The Effect of Iron Supplementation on the Hematological Iron Status and Pb and CD Levels in Erythrocyte, Hair and Urine of Subjects With Suboptimal Iron Status (철분의 구강투여가 철분부족 아동의 혈중철분지표 및 적혈구, 소변, 머리카락의 납 과 카드뮴 수준변화에 미치는 영향)

  • The effect of oral iron supplementation was assessed on blood iron levels and Pb and Cd levels in erythrocytes, hair and urine of 101 Puchon 5th grade school children with suboptimal iron status. Treatment with 25mg of elemental iron per day for 8weeks resulted in a significant increase in the intake of most nutrients in addition to iron. Iron supplementation resulted in significant improvements in hemoglobin, MU, MCH, MCHC, serum ferritin, serum iron, TIBC, and transferrin saturation of subjects(p<0.05 - p<0.01) and cocomitantly lowered Pb and Cd levels in erythrocytes, hair, and urine(p<0.01). Regression analysis showed that only iron intake contributed to significant increases in hemoglobin and serum ferritin. It seems that 25mg of iron supplementation is safe and adequate to improve iron status in school children with suboptimal iron status and it also has the benefit of alleviating Pb and Cd status. Analysis of Hematologic Characteristics of Endangered Korean Native Cattle according to the Age (성장단계별 멸종위기 희소한우의 혈액학적 특성분석)

  • The importance of genetic resource preservation has been highlighted in the literature as a means of maintaining genetic diversity. Investigations for hematologic values and the differential count of white blood cell count (WBC) for Korean indigenous cattle (KIC) and endangered indigenous cattle (EIC) are rarely performed. Therefore, the objective of this study was to investigate the hematologic values of total 40 EIC (White, Black, Mini cattle) and 35 KIC as control by analysis of hematologic characteristics. As a result, the mean values of RBC and platelet of EIC were significantly decreased by age (p<0.05). The mean values of RBC, HCT, MCV and MCHC between EIC and KIC of the same age (2~3 years) showed the statistical significance (p<0.05). Also, in the WBC of EIC, the mean values were decreased according to the age from $13.9{\times}10^3/{\mu}L{\sim}12.7{\times}10^3/{\mu}L$ under 1 year to $9.1{\times}10^3/{\mu}L{\sim}11.5{\times}10^3/{\mu}L$ over 2 years respectively. In the differential count of WBC of EIC (White, Black, Mini cattle), it showed generally the rates of 40.2%, 52.2%, 49.0% lymphocyte and 27.2%, 33.9%, 32.0% segmented neutrophil from 2~3 years respectively. Result of this study will be used for establishing reference range for blood analysis in EIC such as white, black and mini cattle. This study reported hematological values which could serve as baseline information for comparison in conditions of nutrient deficiency, physiological and health status of endangered Korean native cattle. In addition, this study provides a valuable resource for further investigations of the preservation of rare genetic stocks underlying traits of interest in cattle.

  • To establish the hematological reference values in the healthy adults visited our hospitals, following examination were done on 2823 persons by Coulter Counter Model S-plus II ; white blood cell count: (WBC), red blood cell count(RBC), hemoglobin(Hb), hematocrit(Hct), mean corpuscular volume (MCV), mean corpuscular hemoglobin(MCH), mean corpuscular hemoglobin concentration(MCHC), red cell distribution width(RDW), platelet, plateletcrit, mean platelet volume(MPV) and platelet distribution width(PDW). Analysis of Hematologic Characteristics of Korean Native Stripped Cattle Chickso according to the Ages (성장 단계별 희소 한우(칡소)의 혈액학적 분석)

  • Investigations for hematologic values and the differential count of WBC for Korean indigenous cattle (KIC) and Stripped Cattle (SC) are rarely performed. Therefore, when the index of complete blood counts (CBC) analysis of KIC and SC were requested, it had many difficulties to make the results for blood condition since the standard hematologic values of KIC and SC are lacking. The objective of this study was to investigated the hematologic values and the differential count of WBC for blood of total 19 striped cattle (SC) and 187 KIC as a control by estimation analysis of hematologic characteristics. As a result, the mean values of RBC and platelet of KIC were significantly decreased by age (P<0.05). The mean values of RBC, HCT, MCV and MCHC between KIC and SC of the same age (2~3 years) showed the statistical significance (P<0.05). Also, in the WBC of KIC, the mean values were decreased according to the age from $13.8{\times}10^3/{\mu}l$ under 1 year to $9.5{\times}10^3/{\mu}l$ over 5 years. In the differential count of WBC of KIC, it showed generally the rates of 46.2% lymphocyte and 36% segmented neutrophil. Additionally, in comparative analysis between pregnant and non-pregnant group of KIC, the mean values of Hg and HCT in pregnant group were significantly decreased(P<0.05). In conclusion, data obtained from this study may be valuable as a standard for interpretation of the results in hematologic analysis of KIC. Result of this study will be used for establishing reference range for hematologic analysis in SC.

  • The purpose of this experiment was to determine the effects of di(2-ethylhexyl) phthalate(DEHP) on reproductive characteristic, blood hematological and chemical values in mice. The male mice were intraperitoneally injected DEHP in negative control(no treatment), positive control(corn oil, 3ml/kg B.W), 0.5, 1.0 and 10.0mg DEHP/kg B.W and the female mice were injected DEHP in control(corn oil, 3ml/kg B.W), 0.5, 1.0 and 10.0mg DEHP/kg B.W with 5 times for 15 days on 3 days interval. The administration of DEHP in male mice were not affect on body weight, epididymis, vesicular gland and coagulating gland weight. The testis weight were slightly higher in DEHP treatment groups than in control. The semen characteristics(sperm concentration, viability, motility and abnormality) of male mice were not difference in all experimental groups. The RBC, Hb, HT, MCV, MCH, MCHC< PLT, albumin, BUN and total protein of blood hematological and chemical values were not affect the administration of DEHP in mice. The WBC values in 10.0mg DEHP group was slightly difference in all experimental group(P>0.05). The histological evaluation of testis, ovary and affevt the reproductive characteristic, blood hematological and chemical values.

  • The objective of this study was to assess that the effects of DEHP administration on reproductive characteristics and blood hematological and chemical values in pups born after DEHP administration in pregnant mice. DEHP was administrated to pregnant mice by intraperitoneally injection with 0, 0.5, 1.0 and 10.0mg/kg B.W, 5 times at 3 days interval from Day 1 to Day 16 in the gestation period. The body weight and reproductive organ weight(testis, epididymis and coagulating gland) in male pups on 45 day after birth was not affected in all experimental groups, but vesicular gland in DEHP groups was significantly lower than that of control group(P<0.05). The semen characteristics of male pups were not affected in DEHP treatment groups. The WBC, HB, HT, MCH and albumin values in male pups were not affected in all experimental groups, but RBC MCV, MCHC, PLT and total protein values were significantly different among the experimental groups(P<0.05). In female pups, the effects of DEHP administration were not affected the body and uterus weight, but the left ovary in 10.0mg DEHP group was significantly heavier than in control and 0.5mg DEHP group(P<0.05). The WBC, MCV, MCH, MCHC, PLT, albumin, BUN and total protein values in female were not different in all experimental groups. The RBC, HB and HT values were significantly different among the experimental gruop(P<0.05). The historical evaluation of testis in male pups that were grown to 45 days after birth was not different in all experimental groups. The ovary in female pups had many corpus luteum in 10.0mg DEHP group. The endometriosisi of uterus was significantly decreased in DEHP group. There results suggest that low concentration of DEHP administration in pup born after DEHP administration in pregnant mice was not affered on reproductive characteristic, but was affected on blood hematological and chemical values.

Effects of phosalone consumption via feeding with or without sodium bentonite on performance, blood metabolites and its transition to milk of Iranian Baluchi sheep

  • Background: Transfer of pesticides from environment to animal products is inevitable, so the purpose of the present work was to evaluate phosalone consumption via feeding with or without sodium bentonite (SB) on performance, blood metabolites and its transition to milk of Iranian Baluchi sheep. Methods: Twenty Baluchi ewes were divided into four treatments (P1 as control, P2, P3, and P4) of five animals in which phosalone, an organophosphate pesticide, was given via diet (only for P2 and P3) at a dose of 280 mg/sheep/day for 63 consecutive days. The SB (32 g/sheep/day; for P3 and P4) was also evaluated for its ability to reduce deleterious effects of phosalone in the sheep diets. The control group (P1) did not receive any phosalone and SB during the experiment. Sampling was conducted in two periods of time including weeks 5 and 9. Results: Phosalone residues were observed in the milk samples of P2 and P3 groups during two sampling periods. During period 1, the transfer rate of phosalone from feed to milk was 0.23 and 0.02%, respectively for the contaminated diets (P2 and P3), which is relatively similar to period 2 (0.22 and 0.02%). Only 0.34 (period 1) and 0. 36% (period 2) of phosalone residue are excreted in the feces of P2 group following its daily consumption. Transfer of phosalone from feed to milk was affected (P < 0.05) by the dietary inclusion of a commercial SB, as it (SB) decreased excretion of phosalone via milk (P3). The phosalone and SB alone or together had no significant effect (P > 0.05) on the dry matter intake (DMI) and body weight (BW) gain, but feed efficiency, milk production, milk fat, dry matter (DM) and organic matter (OM) digestibility, acetylcholinesterase (AChE) inhibitory activity, hemoglobin (Hb), red blood cell (RBC), serum glutamic pyruvic transaminase (SGPT), serum glutamic oxaloacetic transaminase (SGOT), albumin and mean corpuscular hemoglobin concentration (MCHC) affected by the treatments in period 1 or 2 (P < 0.05). The Hb, RBC, and MCHC were significantly decreased (P < 0.05) by about 9.72, 20.77, and 9.71%, respectively in the group P2 as compared to those of the control group during period 1. The AChE inhibitory activity (period 1 and 2) significantly increased when phosalone administered via the diet (P < 0.05). Conclusions: Although there were no adverse effects on the performance of sheep following the intake of phosalone alone (P2 vs. P1), but other research on the long and short times to the phosalone in high and low doses with more animals is suggested. Overall, compared to the control group, addition of SB in the diet of sheep improved nutrient digestibility, animal performance, and milk health.

Changes in the Blood Components Caused by Water Intake (물 섭취에 의한 혈액 성분 변화에 관한 연구)

  • Although water is an essential component of the human body and is involved in many physiological processes, the effect of a steady and sufficient water intake on blood components has not well elucidated. Therefore, we investigated the changes in hematological parameters, high-sensitivity C-reactive protein (hs-CRP), and immunoglobulin G (IgG) after water intake in 13 healthy adults. They were divided into two groups: The control group (N=4), which consumed water ad libitum, and the experimental group (N=9), which consumed 2 L of water per day. Two weeks later, blood cell counts, hematocrit, and hemoglobin content had increased in the experimental group, although not significantly (p>0.05); however, there was a significant increase in the mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) (p<0.05; and p<0.01, respectively), and a significant decrease in the mean platelet volume (MPV) (p<0.05). Of the immunologic parameters, a non-significant decrease in the concentration of hs-CRP, an indicator of cardiovascular disease risk, was observed (p>0.05). However, there was a dramatic and significant increase in the concentration of IgG (p<0.05). In conclusion, a steady and sufficient water intake may contribute to alleviate anemia by increasing hemoglobin. Additionally, it may decrease the risk of cardiovascular disease by decreasing platelet activation and concentration of hs-CRP. Furthermore, a steady intake of water may improve immune function by increasing the concentration of the components of humoral immunity.

hEPO 형질전환 돼지 후대의 임신기간내 혈액조성 변화

  • 본 연구는 사람 조혈촉진유전자(hEPO)가 도입된 형질전환돼지 Fl 수컷과 암컷간 교배로 F2 생산시, Fl 모돈에 대한 임신기간(약 114일)내 혈액조성(적혈구, 백혈구, Hemoglobin, Hematocrit value, MCV, MCH, MCHC, 혈소판) 변화를 구명하기 위하여 실시하였다. 시험에 이용된 암퇘지는 형질전환 F1 암컷 2마리와 일반돼지 1마리였고 교배와 동시에 임신기간동안(114일) 5일 간격으로 5ml의 혈액을 경정맥(jugular vein)에서 채취하였으며 혈액조성분석은 Celltac MEK 5108K(Nihon Kohden, Japan)를 이용하였는데 얻어진 결과는 다음과 같다. 임신기간중 혈액내 백혈구의 변화는 일반돼지의 경우 13.8($\pm$2.4)$\times$$10^3$/ul 로임신기간내에 일정한 변화양상을 보였으나 형질전환돼지의 경우 29.4($\pm$19.4)$\times$$10^3$와 22.5($\pm$14.1)$\times$$10^3$/ul 으로 일반돼지보다는 높게 나타났으며 임신기간내에 매우 높은 변화 양상을 나타내었다. 또한 적혈구는 일반돼지(7.2$\pm$0.7 $\times$$10^{6}$/ul)에 비하여 형질전환돼지(11.5$\pm$0.5 와 11.9$\pm$0.5 $\times$$10^{6}$ul)에서 매우 높게 나타났으며 변화양상은 일반돼지와 마찬가지로 작게 나타났다. 적혈구와 함께 Hemoglobin(Hb)과 평균혈구용적율(Hematocrit value ;, HCT) 모두 형질전환돼지가 일반돼지보다 높게 나타났으며 변화양상 또한 작게 나타남으로서 사람 조혈촉진유전자(hEPO)가 형질전환된 돼지에서는 지속적으로 발현되어 조혈촉진이 이루어지고 있으며, 조혈촉진에 의해 헤모글로빈 및 적혈구가 생성되어 혈액내 높은 혈구용적율(HCT)을 이루고 있음을 알 수 있었다. 적혈구 평균용적(Mean Corpuscular Volume ; MCV), 평균적혈구혈색소량(Mean Corpuscular Hemoglobin ; MCH), 평균적혈구혈색소농도(Mean Corpuscular Hemoglobin Concentration ; MCHC) 그리고 혈소판(Platelets) 분석결과 형질전환돼지가 일반돼지보다 약간 높은 수치를 나타냈으며 변화양상 또한 유사한 결과를 나타내었다. 이와 같은 시험분석결과를 토대로 사람 조혈촉진유전자(hEPO)가 형질전환된 돼지는 유즙으로 발현할 수 있도록 형질전환 되었음에도 불구하고 헤모글로빈 및 적혈구가 증가함으로서 형질전환돼지 개체의 혈장으로도 사람 조혈촉진인자가 분비하고 있음을 간접적으로 알 수 있었다. 정상돼지 보다 형질전환돼지가 약 30% 높은HCT 수준을 보였으며 이러한 현상은 사람에서는 적혈구증다증(erythrocytosis)으로 분류되고 있다. 이에 대한 고찰은 형질전환돼지 자체의 생리적 문제점(side effects)에 대한 해결과 더불어 기존의 인간질병에 대한 모델동물로서의 이용 가능성을 제시할 수 있는 것으로 사료된다.

Bisphenol A의 투여가 생쥐의 번식효율에 미치는 영향

  • 내분비계 장애물질 중 관찰물질로 지정된 bisphenol A(BPA)의 투여가 생쥐의 번식 효율에 미치는 영향을 규명하기 위하여 웅성 ICR 생쥐에 무처리구, corn oil, 0.05, 0.5 및 5.0mg BPA/kg b.w의 BPA을 3일 간격으로 5회 투여했으며, 자성 ICR 생쥐도 웅성생쥐와 동일하게 3일 간격으로 5회 투여한 후, 웅성은 투여후 2일, 자성은 7일후에 모든검사를 실시하였다. BPA의 투여가 웅성 생쥐의 번식기관무게에 미치는 영향을 조사한 결과는 체중, 정소상체, 정낭선 및 응고선의 무게는 투여구간에 커다란 차이가 없었으며(P>0.05), 정소의 무게는 5.0mg 투여구가 여타구보다 다소 낮은 무게를 나타냈다. BPA의 투여가 웅성 생쥐의 정액성상에 미치는 영향을 조사한 결과, 생존율과 유효 정자수는 각 투여구간에 커다란 차이가 없었으나 총정자수는 각각 35.50, 35.73, 36.33, 33.13 및 29.28$\times$$10^{7}$ 정자/$m\ell$로서 5.0mg 투여구가 여타구보다 통계적으로 유의하게 낮은 결과을 나타냈다(P<0.05). 기형율은 5.0mg 투여구가 23.15%로서 무처리구(18.13%), 대조구(18.58%) 및 0.05mg 투여구(19.68%)보다 통계적으로 유의하게 높은 기형율을 나타냈다(P<0.05). 웅성 생쥐에 BPA의 투여가 혈액의 일반성분에 미치는 영향을 조사한 결과, RBC, Hb, HT, MCV, MCH 및 MCHC는 각 투여구간에 차이가 없었으며, WBC는 BPA 투여구가 무처리구 및 대조구보다 다소 낮은 수치를 나타냈으며, PLT는 BPA 투여구가 무처리구 및 대조구보다 높은 수치를 나타냈다. BPA의 투여가 자성 생쥐의 번식기관무게에 미치는 영향을 조사한 결과, 체중은 투여구간에 커다란 차이가 없었으며 자궁의 무게는 BPA 투여구가 대조구에 비해 다소 논은 무게를 나타냈다. 오른쪽 난소의 무게는 0.5mg 투여구와 5.0mg 투여구가 각각 0.0059 및 0.0061g으로 대조구(0.0081g)와 0.05mg 투여구(0.0071g)보다 낮은 무게를 나타냈으며, 왼쪽 난소의 무게는 각각 0.0076, 0.0076, 0.0058 및 0.0061g으로 0.5mg 투여구가 대조구와 0.05mg 투여구보다 낮은 무게를 나타냈다(P<0.05). BPA의 투여가 자성 생쥐의 혈구화학치에 미치는 영향을 조사한 결과, RBC, Hb, HT, MCV, MCH 및 MCHC는 각 투여구간에 차이가 없었으며, WBC는 BPA 투여구가 대조구보다 다소 높은 수치를 나타냈다. PLT는 0.5mg 투여구와 5.0mg 투여구가 대조구보다 다소 높은 수치를 나타냈다. 정소, 난소 및 자궁의 조직, 병리학적인 검사에서는 모든 투여구간에 커다란 차이가 없었다.
