고준위 방사성폐기물 처분장에서 초기 용기 파손 시나리오의 장기 방사선적 안전성 평가 (Post Closure Long Term Safety of an Initial Container Failure Scenario for a Potential HLW Repository)

  • 고준위 방사성폐기물 처분장에서 적용하고 있는 다중 방벽의 한 부분인 처분 용기는 벤토나이트 완충재의 팽윤과 지압으로부터 폐기물을 역학적으로 안전하게 보호함과 동시에 일정 기간 방사성폐기물의 유출을 억제하는 역할을 한다. 용기는 엄격한 재질 선정과 품질 보증을 거쳐 건전성을 확보하나 보수적인 관점에서 보면 용기 제작 과정이나 수송 중 예상치 못한 사건으로 인해 불량품이 발생할 개연성이 있다. 본 연구에서는 이와 같은 사고 시나리오를 가정할 경우 불량 용기를 포함한 전체 용기에 거치된 방사성폐기물의 시간에 따른 환경 위해도를 평가하였다. 본 연구결과 일부 처분 용기에 초기 파손이 발생하더라도 규제치를 잘 만족하는 것으로 판명되었다.

1-Deoxynojirimycin 생산 균주 Bucillus subtilis MORI 3K-85의 단백질 분석 (Protein Analysis of Bacillus subtilis MORI 3K-85 with Reference to the Biosynthesis of 1-Deoxynojirimycin)

  • In our previous study, we isolated and characterized a 1-deoxynojirimycin (DNJ)-producing bacterium, Bacillus subtilis MORI, from chungkookjang, a Korean traditional food. B. subtilis MORI was subjected to ${\gamma}$-irradiation and the resulting bacteria were screened for increased DNJ production. A mutant was identified that produced 7.6 times more DNJ and named B. subtilis MORI 3K-85. In this study, the protein profiles of both strains were compared by one-dimensional and two-dimensional gel electrophoresis (1-DE and 2-DE, respectively) under both native and denaturing conditions. The 1-DE native-PAGE and 1-DE SDS-PAGE analyses identified 5 and 7 bands, respectively, that were found at higher concentrations in B. subtilis MORI 3K-85 than in B. subtilis MORI. Similarly, 2-DE analyses identified 20 protein spots which were found at higher concentrations in B. subtilis MORI 3K-85. The peptide mass profiles of these 20 proteins were analyzed by MALDI-TOF and compared with peptide sequences of B. subtilis and B. amyloliquefaciens in the MASCOT database. This screening suggested that three dehydrogenases, an aldolase, a synthetase, an isomerase, a reductase, and a peroxidase are elevated in B. subtilis MORI 3K-85. Based on this data, one or more of the elevated 8 enzymes might be related to the DNJ biosynthetic pathway.

Reinterpretation of the protein identification process for proteomics data

  • Introduction: In the mass spectrometry-based proteomics, biological samples are analyzed to identify proteins by mass spectrometer and database search. Database search is the process to select the best matches to the experimental mass spectra among the amino acid sequence database and we identify the protein as the matched sequence. The match score is defined to find the matches from the database and declare the highest scored hit as the most probable protein. According to the score definition, search result varies. In this study, the difference among search results of different search engines or different databases was investigated, in order to suggest a better way to identify more proteins with higher reliability. Materials and Methods: The protein extract of human mesenchymal stem cell was separated by several bands by one-dimensional electrophorysis. One-dimensional gel was excised one by one, digested by trypsin and analyzed by a mass spectrometer, FT LTQ. The tandem mass (MS/MS) spectra of peptide ions were applied to the database search of X!Tandem, Mascot and Sequest search engines with IPI human database and SwissProt database. The search result was filtered by several threshold probability values of the Trans-Proteomic Pipeline (TPP) of the Institute for Systems Biology. The analysis of the output which was generated from TPP was performed. Results and Discussion: For each MS/MS spectrum, the peptide sequences which were identified from different conditions such as search engines, threshold probability, and sequence database were compared. The main difference of peptide identification at high threshold probability was caused by not the difference of sequence database but the difference of the score. As the threshold probability decreases, the missed peptides appeared. Conversely, in the extremely high threshold level, we missed many true assignments. Conclusion and Prospects: The different identification result of the search engines was mainly caused by the different scoring algorithms. Usually in proteomics high-scored peptides are selected and low-scored peptides are discarded. Many of them are true negatives. By integrating the search results from different parameter and different search engines, the protein identification process can be improved.

중소기업의 베트남 진출을 위한 환경분석 및 진입 방안에 관한 연구 (A Study of Environment Analysis and Entry Plan for Medium and Small Enterprises' Vietnam Advance)

  • 최근 우리나라의 기업들은 해외진출을 고려하거나 이미 진출한 경우가 많다. 과거 기업의 해외진출은 대부분 대기업 주도의 해외진출로 이루어졌으나 대외적 환경 변화와 기술 및 자본도입의 자유화 등 경제 개방화로 인해 대기업뿐만 아니라 중소기업도 해외진출을 통한 경쟁력 확보가 필요불가결한 요인으로 인식되고 있다. 우리나라의 경우 중국으로의 진출이 애로를 겪고 있는 중 대안으로서 베트남 진출은 점차 확대되고 있다. 따라서 본 연구에서는 우리나라 기업의 베트남 진출과 관련하여 경제, 정치, 사회적인 환경 분석을 통하여 효율적이고 바람직한 베트남 진출 전략수립을 위한 방안을 제시하고자 하였다. 본 연구에서는 이러한 목적에 따라 이론적 고찰을 통하여 베트남 진출 관련 국내.외 문헌조사와 선행연구를 토대로 하여 이론적 고찰을 행하였으며 진출방안으로서 향후 우리나라 중소기업의 유망진출 분야로는 에너지 산업, 정유 및 석유화학산업, 전력 산업, 기타분야로서 건설 자재 관련 산업, 노동집약적 제조업, 수산업 농산물 재배, 가공업 등을 들 수 있다.
