부산지역 양호교사의 업무분석에 관한 연구 (A Study on the analysis of activities of t?e 5.H.T. (5.H.T. in Pusan City))

  • The purpose of this study was to find out the general characteristics of school health teachers, the status of school health resources and the degrees of self-confident performance for the 124 school health teachers in Pusan City. Data was collected by means of questionaires from Aug. 1986 to Mar. 1987. The data were analyzed by use of percentage, mean, standard deviation, t-test, ANOVA and Pearson-correlation coefficient. The results of this study were as follows: 1 . General Characteristics of School Health Teachers (SHT) 1) The average of age of the SHT was 32.8 years old and 39.5% of them were from 30 to 34 years old. 2) The average for school nursing experience of the SHT was 7.9 years and 37.9% of them were from 5 to 9 years. 3) The 45.2% of them have not the clinical experience. 4) The 74.2% of them were graduated from the 3 years college of nursing. 2. Status of school health resources and nursing activities. 1) The average of student number was 2497.3 and class number was 45.2. 2) The average of school health budget was 1039000 won and 27.7% of school health budget expended on examination cost. 3) Only 29.0% of all schools have organization for school health. 4) The 84.7% of all schools have health clinic separately and 69.1% of schools have less than $33m^2$ sized. 5) The average of clinic visitor number was 2111.8 for 1 year. 6) Major problem was on digestive system. And other problems were skin, respiratory, musculo-skeletal system and dental problem. 7) The number of literal message was 14.4 times for 1 year. 3. The degree of the school health teachers' self-confidence. 한국남부(韓國南部)의 백악기말(白堊紀末) 이후(以後)의 화성활동(火成活動)과 광화작용(鑛化作用)에 대(對)한 판구조론(板構造論)의 적용성(適用性) 연구(硏究)( I ) (Applicability of plate tectonics to the post-late Cretaceous igneous activities and mineralization in the southern part of South Korea( I ))

  • Petrochemical, K-Ar dating, Sand Rb/Sr isotopes, metallogenic zoning, paleomagnetic and geotectonic studies of the Gyongsang basin were carried out to examine applicability of plate tectonics to the post-late Cretaceous igneous activity and metallogeny in the southeastern part of Korean Peninsula. The results obtained are as follows: 1. Bulgugsa granitic rocks range from granite to adamellite, whose Q-Ab-Or triangular diagram indicates that the depth and pressure at which the magma consolidated increase from coast to inland varying from 6 km, 0.5-3.3 kb in the coastal area to 17 km, 0.5-10 kb in the inland area. 2. The volcanic rocks in Gyongsang basin range from andesitic to basaltic rocks, and the basaltic rocks are generally tholeiitic in the coastal area and alkali basalt in the inland area. 3. The volcanic rocks of the area have the initial ratio of Sr^{87}/Sr^{86} varying from 0.706 to 0.707 which suggests a continental origin; the ratio of Rb/Sr changing from 0.079-0.157 in the coastal area to 0.021-0.034 in the inland area suggests that the volcanism is getting younger toward coastal side, which may indicate a retreat in stage of differentiation if they were derived from a same magma. The K_2O/SiO_2 (60%) increases from about 1.0 in the coastal area to about 3.0 in the inland area, which may suggest an increase indepth of the Benioff zone, if existed, toward inland side. 4. The K-Ar ages of volcanic rocks were measured to be 79.4 m.y. near Daegu, and 61.7 m.y. near Busan indicating a southeastward decrease in age. The ages of plutonic rocks also decrease toward the same direction with 73 m.y. near Daegu, and 58 m.y. near Busan, so that the volcanism predated the plutonism by 6 m.y. in the continental interior and 4 m.y. along the coast. Such igneous activities provide a positive evidence for an applicability of plate tectonics to this area. 5. Sulfur isotope analyses of sulfide minerals from 8 mines revealed that these deposits were genetically connected with the spacially associated ingeous rocks showing relatively narrow range of ${\delta}^{34}S$ values (-0.9‰ to +7.5‰ except for +13.3 from Mulgum Mine). A sequence of metallogenic zones from the coast to the inland is delineated to be in the order of Fe-Cu zone, Cu-Pb-Zn zone, and W-Mo zone. A few porphyry type copper deposits are found in the Fe-Cu zone. These two facts enable the sequence to be comparable with that of Andean type in South America. 6. The VGP's of Cretaceous and post Cretaceous rocks from Korea are located near the ones($71^{\circ}N$, $180^{\circ}E$ and $90^{\circ}N$, $110^{\circ}E$) obtained from continents of northern hemisphere. This suggests that the Korean peninsula has been stable tectonically since Cretaceous, belonging to the Eurasian continent. 7. Different polar wandering path between Korean peninsula and Japanese islands delineates that there has been some relative movement between them. 8. The variational feature of declination of NRM toward northwestern inland side from southeastern extremity of Korean peninsula suggests that the age of rocks becomes older toward inland side. 9. The geological structure(mainly faults) and trends of lineaments interpreted from the Landsat imagery reveal that NNE-, NWW- and NEE-trends are predominant in the decreasing order of intensity. 10. The NNE-trending structures were originated by tensional and/or compressional forces, the directions of which were parallel and perpendicular respectively to the subduction boundary of the Kula plate during about 90 m.y. B.P. The NWW-trending structures were originated as shear fractures by the same compressional forces. The NEE-trending structures are considered to be priginated as tension fractures parallel to the subduction boundary of the Kula plate during about 70 m.y. B.P. when Japanese islands had drifted toward southeast leaving the Sea of Japan behind. It was clearly demonstrated by many authors that the drifting of Japanese islands was accompanied with a rotational movement of a clock-wise direction, so that it is inferred that subduction boundary had changed from NNE- to NEE-direction. A number of facts and features mentioned above provide a suite of positive evidences enabling application of plate tectonics to the late Cretaceous-early Tertiary igneous activity and metallogeny in the area. Synthesizing these facts, an arc-trench system of continental margin-type is adopted by reconstructing paleogeographic models for the evolution of Korean peninsula and Japan islands. The models involve an extention mechanism behind the are(proto-Japan), by which proto-Japan as of northeastern continuation of Gyongsang zone has been drifted rotationally toward southeast. The zone of igneous activity has also been migrated from the inland in late-Cretaceous to the peninsula margin and southwestern Japan in Tertiary.

u-헬스케어 시스템에서 멀티 생체신호 측정 무선 단말기 설계 및 구현 (Design and Implementation of Mobiel Terminal with Multi Bio-signal Measurement for u-Healthcare System)

  • 기존의 u-헬스케어 시스템의 생체 신호 측정 단말기는 단말기에서 하나의 생체 신호를 측정하여 평가하는 것이 대부분이고, 복합된 기능의 단말기이더라도 사용자의 선택에 의해서 생체 신호를 측정하도록 되어 있다. 본 논문에서는 이러한 문제점을 해결하고, u-헬스케어 시스템에 적용하기 위한 멀티 생체신호 측정 무선 단말기를 구현하였다. 구현된 단말기는 4개의 적외선 센서를 사용하여 맥박수를 측정하고, 2개의 전극을 이용하여 피부전도도를 측정하고, 3축 가속도 센서를 사용하여 운동량을 측정하였다. 뿐만 아니라, 생체신호 데이터를 무선으로 전송하기 위해 지그비를 이용하여 PC 또는 휴대단말기로 전송할 수 있는 통신 패킷 프레임을 제안한다. 무선 단말기를 구현하여 실제 테스트한 결과 맥박 수는 2회 이내의 오차 범위를 가지고, 운동량은 약 85.6%, 84.7%의 신뢰도를 보였으며, 피부전도도는 사용자의 신체 상태에 따라 변화함을 알 수 있었다.

Activated Phenoloxidase Interacts with A Novel Glycine-rich Protein on the Yeast Two-hybrid System

  • One of the innate immune reactions in invertebrates is the pro-phenoloxidase (pro-PO) activation system that is involved in the generation of superoxide, melanin synthesis, and the subsequent sequestration of foreign matter entering the hemocoel of the invertebrates. However, the molecular mechanism of this biological reaction is still obscure. To expand our understanding of the biological roles of the pro-PO activation system in invertebrates, we performed a yeast two-hybrid screening by using three regions of pro-PO as bait and a yeast two-hybrid cDNA library from Tenebrio molitor larvae as prey We isolated a novel partial cDNA clone that encodes a glycine-rich protein that interacted with the active phenoloxidase (termed phenoloxidase interacting protein, POIP). POIP consists of two domains: One is an N-terminal unique domain and the other is a C-terminal glycine-rich domain. The C-terminal glycine-rich domain showed sequential homology with those of insect antifungal proteins. Also, the yeast two-hybrid screen in a reverse orientation (using POIP as bait) yielded PO, suggesting that the PO-POIP interaction is specific. By using a 315 bP PCR fragment of the N-terminal unique region of POIP, we cloned the full-length cDNA of POIP from the Tenebruo cDNA library constructed by using E. coli injected larvae. The interaction analysis between PO, and a truncated fragment lacking the N-terminal unique region of POIP, indicated that the N-terminal unique region is necessary for interaction between PO and POIP. The expression level of the POIP mRNA is increased by bacterial injection into T. molitor larvae. This suggests that POIP might be engaged in the humoral defense reaction.

  • 전자상거래의 도입으로 인하여 전신송금환(T/T)${\cdot}$우편송금환(M/T)${\cdot}$송금수표(D/D)와 같은 단순송금방식(Remittance), 현금결제방식(COD)과 서류결제방식(CAD), 지급인도조건(D/P)${\cdot}$인수인도조건(D/A)과 같은 추심 결제방식, 신용장(Letter of Credit)방식 등 기존의 무역결제수단이 신용카드(Credit Card), 전자화폐(Electronic Cash), 전자자금이체(EFT; Electronic Fund Transfer), 무역카드(TradeCard), SWIFT의 전자신용장(Electronic L/C), 볼레로 시스템 등의 전자결제시스템으로 전환되었다. 이에 본고는 대표적 전자무역결제시스템인 볼레로와 무역카드를 비교${\cdot}$분석함으로써 이들 시스템의 활용가능성을 모색하고자 하는 것이다.

Aspergillus nidulans mtDNA의 자가복제절편 (Autonomously Mitochondrial Replicating Sequence of Aspergillus nidulans)

  • Aspergillus nidulans 의 DNA 로부터 Saccharomyces cerevisiae에서 스스로 복제가능하고, 형질전환율도 삽입벡터에 비해 10\sup 4\ 배정도 높여주는 절편을 분리한 바 있다. A. nidulans에서 ANRI 의 일부를 포함한 pILJ16-4.5는 삽입벡터인 pILJ16보다 170배 정도 높은 형질전환 효율을 보였으며, S.cerevisiae와 마찬가지로 plasmid 상태로 회수가능했다. A.nidulans 페소내에서 2-3 copy 정도로 염색체와는 별개로 존재하는 것으로 나타났으며, 재형질전환능력도 있었다. ANRI은 미토콘드리아의 DNA에서 유래한 절편으로 밝고, ARS 공통절편과 유사한 절편도 11곳에 존재한다. $\Phi$X174와 SV40 DNA에서 gyrase 가 결합하는 부위의 공통편인 YRTGNYNNY도 6곳에 존재하며, ColE1에서 gyrase가 결합하는 b site와 일치하는 절편도 하나 포함하고 있으며, ABF1 결합 부위이 공통절편과 유사한 절푠$TCN_7ACG$ 도 하나 포함하고 있다. 이를 토대로 ANRI은 A.nidulans의 형질전환 후 회수가 가능한 replicating vector 개발에 이용할 수 있다.

FOX hunting system을 이용한 배추 기능유전자 탐색 (Systematic approaches to identify functional genes using the FOX-hunting system in Chinese cabbage)

  • Full-length cDNAs are essential for the correct annotation of genomic sequences and for the functional analysis of genes and their products. To elucidate the functions of a large population of Chinese cabbage (Brassica rapa) genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the full-length cDNA Over-eXpresser (FOX) gene hunting system. With oligo dT column it purify the each mRNA from the flower organs, leaf and stem tissue. And about 120,000 cDNAs from the library were transformed into $\lambda$-pFLCIII-F vector. Of which 115,000 cDNAs from the library were transformed into T-DNA binary vector, pBigs for transformation study. We used normalized full-length cDNA and introduced each cDNA into Arabidopsis by in planta transformation. Full-length Chinese cabbage cDNAs were expressed independently under the CaMV 35S promoter in Arabidopsis. Selfed seeds were harvested from transgenic Arabidopsis. We had selected 2,500 transgenic plants by hygromycin antibiotic tolerant test, and obtained a number of transgenic mutants. Each transgenic Arabidopsis was investigated in morphological changes, fertility and leaf colour. As a result, 285 possible morphological mutants were identified. Introduced cDNA was isolated by PCR amplification of the genomic DNA from the transgenic mutants. Sequencing result and BLAST analysis showed that most of the introduced cDNA were complete cDNAs and functional genes. Also, we examined the effect of Bromelain on enhancing resistance to soft rot in transgenic Chinese cabbage 'Osome'. The bromelain gene identified from FOX hunting system was transformed into Chinese cabbage using Agrobacterium methods. Transformants were screened by PCR, then RT-PCR and real time PCR were performed to analyze gene expression of cysteine protease in the T1 and T2 generations. The anti-bacterial activity of bromelain was tested in Chinese cabbages infected with soft rot bacteria. The results showed that the over-expressed bromelain gene from pineapple conferred enhanced resistance to soft rot in Chinese cabbage.

만경강 유역의 비점오염물질 유출모의를 통한 새만금 만 유입부의 수질 예측 (Prediction of Water Quality at the Inlet of Saemangeum Bay by using Non-point Sources Runoff Simulation in the Mankyeong River Watershed)

  • This study was carried out to forecast the flow rate and water quality at the inlet of the Saemangeum bay in Korea using the SWMM(Storm Water Management Model) and the WASP(Water Analysis Simulation Program), and to analyze the impacts of pollutant loading from non-point source on the water quality of the bay. The calibration and validation of flow rate and water quality were performed using those from two monitoring points in the Mankyeong river administrated by Korean Ministry of Environment as part of the national water quality monitoring network. When the river flow rate was calibrated and validated using the rainfall intensities during 2011-2012, $R^2$ (i.e., coefficient of determination) was ranged from 0.91 to 0.96. For water qualities, it was shown that $R^2$ of BOD(Biochemical Oxygen Demand) was ranged from 0.56 to 0.86, and $R^2$ of T-N(Total Nitrogen) was from 0.64 to 0.75, and $R^2$ of T-P(Total Phosphorus) was from 0.67 to 0.89. The integrated modeling system showed significant advances in the accuracy to estimate the water quality. Finally, further simulations showed that annual average flow of the river running into the bay was estimated to be $1.439{\times}10^9m^3/year$. The discharged load of BOD, T-N, and T-P into the bay were anticipated to be 618.7 ton/year, 331.5 ton/year, and 40.4 ton/year, respectively.

Electrical Characteristics of a-GIZO TFT by RF Sputtering System for Transparent Display Application

  • 2004년 일본의 Hosono 그룹에 의해 처음 발표된 이래로, amorphous gallium-indium-zinc oxide (a-GIZO) thin film transistors (TFTs)는 높은 이동도와 뛰어난 전기적, 광학적 특성에 의해 큰 주목을 받고 있다. 또한 넓은 밴드갭을 가지므로 가시광 영역에서 투명한 특성을 보이고, 플라스틱 기판 위에서 구부러지는 성질에 의해 플랫 패널 디스플레이나 능동 유기 발광 소자(AM-OLED), 투명 디스플레이에 응용될 뿐만 아니라, 일반적인 Poly-Si TFT에 비해 백플레인의 대면적화에 유리하다는 장점이 있다. 최근에는 Y2O3나 ZrO2 등의 high-k 물질을 gate insulator로 이용하여 높은 캐패시턴스를 유지함과 동시에 낮은 구동 전압과 빠른 스위칭 특성을 가지는 a-GIZO TFT의 연구 결과가 보고되었다. 하지만 투명 디스플레이 소자 제작을 위해 플라스틱이나 유리 기판을 사용할 경우, 기판 특성상 공정 온도에 제약이 따르고(약 $300^{\circ}C$ 이하), 이를 극복하기 위한 부가적인 기술이 필수적이다. 본 연구에서는 p-type Si을 back gate로 하는 Inverted-staggered 구조의 a-GIZO TFT소자를 제작 하였다. p-type Si (100) 기판위에 RF magnetron sputtering을 이용하여 Gate insulator를 증착하고, 같은 방법으로 채널층인 a-GIZO를 70 nm 증착하였다. a-GIZO를 증착하기 위한 sputtering 조건으로는 100W의 RF power와 6 mTorr의 working pressure, 30 sccm Ar 분위기에서 증착하였다. 소스/드레인 전극은 e-beam evaporation을 이용하여 Al을 150 nm 증착하였다. 채널 폭은 80 um 이고, 채널 길이는 각각 20 um, 10 um, 5 um, 2 um이다. 마지막으로 Furnace를 이용하여 N2 분위기에서 $500^{\circ}C$로 30분간 후속 열처리를 실시한 후에, 전기적 특성을 분석하였다.

대장균의 periplasm으로부터 재조합 PGA 단백질의 효율적이고 간단한 방출 방법 (An Efficient Method for the Release of Recombinant Penicillin G Amidase from the Escherichia coli Periplasm)

  • 세제에 의하여 대장균의 periplasm에서 penicillin G amidase (PGA)를 방출하는 방법을 연구하였다. 결과적으로 세제와 lysozyme의 혼합 작용이 효과적인 것으로 나타났다. 세포 투과성의 최적 조건을 알아보기 위하여 세제의 종류, 농도, pH, 반응 시간, 온도 등의 영향을 살펴보았다. 그리하여 대장균에서 재조합 PGA를 periplasm에서 방출하는 모델을 만들 수 있었고 방출된 PGA를 농축할 수 있었다. 실리카 구슬을 이용한 고정화 시스템으로 PGA 용액을 농축할 수 있었으며, 더 이상의 정제 과정 없이 순수하게 추출 할 수 있었다. 고정화된 PGA는 penicillin G 생성의 원료인 6-APA를 생산하는데 사용할 수 있었다. 이 방법은 대장균으로부터 재조합 단백질을 추출하는 간단한 방법이며 고정화 PGA를 이용하여 ${\beta}-lactam$ 항생물질의 산업적 생산 이용될 수 있을 것으로 사료된다.
