Sequence comparisons of 28S ribosomal DNA and mitochondrial cytochrome c oxidase subunit I of Metagonimus yokogawai, M. takahashii and M. miyatai

  • We compared the DNA sequences of the genus Metagonimus: M. yokogawai, M. takahashii, and M. miyatai. We obtained 288 D1 ribosomal DNA (rDNA) and mitochondrial cytochrome c oxidase subunit I (mtCOI) fragments from the adult worms by PCR, that were cloned and sequenced. Phylogenetic relationships inferred from the nucleotide sequences of the 28S D1 rDNA and mtCOI gene. M. takahashii and M. yokogawai are placed in the same clade supported by DNA sequence and phylogenie tree analysis in 28S D1 rDNA and mtCOI gene region. The above findings tell us that M. takahashii is closer to M. yokogawai than to M. miyatai genetically. This phylogenetic data also support the nomination of M. miyatai as a separate species.

  • Heterophyidae과에 속하는 Matagenimus yokegawai, Metagonimus Miyata Type, M. takahashii, Heterophyes disbar, H. heterophyes, H. nocens, Heterophyepsis continua, Pygidiopsis summa, Stellanpchasmusfaltatus, Stittodora lari 등 총 21 Operational Taxonomic Unit(OTU)를 사용하여 수리분류학적 연구를 시도하였다. 형질은 절대 측정치가 아닌 상대적 비율을 주로 사용하였으며, 형질수는 26개를 사용하였다. 각 형질간의 유사도를 측정하여 상관계수를 계산한 후 Ward의 방법에 의한 집괴 분석 및 인자 분석을 수행하였다. 분석 결과 Metagonimus 속(genus) 그룹은 Metagonimus yokogawai, Metagonimus Miyata Type, M. takahashii 등 3개의 표현군으로 구분이 되었으며, Metagenimus Miyata Type은 M. takahashii 의 아종 수준으로 분류되었다. Heterophyes 속 그룹은 Heterophyes dispar, H. heterophyes, Jf. nocens 등 명확한 3개의 표현 군으로 구분되며, H. nocens는 H. heterophyes의 아종이 아닌 뚜렷한 종의 수준에서 분류됨을 나타내었다. 그 외의 종들도 역시 뚜렷한 표현 군으로 분류되었으며, 이로부터 흡충류에 있어서도 수리분류학적 방법에 의해 분류가 명화한 분류군의 한계를 규정하는데 많은 도움을 줄 수 있을 것으로 사료된다.

A cytogenetic study on human intestinal trematodes of the genus Metagonimus (Digenea: Heterophyidae) in Korea

  • In order to analyze chromosome numbers and karyotypes of intestinal trematodes belonging to the genus, Metagonimus, the gonad tissues of M. takahashii, M. miyatai, and M. yokogawai were prepared and examined. The number of bivalents in the first meiotic division of M. takahashii was nine (n=9). The diploid number of M. miyatai was observed to be eighteen (2n=18) and their chromosomes consisted of one pair of metacentric, 7 pairs of submetacentric, and one pair of telocentric chromosomes. The diploid number of M. yokogawai was thirty-two (2n=32) and the chromosome complements were composed of two pairs of metacentric, 11 pairs of submetacentric, and three pairs of subtelocentric chromosomes. These results could be a supporting evidence for the validity of the new species, M. miyatai, distinct from M. yokogawai.

Metagonimus miyatai sp. nov. (Digenea: Heterophyidae), a new intestinal trematode transmitted by freshwater fishes in Japan and Korea

  • Metagonimus miyatai n. sp. (Digenea: Heterophyidae) is described based on adult flukes collected from experimental dogs and hamsters fed with metacercariae encysted in the sweetfish, Plecoglossus altivelis, dace, Tribolodon hakonensis and T. taczanowskii, common fat-minnow. Morocco steindachneri pale chub, Zacco platypus, and dark chub, Zacco temmincki, and on those collected from naturally infected humans. The new species was morphologically compared with M. yokogawai and M. takahashii obtained from experimental animals fed with the sweetflsh and the crucian carp, Cnrossius caressius, respectively. The uterine loops of M. miyatai reached near the posterior end of the body through the space between the two testes, whereas those of M. yokogawai, occupied only the space between the acetabulum and anterior border of two testes. This uterine tubule distribution was similar to that of another closely related species, M. takahashii. However vitellaria of M. miyatai ended in front of the posterior end of the left testis, while those of M. takahcshii reached the posterior end of the left testis and ran it over. By raising M. miyatai as a new species, differentiation of M. yokogawai and M. takahashii became very clear. A key to the species of the genus Metagonimus in the Far East has been proposed.

Metagonimus yokogawai (Trematoda: Heterophyidae): From Discovery to Designation of a Neotype

  • Metagonimus yokogawai (Katsurada, 1912) Katsurada, 1912 (Trematoda: Heterophyidae) is parasitic in the small intestine of mammals including man and birds in Far Eastern Russia, Korea, Japan, China, and Taiwan. In the present study, the metacercariae and adults of M. yokogawai were redescribed to designate a neotype of this fluke together with reviews of previous studies including study histories from the first discovery to now. We particularly, attempted to review the study histories and morphological descriptions of M. yokogawai for the species validity, and compared with the morphological characteristics and life cycles of the closely related species, Metagonimus takahashii and Metagonimus miyatai. Finally, we proposed a differential key for the 8 known Metagonimus species distributed in East Asia. Metacercariae were obtained from the body muscles of sweetfish (Plecoglossus altivelis) collected in the Asahi River at Takebe-cho, Kita-ku, Okayama City, Okayama Prefecture, Japan. Adults were recovered from the small intestine of Syrian golden hamsters, to which the metacercariae had been fed 14 days before. A neotype was selected out of the present adult specimens. The Asahi River at Takebo-cho became the type locality of M. yokogawai. In conclusion, the present review shows that M. yokogawai, M. takahashii, and M. miyatai are valid and discriminated by means of morphological characteristics.

Surface ultrastructure of Metagonimus takahashii metacercariae and adults

  • A scanning electron microscopic study was performed on the surface ultrastructure of metacercariae and adults of Metagonimus takahashii. Metacercariae were collected from the scale of crucian carp (Carassius auratus) , and adult flukes were harvested 1-4 weeks after infection to rats. In excysted metacercariae. the oral sucker had type I (numerous) and type II (seven in total) sensory papillae. Tegumental spines were dense and digitated into 5-7 points on the surface anterior to the ventral sucker, but became sparse and less digitated posteriorly toward the end of the body In adults, seven type II sensory papillae were characteristically arranged around the lip of the oral sucker, and on the inner side of the lip four small and two large type I sensory papillae were symmetrically seen on each side (12 in total). Tegumental spines on anterior two-thirds of the body. were digitated with 9-12 tips ventrally and 8-13 tips dorsally. Sperms entering into the Laurer's canal were observed. The results show that the surface ultrastructure of M. takuhashii is generally similar to those of M. yokogawai and M. miyatai except for the digitation of tegumental spines.

Heterophyid metacercarial infections in brackish water fishes from Jinju-man (Bay), Kyongsangnam-do, Korea

  • Heterophyid metacercarial infections in brackish water fishes, i.e., perch, shad, mullet, redlip mullet, and goby, of Jinju-man (Bay), Kyongsangnam-do, Korea, were investigated using a digestion technique. Among 45 perch (Lateolabrax japonicus), the metacercariae of Heterophyopsis continua were found in $55.6\%$ (18.5 metacercariae per fish), Stictodora spp. in $28.9\%$ (3.6), and Metagonimus takahashii in $6.7\%$ (17.0). The metacercariae of H. continua were detected in 23 $(65.7\%)$ of 35 shad (Konosirus punctatus). Among 15 mullet (Mugil cephalus), the metacercariae of Pygidiopsis summa were found in $100\%$ (105.9 metacercariae per fish), Heterophyes nocens in $40.0\%$ (8.5), H. continua in $13.3\%$, and Stictodora spp. in $6.7\%$. Among 12 redlip mullet (Chelon haematocheilus), the metacercariae of P. summa were detected in $91.7\%$ (1,299 metacercariae per fish), H. nocens in $16.7\%$, and Stictodora spp. in $16.7\%$. Among 35 gobies (Acanthogobius flavimanus), the metacercariae of Stictodora spp. were found in $82.9\%$ (44.5 metacercariae per fish), and H. continua in $25.7\%$ (2.8). The adult flukes recovered from a cat experimentally infected with metacercariae from perches were compatible with M. takarashii. The results confirm that the estuarine fish in Jinju-man (Bay) are heavily infected with heterophyid metacercariae. Furthermore, it was found for the first time that the perch acts as a second intermediate host for M. takahashii.

섬진강산 황어에 피낭한 Metagonimus 속 선충의 세 가지 형태학적 유형 (Three morphological types ot the genus Metagonimus encysted in the dace, Tribolodon taczanowskii, caught from the Sumjin River)

  • 섬진강 유역(경남 하동군)산 황어(Tribelodon taczanowskii)로부터 Metagonimus속 피낭유충을 다수 검출하고 실험적으로 흰쥐와 햄스터에 감염시킨 후 소장으로부터 성충을 얻어 관찰한 바 Saito(1984)의 분류에 따라 Metagonimus Yokogawa형 (M. yokogawai), Miyata형 및 Koga형 등 3가지로 나눌 수 있었다. 실험감염 결과, 흰쥐 4마리 및 햄스터 4마리로부터 Metagonimus 충체 총 1,624마리를 회수하였고 그 중 Yokogawa형은 820마리, Miynta형은 688마리, Koga형은 46마리이었다. 나머지 충체 70마리는 유형을 결정하기 어려웠다. Yokgawa형과 Miyata형의 주요 감별정은 두 고환의 위치, 자궁관의 분포 및 충란의 크기 등이었고, Miyata형과 Koga형의 감별점은 두 고환의 위치, 난환선의 분포, 충란의 크기 등이었다. 이들 유형 특히 Miyata 형의 별종 가능성에 대해서는 많은 증거가 얻어질 때까지 이를 유보해야 할 것으로 생각되고, Koga형은 중간숙주의 차이 이외에는 형태학적으로 Takahashi형 (M. takahashii)과 전혀 차이가 인정되지 않으므로 이와 동일한 것으로 취급하는 것이 옳을 것으로 판단되었다. 이상의 결과로 섬진강산 황어에는 3가지 유형의 Metagonimus가 피낭하고 있음을 확인하였다.

강원도에 분포하는 Metagonimus속 장흡충과 제2중간숙주에 대하여 (Intestinal flukes of genus Metagonimus and their second intermediate hosts in Kangwon-do)

  • 강원도의 강 하천유역에 있어서 주민의 Metqgonimus속 흡충의 감염률, 어류의 피낭 유충 자연감염 및 동물감염 실험에서 얻은 성충의 형태 등을 관찰하였다 주민의 감염률은 평균 7.8%(1,067명 중 83명 양성)였다 성별로는 남자 11.4%. 여자 3.2%였다. 강별로는 영서지역에서 섬강유역 7.3%, 주천강유역 6.3%, 평창강유역 12.8% 동강유역 3.8% 홍천강유역 9.8%와 영동지역 삼척 오십천유역이 8.0% 등이었다. 조사한 어류의 Metoqonimus속 피낭유충 감염률은 평균 81.0%(318마리 중 256마리)였고 감염어종은 영서지역에서 피라미(Zacco platypus), 갈겨니 (Zacco temmincki) . 끄리 (Opsarrichthvs binens) . 몰개(Squalidis sp. ). 붕어 (Carossius corossius) 등이었다 영동지역 삼척 오십천에서는 은어(Pruoglossus altivelis), 황어(Ribolodnn hokonensis)였다. 퍼라미와 끄리에서 분리한 피낭유충을 흰쥐와 개에 실험감염시켜 얻은 성충체는 대부분이 MetogonimMlyata형이고 부분적으로 M. tukqhoshii가 혼재하여 있었다. 영동지역 삼척 오십천 은어에서는 M. Vokogawni만이 검출되었고 황어에서는 M. Vokogawci와 MetoEonim Miyata형이 혼재하여 있었다. 이상으로 강원도에 분포하는 Metagonimus속 흡충은 영서내륙지역에 서는 주로 피라미와 끄리를 감염원으로 하는 Metagonimus miyata형 흡충과 M. takahashii흡충이 분포하였다. 영동지역(삼척 오십천)에서는 주로 은어. 황어를 감염원으로 하는 M. yokogawai흡충 이 분포하며 일부 황어에서는 Metagonimus Miyata형도 혼재할을 알 수 있었다.

Comparative Morphology of Minute Intestinal Fluke Eggs That Can Occur in Human Stools in the Republic of Korea

  • The egg morphology of minute intestinal flukes (MIF) that can occur as human infections in the Republic of Korea, i.e., Metagonimus yokogawai, M. miyatai, M. takahashii, Heterophyes nocens, Heterophyopsis continua, Stellantchasmus falcatus, Stictodora fuscata, Pygidiopsis summa, and Gymnophalloides seoi, was studied in comparison with Clonorchis sinensis. The adult worms were obtained from residents of endemic areas, and their intrauterine eggs were studied and measured using light microscopy; the length, width, length-width ratio (LWR), and Faust-Meleney index (FMI). Several specimens were processed for scanning electron microscopy (SEM), and before gold-coating, the uterine portion of each fluke was etched with a sharp pin in order to expose the eggs. The MIF eggs were ovoid, pyriform, or elliptical with a size range of $21-35{\times}12-21{\mu}m$. S. fuscata eggs revealed the highest FMI (largest in the area) and lowest LWR, whereas P. summa eggs showed the lowest FMI and medium LWR. SEM revealed that G. seoi and S. fuscata had remarkably clean shell surface lacking the muskmelon-like structure which is prominent in C. sinensis eggs. In Metagonimus spp., H. continua, H. nocens, and S. falcatus eggs, minute surface ridges were recognizable though less prominent compared with C. sinensis. On the surface of P. summa eggs, thread-like curly structures were characteristically seen. The results revealed that important differential keys for MIF eggs include the length, width, area (FMI), shape of the eggs, and the extent of the muskmelon-like structure or ridges on their shell surface and operculum.
