는 HMAC[2. 3]을 일반적으로 사용하였다. 그러나 일방향 해시 함수는 e-seal과 같은 제한된 자원을 갖는 환경에 적합하지 않다. 따라서, 본 논문에서는 e-seal 보안 프로토콜을 위한 효율적인 PRF을 제안한다. 기존의 일방향 해시 함수 기반이 아닌 블록 암호화 알고리즘을 기반으로 하는 MAC을 이용하여 PRF을 보다 효율적으로 구현하였고, 블록 암호화 알고리즘은 AES를 선택 합성체 $GF((2^4)^2)$을 통해 하드웨어 모듈을 최적화 하였다. AES를 기반으로 하는 MAC은 HMAC에 비해 면적 및 처리율에서 뛰어난 결과를 보여주었다.<0.01).이상의 연구 결과, cook-chill생산 시 녹차 추출물의 첨가가 미생물적 품질유지에 효과가 있다고 사료되는 바 본 연구결과를 기초로 급식소에서 음식 생산 시 녹차 추출물 및 천연 항균성 물질 첨가에 따른 미생물적 품질 및 관능적 품질검사를 통한 레시피 개발에 관한 지속적인 연구가 수행되어야 하겠다.다.다리다보니 점심시간을 활용할 수 없게 되는 문제점에 대한 재검토가 필요하다. 따라서 차후 학교급식의 안전성 확보를 위한 급식환경 개선의 일환으로 식당공간 확보 시 신속한 시간 내에 급식이 가능하도록 넓은 공간과 쾌적한 환경의 식당 조성에 대해 관심을 기울여야 할 것으로 사료된다. 이상 여부를 반영하는 임상증상의 빈도가 높은 청소년기 남녀 중학생의 경우 아침과 저녁의 결식빈도 및 외식과 간식의 빈도가 높았고, 아침식사의 질과 체형만족도가 낮은 것으로 나타나 청소년의 건강과 식습관 및 체형만족도가 상호 관련성이 높은 것으로 나타났다. 따라서 본 연구 결과는 성장기 청소년의 건강 유지를 위하여 바람직한 식습관의 중요성을 재인식할 수 있었으며, 올바른 식습관 확립을 위한 영양교육의 중요성이 재확인되었다.경제적일 것으로 판단된다.er 90 % of good relative dynamic modulus of elasticity due to fineness of formation caused by the increase of the unit powder content and the improvement of flowability, without regard to the replacement of crushed stone fines. Effects of Drinking Ion Water on Performance, Egg Quality and Blood Biochemical Composition of Laying Hens under High Temperature Conditions (고온환경에서 이온수의 급수가 산란계의 생산성, 계란 품질 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate the effect of drinking ion water on the performance, egg quality, and serum biochemical properties of laying hens in a high-temperature environment. A total of 180 laying hens with equal egg production rates were randomly divided into three treatments with five replicates. Treatments were applied from 24 to 27 weeks of age and included the supply of ion water (IW), tap water (TW), and cold water (CW). The results showed that the egg production rate, egg weight, and feed intake of hens were significantly (P<0.01) improved in the IW group compared with those in the TW group but did not differ from those in the CW group. Egg quality parameters such as albumen height, Haugh unit, and eggshell thickness were found to be higher in the IW than in the TW group (P<0.05); however, no significant differences were observed in comparison with the CW group. Serum Ca and P concentrations tended to increase (P<0.10) in the IW group, and serum high-density lipoprotein cholesterol and albumin levels were greater (P<0.05) in response to IW than those to TW. This study provides evidence that the supply of IW can relieve the high-temperature stress during the summer. In particular, because the improved effects of the IW were confirmed to be similar to those of CW, it shows potential as a method to manage the drinking water of hens in the summer season along with the supply of CW.

  • 오존 경보제와 예보제의 실시가 오존 오염에 관한 일반인의 관심을 증폭시키는 중요한 계기가 되었음에도 우리 연구계와 정책 당국의 대응은 이에 훨씬 못미치고 있다. 1990년 이래 학회지 관련 논문 편수는 연간 1∼3편에 그치고 있고, 다른 이차오염 현상과 달리 정책 당국은 여전히 지원을 고려하지 않고 있다. 오존 연구가 뿌리를 내리지 못한 가운데 강행된 경보제와 예보제의 와중에 책임을 떠맡은 지방자치단체는 조급하게 노련한 예보 모델을 찾고 있고 충분한 검토도 없이 외국의 사례에서 대책을 구하고 있다. 논란이 되고 있는 오존 예보의 부정확성은 모델 선택의 문제가 아니라 모델 이용이 잘못된 때문이다. 현재 오존 대책의 일환으로 검토되고 있는 차량 부제 실시와 같은 단기 대책은 역효과를 낼 수 있다는 연구결과도 있다. 선진국의 오존 연구는 도시 규모에서 지역 규모로 확대되고 있고, 구츰 등 액상반응의 효과 규명과 1 kin 이하 미세 변화 모델링이 시도되고 있고, 지구 단위 대류권 실험의 일환으로 태평양 상공의 오존과 오존 전구물질의 대기화학이 조사되고 있다. 우리는 우선 우리의 오존 문제를 정확히 이해하도록 노력하여야 한다 합리적 계획 아래 체계적으로 접근하여 갈 때 당장은 아니지만 우리의 오존에 대한 이해가 깊어갈수록 예보의 정확성은 향상될 수 있다. 많은 비용과 노력이 필요한 오존 대책의 시행은 충분히 신중하여야 한다. 서울과 같은 대도시라면 적합한 수치모델을 구비하여 효과를 점검할 수 있어야 한다. 일부에서는 예보의 정확성을 높일 수 있는 방안의 하나로 수치모델 이용을 거론하고 있으나 수치모델은 매일의 예보와 같이 일상적 목적을 위하여 사용될 수 있는 모델이 아니며, 현재와 같이 기초가 갖추어져 있지 않은 상황에서는 더욱 그러하다. 정상적 모델 이용이 가능할 수 있도록 배출원 자료 체계를 갖추어야 하고, 서을 등 특징적 지역에 대하여서는 집중적 현장 조사를 실시하여야 한다.3)와 NAS(National Academy of Sciences, 1983), 미국에서 발행되는 정부 지침서 (Federal Register)에 고시된 내용 등을 토대로 하였다. 연구는 당면현실로 다가온 정보화 및 세계화의 기업환경에서 예견되는 몰입(committment)의 약화에 대한 치유방안으로서, 정보화된 경영모의게임의 기업 교육훈련 시스템(Business Training System)으로의 발전 가능성을 제시한다 하겠다.암시하며, 따라서 우리 교육문화에 맞는 재택수업 형태의 개발이 시급함을 제시한다고 하겠다.column density of HCaN is (1-3):n1014cm-2. Apparent digestibility coefficients of the extruded pellet diets containing various fish meals for olive flounder, Paralichthys olivaceus

  • Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, energy, essential amino acids, and fatty acids in extruded pellets containing various fish meals were determined for olive flounder (Paralichthys olivaceus). Eight extruded pellet diets were prepared to contain different fish meals (herring fish meal, anchovy fish meal, mackerel fish meal, sardine fish meal-A, sardine fish meal-B, tuna fish meal, pollock fish meal-A, and pollock fish meal-B) designated as HM, AM, MM, SM-A, SM-B, TM, PM-A, and PM-B, respectively. Chromic oxide ($Cr_2O_3$) was used as an inert indicator at a concentration of 0.5 % in the diet. Feces were collected from triplicate groups of fish ($151{\pm}4.0g$) using a fecal collection column attached to the fish rearing tank for 4 weeks. Dry matter ADCs of the MM, SM-A, SM-B, and PM-A diets were higher than those of all the other dietary groups, and the lowest digestibility of dry matter was observed in the PM-B diet. Fish fed the MM, SM-A, and PM-A diets showed significantly higher ADC of protein than those fed the AM, SM-B, TM, and PM-B diets. Lipid ADC of PM-B was significantly lower than that of the other diets. Energy ADCs of fish fed the MM, SM-A, and PM-A diets were significantly higher than those of the other diets. The availability of essential amino acids in the MM, SM-A, and PM-A diets were generally higher than that of the other fish meal diets, while TM showed the lowest values among all the experimental diets. ADCs of fatty acids in the AM, MM, SM-A, and PM-A diets were generally higher than those of fatty acids in the other diets, and the lowest values were recorded for the PM-B diet. These results provide information on the bioavailability of nutrients and energy in various fish meals which can be used to properly formulate practical extruded feeds for olive flounder.

  • The behavior of peptide or protein solutes in saline aqueous solution is a fundamental topic in physical chemistry. Addition of ions can strongly alter the thermodynamic and physical properties of peptide molecules in solution. In order to study the effects of added ionic salts on protein conformation and dynamics, we have used the molecular dynamics (MD) simulations to investigate the behavior of Staphylococcus aureus Hfq protein under two different ionic concentrations: 0.1 M NaCl and 1.0 M NaCl in presence and absence of RNA (a hepta-oligoribonucleotide AU5G). Hfq, a global regulator of gene expression is highly conserved and abundant RNA-binding protein. It is already reported that in vivo the increase of ionic strength results in a drastic reduction of Hfq affinity for $Q{\beta}$ RNA and reduces the tendency of aggregation of Escherichia coli host factor hexamers. Our results revealed the crucial role of 0.1 M NaCl Hfq system on the bases with strong hydrogen bonding interactions and by stabilizing the aromatic stacking of Tyr42 residue of the adjacent subunits/monomers with the adenine and uridine nucleobases. An increase in RNA pore diameter and weakened compactness of the Hfq-RNA complex was clearly observed in 1.0 M NaCl Hfq system with bound RNA. Aggregation of monomers in Hfq and the interaction of Hfq with RNA are greatly affected due to the presence of high ionic strength. Higher the ionic concentration, weaker is the aggregation and interaction. Our results were compatible with the experimental data and this is the first theoretical report for the experimental study done in 1980 by Uhlenbeck group for the present system.

Effects of Tumeric (Curcuma aromatica Salab.) Extract on Shelf Life of Cooked Rice (강황 추출물이 쌀밥의 저장성에 미치는 영향)

  • This studies were engaged to investigate the potential us of Curcuma aromatica Salab. as a food ingredients. The antimicrobial activity of Curcuma aromatica Salab. extract (CE) and effect on shelf life of the cooked rice were examined. The antimicrobial activity of CE showed apparently against Bacillus cereus, B. subtilis and B. megaterium. The minimum inhibitory concentration (MIC) of CE was 0.15% for B. cereus md 0.2% for B. subtilis and B. megaterium. The antimicrobial activity of CE remained relatively high after heat treatments(30 min at 80 and $100^{\circ}C$, 15 min at $121^{\circ}C$). The shelf life of cooked rice containing above 0.05% CE increased more 1 or 2 days than that of control at $30^{\circ}C$, The texture of cooked rice was improved by addition of above 0.01% CE. The sensory quality of cooked rice with CE did not show significant difference between 0.00% and 0.05% in taste and overall acceptability but decreased above 0.10% in taste, color, flavor and overall acceptability.

Expression of mRNAs characteristic of cartilage and bone in the developing mandibular condyle of mice (발육중인 생쥐 하악 과두에서 연골 및 골의 특이 유전자 발현)

  • It has not been elucidated whether the initiation of condylar development of the mandible is related with the periosteum of the mandible, or if it derives from a separate programmed blastema not related with the mandible. Also, although the mandibular condylar cartilage is known to promote growth, few studies have dealt with molecular-biologic mechanisms such as the expression of specific genes according to the differentiation of the mandibular condyle. To elucidate the unique cellular characteristics, development, and differentiation process of the mandibular condyle, an examination of expressions of genes characteristic of cartilage and bone were carried out using RT-PCR and mRNA in situ hybridization. 1. Type? collagen mRNA was detected with type II collagen mRNA in the differentiation and growth process of the cartilage of the mandibular condyle. TypeII collagen mRNA was demonstrated in the whole resting md upper part of the poliferative zone, whereas type II collagen mRNA was observed in the resting, proliferative and upper hypertrophic cartilage zone of the mandibular condyle. 2. The condylar cartilage rapidly increased in size due to the accumulation of hypertrophic chondrocytes as characterized by the expression of type II collagen mRNA during postnatal development. 3. BMP-4 mRNA was present in the anlage of the future condylar process and also in the ossifying mandibular body. 4. IHH mRNA was limited exclusively to the lower part of the proliferative zone and the upper part of the hypertrophic cartilage zone during condylar development. These findings were different from those in the growth-plate cartilage of the long bone, indicating a characteristic feature of the differentiation of the chondrocytes in the condylar cartilage present in prenatal and postnatal development. Furthermore, it was also suggested that chondroblasts of condylar cartilage rapidly differentiate into hypertrophic chondrocytes with increased functional Load force such as muscle activity and mastication.
