Microcystin-LR (MC-LR) is a cyanobacterial hepatotoxin mainly produced by Microcystis aeruginosa. The current study examined the effects of a single intraperitoneal dose of MC-LR in rats. Female Sprague-Dawley rats were intraperitoneally injected with MC-LR ($100{\mu}g/kg$ body weight) and they were sacrificed at 0, 20, 40, 80, 160 min, or 12 h after injection. Clinically, animals showed lethargy and had ruffled hair beginning at 40 min post injection. In the gross findings, liver was enlarged and its color was changed into dark red beginning at 40 min post injection. Microscopically, dissociation of centrilobular hepatocytes and hemorrhage was observed in the hepatic central legions and such pathological changes were then extended to the portal regions of liver by time course manner. Interestingly at 80 min after MC-LR injection, the entrapped eosinophilic materials that may be necrotic fragments of dissociated hepatocytes were found in the capillaries of lung and renal glomerulus. Ultrastructurally, microvilli of the hepatocytes were disrupted or lost at all time points. Furthermore, the Disse space and gap junctions were widened beginning at 40 min post injection. These results suggest that liver is the major target organ of MC-LR and isolated hepatocytes by the effects of such hepatotoxin may secondarily reduce the physiological function of lung and kidney.
To determine the content of hazard microcystin (MC) in Han River system and Ecological Ponds in Seoul City and Kyunggi district, a most toxic derivative, microcystin-LR (MCLR) of 15 samples of 7 ponds, 4 rivers and 4 reservoirs during low precipitation and cold season in 2003 were analyzed by ELISA method. With the change of water temperature ($0.4\;{\sim}\;21.9^{\circ}C$), cyanobacteria including Microcystis aeruginosa dominated the cold phytoplankton community in small ecological ponds such as Kyungbokgung Kyunghyaeru (KBP), Seokchon reservoir (SCR), Yangsoori Ryukgakji (YSS), having the long residence time. Contents of MCLR (the detection limit; $0.05\;{\mu}g\;L^{-1}$) were high in cyanobacteria-rich sites, especially, Microcystis aeruginosa. Total MCLR, cell extracted type plus dissolved type, were $1.39\;{\mu}g\;L^{-1}$ in KBP, $0.55\;{\mu}g\;L^{-1}$ in SCR and $0.59\;{\mu}g\;L^{-1}$ in YSS, in the first sampling having a high temperature (>$20^{\circ}C$), while some detected only in YSS during the cold season. As expected, the MCLR content was correlated with Microcystis aeruginosa (r = 0.526 for cell extracted type, r = 0.433 for dissolved type). Therefore, low concentration of MCLR detected in small ponds and Han river system in Seoul metropolitan city and Kyunggi district, maybe hardly affect human recreation activity, especially the drinking water supply.
Different detection characteristics of fluorescence immunochromatography method and high performance liquid chromatography (HPLC) method for the analysis of cyanobacterial toxins were studied. In particular, low and high limits of detection, detection time and reproducibility and detectable microcystin species were compared when fluorescence immunochromatography method and high performance liquid chromatography method were applied for the detection of microcystin (MC), a cyclic peptide toxin of the freshwater cyanobacterium Microcystis aeruginosa. A Fluorescence immunochromatography assay system has the unique advantages of short detection time and low detection limit, and high performance liquid chromatography detection method has the strong advantage of individual quantifications of several species of microcystins.
This study examined the special technique of photocatalytic degradation (RPODisk) for removal of taste and odor causing materials, algae, and algal toxin. The RPODisk was effective for removal of these troublesome contaminants. It outperformed the fixed media and the UV irradiation for geosmin removal. The RPODisk performance was comparable to the combination of the UV irradiation with TiO2. The RPODisk performance was affected by the rotating speed. The faster the speed was, the better the performance. The RPODisk was also effective for removal of algae and algal toxin. The algal activity reduced by 80% after 30 mins of the treatment. More toxic microcystin (MC)-LR was more difficult to remove than MC-RR. The times for 50% removal were 23.7 mins for MC-LR and 14.1 mins for MC-RR. Almost 100 mins of the contact time was required to completely remove MC-LR at the rotating speed of 260 rpm.

Microcystin-LR (MC-LR) is one of most abundant microcystins, and is derived from blue-green algae bloom. Advanced oxidation processes (AOPs) are effective process when high concentrations of MC-LR are released into a drinking water treatment system from surface water. In particular, UV-based AOPs such as UV, $UV/H_2O_2$, $UV/O_3$ and $UV/TiO_2$ have been studied for the removal of MC-LR. In this study, UV-LED was applied for the degradation of MC-LR because UV lamps have demonstrated some weaknesses, such as frequent replacements; that generate mercury waste and high heat loss. Degradation efficiencies of the MC-LR (initial conc. = $100{\mu}g/L$) were 30% and 95.9% using LED-L (280 nm, $0.024mW/cm^2$) and LED-H (280 nm, $2.18mW/cm^2$), respectively. Aromatic compounds of natural organic matter changed to aliphatic compounds under the LED-H irradiation by LC-OCD analysis. For application to raw water, the Nak-dong River was sampled during summer when blue-green algae were heavy bloom in 2016. The concentration of extracellular and total MC-LR, geosmin and 2-MIB slightly decreased by increasing the LED-L irradiation; however, the removal of MC-LR by UV-LED (${\lambda}=280nm$) was insufficient. Thus, advanced UV-LED technology or the addition of oxidants with UV-LED is required to obtain better degradation efficiency of MC-LR.
Objectives: Microcystin (MC) produced during cyanobacterial blooms is a worldwide problem presenting a serious health threats to humans and ecosystems. During July through October of 2013, the Ilwol Reservoir experienced a high biomass of phytoplankton (maximum $211.7mg/m^3$ of Chlorophyll-a) containing the toxigenic cyanobacterium Oscillatoria sp. The aim of this study is to analyze MC concentration in the reservoir water, as well as in representative fish species (Carassius cuvieri, Carassius auratus, Channa argus). We also evaluated the human health risk of exposure to MCs accumulated in the fish. Methods: Concentrations of MCs in the water and fish samples were analyzed by liquid chromatography with a triple quadrupole tandem mass spectrometer (LC/MS/MS) and enzyme-linked immunosorbent assay (ELISA). Results: The total levels of four MC variants, including MC-LR, MC-RR, MC-YR and MC-LA were below the WHO drinking water guideline limit (1 ug MC-LR per liter) both for the dissolved and particulate fraction present in the water samples. The mean MC concentrations in the livers of all species were significantly higher than in the gills (p < 0.01) and muscles (p < 0.05). The values of estimated daily intake of MCs in muscles, the edible part of the fish, would be only $0.005-0.015{\mu}g/kg{\cdot}day$, much lower than WHO's provisional tolerable daily intake of $0.04{\mu}g/kg{\cdot}day$. Conclusion: This study suggests that, owing to the spatial distribution or temporal variation of MC, there is a need for careful monitoring of cyanotoxin in reservoir water and aquatic animals to protect public health.

Contamination of microcystins, a family of heptapeptide hepatotoxins, in eutrophic water bodies is a worldwide problem. Due to their poisoning effects on animals and humans, there is a requirement to characterize and quantify all microcystins present in a sample. As microcystins are, for most part, intracellular toxins produced by some genera of cyanobacteria, lysing cyanobacterial cells to release all microcystins is considered an important step. To date, although many cell lysis methods have been used, little work has been conducted comparing the results of those different methods. In this study, various methods for cell lysis and toxin extraction from the cell lysates were investigated, including sonication, bead beating, freeze/thaw, lyophilization and lysing with TritonX-100 surfactant. It was found that lyophilization, followed by extraction with 75% methanol, was the most effective for extracting toxins from Microcystis aeruginosa cells. Another important step prior to the analysis is removing impurities and concentrating the target analyte. For these purposes, a C18 Sep-Pak solid phase extraction cartridge was used, with the percentage of the eluent methanol also evaluated. As a result, methanol percentages higher than 75% appeared to be the best eluting solvent in terms of microcystin-leucine-arginine (MC-LR) recovery efficiency for the further chromatographic and mass spectrometric analyses.
The effect of permanganate oxidation was investigated as water treatment strategy with a focus on comparing the reaction characteristics of NaOCl and sodium permanganate (NaMnO4) in algae (Monoraphidium sp., Micractinium inermum, Microcystis aeruginosa)-contained water. Flow cytometry explained that chlorine exposure easily damaged algae cells. Damaged algae cells release intracellular organic matter, which increases the concentration of organic matter in the water, which is higher than by NaMnO4. The oxidation reaction resulted in the release of toxin (microcystin-LR, MC-LR) in water, and the reaction of algal organic matter with NaOCl resulted in trihalomethanes (THMs) concentration increase. The oxidation results by NaMnO4 significantly improved the concentration reduction of THMs and MC-LR. Therefore, this study suggests that NaMnO4 is effective as a pre-oxidant for reducing algae damage and byproducts in water treatment process.

최근 화두가 되고 있는 환경문제 중 하나로 녹조현상을 꼽을 수 있다. 녹조란 남세균이 대량 증식함으로써 물빛이 녹색으로 변하는 현상으로, 영양염류 및 수온 등 이화학적 요소뿐만 아니라체류시간과 같은 수리학적 요인까지 모두 충족되었을 때 발생한다. 심하면 고밀도의 스컴(scum)을 형성하며 독소와 악취를 동반하기도 한다. 유해 남세균이 생성하는 마이크로시스틴(microcystin, MC)이 함유된 물을 입 또는 코로 섭취시 간을 손상시킨다는 보고가 있으며, 최근 해외에서는 MC가 미세먼지처럼 공기 중에 떠다니다 수변에서 생활하는 사람의 호흡기로 들어가 건강 피해를 줄 수 있다는 연구가 속속 나오고 있다. 본 연구는 우리나라 최초의 수질개선용 댐인 영주댐을 연구 대상으로 삼아 수질 모델링을 구축하고 영주댐 운영에 따른 댐 상·하류 조류 변화를 정량적으로 분석하였다. 조류의 강도를 추정하는데 클로로필-a 농도를 사용하였으며, 분석 도구로는 국립환경과학원이 수질예측 및 평가 시 사용하는 EFDC(Environmental Fluid Dynamics Code) 모형을 활용하였다. 대상 구간의 실제 폭, 하상고 분포 등을 고려하여 수표면 격자망을 구현하였으며, 환경부에서 제공하는 수위 및 DO, TN, T-P, 클로로필-a 등을 활용하여 EFDC 모형의 수리 및 수질 재현성 검토를 하였다. 검·보정된 EFDC 모형으로 영주댐의 방류량 변화 및 댐의 개방과 같은 수리학적 요인을 제어하여 특정 지점의 조류 변화를 분석하였다.

Cyanobacteria have been used as pollution indicator species in freshwater ecosystems, and identifying their fluctuations can be an important part about management of surface waters globally. Cyanotoxins produced by cyanobacteria are directly or indirectly a threat to human and environmental health. In order to confirm the potential risk of these cyanotoxins, the fluctuations of phytoplankton and phylogenetic analysis of cyanotoxin synthetase genes were conducted at each point in the Yeongsan River water system in Gwangju from November 2021 to October 2022. Diatoms which grow well in winter were dominant at 99.4 ~ 99.5%, and diatoms and green algae were dominant from the spring to autumn when the water temperature rises. Stephanodiscus spp. were dominant at 92.7 to 97.5 % at all sites in the winter, and Aulacoseira spp., which grow in warm water temperatures, were dominant in summer and autumn. Microcystis aeruginosa was dominant at 25.2% in summer only at site 5. mcyB and anaC have been detected as cyanotoxin synthetase genes. The phylogenetic tree of anaC could be divided into two groups (Group 1 & Group 2). Group 1 contained Aphanizomenon sp. and Cuspidothrix issatschenkoi. It is combined with Aphanizomenon sp. and Cuspidothrix issatschenkoi, which are known to produce cyanotoxins.
