Microwave를 이용한 식물세포배양으로부터 paclitaxel 추출 (Microwave-assisted extraction of paclitaxel from plant cell cultures)

  • 본 연구에서는 식물세포배양액에서 회수한 식물세포인 biomass로부터 항암제 paclitaxel을 효율적으로 회수하기 위해, microwave를 이용한 추출 (MAE) 방법을 도입하여 기존의 유기용매 추출 (CSE) 방법과 추출효율을 비교하였다. CSE 방법에서 추출온도 25, $30^{\circ}C$에서는 추출시간 (3, 6, 10 min)에 대해 추출효율이 온도에 영향을 받지 않은 반면, 40, $50^{\circ}C$에서는 추출시간 10 min에서 paclitaxel 수율이 감소하는 경향을 보여 온도에 영향을 받음을 알 수 있었다. CSE 방법의 경우 총 4회의 추출로 식물세포 내 대부분의 paclitaxel을 회수 (>99%) 가능한 반면 MAE 방법의 경우 1회의 추출로 식물세포 내 대부분의 paclitaxel을 회수 (>99%) 가능하였다. MAE 방법이 CSE 방법보다 paclitaxel를 회수하는데 있어서 높은 추출효율, 공정시간 단축, 에너지 절감 효과가 있음을 알 수 있었다. 또한 MAE 공정에서 최적의 biomass/methanol의 비율은 1/1 (w/v)임을 알 수 있었다.

Effect of Microwave Assisted Water Extraction on Insoluble Phenolic Compounds through Bioactivity of Fagopyrum esculentum

  • Fagopyrum esculentum (Buckwheat) is a globally used alternative crop that contains several useful substances with various effects; however, many of these substances (rutin, quercetin, etc.) are water insoluble. To extract these substances, alcohols is required, which is inconvenient because these solvents cause diverse problems. Many studies are underway to achieve effective extraction of these substances with water. Among of these studies, microwave assisted water extraction (MAE) has been performed extensively. In this study, we performed the extraction in various solvents and/or microwave from Fagopyrum esculentum. The analysis of the content of useful substances and the bioactivity were performed and shown to increase in MAE. Liquid chromatography-mass was performed in order to identify of the useful water-insoluble substances. Catechin, quercetin, and rutin, which are all insoluble in water, were hardly extracted with water even on heating (4.4 ppb, 3.9 ppb and 60.3 ppb, respectively). However, MAE was found to extract much more of these substances than water (1204 ppb, 110.8 ppb and 2946 ppb, respectively). Although less efficient than alcohols, MAE showed much higher efficiency than simple water extraction. These results indicate that water extraction using microwave technology is effective in cases where it is difficult to extract useful substances using water.

  • 고속 움직임 추정 방법들에서는, 움직임 추정 오차는 최적 탐색점에서 멀어질수록 단조 증가한다는 성질을 이용하여, 계산량을 줄인다 본 논문에서는 먼저, 참조 탐색점들의 MAE (mean absolute error) 차가 클 때에, 대부분의 움직임 벡터는 작은 MAE의 참조 탐색점 방향에서 나타난다는 통계적 특성을 조사하였다 그러므로, 이 특성을 이용하여 탐색점 수를 줄일 수 있는 고속 움직임 추정 방법을 제안한다 모의 실험 결과, 기존의 고속 움직임 추정 방법들에 비해서는 비슷한 성능을 유지하면서 계산량을 줄일 수 있었다.

Comparison of Gut Microbiota between Lean and Obese Adult Thai Individuals

  • Current reports suggest that obesity is a serious global health issue. Emerging evidence has predicted strong links between obesity and the human gut microbiota. However, only a few such studies have been conducted in Asia, and the gut microbiota of lean and obese adult Asians remains largely unexplored. Here, we investigated the potential relationship between gut microbiota, body massindex (BMI), and metabolic parameters in adults from Thailand, where obesity is increasing rapidly. Fecal and blood samples were collected from 42 volunteers who were allocated into lean, overweight, and obese groups. The fecal microbiota was examined by quantitative PCR analysis. Bacteroidetes, Firmicutes, and Staphylococcus spp. and methanogens were most abundant in lean volunteers. Overweight volunteers majorly harbored Christensenella minuta and Akkermansia muciniphila, ${\gamma}-Proteobacteria$, and bacteria belonging to the genus Ruminococcus. Methanogens and bacteria belonging to the phylum Bacteroidetes were negatively correlated with adiposity markers (BMI and waist circumference), but positive correlated with high-density lipoprotein, suggesting that they can be used as leanness markers. While some of our results agree with those of previous reports, results regarding the contributions of specific taxa to obesity were inconsistent. This is the first study to report the adult gut microbiota in Southeast Asian populations using molecular techniques and biochemical markers and provides a foundation for future studies in this field.

태양광 에너지 예측을 위한 기상 데이터 기반의 인공 신경망 모델 구현 (Solar Energy Prediction Based on Artificial neural network Using Weather Data)

  • 태양광발전시스템은 태양광으로부터 에너지를 생산하는 발전기술이며, 신재생 에너지 기술 중 가장 빠르게 성장하고 있다. 태양광 발전 시스템은 부하에 안정적으로 에너지를 공급하는 것이 가장 중요시 된다. 그러나 날씨 및 기상 조건에 따라 에너지 생산이 불안정하기 때문에 에너지 생산량에 대한 정확한 예측이 필요하다. 본 논문에서는 강수량, 장 단파 복사선 평균, 온도 등 15가지 종류의 기상 데이터를 사용하여 태양광 에너지를 예측하는 인공 신경망(ANN)을 구현하고 성능을 평가한다. 인공 신경망은 은닉층을 구성하고 오버피팅을 방지하기 위한 페널티 ${\alpha}$와 같은 파라미터를 조절하여 구현한다. 예측모델의 정확도와 타당성을 검증하기 위해 성능지표로 MAPE(Mean Absolute Percentage Error)와 MAE(Mean Absolute Error)를 사용한다. 실험 결과 Hidden Layer $Sizes=^{\prime}16{\times}10^{\prime}$을 사용하였을 때 MAPE=19.54와 MAE=2155345.10776로 나타났다.

Freshwater Snail Diversity in Mae Lao Agricultural Basin (Chiang Rai, Thailand) with a Focus on Larval Trematode Infections

  • The aim of this study was to conduct a freshwater snail survey in Mae Lao agricultural basin to assess the diversity with a focus on habitat types and their larval trematode infections. Snails were collected and examined in 14 sites of Mae Lao agricultural basin from August 2016 to October 2017. A total of 1,688 snail individuals were collected and classified into 7 families, 8 genera, and 12 species. Snail diversity and habitat types were higher in rice paddies than irrigation canals and streams. The most abundant species was Bithynia siamensis siamensis, representing 54.6% of the sample. Three species of snails act as first intermediate host were found with cercarial infections. They were Filopaludina sumatrensis polygramma, B. s. siamensis, and Melanoides tuberculata. The cercariae were categorized into 7 types; echinostome, monostome, gymnocephalous, virgulate, parapleurolophocercous, pleurolophocercous and megalurous cercariae. Parapleurolophocercous cercariae constituted the most common type of cercariae recovered, contributing 41.2% of all infections in snails. Echinostome metacercariae infections were found in 6 snail species with 7.6% prevalence. In addition, the metacercaria of avian trematode, Thapariella sp. were found in Filopaludina spp. snails and B. funiculata with a prevalence of 0.5%. This is the first report for Thapariella metacercariae in the snail host, B. funiculata, and also confirmed that viviparid and bithyniid snails act as the second intermediate hosts of this trematode. This work will provide new information on the distribution and intermediate host of trematode in this area.

An optimized microwave-assisted extraction method for increasing yields of rare ginsenosides from Panax quinquefolius L.

  • Background: Rare ginsenosides in Panax quinquefolius L. have strong bioactivities. The fact that it is hard to obtain large amounts of rare ginsenosides seriously restricts further research on these compounds. An easy, fast, and efficient method to obtain different kinds of rare ginsenosides simultaneously and to quantify each one precisely is urgently needed. Methods: Microwave-assisted extraction (MAE) was used to extract nine kinds of rare ginsenosides from P. quinquefolius L. In this article, rare ginsenosides [20(S)-Rh1, 20(R)-Rh1, Rg6, F4, Rk3, 20(S)-Rg3, 20(R)-Rg3, Rk1, and Rg5] were identified by high performance liquid chromatography (HPLC)-electrospray ionization-mass spectrometry. The quantity information of rare ginsenosides was analyzed by HPLC-UV at 203 nm. Results: The optimal conditions for MAE were using water as solvent with the material ratio of 1:40 (w/v) at a temperature of $145^{\circ}C$, and extracting for 15 min under microwave power of 1,600 W. Seven kinds of rare ginsenosides [20(S)-Rh1, 20(R)-Rh1, Rg6, F4, Rk3, Rk1, and Rg5] had high extraction yields, but those of 20(S)-Rg3 and 20(R)-Rg3 were lower. Compared with the conventional method, the extraction yields of the nine rare ginsenosides were significantly increased. Conclusion: The results indicate that rare ginsenosides can be extracted effectively by MAE from P. quinquefolius L. in a short time. Microwave radiation plays an important role in MAE. The probable generation process of rare ginsenosides is also discussed in the article. It will be meaningful for further investigation or application of rare ginsenosides.

마이크로웨이브 추출조건에 따른 섬쑥부쟁이 추출물의 총 폴리페놀 함량 및 전자공여 작용 변화 (Changes of Total Polyphenol Content and Electron Donating Ability of Aster glehni Extracts with Different Microwave-assisted Extraction Conditions)

  • 마이크로웨이브 추출방법과 환류냉각 추출방법을 비교한 결과, 물과 메탄올 및 에탄올 혼합용매로 추출한 경우 마이크로웨이브 추출 방법에 의하여 추출시간을 단축시키면서 환류냉각 추출방법에서와 같은 수준의 추출율과 총 폴리페놀 함량을 갖는 섬쑥부쟁이 추출물을 얻을 수 있었다. 마이크로웨이브 추출시 최적 마이크로웨이브 에너지는 $120{\sim}150$ W였고 추출시간의 경우 $4{\sim}8$분이 적당하였다. 추출에 사용한 용매들 가운데 에탄올, 메탄올보다 물 그리고 물과 에탄올 또는 메탄올 혼합용매를 사용한 추출물의 추출율, 총 폴리페놀 함량 및 전자공여 효과가 높은 것으로 나타났다.
