8-60hIPP5m-Induced G2/M Cell Cycle Arrest Involves Activation of ATM/p53/p21cip1/waf1 Pathways and Delayed Cyclin B1 Nuclear Translocation

  • Protein phosphatase 1 (PP1) is a major serine/threonine phosphatase that controls gene expression and cell cycle progression. The active mutant IPP5 ($8-60hIPP5^m$), the latest member of the inhibitory molecules for PP1, has been shown to inhibit the growth of human cervix carcinoma cells (HeLa). In order to elucidate the underlying mechanisms, the present study assessed overexpression of $8-60hIPP5^m$ in HeLa cells. Flow cytometric and biochemical analyses showed that overexpression of $8-60hIPP5^m$ induced G2/M-phase arrest, which was accompanied by the upregulation of cyclin B1 and phosphorylation of G2/M-phase proteins ATM, p53, $p21^{cip1/waf1}$ and Cdc2, suggesting that $8-60hIPP5^m$ induces G2/M arrest through activation of the ATM/p53/$p21^{cip1/waf1}$/Cdc2/cyclin B1 pathways. We further showed that overexpression of $8-60hIPP5^m$ led to delayed nuclear translocation of cyclin B1. $8-60hIPP5^m$ also could translocate to the nucleus in G2/M phase and interact with $pp1{\alpha}$ and Cdc2 as demonstrated by co-precipitation assay. Taken together, our data demonstrate a novel role for $8-60hIPP5^m$ in regulation of cell cycle in HeLa cells, possibly contributing to the development of new therapeutic strategies for cervix carcinoma.

  • In the present study, the effect of cysteine and NT or bisphenol A(BP) on in vitro aturation(IVM) of porcine oocytes were examined. COCs was cultured in NCSU-23 medium supplement with 10% FCS which had previously been covered with mineral oil and equilibrated in a humidified atmosphere of 5% $CO_2$ and 95% air at $38^{\circ}C$. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 medium supplement with 0.5~10.0 mM cysteine were $34.0{\pm}3.2%$, $36.0{\pm}3.5%$, $48.0{\pm}3.8%$, $22.0{\pm}3.2%$, respectively. The IVM rate of oocytes cultured in NCSU-23 medium supplement with 0.5~5.0mM NT for 48 hrs were $24.0{\pm}4.2%$, $18.0{\pm}4.9%$, $8.0{\pm}2.2%$, respectively. NT affects oocyte in vitro maturation rate in a dose-dependent. This result were significantly lower than the control group. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 medium supplement with 1.0 mM NT+5.0 mM cysteine($38.0{\pm}4.3%$) were significantly higher than that of NT treatment. The IVM rate of oocytes cultured in NCSU-23 medium supplement with 0.05~5.0 mM BP for 48 hrs were $20.0{\pm}4.7%$, $10.0{\pm}5.3%$, $6.0{\pm}3.2%$, respectively. The IVM rate of oocytes cultured in NCSU-23 medium supplement with BP was significantly lower cultured non supplement of BP ($44.0{\pm}3.5%$). BP affects porcine oocyte maturation rate in a dose-dependent manner. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 medium supplement with 1.0 mM BP+5.0 mM cycteine ($32.0{\pm}3.2%$) were increased than that of BP treatment.

Mass Size Distribution of Atmospheric Aerosol Particles Collected at 865 m High of Jirisan National Park (지리산 국립공원 해발 865 m 지점에서 대기에어로졸입자의 입경별 질량농도 특성)

  • Aerosol mass size distributions were investigated at 865 m high the of Jirisan national park. A nanosampler cascade impactor was used to collect aerosols. The atmospheric aerosol particles had a unimodal mass size distribution, which peaked at $0.5-1.0{\mu}m$, and a mass aerodynamic diameter of $1.13{\mu}m$. The annual average concentrations of TSP, $PM_{10}$, $PM_{2.5}$, $PM_1$, $PM_{0.5}$ and $PM_{0.1}$ were $20.9{\mu}g/m^3$, $19.3{\mu}g/m^3$, $14.9{\mu}g/m^3$, $10.7{\mu}g/m^3$, $5.3{\mu}g/m^3$, $1.2{\mu}g/m^3$, respectively. TSP concentrations were below $30{\mu}g/m^3$ during the sampling period. On average $PM_{10}$, $PM_{2.5}$, $PM_1$, $PM_{0.5}$ and $PM_{0.1}$ made up 0.91, 0.70, 0.41, 0.19 and 0.07 of TSP, respectively. The annual average of PM2.5/PM10 ratio was 0.77.

  • The synthesis and characterization of the mononuclear group 13 heterocyclic carboxaldehyde methyldithiocarbazate complexes Me2M[NC5H4CRNNC(S)SCH3] (M=Al, R=H(1); M=Ga, R=H(2); M=Al, R=CH3(3); M-Ga, R=CH3(4); M=In, R=CH3(5)) are described. Compounds 1-5 were prepared by the reaction of MMe3 (M=Al, Ga, In) with 2-formy or 2-acetylpyridine-S-methyldithiocarbazate in toluene. These compounds 1-5 have been characterized by microanalysis, NMR (1H, 13C) spectroscopy, mass spectra, and single-crystal X-ray diffraction. X-ray single-crystal diffraction analyses reveal that 4-5 are mononuclear metal compounds with coordination number of 5 and N,N,S coordination mode.

  • This paper describes the design of a two stage 1V power supply SiGe Low Noise Amplifier operating at 5.25 GHa for 802.lla wireless LAN application. The achieved performance includes a gain of 17 ㏈, noise figure of 2.7㏈, reflection coefficient of 15 ㏈, IIP3 of -5 ㏈m, and 1-㏈ compression point of -14㏈m. The total power consumption of the circuit was 7 mW including 0.5mW for the bias circuit.

Purification and Properties of Intracellular Cytosine Deaminase from Chromobacterium violaceum YK 391

  • Cytosine deaminase (cytosine aminohydrolase, EC 3.5.4.1) stoichiometrically catalyzes the hydrolytic deamination of cytosine and 5-fluorocytosine to uracil and 5-fluorouracil, respectively. The intracellular cytosine deaminase from Chromobacterium violaceum YK 391 was purified to apparent homogeneity with 272.9-fold purification with an overall yield of $13.8\%$. The enzyme consisted of dimeric polypeptides of 63 kDa, and the total molecular mass was calculated to be approximately 126 kDa. Besides cytosine, the enzyme deaminated 5-fluorocytosine, cytidine, 6-azacytosine, and 5-methylcytosine, but not 5-azacytosine. Optimum pH and temperature for the enzyme reaction were 7.5 and $30^{\circ}C$, respectively. The enzyme was stable at pH 6.0 to 8.0, and at 30T for a week. About $70\%$ of the enzyme activity was retained at $60^{\circ}C$ for 5 min. The apparent $K_{m}$ values for cytosine, 5-fluorocytosine, and 5-methylcytosine were calculated to be 0.38 mM, 0.87 mM, and 2.32 mM, respectively. The enzyme activity was strongly inhibited by 1 mM $Hg^{2+},\;Zn^{2+},\;Cu^{2+},\;Pb^{2+},\;and\;Fe^{3+}$, and by o-phenanthroline, $\alpha,\;{\alpha}'$-dipyridyl, p-choromercuribenzoate, N-bromosuccinimide, and cWoramine­T. In addition, the enzyme activity was strongly inhibited by I mM 2-thiouracil, and weakly inhibited by 2-thiocytosine, or 5-azacytosine. Finally, intracellular and extracellular cytosine deaminases from Chromobacterium violaceum YK 391 were found to have a different optimum temperature, apparent $K_{m}$ value, and molecular mass.

Effects of Glutamine, Glucosamine and Glutathione on the In Vitro Maturation of Porcine Oocytes

  • This study was carried out to investigate the effects of the supplementation of glutamine, glucosamine and glutathione on the porcine oocytes on IVM rates. Cocs were incubated in NCSU-23 supplemented with at $2.0{\sim}10.0\;mM$ glucosamine, $0.5{\sim}4.0\;mM$ glutamine and $0.1{\sim}1.0\;mM$ glutathione for 48 hrs. Oocytes were transferred to 50 ul drops of maturation medium covered with mineral oil and cultured in a $CO_2$ incubator ($38^{\circ}C$, 5% $CO_2$, 95% air). The IVM rates of oocytes cultured in NCSU-23 supplemented with 0.5, 1.0, 2.0 and 4.0 mM glutamine for 48 hrs were $46.0{\pm}4.5%$, $52.0{\pm}4.8%$, $50.0{\pm}4.2%$ and $44.0{\pm}4.5%$, respectively. The IVM rates of oocytes cultured in NCSU-23 supplement with 2.0, 5.0, 7.0, 10.0 mM glucosamine for 48 hrs were $44.0{\pm}4.5%$, $42.0{\pm}4.5%$, $38.0{\pm}4.6%$ and $24.0{\pm}4.8%$, respectively. The IVM rates of oocytes cultured in NCSU-23 supplemented with glucosamine were no significantly increased compare to the control ($42.5{\pm}4.0%$). The IVM rate of oocytes cultured in NCSU-23 supplemented with 3.0, 5.0, 7.0, 10.0 mM glutathione for 48 hrs were $40.0{\pm}3.2%$, $54.0{\pm}4.2%$, $48.0{\pm}4.5%$, $44.0{\pm}4.8%$, respectively. The IVM rate of oocytes cultured in NCSU-23 supplemented with glutamine and glutathione were significantly increased co~pared to those control ($42.5{\pm}4.0%$). Glucosamine did not affect the IVM rates of oocytes. IVM rates of oocytes cultured in NCSU-23 medium for 48 hrs were significantly increased compared to the cultured for 40 hrs.

Simultaneous traveltime inversion of surface and borehole seismic data in Pungam basin (풍암분지 시험시추공 주변에서의 지표 및 시추공 초동주시 동시역산)

  • Velocity structures were defined in the vicinty of the 140-m deep test borehole in the pungam basin through simultaneous inversion of surface seismic refraction and far-ofset VSP traveltime data. Seismicenergy generated at the surface by a seisgun was recorded both at 42 surface locations at 3-m intervalsalong the profiles in the N20E and its orthogonal directions and at 71 m depth in the borehole. Forthe ofset VSP study, seismic energy was generated by a 5 kg sledgehamer at the surface in the horizontal ofset range of -19.5∼+19.5 m from the borehole. The seismic signals were detected at 9∼99 m depths with 1∼2 m intervals and recorded for 204 ms per shot. After shot static corrections,first-arrival times picked from both the surface refraction and borehole records were simultaneouslyinverted to yield velocity tomograms. The tomograms indicate that a 1.5 m thick soil layer with velocities les than 500 m/s overlies basements having a velocity range of 3,067 ∼5,717 m/s. Within the basements,∼4 m and deeper than 71 m. The high-velocit yzones may be due to conglomerates intercalated with sandstones and siltstones. No evidence for large-scale fracture zones or faults is detected near the borehole

  • In order to establish the basic data for the active use of the LED light source in the aquaculture industry, the experiment about the behavior of rockfish (Sebastes inermis) corresponding to the LED light was conducted by image analysis. LED lights for the experiment were one red light (wave length: 622 nm; light power: 811 mW) and two green lights (wave length: 518 nm; light power: 648 mW, wave length: 518 nm; light power: 810 mW). Behavior of the rockfish was analyzed using a moving distance (MD5) for 5 minutes where MD5s during the period of feeding were 20.0 m, 19.6 m, 16.3 m and 20.5 m in the ch1, ch2, ch3 and ch4. At the sunrise, mean MD5 of the entire channel about right before and right after were 6.3 m and 8.2 m. At the sunrise, mean MD5s of the entire channel about right before and right after were 13.6 m and 12.0 m. In the ch1, ch2 and ch3, mean MD5s during the period of 24 hours were 13.2 m, 9.6 m and 6.8 m at experiment 1and 5.2 m, 3.8 m and 2.9 m in the experiment 2.

Effect of Gamma Energy of Positron Emission Radionuclide on X-Ray CT Image (양전자 방출 핵종(18F)의 감마에너지가 X선 CT영상에 미치는 영향)

  • This study is aimed to assess the effect of the gamma ray of 511keV energy which is emitted from phantom where the positron emission radionuclide was injected on X-ray CT image. As a scanning method, the CT number and pixel value of the reference image where water was injected(0 mCi), and those acquired by changing the capacity of 18F(Fluorine), positron emission radionuclide, into 1 mCi, 2 mCi, 5 mCi, and 10 mCi were measured. As a result of measuring the CT number(HU) of the phantom image where the positron emission radionuclide($^{18}F$) was injected, there were reference water ($-7.58{\pm}0.66$ HU), 1 mCi($-9.85{\pm}0.50$ HU), 2 mCi($-10.27{\pm}0.21$ HU), 5 mCi($-11.31{\pm}0.66$ HU), and 10 mCi($-13.47{\pm}0.38$ HU). Compared with the image where it was filled with water, there was a reduction of 5.89 Hu in 10 mCi, 3.73 in 5 mCi, 2.69 HU in 2 mCi, and 2 HU in 1 mCi. As for the pixel value of the phantom image, there were reference water ($-2.70{\pm}0.75$), 1 mCi($-4.72{\pm}0.58$), 2 mCi($-6.01{\pm}0.78$), 5 mCi($-6.10{\pm}0.84$), and 10 mCi($-8.20{\pm}0.60$). Compared with the reference image, there was a reduction of 5.50 in 10 mCi, 3.40 in 5 mCi, 3.10 in 2 mCi, and 2.02 in 1 mCi. Through this experiment, it was indicated that, with the increase in the dose of the positron emission radionuclide($^{18}F$), the CT number and the pixel value of the image reduced proportionally, and the width of reduction showed a similar value, too. Accordingly, according to the degree of change in X-ray CT image due to the positron emission radionuclide in the quality control item of PET/CT, the proper standard should be established and it should be periodically managed.
