Effect of Fermented Scutellariae Radix Extract on Production of Inflammatory Mediator in LPS-stimulated Mouse Macrophages (황금(黃芩) 발효 추출물이 LPS로 유발된 마우스대식세포에서 염증인자증가에 미치는 영향)

  • Objectives : This study aims at examining the immuno-modulating activity in the fermentative extract of the root of Scutellaria baicalensis Georgi (Scutellariae Radix) on the production of inflammatory mediator in LPS-stimulated RAW264.7 mouse macrophages. Method : Measurements were done for the influences on the cell viability, generation of hydrogen peroxide in cells and nitric oxide (NO) generation using the macrophage of mouse with the specimen SBS as the fermentative extract of Scutellariae Radix (SBS) with Saccharomyces cerevisiae STV89. Result : As a result of carrying out MTT assay to check the cellular toxicity of the fermentative extract of Scutellariae Radix, any excessive toxicity to the macrophage did not occur from treatments by concentration for SBS. SBS increased the generation of hydrogen peroxide in the macrophage. SBS suppressed the NO generated in macrophages and SBS concentration higher than $25{\mu}g/mL$ significantly suppressed the increased NO generated in LPS-stimulated macrophages. SBS concentration higher than $25{\mu}g/mL$ significantly suppressed the generation of IL-6, IL-10, IL-12p40 and MCP-1 in LPS-stimulated macrophages. Conclusion : Our findings indicate that SBS has an immuno-modulating activity in macrophage activation through suppressing the generation of inflammatory substances, NO, IL-6, IL-10, IL-12p40 and MCP-1.

Anti-inflammatory Effect of Angelicae Gigantis Radix Water Extract on LPS-stimulated Mouse Macrophages (Lipopolysaccharide로 유발된 마우스 대식세포의 염증매개성 Cytokine 생성증가에 대한 참당귀 물추출물의 효능 연구)

  • Objectives : The purpose of this study was to investigate the effects of Angelicae Gigantis Radix Water Extract(AG) on the production of proinflammatory mediators in RAW 264.7 cells stimulated with lipopolysaccharide(LPS). Method : RAW 264.7 cells were cotreated with AG(50 and 100 ug/mL) and lipopolysaccharide(LPS; 1 ug/mL) for 24 hours. After 24 hour treatment, using Bead-based multiplex cytokine assay, concentrations of various cytokines such as interleukin(IL)-6, IL-$1{\beta}$, IL-10, tumor necrosis factor-alpha(TNF-${\alpha}$), granulocyte colony-stimulating factor(G-CSF), granulocyte macrophage colony-stimulating factor(GM-CSF), interferon inducible protein-10(IP-10), leukemia inhibitory factor(LIF), lipopolysaccharide-induced chemokine(LIX), monocyte chemoattractant protein-1(MCP-1), macrophage colony-stimulating factor(M-CSF), macrophage inflammatory protein(MIP)-$1{\alpha}$, MIP-$1{\beta}$, MIP-2, Regulated on Activation, Normal T cell Expressed and Secreted(RANTES) and vascular endothelial growth factor(VEGF) were measured. Result : AG significantly inhibited LPS-induced production of TNF-${\alpha}$, MIP-$1{\alpha}$, G-CSF, RANTES, IL-10, and M-CSF from LPS-stimulated RAW 264.7 cells at the concentrations of 50 and 100 ug/mL. AG significantly inhibited LPS-induced production of MIP-$1{\beta}$, MIP-2, GM-CSF, and IL-6 from LPS-stimulated RAW 264.7 cells at the concentrations of 50 ug/mL. AG significantly inhibited LPS-induced production of VEGF from LPS-stimulated RAW 264.7 cells at the concentrations of 100 ug/mL. But AG did not show any significant effect on the production of MCP-1, LIF, LIX, IP-10 and IL-$1{\beta}$ from LPS-induced RAW 264.7 cells. Conclusion : These results suggest that AG has anti-inflammatory effect related with its inhibition of proinflammatory mediators such as TNF-${\alpha}$, MIP-$1{\alpha}$, G-CSF, RANTES, IL-10, MIP-$1{\beta}$, MIP-2, GM-CSF, IL-6, VEGF and M-CSF in LPS-induced macrophages.

Anti-inflammatory Effect of Coptidis Rhizoma Extract (황련(黃連) 추출물의 항염효능에 관한 연구)

  • Objectives : This research has been done to investigate the anti-inflammatory effect of Coptidis Rhizoma extracts. Method : Coptidis Rhizoma was extracted by $100^{\circ}C$ water. The extract (CC : Extract of Coptis chinensis rhizome) was used to examine its effects on the cell viability of mouse macrophage Raw 264.7 cell line. Also the production of nitric oxide (NO), the c-jun N-terminalkinase (JNK) activation and the production of cytokines such as (IL)-5 were evaluated in lipopolysaccharide (LPS)-stimulated Raw 264.7 cells. After the CC and LPS were applied to Raw 264.7 cells which were cultured for 24 hours, the MTT assay was performed. Result : The CC extracts didn't affect the viability of macrophage cells. However, the extracts inhibited the NO production and the JNK activation significantly in LPS-stimulated macrophage cells treated with 100 and $200{\mu}g/mL$ concentrations. The CC extract, also, impeded the production of inflammation-related factors and cytokines such as KC, VEGF, MCP-1, GM-CSF, IL-$1{\alpha}$, IL-5, IL-6, and IL-12p40 in LPS-stimulated macrophage cells at the concentration higher than $25{\mu}g/mL$. The production of basic-FGF concentration of 50 and $100{\mu}g/mL$, the production of IP-10 at $100{\mu}g/mL$, and the production of IFN-${\gamma}$ at $25{\mu}g/mL$, respectively. Conclusion : The CC prepared using $100^{\circ}C$ water showed the significant anti-inflammatory effect such as the inhibition not only on the production of NO, KC, VEGF, MCP-1, GM-CSF, IL-$1{\alpha}$, IL-5, IL-6, and IL-12p40 in LPS-stimulated macrophage cells at or higher than the concentration of $25{\mu}g/mL$, but also on the JNK activation at 100 and $200{\mu}g/mL$.

The Low Height Looping Technology for Multi-chip Package in Wire Bonder (와이어 본더에서의 초저 루프 기술)

  • Recent new packages such as MCP(Multi-Chip Package), QDP(Quadratic Die Package) and DDP(Dual Die Package) have stack type configuration. This kind of multi-layer package is thicker than single layer package. So there is need for the low height looping technology in wirebonder to make these packages thinner. There is stiff zone above ball in wirebonder wire which is called HAZ(Heat Affect Zone). When making low height loop (below $80\;{\mu}m$) with traditional forward loop, stiff wire in HAZ(Heat Affected Zone) above ball is bended and weakened. So the traditional forward looping method cannot be applied to low height loop. SSB(stand-off stitch) wire bonding method was applied to many packages which require very low loops. The drawback of SSB method is making frequent errors at making ball, neck damage above ball on lead and the weakness of ball bonding on lead. The alternative looping method is BNL(ball neckless) looping technology which is already applied to some package(DDP, QDP). The advantage of this method is faster in bonding process and making little errors in wire bonding compared with SSB method. This paper presents the result of BNL looping technology applied in assembly house and several issues related to low loop height consistence and BNL zone weakness.

Interzonal Comparative Analysis of the Wintering Habitat of Spot-billed Duck (Anas poecilorhyncha) (흰뺨검둥오리의 지역간 월동서식지이용 비교분석)

  • This study was conducted using the Wild -Tracker (WT-300, GPS-Mobile Phone Based Telemetry KoEco) to understand the habitats of the spot-billed duck wintering in urban and rural areas and provide the results as the basic data for the protection and management of the habitats of the waterbirds in Korea. Study areas consisted of the Anseong stream in Gyeonggi-do and the Sansu reservoir in Haenam. Five spot-billed ducks were captured by region, and we attached Wild-Tracker to each of the spot-billed ducks. We analyzed the tracking location data using ArcGIS 9.x and calculated Kernel Density Estimation (KDE) and Minimum Convex Polygon (MCP). The average home-range measured by MCP was $250.8km^2$(SD=195.3, n=5) in Anseong and was $89.1km^2$ (SD=69.6, n=5) in Haenam. 50% home-range measured by KDE was $21.8km^2$ (SD=26.9, n=5) in Anseong and $3.5km^2$ (SD=2.2, n=5) in Haenam, indicating a narrow home range in Haenam. During the winter season, both wetland and paddy field were mostly used as habitats in Anseong and Haenam. While the paddy field utilization rate was high in the daytime in Haenam, it was high in the nighttime in Anseong. By late winter, Haenam's day time paddy field utilization rate and Anseong's night time paddy field utilization rate increased.

  • This paper introduces a radio-wave interference assessment of wind turbines that were planned to be installed at Homi-Cape in Pohang region where wind resource has been evaluated worthwhile developing a wind farm. In that area, AM radio station with two antennas and a harbor radar facility are located so that radio-wave coupling is inevitable if the wind farm is designed without considering radio-wave environmental impact. A low-frequency analysis using MoM (Method of Moment) is used to examine interference effect caused by wind turbines and an optimal layout minimizes coupling effect is presented.

Microstructure of W-Cu Composite Powders with Variation of Milling Method during Mechanochemical Process (기계화학적 공정의 밀링 방법에 따른 W-Cu 복합분말의 미세조직)

  • Recently, the fabrication process of the W-Cu nanocomposite powders has been studied to improve the sinterability through the mechanical alloying and reduction of W and Cu oxide mixtures. In this study. the W-Cu composites were produced by mechanochemical process (MCP) using $WO_3-CuO$ mixtures with two different milling types of low and high energy, respectively. These ball-milled mixtures were reduced in $H_2$ atmosphere. The ball-milled and reduced powders were analyzed through XRD, SEM and TEM. The fine W-Cu powder could be obtained by the high energy ball-milling (HM) compared with the large Cu-cored structure powder by the low energy ball-milling (LM). After the HM for 20h, the W grain size of the reduced W-Cu powder was about 20-30 nm.

  • Bi-2212 HTS tube was fabricated by centrifugal forming process(CFP). As a variation of melt casting process(MCP) or centrifugal casting technique, the centrifugal forming process is a flexible method for manufacturing Bi-2212 bulk tubes and has been optimized to achieve smooth surface and uniform thickness. At this process, the slurry was prepared in the mixing ratio of 10:1 between Bi-2212 powder and binder and initially charged into the rotating mold under the speed of 300~450 rpm Heat-treatment was performed at the temperature ranges of 860 ~ $890^{\circ}C$ in air for partial melting. The HTS tube fabricated by centrifugal forming process at $890^{\circ}C$ under the rotating speed of 450 rpm was highly densified and the plate-like grains with more than 20$\mu$m were well oriented along the rotating axis. The measured Tc and Jc at 10K were around 85K and 3,000A/cm2 respectively.

Comparison of Movement Distance and Home Range Size of Gold-spotted Pond Frog (Pelophylax chosenicus) between Rice Paddy and Ecological park - Focus on the Planning Alternative Habitat - (논과 생태공원에서 금개구리 이동 거리 및 서식영역 크기 비교 - 대체서식지 조성 중심으로 -)

  • The movement distance and home range size of Pelophylax chosenicus were identified in the rice paddy and ecological park as alternative habitats from July to November 2017. A total of 39 frogs were tracked by radio tracking method. As a result, the average move distance in the population of rice paddy was 11.7 ± 1.9 m (n = 64) and the population of ecological park was 24.7 ± 4.3 m (n = 39). The move distance between the two populations was significantly different. The mean MCP of the population of rice paddy was 181.2 ± 110.8 m2 (n = 11) and the population of ecological park was 416.1 ± 276.2 m2 (n = 10), but there was no significant difference. The population area of rice paddy was 4,160 m2 (Kernel density 95%) and the core area was 1,080 m2 (Kernel density 50%). The population area (Kernel density 95%) of ecological park was 5,391 m2 and the core area (Kernel density 50%) was 736 m2. This study shows that it is appropriate to construct the area of alternative habitat for P. chosenicus at least 1.33 ha, and it is more advantageous for the ecological park to be constructed than the paddy field with high development pressure and human interference. If the rice paddies were to be abandoned for several years, or to be used traditional farming methods, such as refraining from using agricultural machinery and chemicals, they could be used as alternative habitat for P. chosenicus.

Study on the Home-range and Winter Habitat Pintail using the Wild-Tracker (WT-300) in Korea (WT-300을 이용한 월동기 고방오리(Anas acuta)의 행동권 및 서식지 이용연구)

  • Pintail (Anas acuta) is the major wintering bird in South Korea and known as a major mediator of the highly pathogenic avian influenza (HPAI). Pintail migrates long distances between Russian Siberia and Korea. This species prefers a rice paddy area as their winter habitat. The purpose of this study is to provide the data necessary for the conservation and management of bird habitats in Korea by understanding the wintering home-range and habitat of pintail in Korea. We captured six pintails using a cannon-net in the winter of 2015 and attached the GPS-mobile phone based telemetry (WT-300) on them to study the wintering home-range and wintering habitat. We analyzed the tracking location data using ArcGIS 9.0 Animal Movement Extension and calculated Kernel Density Estimation (KDE) and Minimum Convex Polygon (MCP). The average home-range in the wintering ground analyzed by MCP was $677.3km^2$ (SD=130.2, n=6) while the maximum and minimum were $847.7km^2$ and $467.5km^2$, respectively. Extents of home-range analyzed by KDE were $194.7km^2$ (KDE 90%), $77.4km^2$ (KDE 70%), and $35.3km^2$ (KDE 50%). The pintails mostly used both sea and paddy field as habitat in the winter season and utilized paddy fields more during the nighttime and than the daytime. We concluded that the home-range and habitat of pintails in the winter could be used as the reference data for the preservation of species, management of habitats, and coping with a breakout of HPAI.
