산개성단 M38 (NGC 1912) 영역의 새로운 변광성 I: δ Scuti와 γ Doradus 변광성 (NEW VARIABLE STARS IN THE REGION OF THE OPEN CLUSTER M38 (NGC 1912) I: δ SCUTI AND γ DORADUS TYPE STARS)

  • From the short-period variability survey (SPVS) in Bohyunsan Optical Astronomy Observatory, we obtained time-series BV CCD images in the region of an open cluster M38 (NGC 1912) for 23 nights from February 1, 2007 to December 7, 2008. We found 18 $\delta$ Scuti stars and 2 $\gamma$ Doradus stars. Among them, only 3 $\delta$ Scuti stars were known ones. From the color-magnitude diagram (CMD) of M38, most of them have been located in the instability strip. The two $\gamma$ Doradus stars have been located within $\gamma$ Doradus strip of M38. But, only four $\delta$ Scuti stars were located within radius 10' from the center of M38. To confirm of their membership for M38, we need more study, such as, proper motion by spectroscopic observation.

임파구 CD38의 효소학적 연구 (Enzymatic study on lymphocyte CD38)

  • Murine CD38 is a 42 kDa type II glycoprotein expressed on cell surface of both B and T lymphocytes. CD38 is a multifunctional enzyme that catalyzes the formation and hydrolysis of cyclic adenosine diphosphoribose (cADPR): ADP-ribosyl cyclase activity of CD38 catalyzes the formation of cADPR from NAD and cADPR hydrolase activity of CD38 catalyzes the hydrolysis of cADPR to ADP-ribose (ADPR). And also, CD38 has the catalytic activity of NAD glycohydrolase (NADase) which catalyzes the hydrolysis of catalyzes the formation and hydrolysis of cyclic adenosine diphosphoribose (cADPR): ADP-ribosyl cyclase activity of CD38 catalyzes the formation of cADPR from NAD to ADPR. In this study, we attempted to purify CD38 from mouse lymphocytes by using the immobilized anti-CD38 monoclonal antibody. The single step immuno-affinity column chromatography resulted in homogeneous purification, showing a single protein of 42 kDa on a SDS polyacrylamide gel. We have investigated the effects of various inhibitors on the enzyme activities of the purified CD38. Cibacron blue (0.5 mM) inhibited all three enzyme activities of CD38, NADase, ADP-ribosyl cyclase and cADPR hydrolase activities. ADPR (2 mM) showed inhibitory effect on both cADPR hydrolase activity and NADase, but not on ADP-ribosyl cyclase activity. However, ATP (2 mM) inhibited only cADPR hydrolase activity. $Zn^{2+}$ (1 mM) showed similar inhibitory effect as that of ADPR, but activated cyclase activity These results suggest that CD38 has three different catalytic activity domains which might be differentially regulated by their specific inhibitors.

Effect of Slaughter Age on Beef Color Stability during Display of Four Muscles from Japanese Black Steers

  • Effect of slaughter age (24, 28 and 38 months of age) on beef color stability during display of m. serratus ventralis, m. psoas major, m. semitendinosus and m. longissimus thoracis from Japanese Black steers was studied. Steak samples from muscles were over-wrapped with PVC film and displayed under fluorescent lights at $4^{\circ}C$ for 12 days. Percentages of metmyoglobin of steak samples were determined at days 0, 3, 6, 9 and 12. The percentage of metmyoglobin of m. psoas major at day 3 of display in the 24 months group was lower (p<0.05) than that in the 38 months group. The percentage of metmyoglobin of m. semitendinosus at day 6 of display in the 38 months group was higher (p<0.05) than that in the other groups. The percentage of metmyoglobin of m. longissimus thoracis at day 3 of display in the 24 months group was lower (p<0.01) than that in the other groups. The percentage of metmyoglobin of m. longissimus thoracis at day 6 (p<0.01), 9 (p<0.01) and 12 (p<0.05) of display in the 38 months group were higher than those in the other groups. Crude fat concentration in m. longissimus thoracis increased (p<0.05) after 28 months of age. $\alpha$-Tocopherol concentration in m. serratus ventralis in the 38 months group was higher (p<0.001) than that in the other groups. In m. psoas major the $\alpha$-tocopherol concentration in the 38 months group was higher (p<0.05) than that in the 24 months group. The $\alpha$-tocopherol concentration in m. longissimus thoracis increased (p<0.001) with age. These results suggested that in spite of increase in both the crude fat and the $\alpha$-tocopherol concentrations in m. longissimus thoracis, the beef color stability during display became short with age.

  • 씨스트선충과의 한국 2 미기록속 및 종, Globodera hypolysi, Sarisodera hydrophila를 팔공산에서 채집하였다. 한국산 G. hypolysi는 원 기재와 비슷하나 유충의 구침 길이가 약간 짧고 (21-24 vs.24~25$\mu$m); 씨스트 항문에서 탕까지 거리가 약간 짧았다 (23~54 vs.25~80$\mu$m).한국산 S. hydrophila는 모든 측정치가 원 기재에 비하여 약간 짧았다; 암컷 체장(367.2~597.6$\mu$m vs 359~ 688 mm); 구침장 (37~44$\mu$m vs 38~46$\mu$m); 수컷 체장 (662~1198$\mu$m vs 579-1,405$\mu$m); 구침장 (37~ 43.8$\mu$m vs 38~46$\mu$m). 제2령유충: 체장 (460~536$\mu$m vs 466~558$\mu$m); 구침장이 짧았다 (32~38$\mu$m vs 39~43$\mu$m).

Characteristics of the Molecular Epidemiology of CTX-M-Producing Escherichia coli Isolated from a Tertiary Hospital in Daejeon, Korea

  • The aims of this study were to characterize the molecular epidemiological profiles of CTX-M-producing uropathogenic Escherichia coli isolates from a tertiary hospital in Daejeon, Korea, and to investigate the genetic diversity and compare the prevalence of sequence types (STs) in different areas. Extended spectrum β-lactamase-producing E. coli strains isolated from urine were analyzed for CTX-M, integrons, and insertion sequence common regions (ISCRs) by PCR and sequencing. Multilocus sequence typing (MLST), pulsed-field gel electrophoresis (PFGE), phylogenetic analysis, and rep-PCR were also used for molecular typing of the isolates. Of 80 CTX-M producers, 31 and 46 expressed CTX-M-15 and CTX-M-14, respectively. MLST analysis indicated that the most prevalent ST was ST131 (n = 34, 42.5%), followed by ST38 (n = 22, 27.5%), ST405 (n = 8, 10.0%), and ST69 (n = 6, 7.5%). Most CTX-M producers harbored class 1 integrons. ST131 strains belonged to phylogenetic group B2 and showed identical rep-PCR patterns, whereas ST69, ST38, and ST405 strains belonged to phylogenetic group D; the ST38 and ST405 strains displayed the same rep-PCR pattern, respectively. ST131 and ST38 isolates showed 21 and 19 distinct types, respectively, by PFGE. In Daejeon, D-ST38 CTX-M-14 producers were relatively more prevalent than in other countries and Korean cities. Our results indicate that CTX-M-producing E. coli isolates belonged mostly to ST131 or ST38 and were more related to hospital-onset than to community-onset infections and that the blaCTX-M gene may vary according to the ST.

마그네슘의 폭발특성에 미치는 평균입경의 영향 (Effect of Mean Diameter on the Explosion Characteristic of Magnesium Dusts)

  • 본 연구에서는 마그네슘(Mg)분진의 폭발특성에 미치는 평균 입경의 영향을 조사하였다. 이를 위해 20-L분진 폭발시험장치와 10 kJ의 에너지를 갖는 화학 점화기를 사용하여 실험을 수행하였다. 실험에 사용한 Mg분진은 평균입경이 서로 다른 3가지 시료(38, 142, $567{\mu}m$)를 대상으로 분진농도를 $2250g/m^3$까지 조사하였다. Mg분진의 폭발하한농도(LEL)는 평균입경 38, $142{\mu}m$에 있어서 각각 30, $40g/m^3$ 이 얻어졌다. LEL은 입경 증가에 의해 감소 경향을 나타냈는데 Mg분진은 입경이 증가할수록 폭발 확률이 감소함을 의미한다. 최대폭발압력($P_m$)과 폭발지수($K_{st}$)는 입경이 증가하면 감소하였지만, 평균입경 $567{\mu}m$의 Mg분진의 경우에는 5 kJ의 착화에너지에서는 폭발 현상이 관찰되지 않았다.

  • Next to Paper I (Jeon 2009a), time-series BV CCD images of the open cluster M38 were taken for 4 nights on December, 2009. The observations have been carried out for total 27 nights. In addition to the 20 variable stars in the Paper I, the discovery of 44 new variable stars has been presented in this paper: $6{\delta}$ Scuti stars, $2{\gamma}$ Doradus stars, 18 eclipsing binaries and 18 semi-long periodic and/or slow irregular type variable stars. For the V photometry of the ${\delta}$ Scuti and ${\gamma}$ Doradus stars, multi-frequency analysis was performed using the Discrete Fourier Transform and linear least-square fitting. The period search for the eclipsing binaries and the semi-long periodic and/or slow irregular type variable stars was performed by phase fitting method. As a result, the periods for 23 variable stars among the 44 ones were defined.

Effect of p38 inhibitor on the proliferation of chicken muscle stem cells and differentiation into muscle and fat

  • Objective: Inhibiting the p38 mitogen-activated protein kinase (MAPK) signaling pathway delays differentiation and increases proliferation of muscle stem cells in most species. Here, we aimed to investigate the effect of p38 inhibitor (p38i) treatment on the proliferation and differentiation of chicken muscle stem cells. Methods: Chicken muscle stem cells were collected from the muscle tissues of Hy-line Brown chicken embryos at embryonic day 18, then isolated by the preplating method. Cells were cultured for 4 days in growth medium supplemented with dimethyl sulfoxide or 1, 10, 20 μM of p38i, then subcultured for up to 4 passages. Differentiation was induced for 3 days with differentiation medium. Each treatment was replicated 3 times. Results: The proliferation and mRNA expression of paired box 7 gene and myogenic factor 5 gene, as well as the mRNA expression of myogenic differentiation marker gene myogenin were significantly higher in p38i-treated cultures than in control (p<0.05), but immunofluorescence staining and mRNA expression of myosin heavy chain (MHC) were not significantly different between the two groups. Oil red O staining of accumulated lipid droplets in differentiated cell cultures revealed a higher lipid density in p38i-treated cultures than in control; however, the expression of the adipogenic marker gene peroxisome proliferator activated receptor gamma was not significantly different between the two groups. Conclusion: p38 inhibition in chicken muscle stem cells improves cell proliferation, but the effects on myogenic differentiation and lipid accumulation require additional analysis. Further studies are needed on the chicken p38-MAPK pathway to understand the muscle and fat development mechanism.

간상피세포에서 BHT와 propyl gallate에 의한 gap junctional intercellular communication 억제 효과 (Inhibition of Gap Junctional Intercellular Communication in Rat Liver Epithelial Cells Induced by BHT and Propyl Gallate)

  • 본 연구에서는 정상 세포인 간상피세포를 이용하여 BHT와 PG가 GJIC에 미치는 효과 및 그 작용 기전을 살펴보았다. BHT와 PG를 WB-F344세포에 각각 1.0mM, 0.75mM 이상 처리한 결과, 세포 생육이 80%이하로 저하되어 세포 독성을 보였고, BHT와 PG의 GJIC에 대한 억제효과는 이보다 낮은 농도인 0.6mM, 0.1mM에서 나타났으며 처리에 따른 Cx43 단백질의 발현 및 인산화를 측정한 결과, BHT와 PG 모두 농도의존적으로 Cx43의 인산화가 증가하였고, Cx43의 구조적 변화와 관련된 MAP kinase는 주요 biomarker인 ERK, p38, JNK의 발현 및 인산화를 측정한 결과, 농도의존적으로 ERK와 p38의 인산화가 증가하는 것으로 나타났다. 이는 WB-F344세포에 BHT와 PG의 처리가 세포독성을 나타내기 전 농도에서 GJIC의 억제하였음을 의미하며 이는 식품첨가물의 안전성 평가에도 활용될 수 있을 것으로 기대된다.

Rapamycin reduces orofacial nociceptive responses and microglial p38 mitogen-activated protein kinase phosphorylation in trigeminal nucleus caudalis in mouse orofacial formalin model

  • The mammalian target of rapamycin (mTOR) plays a role in various cellular phenomena, including autophagy, cell proliferation, and differentiation. Although recent studies have reported its involvement in nociceptive responses in several pain models, whether mTOR is involved in orofacial pain processing is currently unexplored. This study determined whether rapamycin, an mTOR inhibitor, reduces nociceptive responses and the number of Fos-immunoreactive (Fos-ir) cells in the trigeminal nucleus caudalis (TNC) in a mouse orofacial formalin model. We also examined whether the glial cell expression and phosphorylated p38 (p-p38) mitogen-activated protein kinases (MAPKs) in the TNC are affected by rapamycin. Mice were intraperitoneally given rapamycin (0.1, 0.3, or 1.0 mg/kg); then, 30 min after, 5% formalin (10 μl) was subcutaneously injected into the right upper lip. The rubbing responses with the ipsilateral forepaw or hindpaw were counted for 45 min. High-dose rapamycin (1.0 mg/kg) produced significant antinociceptive effects in both the first and second phases of formalin test. The number of Fos-ir cells in the ipsilateral TNC was also reduced by high-dose rapamycin compared with vehicle-treated animals. Furthermore, the number of p-p38-ir cells the in ipsilateral TNC was significantly decreased in animals treated with high-dose rapamycin; p-p38 expression was co-localized in microglia, but not neurons and astrocytes. Therefore, the mTOR inhibitor, rapamycin, reduces orofacial nociception and Fos expression in the TNC, and its antinociceptive action on orofacial pain may be associated with the inhibition of p-p38 MAPK in the microglia.
