제주 흑우 집단에서 Indel, Microsatellite 마커와 MC1R 유전자형을 이용한 친자 확인 (A Parentage Test using Indel, Microsatellite Markers and Genotypes of MC1R in the Jeju Black Cattle Population)

  • This study was carried out to examine a molecular marker system for parentage test in Jeju Black cattle (JBC). Based on the preliminarily studies, we finally selected for construction of a novel genetic marker system for molecular traceability, identity test, breed certification, and parentage test in JBC and its related industrial populations. The genetic marker system had eight MS markers, five indel markers, and two single nucleotide polymorphisms (SNPs; g.G299T and g.del310G) within MC1R gene which is critical to verify the breed specific genotypes for coat color of JBC differing from those of exotic black cattle breeds such as Holstein and Angus. The results showed lower level of a combined non-exclusion probability for second parent (NE-P2) of $4.1202{\times}10^{-4}$ than those previously recommended by International Society of Animal Genetics (ISAG) of $5.000{\times}10^{-4}$ for parentage, and a combined non-exclusion probability for sib identity (NE-SI) of $2.679{\times}10^{-5}$. Parentage analysis has been successfully identified the JBC offspring in the indigenous population and cattle farms used the certified AI semens for production using the JBC-derived offspring for commercial beef. This combined molecular marker system will be helpful to supply genetic information for parentage test and traceability and to develop the molecular breeding system for improvement of animal productivity in JBC population.

전기 근육자극요법이 노인의 만성무릎통증과 우울에 미치는 효과 (The Effect of Electrical Muscle Stimulation Therapy on Chronic Knee Pain and Depression for Aged)

  • Purpose: This study was to examine the effect of electrical muscle stimulation therapy on chronic knee pain and depression for the aged. Method: Design was one-group pretest-posttest design. Samples were 32 elderly of 60 years old and above with chronic knee pain and depression. Measures were the S-F McGill Pain Questuionnaire for chronic knee pain and the Korean Elderly Depression Scale for depression. Electrical muscle stimulation therapy, experimental treatment, was applied for 12 weeks, 3 times/week, 15 min/time. Data were collected from January 2005 to May 2005. Data were analyzed using SPSS PC+ 12 version. Results: After receiving electrical muscle stimulation therapy, chronic knee pain (t=-88.034, P=.000) and depression (t=-114.659, P=.000) were significantly decreased. Conclusion: Electrical muscle stimulation therapy can be a better effective primary nursing intervention on chronic knee pain and depression for community dwelling elders.

온라인 열람목록의 이용행태에 관한 연구 (A Study on the Searching Behavior of OPAC Users)

  • 온라인 열람목록의 이용자 인터페이스 특성이 이용행태에 미치는 영향을 파악하고, 이용자에게 보다 친근한 인터페이스를 평가하여 시스템 설계의 방향을 제시하는 데에 본 연구의 목적을 두었다. 이용자 인터페이스에 있어서 뚜렷한 특성상의 차이를 보이는 두 시스템을 대상으로 하여 $2\times2$ 교차계획법으로 이용행태연구를 위한 실험연구를 수행하였다. 65명의 탐색자가 실험에 참여하였다. $2\times2$ 교차계획법에서의 이월효과와 시스템효과, $\chi^2$검증, t 검증 매크네마 검증, 최대우도법을 통한 주변대칭성 검증, 인자분석, 회귀분석, 분산분석 등의 통계기법을 검증해야 할 가설과 데이터의 성격에 따라 적절히 적용하여 분석하였다.

건조수축에 따른 균열 방지를 위한 콘크리트 슬래브의 정량적 줄눈 간격 산정 (Quantitative Estimation of Joint Spacing for Concrete Slab to Prevent Cracking of Drying Shrinkage)

  • 콘크리트 슬래브의 수축으로 인해 발생하는 응력에 의한 무분별한 균열을 억제하기 위해 줄눈을 설치한다. 그러나 설치된 줄눈의 간격은 구체적인 산출 근거나 정량적인 기준 없이 경험적으로 나누어지고 있다. 따라서 이 연구에서는 콘크리트 슬래브의 수축 응력에 의한 변형량을 측정하고 이에 따른 정량적인 평가 기준을 제시하고자 한다. 노출된 환경 조건은 온도 $15^{\circ}C$, 상대습도 60%를 적용하였다. 건조수축 실험은 현재 슬래브 설계에 많이 사용되는 설계 강도 30 MPa 및 40 MPa 배합을 사용하였고 그 결과를 기존 건조수축 모델에 적용하였다. 그 결과 ACI 209R 모델에 가장 일치하는 것으로 나타났으며 이를 바탕으로 콘크리트 슬래브의 정량적인 줄눈 간격을 산정하였다.

수문컴포넌트별 오염부하 산정이 가능한 WAPLE4의 개발 (Development of Pollutant Load Estimation System for Hydrologic Component, WAPLE4)

  • 하천유량은 기저유량과 직접유량으로 구성되어 있으며 기저유량은 갈수기 하천유량의 대부분을 차지하므로 직접유출과 기저유출의 분리는 중요하다. 또한, T-N, T-P는 기저유출에 영향을 많이 받는 수질 항목으로 기저유출과 직접유출에 의한 오염부하량을 정확히 분석해야한다. 따라서, 기저유출의 오염부하량 산정을 위해 기존의 WAPLE 2의 단점을 개선한 WAPLE 3가 개발되었으며, WAPLE 3는 유량 곡선의 하강부 변곡점에 붙는 특성을 가지고 있는 Baseflow filter program(BFlow) pass 1값을 사용하여 기저유량을 분리해 기저유출 오염부하량을 산정한다. WAPLE 3는 하천유량 중 기저유출을 어느 정도 분리하는지 결정하는 filter parameter 값을 Nathan과 McMahon가 제시한 최적값인 0.925를 사용하였다. 그러나 지형과 강우량 등에 따라 하천 유량에서의 기저유출 비율은 달라지기 때문에 이러한 한계점을 극복하기 위해 WAPLE 4를 개발하였다. WAPLE 4는 filter parameter 값을 사용자가 변경할 수 있게 개발하여 강우에 의한 유량변동 특정이 고려된 기저유량 및 오염부하를 산정하여 결과에 대한 정확도를 높였다. 또한, WAPLE 4는 강우시 오염부하량 산정에 탁월한 Numeric Integration(NI) 방법을 사용하여 직접유량, 기저유량의 오염부하량 및 유량가중평균농도(FWMC) 산정이 가능하도록 하였다. 본 연구의 결과는 오염총량제 및 기저유량 관리를 통한 유량 관련 정책 수립 시 기초자료로 활용될 수 있을 것으로 판단된다.

Ginseng root-derived exosome-like nanoparticles protect skin from UV irradiation and oxidative stress by suppressing activator protein-1 signaling and limiting the generation of reactive oxygen species

  • Background: Recently, plant-derived exosome-like nanoparticles (PDENs) have been isolated, and active research was focusing on understanding their properties and functions. In this study, the characteristics and molecular properties of ginseng root-derived exosome-like nanoparticles (GrDENs) were examined in terms of skin protection. Methods: HPLC-MS protocols were used to analyze the ginsenoside contents in GrDENs. To investigate the beneficial effect of GrDENs on skin, HaCaT cells were pre-treated with GrDENs (0-2 × 109 particles/mL), and followed by UVB irradiation or H2O2 exposure. In addition, the antioxidant activity of GrDENs was measured using a fluorescence microscope or flow cytometry. Finally, molecular mechanisms were examined with immunoblotting analysis. Results: GrDENs contained detectable levels of ginsenosides (Re, Rg1, Rb1, Rf, Rg2 (S), Gyp17, Rd, C-Mc1, C-O, and F2). In UVB-irradiated HaCaT cells, GrDENs protected cells from death and reduced ROS production. GrDENs downregulated the mRNA expression of proapoptotic genes, including BAX, caspase-1, -3, -6, -7, and -8 and the ratio of cleaved caspase-8, -9, and -3 in a dose-dependent manner. In addition, GrDENs reduced the mRNA levels of aging-related genes (MMP2 and 3), proinflammatory genes (COX-2 and IL-6), and cellular senescence biomarker p21, possibly by suppressing activator protein-1 signaling. Conclusions: This study demonstrates the protective effects of GrDENs against skin damage caused by UV and oxidative stress, providing new insights into beneficial uses of ginseng. In particular, our results suggest GrDENs as a potential active ingredient in cosmeceuticals to promote skin health.

Regulation of ADAMTS-2 by 1,25-Dihydroxyvitamin D3 in Osteoblastic Cells

  • Biosynthetic processing of fibrillar procollagens is essential for producing mature collagen monomers that polymerize into fibrils by a self-assembly process. The metalloproteinase ADAMTS-2 is the major enzyme that processes the N-propeptide of type I procollagen in the skin and also of type II and type III procollagens. Mutations in the ADAMTS-2 gene cause dermatospraxis in animals and Ehlers-Danlos syndrome VIIC in humans, both of which are characterized by the accumulation of type I pN-collagen and the formation of abnormal collagen fibrils in the skin. Despite its importance in procollagen processing, little is known about the regulation of ADAMTS-2 expression. Here, we demonstrate that ADAMTS-2 can be regulated by 1,25-dihydroxyvitamin D3, an inducer of type I procollagen synthesis. This steroid hormone induced ADAMTS-2 mRNA ${\sim}3-fold$ in MG-63 human osteosarcoma cells and MC3T3-E1 murine osteoblastic cells. This induction was dose- and time-dependent in MG-63 cells. In contrast, secreted ADAMTS-2 protein was increased only 1.4-fold with 1,25-dihydroxyvitamin D3. Finally, 1,25-dihydroxyvitamin D3 in the presence of ascorbate increased levels of secreted ADAMTS-2 1.9-fold over ascorbate treatment alone, which did not appreciably change ADAMTS-2 expression. These data indicate that the regulation of ADAMTS-2 is coupled with the synthesis of type I procollagen through 1,25-dihydroxyvitamin D3 signaling and may involve translational or posttranslational control.

덱사메타존이 골아유사세포의 성장과 분화에 미치는 영향 (The Effects of Dexamethasone on Growth and Differentiation of Osteoblast-like Cell)

  • The ultimate aim of periodontal treatment is periodontal regeneration, which necessiates the regeneration of bone tissues. To evaluate the effects of Dex growth and differentiation of MC3T3-E1 cells, cells were seeded in alpha-modified eagle medium containing 10% fetal bovine serum, 10mM beta-glycerophosphate , $50{\mu}g/ml$ of ascorbic acid, with or without $10^{-7}M$ Dex and examined cell proliferation activities, alkaline phosphatase activities, and bone nodule formation until 25days. The results were as follows : 1. In Dex group, cell proliferation activities were lower until 15 days compared to control group. Bone nodules formation were showed at 10 days. 2. In the time-response effect, ALP activities were increased until the 10 days in control groups thereafter decreased and ALP activities of Dex group were lower aspect than control group until the 10 days In this study, bone nodule formation of osteoblast-like cells were accelerated by Dex and cell proliferation activities, ALP activity of Dex group showed lower than control group. Dex was considered that it did suppress initial growth, but accerelate mineralization of osteoblast-like cells.

초등학교 저학년 학부모의 성지식과 성교육에 대한 인식 및 실태 비교 (Sexual Knowledge and Perception and Current Status of Sex Education among Parents of First and Second Grade Elementary Schoolers)

  • Purpose: This study was done to identify sexual knowledge and perception and current status of sex education among parents of first and second grade elementary school age children. Methods: Participants in the study were recruited from parents with a child in the lower grades and who resided in Seoul. From January 1 to February 28, 2011, data were collected using a self-report questionnaire. Data were analyzed using McNemer test, paired t-test, ANONA, and Scheffe test. Results: Mean sexual knowledge scores for fathers and mothers were $27.74{\pm}4.94$, and $28.62{\pm}4.70$ respectively. Parent's sexual knowledge correlated with their occupation, education level, and family's monthly income. Conclusion: Study results showed that both parents are aware of the need for sex education. However, at home, mothers have a more active attitude towards sex education and conduct the education more often than fathers.

혈관 플라그 형성 저해단백질에 대한 아연의 기능 (Role of zinc for calcification inhibitor protein in vascular smooth muscle cell plaque formation)

  • Purpose: Zinc, a biomineral present within and outside cells, manages various cellular mechanisms. In this study, we examined whether zinc was involved in vascular smooth muscle cell (VSMC) calcification via regulation of calcification inhibitor protein, osteopontin (OPN). Methods: Rat aorta cell line (A7r5 cells) and primary vascular smooth muscle cells (pVSMCs) from rat aorta were cultured with phosphate (1-5 mM) and zinc ($0-15{\mu}M$) as appropriate, along with osteoblasts (MC3T3-E1) as control. The cells were then stained for Ca and P deposition for calcification examination as well as osteopontin expression as calcification inhibitor protein was measured. Results: Both Ca and phosphate deposition increased as the addition of phosphate increased. In the same manner, the expression of osteopontin was upregulated as the addition of phosphate increased in both cell types. When zinc was added, Ca and P deposition decreased in VSMCs, while it increased in osteoblasts. Conclusion: The results imply that zinc may prevent VSMC calcification by stimulating calcification inhibitor protein OPN synthesis in VSMCs.
