Optimal Condition for Mycelial Growth of Beauveria bassiana and Its Extracellular Enzyme Activity (백강균(Beauveria bassiana)의 균사체 최적 배양조건 및 효소활성)

  • The optimum temperature and pH for mycelial growth of B. bassiana DGUM 34001 were $24^{\circ}C$ and pH 7.0, respectively. Among the complex media used, mushroom complex medium (MCM) was the most favorable for mycelial growth. When Czapek-Dox medium was used as a minimal medium, glucose was an excellent source for carbon and energy. Soytone and sodium phosphate were favorable constituent for culture medium as a source of organic nitrogen and phosphorus, respectively. When the fungus was grown in MCM broth, the specific activity of extracellular enzyme of ${\alpha}$-amylase, lipase, chitinase, CMCase and pretease were 297.0, 0.058, 0.33, 0.21 and 22.8 units/mg protein, respectively. When various sources of organic nitrogen and chitin were supplemented to determine the production of enzymes, casein and soluble chitosan enhanced the production of extracellular protease and chitinase.

Via Formation in Dielectric Layers Made of Photosensitive BCB (감광성 BCB를 이용한 절연막층에서의 비아형성)

  • Via for achieving reliable fabrication of MCM(Multichip Module) substrate was formed on photosensitive BCB layer. The MCM substrate consists of photosensitive BCB(Benzocyclobutene) interlayer dielectric and copper conductors. In order to form the vias in the photosensitive BCB layer, the process of forming the BCB layer and its via forming plasma etch using C$_2$F$\_$6//O$_2$ gas were evaluated. The thickness of the BCB layer after hard bake was shrunk down to 40% of the original. The resolution of vias formed on the BCB was 15㎛ and the slope after develop was 85 degree. AES analysis was done on two vias, one is etched in C$_2$F$\_$6/O$_2$ gas and the other isnot etched. On the via etched in C$_2$F$\_$6//O$_2$, native C was detected and the amount of native C was reduced after Ar sputter. On the via not etched in C$_2$F$\_$6//O$_2$, organic C was detected. As a result of AES, BCB residue was not removed by Ar sputter, so plasma etch is necessary for achieving reliable vias.

Optimal Conditions for the Laccase Production from Fomitopsis pinicola Mycelia (Fomitopsis pinicola 균사체로부터 Laccase의 최적생산조건)

  • The culture conditions to maximize the production of laccase (EC 1.10.3.2) from Fomitopsis pinicola mycelia were investigated. Among the tested media for the enzyme production, mushroom complete medium (MCM ; 2% dextrose, 0.2% peptone, 0.2% yeast extract, 0.05% $KH_2PO_4$, and 0.05% $MgSO_4{\cdot}7H_2O$) showed the highest activity of the enzyme. To optimize the culture condition for the laccase activity, influence of various carbon and nitrogen sources was investigated in MCM. Among various carbon and nitrogen sources, 2% glucose and 0.4% peptone showed the highest production of the enzyme, respectively. For the phosphorus and inorganic source, 0.05% $NaH_2PO_4$ and 0.05% $CaCl_2$ were best for the enzyme activity. The enzyme production was reached to highest level after the cultivation for 8 days at $25^{\circ}C$. Native polyacrylamide gel electrophoresis (PAGE) followed by the laccase activity staining using 2, 2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) as the substrate was performed to identify the laccase under culture conditions studied. Zymogram analysis of the culture supernatant showed a laccase band with molecular mass of 52 kDa. The optimum pH and temperature for the enzyme activity were $80^{\circ}C$ and pH 3.0.

  • Propylene epoxidation by $H_2O_2$ (30% aqueous) as oxidant was studied in a semi-batch reactor using TS-1 catalyst: Effects of reaction temperature, time, pressure, solvent, catalyst and $H_2O_2$ concentration on $H_2O_2$ conversion (limiting reagent) and product distribution were investigated. Potential inhibition by propylene oxide on the epoxidation rate was also examined. Ti-MCM-22 with MWW zeolytic structure was found to exhibit better performance than TS-1 with MFI structure, provide that a proper choice of solvent(acetonitrile) is made.

  • Thermodynamic analysis on carbon dioxide reforming of methane was performed using a computer program which can handle condensed species in the products, and the reforming experiments were conducted over $Al_2O_3$, $La_2O_3$, ZSM-5, MCM-41, KIT-1 supported nickel catalysts, and a commercial ICI 46-1. It was estabished that a system which consists of $CH_4$, $CO_2$, CO, $H_2$, $H_2O$, and C is appropriate for theoretical equilibrium calculations and addition of water vapor or oxygen was found to diminish the contribution of carbon dioxide in reforming. Silicate molecular sieve-supported catalysts such as Ni/ZSM-5, Ni/MCM-41, Ni/KIT-1 were effective for high $CH_4$ and $CO_2$ conversions as well as for high CO yield. Coke formation was suppressed when CaO was added as a promoter. Ni/Ca/KIT-1 which contains 10% Ni with 3% Ca showed conversion approaching equilibrium levels above $650^{\circ}C$ and maintained constant activity over 20 h. Despite increased space velocity, relatively high conversion and CO yield were observed.

The Cultural Characteristics and Antibacterial Activities of Cordyceps militaris and Paecilomyces tenuipes (번데기동충하초와 눈꽃동충하초의 배양특성 및 항균활성)

  • The cultural characteristics and antibacterial activities of Cordyceps militaris and Paecilomyces tenuipes were compared. The mycelial growth was the highest on MCM (Mushroom Complete Medium) for C. militaris and on YMA (Yeast Malt Agar) for P. tenuipes. But the mycelial density on MMM (Mushroom Minimal Medium) was lower than other on media. The optimum mycelial growth was observed at $25^{\circ}C$. C. militaris was low mycelial growth when it was transferred over 5 times generation. The carbon source for the optimum mycelial growth was fructose of monosaccharide, maltose of disaccharide and dextrin of polysaccharide. The calcium nitrate of organonitrogen was found the best mycelial growth on C. militaris, while the sodium nitrate observed to be well for mycelial growth on P. tenuipes. The ammonium tartrate was observed to be the best among the inorganonitrogen used for mycelial growth. Antibacterial activities were found out just C. militaris against Bacillus cereus of Gram (+).

Excellent strain selection and optimal mycelial growth condition of Pleurotus cornucopiae (노랑느타리벗섯(Pleurotus cornucopiae)의 우수균주 선발 및 최적 균사배양)

  • The results of examining selection of optimal mycelial growth condition excellent strain for determining the condition of Pleurotus cornucopiae are as follows. 1) Mycelial growth and density of P. cornucopiae were the highest in the medium of MCM(87.8mm/10d) followed by the order of PDA, YM, MEA and Czapek. 2) Optimal temperature for the mycelial growth and density of P. cornucopiae was shown to be $25^{\circ}C$, but the hyphae were dead at $5^{\circ}C$. The mycelial growth and density of KNAC2003 strain was the highest at $25^{\circ}C$(87.8mm/10d) followed by the order of 25, 30, 20, 35, 10 and $5^{\circ}C$. 3) Optimal pH for the mycelial growth and density was revealed to be 6.0(88.3mm/10d), but the mycelial growth and density were shown to be retarded in above or below pH 6.0.
