Root-knot Nematode Species Distributing in Greenhouses and Their Simple Identification Scheme (시설원예지에 분포하는 뿌리혹선충의 종류 및 간이 동정법)

  • Species and races of root-knot nematodes in greenhouses in southern Korea were investigated and a simple identification scheme was provided. Among 23 populations of root-knot nematodes, Meloidogyne arenaria race 2 was 59%, M. incognita race 1 was 23%, and an unknown race of M. incognita was 18%. Total length of M. arenaria juveniles was 411㎛(306-503㎛) and that of M. incognita was 384㎛(312-488㎛); however, the ranges of two species were overlapped and could not be used to distinguish the two species. Excretory pore in female head was a consistent character to differentiate M. arenaria and M. incognita.

Resistance of Soybean Cultivars to Root-Knot Nematode Species (Meloidogyne incognita and M. hapla) in Korea (뿌리혹선충(Meloidogyne incognita와 M. hapla)에 대한 우리나라 콩 장려품종의 저항성 검정)

  • Two species of root-knot nematode, Meloidogyne incognita and M. hapla, were used in tests of soybean cultivars for resistance. Sixteen soybean cultivars were planted in the 20-cm clay pot and inoculated with each of M. incognita and M. hapla. Four soybean cultivars, Eundaedu, Baegcheon, Dongbugtae and Danyeobkong were resistant; Kwangdu, Hwanggeumkong, and Buseog were moderate, and the rest nine cultivars were susceptible to M. incognita. Jangyeobkong only was resistant, Danyeobkong, Hill, Dongbutae, Hamandaerip and Chungbugbaeg were moderate, and the rest ten cultivars were susceptible to M. hapla. There were no cultivars resistant to both nematodes although Dongbugtae and Danyeobkong were resistant to M. incognita and moderately resistant to M. hapla.

  • M. incognita are plant-parasite nematode that cause severe damage to the crops. P. lilacinus are renowned for inhibitation of development of M. incognita's egg. We make a study for enzymatic examining the cause closely that P. lilacinus suppress development of M. incognita's egg by parasiting. The research result is explained the place below. 1. The egg that is exposed to co-enzymes which are cultured in the broth culture starts to change the membrane of egg from 3days. And in 10days, that membrane completely disappear. These are observed through light microscope. Therefore, we know that M. incongnita are controlled by extracellular lytic enzymes that are produced by P. lilacinus. 2. Through scanning electron microscope, we can find that the egg that is attacked by P. lilacinus loses it's membrane gradually, and that loss of the membrane causes transform, which suppresses the development of egg.

Resistance of Commercial Pepper Cultivars to Root-knot Nematodes (시판 고추품종에 대한 뿌리혹선충 저항성 검정)

  • Ninety two pepper (Capsicum annuum L.) cultivars were screened for resistance to two common species of root-knot nematodes, Meloidogyne incognita and M. arenaria. All 92 pepper cultivars were resistant to M. arenaria (0-3 eggmass/plant) but were susceptible to M. incognita (76-678 eggmass/plant). Susceptibility to M. incognita were slightly differed; 'Geomok' and 'Shintaepung' had relatively less eggmass (<100 eggmass/plant) when compared to those 29 very susceptible culivars such as 'Bulggotcheoreum' (>300 eggmass/plant). Therefore, pepper is highly recommended as a high-valued rotation crop to only those greenhouses infested with M. arenaria, but should restrict for M. incognita.

  • To select resistant oil seed crops against two species of root-knot nematodes, Meloidogyne incognita and M. arenaria, 10 cultivars of sesame (Sesamum indicum L.) and 10 cultivars of perilla (Perilla frutescens var. japonica) were screened in greenhouse pot test. All sesame cultivars tested were resistant to M. incognita but susceptible to M. arenaria. While, perilla was resistant to both Meloidogyne species. Therefore, perilla cultivars could be used as rotation crop in greenhouse infested with both M. incognita and M. arenaria. But, sesame cultivars only can be used as a rotation crop in greenhouse infested with M. incognita but not for M. arenaria.

  • Egg masses of the root- knot nematodes ( Meloidogyne incognita, M. arenaria and M. haply) were exposed to two different regimes of soil moisture (459 and 2459) and temperature ( -2$^{\circ}C$ and 33$^{\circ}C$), quite extreme condition in their natural environment, and their survival rate was compared. Three species did not show any difference in the rate when exposed to either soil moisture for 25 days, with the rate in the range of 8.6% to 10.4%. In response to temperature treatment, however, they differed : the best survival rate was obtained from M. incognita at high temperature ( 33"C) and from M, hapla at low temperature (-2$^{\circ}C$) plot. The third species (M. arenaria) was intermediate in both temperature regimes.imes.

Comparisons of Pathological Responses in Carrot to Root-knot Nematodes

  • Carrot (Dacus carota var. sativus) is one of the top-ten most economically important vegetable crops produced worldwide, and the root-knot nematodes, Meloidogyne spp., are one of the most important pests in the carrot. In Korea, M. hapla and M. incognita are presumed to be the major root-knot nematodes distributing mostly in open carrot fields and greenhouses, respectively. In our study, currently-developed and commercial carrot cultivars and the parental lines were examined for their pathological responses to M. incognita and M. hapla 7 weeks after inoculation with about 1,000 second-stage juveniles (J2) of the nematodes. All the carrot cultivars and lines showed susceptible responses to both nematodes with the gall index (GI) of 2.4-4.4, which were always higher on the carrot plants infected with M. incognita than M. hapla. Gall sizes were remarkably larger with more serious reduction of the root growths in the plants infected with M. incognita than M. hapla, suggesting the carrot lines examined in our study were more susceptible to the former than the latter. In the infection sites of the root tissues, giant cells were more extensively formed, occupying larger stellar regions with the prominent destruction of adjacent xylem vessels by M. incognita than M. hapla. All of these results suggest M. incognita affect more seriously on the carrot plants that are grown in greenhouses, compared to M. hapla that has a major distribution in open carrot fields, which would be used for determining cropping systems based on target nematode species, their damage and pathological characteristics.

Effects of Root-knot Nematodes, Meloidogyne hapla, M. incognita, M. arenaria and M. javanica on Growth and Yield of Tomato (토마토에 대한 뿌리혹선충의 주요 종별 피해정도)

  • The effects of infection severity of susceptible tomato varieties, Rutgers and Boksu 2, by root-knot nematode, meloidogyne hapla, M. arenaria or M. javanica on plant growths and tomato yields were investigated. The inoculum levels of each nematode species were 0, 1,000 and 10,000 nematodes per 42cm diam. pot. Tomato yield was reduced by the nematode infection. Severity of infection was on the decreasing order of M. javanica, M. incognita, M. arenaria, M. hapla, ranging from $80\%$ by M. javanica to only $7\%$ by M. hapla. Yield reduction by infection of M. hapla was more prominent in Boksu 2 than in Rutgers. However the results were vice versa for the other nematode species, The top fresh-weight of Rutgers inoculated with 10,000 nematodes was greater than root weight, regardless of the nematode species, whereas plant height and top fresh weight decreased with increasing root weight when inoculated with inoculum density of 1,000/pot.

  • In order to investigate the distribution and density of root-knot nematodes in economic crops, samples were collected from 3,226 fields of 18 economic crops. Nematodes were extracted using a modified Baermann's funnel technique. Identification of races was based on the differential host-test for Meloidogyne spp. by Tayler and Sasser. M. hapla was dominant in fields in the middle parts of Korean peninsula; M. incognita, in the southern parts, and distribution of M. javanica was limited to Jeju island and southern seaside areas. Two races of M. arenaria identified in this study were races 1 and 2. The three races identified in M. incognita were races, 1, 2 and 3. Of these races, race 1 of M. arenaria and race 3 of M. icognita were identified for the first time in Korea.

  • The effect of biological control of the root-knot nematode, Meloidogyne incognita, on cucumber and lettuce was evaluated with green manure crop species in greenhouse. Nematicidal effect of sudan grass cultivation in cucumber greenhouse was comparable to that of chemical treatment with fosthiazate GR, showing the high activity of 88.6%. Sudan grass cultivation in lettuce greenhouse significantly reduced the number of M. incognita in soil, showing 93.5% of nematiidal activity. In addition, since growth of sudan grass was superior to other green manure crop species, it is considered that cultivation of sudan grass is proper to control M. incognita in greenhouse.
